A new disease, the slippery scar, was investigated in cultivated bags of Auricularia polytricha. This fungus was isolated from the infected mycelia of cultivated bags. Based on morphological observation, rDNA-internal transcribed spacer and 18S sequence analysis, this pathogen was identified as the Ascomycete Scytalidium lignicola. According to Koch's Postulation, the pathogenicity of S. lignicola to the mycelia of A. polytricha was confirmed. The parasitism of this fungus on mushroom mycelia in China has not been reported before.

We conducted an analysis by comparing the nutritional contents of Auricularia auricula-judae(black), Auricularia polytricha and Auricularia auricula-judae (brown). In nutritional contents of three strains of Auricularia spp., four free sugars, seven organic acids and 24 amino acids were detected. Auricularia auricula-judae (black) was highly contained free sugar, organic acid and amino acid. There was the most prevalent Vitamin $D_2$ content in Auricularia auricula-judae (black) of Auricularia spp. Dietary fiber of three strains showed contents of about 60% but were not significantly different. ${\beta}$-glucan contents of Auricularia auricula-judae (brown) contained the highest contents with $25.21{\pm}0.37%$ and showed significant differences between Auricularia polytricha and Auricularia auricula-judae (black). Total polyphenol contents of Auricularia polytricha showed the highest contents, followed by Auricularia auricula-judae (brown) and Auricularia auricula-judae (black).
To screen biologically active components of the higher fungi of Korea, the dried carpophores of Auricularia polytricha were extracted with water. The extract was examined for acute toxicity in ICR mice. A low molecular weight toxin of this fungus was purified by acetone precipitation followed by cellulose, silica gel and LH-20 Sephadex column chromatography. Major symptoms of this toxin were eye extrusion, hair erection, trembling of head, paralysis, rapid running or moving before death and depression of respiration. The median lethal doses of the total extract were 1.25 g/kg and 4.18 g/kg by i.p. and p.o. administrations, respectively. The amounts of one mouse lethal unit (MLU) of the total extract and final fraction that killed a 20-g mouse within 30 minutes were 28.5 mg/mouse and 12 mg/mouse, respectively.

The purpose of this study is to discover a food material having anti-obesity effects and to disseminate information on the effects of the material to people who are interested in anti-obesity. For this study, 11 kinds of Auricularia (wood ear) spp., including 8 strains of Auricularia auricula-polytricha, and 3 strains of A. auricula-judae, were presented by the Jeollanamdo Agricultural Research & Extension Services. 3T3-L1 (preadipocyte cell) was used for identifying the inhibition effect on adipocyte differentiation. As a result, this study found that all the extracts had slightly different degrees of inhibition effects on adipocyte differentiation. Among the A. auricula-polytricha strains, strain 21001 showed the most significant effect (4.58%), and the inhibition effect of strain 21002 (4.43%) was the greatest among A. auricula-judae strains. Overall, the inhibition effect of A. auricula-polytricha strains was greater than that of A. auricula-judae strains. The results of mRNA and protein analysis also demonstrated that the inhibition effect of A. auricula-polytricha 21001 was superior to that of any other strains. An in vivo study using 56 ICR mice (6w, male), was performed for 4 weeks. A. auricula-polytricha 21001, which exhibited the most significant effect in the in vitro study was used to compose six different types of feeds. Daily body weight gain of the high-fat diet containing 0.2% 21001 extract feeding group was $0.22{\pm}0.08g$ (*p < 0.05), and it was 31.25% lower than that of the high-fat diet feeding group ($0.32{\pm}0.06$). Internal organ weight measurement and blood analysis were performed immediately after animal sacrifice. The results proved that treatment with more than 0.1% of A. auricula-polytricha strain 21001 could significantly reduce (more than *p < 0.05) the weight of liver and epididymal fat, and levels of glucose, total cholesterol, AST, and ALT in blood.
An attempt was made to determine the volatile flavor components of Auricularia polytricha mushroom. Essential oils from the dried mushroom were isolated by a simultaneous steam distillation-extraction(SDE) method using diethyl ether as a solvent. Total 30 components were identified bt GC-MS from the in essential oils including 11 acids, 10 alcohols, 5 aldehydes, 3 ketons, 1 other components. The major volatile components were heaxadecanoic acid(16.74%), benzeneethanol(7.77%), pentadecanoic acid(7.59%), dihydro-5-penhtyl-2-(3H)-furanone acid(4.28%), tetradecanoic acid(3.37%), pentanoic acid(3.38%) and 1-octen-3-ol(1.26%).
Twenty-one different fungi were tested for their ability to decolorize a wide range of structurally different dyes. Twenty fungal strains were isolated from fruiting bodies which were collected at the Kwangneung National Arboretum, Korea. One fungal strain were isolated from a rotting wood at Soongsil University, Korea. Nine kinds of dyes were used: three anthraquinone dyes and six azo dyes. The five fungal strains, Laetiporus sulphureus, Polyporus arcularius. Auricularia polytricha, Stereum ostrea, and Bjerkandera sp. UK-l showed decolorization ability. Except Auricularia polytricha, the four fungal strains were wood rotting fungi, and belonged to Aphyllophorales. Bjerkandera sp. UK-I, which was a white rot fungus, could decolorize all kinds of dyes tested in this study, indicating this fungus is one of candidates for applying in biological methods of dye waste treatment.
Polysaccharides have attracted great attention for their wide range of applications in biological and medical fields. In this paper, the interaction of polysaccharides with human serum albumin (HSA) was systematically investigated by fluorescence (FL) spectroscopy and circular dichroism (CD) spectra under different conditions. The Stern-Volmer quenching constants ($K_a$) at different ionic strength and pH were calculated, and information of the structural features of HSA was discussed. FL and CD results indicate that both hydrophobic and electrostatic interactions play important roles during the binding process. The quenching of the fluorescence resulting the binding of polysaccharides and HSA is static.
To screen biologically active components of the higher fungi of Korea, the carpophores of Auricularia polytricha, a well-known edible mushroom, were extracted with 0.14M NaCl solution. The extractive was successively fractionated by adding ammonium sulfate in various amounts, the respective precipitates being weparated by centrifugation, dialyzed and freeze-dried. When a dose of 60mg/kg of each was, i.p., injected into ICR mice, the fraction shich was precopitated at 20% (NH/sub 4/)/sub 2/SO/sub 4/ showed the highest toxicity, killing seven mice within two days. The fraction obtained at 40% (NH/sub 4/)/sub 2/SO/sub 4/ showed the second highest toxicity. The two fractions were named auratoxin I and II after the genus name. The symptoms of the intoxication were convulsion during the first 30 minutes after the injection, then sleeping within an hour, and tremor, lacrimation, nasal and ophthalmic bleeding, congestion and death in 24 hours. Particularly the spleen of the mice was fonud to be enlarged remarkably. The chemical analysis of the toxins showed that auratoxin I consisted of 4.4% protein and 84.5% polysaccharide and that auratoxin II 35.8% protein and 48.0% polysaccharide.

However, they Nere shown to have nearly identical composition by physicochemical and 6.8% protein. The polysaccharide moiety was found to have 12.3% $\alpha$-linkage and 87.7% $\beta$-linkage and to be a heteromannoglucan consisting of 45.1% glucose, 435 mannose and 11.0% xylose. The protein moiety contained ten amino adids. The molecular weight of the toxin was $1.5\times10^6$ dalton by Sepharose CL-4B gel filtration. The modian lethal doses of auritoxin in mice were 56.4, 157.2 and 454.6 mg/kg by i.p., s.c. and p.o.administrations, respectively. The signs of intrxication were convulsion during the first 30 minutes after the injection, coma or sleeping within an hour, termor, lacrimation, nasal bleeding congestion, and death in 24 hours. Smong the various organs, the spleen was found to be enlarged remarkably. Human platelet aggregation was inhibited by the addition of auritoxin. The activity of malic dehydrogenase in vitro was inhibited by the toxin.
