Aureobasidium pullulans ATCC 9348의 Pullulan 생산과 균체 형태의 변화 (Pullulan Production and Morphological Change of Aureobasidium pullulans ATCC 9348)

  • The pullulan production and morphological change of Aureobasidium pullulans ATCC 9348 were investigated both in batch fermentation and in continuous fermentation. The best carbon source for pullulan production was sucrose among seven different carbon sources. The pullulan production of A. pullulans was increased with increasing the carbon to nitrogen ratio of the medium using sucrose as a carbon source. In batch fermentation, production of pullullan occurred following exhaustion of the nitrogen source from the medium. The continuous fermentation showed that the pullulan production was closely parallelled with cell growth and was most effective at a dilution rate of 0.06~0.07 hr$^{-1}$-. The ratio of yeast-like cells(blastospores) of A. pullulans increased with the increase of growth rate, and reached 100% over the growth rate of 0.07 hr$^{-1}$. The growth rate, within a certain range, affected not only on the cell morphology, but on the specific pullulan productivity of A. pullulans.

  • 본 연구는 알코올 발효능이 우수한 야생 효모를 막걸리 발효에 응용할 목적으로 충남 예산군 예당저수지 야생화에서 분리한 비병원성 야생 효모들 중 에탄올 생산 우수 효모로 선발한 Aureobasidium pullulans P-1의 균학적 특성과 막걸리 발효 조건을 조사하였다. A. pullulans P-1는 출아에 의해 영양증식을 하였고 자낭포자를 생성하는 유포자 효모로서 내당성과 에탄올 내성이 강한 호염성 야생 효모이었다. A. pullulans P-1의 yeast extract-peptone-dextrose 배양액을 주모로 증자미와 입국과 물이 혼합된 술밑에 5% 첨가한 후 $25^{\circ}C$에서 1~10일간 발효시키면서 발효 중의 이화학적 성질의 변화를 조사한 결과, 에탄올 함량은 $25^{\circ}C$로 10일간 발효시켰을 때 가장 많은 8.45%를 생성하였고 관능 특성이 우수하였으며 항고혈압 활성 안지오텐신 전환효소 저해활성이 71.1%로 높았다.

  • A study on the removal of mercury by Saccharomyces cerevisiae and Aureobasidium pullulans was done, in which the model of adsorption isotherm and adsorption rate was proposed. The adsorption isotherm of mercury by S. cerevisiae was accorded with Langmuir model but A. pullulans was followed to Freundlich model. The amount of mercury removed by A. pullulans was higher than that of S. cerevisiae, but the adsorption rate of mercury by A. pullulans was slower than that of S. cerevisiae. In a rapid adsorption process, therefore, it is more useful to use S. cerevisiae as a biosobent.

  • Aureobasidium pullulans produced three kinds of extracellular polyols e.g. glycerol, mannitol, and sorbitol from either sucrose, glucose, fructose or mannose. Sorbitol was selectively produced when urea was used as a sole nitrogen source, and the amounts of sorbitol produced rapidly reached a plateau after 50 h where its maximum quantity was about 20 g/l with sucrose.

  • Extracellular fructosyl transferase from Aureobasidium pullulans C-23 was characterized. The molecular weight of the isolated enzyme was determined to be approximately 170,000 by SDS polyacrylamide gel electrophoresis. The enzyme has the pI value of about 3.7. The enzyme was almost completely inhibited by 5mM $Hg^{2+}$ , but was not significantly affected by other cations tested. The enzyme was inactivated by treatment of tryptophan-specific reagent N-bromo- succinimide and tyrosine-specific reagent iodine. The substrate sucrose showed protective effect on the inactivation of the enzyme by the both reagents. These results suggest that tryptophan and tyrosine residues are probably located at or near active site of the enzyme.

  • For the production of pullulan from the high concentration of sugar, the utilization of sugars by a pullulan-producing fungus, Aureobasidium pullulans was examined. A. pullulans showed the different utilization patterns for sugars such as sucrose, maltose, and maltotriose. Especially for maltotriose, the hydrolysis of sugar was accompanied by a transferase activity. Glucose and maltose showed the inhibitory effect on the cell growth and the pullulan production at the sugar concentration higher than 0.28M, but sucrose showed the inhibitory effect at the sugar concentration higher than 0.14M. Among the sugars examined, sucrose gave the best result for the pullulan production. 27.5g/l of pullulan was obtained from 5% sucrose.

  • A fungal strain was isolated as a pullulan-producer from plant leaves and identified as Aureobasidium pullulan GM21. With A. pullulans GM21, culture conditions were optimized for the pullulan production and the changes of the molecular weight of pullulan produced were investigated according to the culture conditions we obtained maximum conversion yield of pullulan about 58-60%(40.8-42.0g/l) from 7% sucrose at 25C, initial pH 7.5 by the batch cultivations either in Erlenmeyer flask or in jar fermentor.

  • Xinlei studies on the production of pullulan by Aureobasidium pullulans using batch culture in a 15L bioreactor were carried out. The mathematical models were obtained in this study, which provided a reasonable description for the biomass, the product, and the substrate variation with time. The values frets the mathematical models were satisfactorily coincided with the experimental data for the biomass of A. pullulans, the production of pullulan and the utilization of sucrose as the sole carbon source.

Draft Genome Sequence of Aureobasidium pullulans Strain MHAU2101, a Biological Control Agent against Fire Blight from Korea

  • In this study, we present the draft genome of Aureobasidium pullulans strain MHAU2101, which is the first strain to effectively control fire blight caused by Erwinia amylovora in Korea. The genome of strain MHAU2101 was composed of 28,669,322 base pairs, with a C+G content of 50.4%. The assembly comprised 17 contigs and had 99.22% completeness. The results of this study will be a valuable resource for future research on the biocontrol mechanism of A. pullulans strain MHAU2101.

국내 배나무 꽃에서 분리한 Aureobasidium pullulans MHAU2101의 화상병 발생 억제 효과 (Inhibitory Effects of Aureobasidium pullulans MHAU2101 Isolated from Domestic Pear Blossom Against Fire Blight)

  • 본 연구는 국내 배나무 꽃에서 분리한 효모의 형태학적 특성 및 유전자 염기서열 분석을 통해 종을 동정하고, 사과와 배의 미숙과와 꽃사과의 꽃을 대상으로 화상병 발병 억제효과를 확인하고자 수행하였다. 효모의 형태적 특성을 조사한 결과 기존 A. pullulans의 특성과 일치하였으며, large-subunit (LSU) 26S ribosomal DNA의 D1/D2 영역과 internal transcribed spacer (ITS) 영역의 염기서열을 분석한 결과 A. pullulans (MHAU2101)로 동정되었다. E. amylovora를 미숙과에 접종하기 전에A. pullulans MHAU2101을 처리할 경우 사과와 배의 미숙과에서 모두 화상병 병징이 나타나지 않았다. 꽃사과 꽃에서 화상병 방제 효과를 검정한 결과, A. pullulans MHAU2101 처리구는 무처리구에 비해 꽃 감염률이 유의하게 낮았으며, 스트렙토마이신 수화제 대비 64%의 방제효과를 보였다. 또한A. pullulans MHAU2101 처리구는 무처리구와 비교하여 암술과 화통 모두E. amylovora의 밀도가 낮았으며 특히 화통에서 밀도가 낮은 특징이 있었다. 따라서 본 연구 결과를 통해 국내 배나무 꽃에서 분리한A. pullulans MHAU2101이 화상병 발병을 억제하는데 효과가 있음을 확인하였다.
