Atmospheric Bioaerosol, Bacillus sp., at an Altitude of 3,500 m over the Noto Peninsula: Direct Sampling via Aircraft

  • This work focuses on the analysis of bioaerosols in the atmosphere at higher altitudes over Noto Peninsula, Japan. We carried out direct sampling via aircraft, separated cultures, and identified present isolates. Atmospheric bioaerosols at higher altitudes were collected using a Cessna 404 aircraft for an hour at an altitude of 3,500 m over the Noto Peninsula. The aircraft-based direct sampling system was devised to improve upon the system of balloon-based sampling. In order to examine pre-existing microorganism contamination on the surface of the aircraft body, bioaerosol sampling was carried out just before takeoff using the same method as atmospheric sampling. Identification was carried out by a homology search for 16S or 18S rDNA isolate sequences in DNA databases (GenBank). Isolate sampling just before takeoff revealed Stretpomyces sp., Micrococcus sp., and Cladosporium sp. One additional strain, Bacillus sp., was isolated from the sample after bioaerosol collection at high altitude. As the microorganism contamination on the aircraft body before takeoff differed from that while in the air, the presence of additional, higher atmosphere-based microorganisms was confirmed. It was found that Bacillus sp. was floating at an altitude of 3,500 m over Noto Peninsula.

Andersen 시료채취기를 이용한 도시대기중 부유입자상 물질의 입도분포 측정 (Determination of Size Distribution of Atmospheric Paticulates in Urban Air Using Andersen Sampler)

  • Mass size distribution of atmospheric particulates were measured using an Andersen using an Andersen sampler in urban air. The atmospheric particulates were fractionated in eight stages of an Andersen sampler operating at 28.4$\ell/min$ and collected on polyester sheets. A quartz filter was placed behind the last stage collect permeated partculates. The size distribution of atmospheric particulates were divided around 1-2 $\mu$m into two groups, coarse and fine particulates regardless of sampling times. The variation of course particulates concentration was higher than fine particulates among sampling times. Different meterorological conditions and natural phenomena brought high variation of course particulates' concentration. The rain caused removal of coarse particulates seriously and the yellow and may take part in a increase of the course particulates in spring. The average concentration of atmospheric particulates to be collected by Andersen sampler was 170.8 $\mug/m^3$ during 3 times of sampling. Among them the average concentration of atmospheric particulates which could penetrated under a bronchi and alveoli were 70.4$\mug/m^3$ and 36.6$\mug/m^3$ respectively.

개별입자 분석을 위한 대기에어로졸의 시료채취법 (Sampling Method for Individual Particle Analysis of Atmospheric Aerosol)

  • In this study, the most suitable sampling methods for the bimodal mass distribution characteristics and individual particle analysis of atmospheric aerosols were investigated. Samples collected in Quartz, Teflon, and Nuclepore filters were analyzed for individual particles using scanning electron microscopy with an energy-dispersive X-ray spectrometer (SEM/EDS). Then, the pore diameter of the filter and the collection flow rate were determined using the theoretical collection efficiency calculation formula for two-stage separation sample collection of coarse and fine particles. The Nuclepore filter was found to be the most suitable filter for identifying the physical and chemical characteristics of atmospheric aerosols since it was able to separate the sample and count the different sized particles better than either Quartz or Teflon. Nuclepore filters with 8.0 ㎛ and 0.4 ㎛ pores were connected in series and exposed to a flow rate of 16.7 L/min for two-stage separation sampling. The results show that it is possible to separate and collect both coarse and fine particles. We expect that the proposed methodology will be used for future individual particle analysis of atmospheric aerosols and related research.

대기중 휘발성 유기화합물의 채취 및 분석 방법 비교 (Comparison of Sampling and Analysis Methods for Volatile Organic Compounds in Ambient Air)

  • A field comparison study was carried out to quantify differences among various sampling and analytical methods for volatile organic compounds (VOCs) at a site in Vlsan in June 1997. Air sampling containers (SUMMA canisters) were used by the Korea Institute of Science and Technology (KIST) and adsorption tubes (carbotrap) were used by Yeungnam University (YN Univ.) for sampling ambient air. Duplicate samples obtained by KIST were analyzed by KIST with a GC-MS system for aromatics and halogenated hydrocarbons and by Atm AA with a GC -FID system for C2∼C9 hydrocarbons, respectively. The adsorption tube samples were analyzed by YN Univ. with a GC-FID system for aromatics. VOC levels for the duplicate canister sampls analyzed by KIST and Atm AA were in good agreement. Concentrations of aromatics by the adsroption tube method were generally higher than those by the canister sampling method by factor of 1.5 to 2.0. Differences between the two sampling methods were discussed.

폴리우레탄 폼 수동형 공기시료채취기를 이용한 대기 중 다이옥신/퓨란 농도 측정 (Measurement of Atmospheric PCDD/Fs Concentrations Using Polyurethane Foam Disk Passive Air Samplers)

  • Objectives: This study was conducted to evaluate the use of polyurethane foam disk passive air samplers (PUF PAS) for better measurement of atmospheric polychlorinared dibenzo-p-dioxins/furans (PCDD/Fs) concentrations compared to PUF PAS combined with high volume air samplers (HVS). Methods: Air samples were collected by a low volume air sampler (LVS) and PUF PAS. A total of two pairs were continuously collected for six months, but the PUF was replaced every two months. Results: A good correlation was shown ($R^2=0.8595$, p<0.0001) between atmospheric PCDD/Fs concentration measured by the LVS and PUF PAS. The average air sampling rate ($1.5m^3/day-sampler$) of all PCDD/Fs congeners showed a middle of the means which were measured using a HVS by other researchers in different cities. In addition, the air sampling rates of the LVS for each congener made less difference than did those of the HVS. Conclusion: It was found that measurements using the LVS were less influenced by atmospheric peak PCDD/Fs concentrations. However, trace POPs such as PCDD/Fs may involve relatively large analytical errors in measurement, and as a result the air sampling rate of the respective PCDD/Fs isomer is also likely to involve errors. The method of using a regression straight line between the concentrations obtained from the LVS and those from the PUF PAS was judged higher than the method using the air sampling rate, since the former compensated for the experimental errors in the process of evaluation of atmospheric PCDD/F concentrations using the PUF PAS.

국내 대기 중 독성 휘발성 유기화합물의 오염 특성(I) - 측정 방법론 평가 - (Characteristics of Atmospheric Concentrations of Toxic Volatile Organic Compounds in Korea ( I ) - Evaluation of Sampling and Analytical Methodology)

  • This study was designed to investigate the characteristics of atmospheric concentrations of toxic volatile organic compounds (VOCs) in Korea. Target compounds included 1,3-butadiene, aromatics such as BTEX, and a number of carbonyl compounds. In this paper, as the first part of the study, the performance of sampling and analytical methods was evaluated for the measurement of selected VOCs and carbonyl compounds in the ambient air. VOCs were determined by the adsorbent tube sampling and automatic thermal desorption coupled with GC/MSD analysis, while carbonyls by the DNPH-silica cartridge sampling with HPLC analysis. The methodology was investigated with a wide range of performance criteria such as repeatability, linearity. lower detection limits, collection efficiency, thermal conditioning, breakthrough volume and calibration methods using internal standards. In addition, the sampling and analytical methods established in this study were applied to real field samples duplicately collected in various ambient environments. Precisions for the duplicate samples appeared to be comparable with the performance criteria recommended by USEPA TO-17. The overall precision of the sampling and analytical methods was estimated to be within 20 ∼ 30% for major aromatic VOCs such as BTEX, whereas the precision for major carbonyl compounds such as formaldehyde and acetaldehyde was within 10 ∼ 20% for field samples. This study demonstrated that the adsorbent sampling and thermal desorption method can be reliably applied for the measurement of BTEX in ppb levels frequently occurred in common indoor and ambient environments.

대기 중 오염물질의 시료채취시 관측오차 저감방법에 대한 연구 : 6구형 매니폴더를 장착한 MFC system의 개발과 평가 (Methodological Approaches to Reduce Uncertainties Associated with Air Sampling : Development and Assessment of a Six-port Manifold MFC System)

  • In order to develop a confident sampling technique, we designed and constructed a 6-port manifold MFC sampling system for collecting gaseous pollutants in air. Using this instrumentation, we tested the performance criteria of MFC system in terms of: (1) flow rate; (2) MFC-to-MFC variability; (3) tube-to-tube variability; and (4) time. It was interesting to find that the later two factors did not show any significant variations, while the former two show substantially large variations. However, as most of those variabilities are consistent enough to form systematic patterns, we were able to explain the occurrence patterns of all those MFC biases in terms of those four major variables. The overall results of our experiment suggest that one needs to use correction factor for each MFC unit under a given flow rate to maintain optimal accuracy and precision for sampling of those pollutants.

수산화바륨을 이용한 이산화탄소 확산측정법의 개발 및 평가 (Development and Evaluation of a Carbon Dioxide Diffusive Sampling Method using Barium Hydroxide)

  • This study was aimed at developing and evaluating a diffusive sampling method using a barium hydroxide solution as an absorbent for measuring carbon dioxide ($CO_2$) in ambient air. The collected $CO_2$ concentration was calculated by the change of conductivity resulted in the reaction of $CO_3{^{2-}}$ and $Ba^{2+}$ in aqueous solution. The sampling rate for the diffusive sampler was determined 0.218 mL/min, as obtained from the slope of the linear correlation between the $CO_2$ mass collected by the diffusive sampler and the time-weighted $CO_2$ concentration with the active sampling method. The unexposed blank sampler sealed in aluminium foil-polyethylene laminated packets has remained stable during at least one-month storage period. A good correlation was observed between the diffusive sampler and active sampler with a coefficient of determination of 0.956. This diffusive sampler would be suitable for the indoor $CO_2$ concentration monitoring.

대기 중 다환방향족 탄화수소의 측정을 위한 시료포집방법의 비교평가 (Evaluation of Sampling Methodology for the Measurement of Polycyclic Aromatic Hydrocarbons in the Atmosphere)

  • This study was carried out to investigate the influence of different sampling methods on the measured concentrations of polycyclic aromatic hydrocarbons (PAH) both in the vapor and particulate phases, and to evaluate the effects of ambient temperature and sampling duration on the losses of PAH associated with particle samples due to volatilization. The experimental protocol of this study is consisted of two parts. The first part is related to the comparison of PAH concentrations measured by 4 different sampling systems, each of which involves different sampling principles for comparison purposes, including a medium-volume sampler with XAD-2 adsorbent, a high-volume sampler with polyurethane foam (PUF), two identical low-volume samplers: one with XAD-2 and the other with PUF, respectively. The second part of this study is to quantitatively estimate the losses of particulate PAH samples by volatilization during sampling, using two identical low-volume samplers: one was used for changing the filters every 3 hrs, 6 hrs, 12 hrs, and 24 hrs sampling, while the other was maintained for continuous 48 hours sampling without changing the filter. The concentrations of volatile PAH including 2-3 rings appeared to be significantly affected by the type of adsorbent. Measured levels of these lower-molecular weight PAH by XAD-2 adsorbent were much higher than those by PUF for both high-volume and low-volume sampling. PUF was found to give rise to unknown components that interfered with the PAH analysis, even after extensive clean-up. In addition, the retention efficiency of PUF for lower molecular weight PAH was subject to a large variation, being significantly influenced by sampling conditions such as ambient temperature. However, the effect of sampling methods with different adsorbents on the measured levels of semi-volatile compounds including 4 rings PAH such as fluoranthene, pyrene, BaA and chrysene, was not so much significant as more volatile PAH compounds. It was also clear from this study that volatilization losses of the semi-volatile PAH collected on the filters were inevitably occurred during prolonged sampling, and hence the results obtained from conventional sampling methods may not be expected to yield an accurate distribution of PAH between the vapor and particulate phases.
