Genetic assessments of rare and endangered species are among the first steps necessary to establish the proper management of natural populations. Transcriptome-derived single-sequence repeat markers were developed for the Korean endangered species Astilboides tabularis (Saxifragaceae) to assess its genetic diversity. A total of 96 candidate microsatellite loci were isolated based on transcriptome data using Illumina pair end sequencing. Of these, 26 were polymorphic, with one to five alleles per locus in 60 individuals from three populations of A. tabularis. The observed and expected heterozygosity per locus ranged from 0.000 to 0.950 and from 0.000 to 0.741, respectively. These polymorphic transcriptome-derived simple sequence repeat markers would be invaluable for future studies of population genetics and for ecological conservation of the endangered species A. tabularis.
Tabulis [Astilboides tabularis (Hemsl.) Engl.] has the potential for becoming a useful horticultural crop. This study was conducted to classify the seed dormancy types and to identify the germination conditions that improve the horticultural usefulness of this plant. We found that A. tabularis (Hemsl.) Engl. produced undifferentiated embryos and had a low germination rate of under 50%. In addition, water submersion led to moisture absorption. Therefore, A. tabularis seeds were designated as morphologically dormant. The germination rate was highest (86.3%) at $30^{\circ}C$ under light conditions. Higher temperatures generally led to an acceleration of the germination process, regardless of light condition. To improve the germination rate, seeds were submerged in various concentrations of growth regulators, such as $GA_3$ and kinetin, as well as minerals such as $KNO_3$ and KCl. An analysis of germination characteristics at $30^{\circ}C$ under light conditions revealed that treatment with 200 and $500mg{\cdot}L^{-1}GA_3$ led to an excellent germination rate of 97.0%. Treatment with $100mg{\cdot}L^{-1}GA_3$ led to a better germination rate than the control as well. However, treatment with kinetin, $KNO_3$, and KCl had no effect on germination, regardless of concentration.

Astilboides tabularis (A. tabularis) has been thought to be material for functional food and could help to prevent diabetes for centuries. A. tabularis has been reported to contain phytochemicals (catechin, chlorogenic acid, and rutin), which have antioxidant and anti-inflammatory effects. We focused on anti-inflammatory effects of ethyl acetate fraction of A. tabularis (EAT) through NF-κB signaling pathway. Phytochemicals of EAT were analyzed by total flavonoid and phenolics assay, its content was 219.2 ± 2.01 and 524.7 ± 0.80 mg/g respectively. EAT significantly reduced the expression of iNOS and COX-2, which caused the production of nitric oxide (NO). EAT suppressed mRNA levels of iNOS, COX-2, and cytokines (IL-6, IL-1β, and TNF-α). EAT suppressed the expression of p-p65 through the inhibition of phosphorylation of IkB-a, and the result of immunofluorescence showed that inhibited the translocation of p65 from the cytoplasm to the nucleus.
The environmental characteristics of Astilboides tabularis were investigated to compile basic data for conservation and restoration. Natural habitats, according to investigations, were mostly located on the slopes of mountains facing north at an altitude of 432 m to 1,413 m above sea level with angles of inclination ranging from 16 degree to 49.5 degree. A total of 246 vascular plant taxa were identified from 12 quadrats in 11 habitats. The importance value of Astilboides tabularis is 20.40%, and 6 highly ranked species such as Dryopteris crassirhizoma (4.56%), Rodgersia podophylla (3.72%), Astilbe rubra (2.81%), Carex siderosticta (2.67%), Ainsliaea acerifolia (2.65%), and Meehania urticifolia (2.46%) are considered to be an affinity with Astilboides tabularis in their habitats. Average species diversity was 1.23, dominance and evenness were found to be 0.11 and 0.86, respectively. The litter depth above the soil surface was 2 to 10 cm, and soil depth was 20.8 to 67.5 cm. The soil texture of habitats were divided three types such as loam, silt loam, and sandy loam. The soil pH was 5.63 to 7.74, and the organic matter and nitrogen contents were ranged from 8.88 to 42.55%, and 1.40 to 6.58 mg/g, respectively.
This study was carried out to classify and describe the vegetation of algific talus in Korea by methods of the ZM school of Phytosociology. A part of slope or a hollow at the foot of talus in which cool air exhausts from underground through gaps of accumulated rock debris is called algific talus. The most important thing for conserving plant diversity in an area is to make an overall inventory of the plant species inhabiting the area. In particular, limestone areas are known for their high plant diversity and distribution of specific plants. Despite that, only a few have been designated as protected areas. This study investigated the vascular plants distributed in Goyangsan Mtn. and Munraesan Mtn., located in limestone areas of the central part of the Korean Peninsula. A field survey was conducted eight times from April to October 2021. As a result, we identified a total of 654 taxa comprising 113 families, 357 genera, 592 species, 15 subspecies, 44 varieties, and 3 formulas. They included four endangered wild plant species: Astilboides tabularis, Eleutherococcus senticosus, Cypripedium macranthos, and Epilobium hirsutum. Endemic plants in Korea were identified as 32 taxa. Floristic target plants were identified as 168 taxa, specifically 5 taxa of grade V, 41 taxa of grade IV, and 36 taxa of grade III. The red data plants included 2 taxa as "Endangered (EN)", 7 taxa as "Vulnerable (VU)", and 7 taxa as "Near threatened (NT)". A total of 41 taxa of naturalized plants were identified, and 4 of them were invasive alien plants. The surveyed vicinity of Goyangsan Mtn. and Munraesan Mtn. showed high plant diversity and contained core habitats for distribution of an endangered wild plant, Astilboides tabularis,in the limestone area. Moreover, both mountains contained a small population of Cotoneaster integerrimus. These findings confirm that the area has conservation values. Therefore, we propose to identify areas with high plant diversity and designate them as special protected areas.

In this study, the vascular plants of the Mt. Deokhangsan protected area were surveyed for the purpose of forest genetic resources conservation in Gangwon-do, South Korea. The flora on Mt. Deokhangsan were classified as follows 537 taxa, 90 families, 302 genera, 479 species, 4 subspecies, 51 varieties, and 3 forms. Among the taxa, 19 were Korean endemic plants and 17 were rare and endangered plants. Plants as indicators of limestone areas were found in 33 taxa. Naturalized plants were found in 27 taxa and species threatened by climate change were found in 33 taxa, including 11 endemic plant taxa and 22 northern plant taxa. Plants endemic to Korea and northern plants were mainly found at the study site, whereas southern plants were not found. Thus, Mt. Deokhangsan apparently has an environment suitable for the growth of northern plants.
