삼면이 바다인 우리나라의 연안에 번식력과 포식성이 매우 강한 아무르불가사리의 개체수가 매년 급증하고 있다. 더욱이 아무르불가사리는 살아있는 어패류를 먹기 때문에 양식 어민들의 피해가 매우 크다. 따라서 불가사리를 구제하거나 불가사리의 개체수의 변화 및 이동 경로 파악 등을 위하여 수중 영상에서 불가사리 개체를 인식하는 기술은 매우 중요하다고 할 수 있다. 본 논문에서는 아무르불가사리의 형태적 특징 분석을 이용한 개선된 인식 방법을 제안하였다. 제안한 인식 방법은 아무르불가사리의 형태적 특징인 오목과 볼록에 대한 추출 정보가 부족한 경우를 위해 제안된 개선된 인식 방법이다. 바다 속에서 촬영되는 불가사리의 영상에서 불가사리의 형태적 특징을 모두 추출하는 것은 매우 어렵다. 따라서 제안하는 인식 방법은 다양한 아무르불가사리 영상에서 개체를 인식하는데 효과적이다. 실험결과, 제안한 기법의 인식률은 92.5%로 기존 기법보다 우수한 성능을 보였다.
패턴을 이용한 불가사리 특징 검출은 불가사리의 오목 특징과 볼록 특징을 모두 검출하기 어려우며 또한, 오목과 볼록을 구분 할 수도 없다. 오목과 볼록은 아무르불가사리의 중요한 구조적 특징으로서 반드시 찾아야 할 특징이며 오목과 볼록을 분류함으로서 차후 불가사리 인식에서도 필요하다. 따라서 본 논문에서는 아무르불가사리의 주요 특징인 오목과 볼록 특징을 추출하는 기법을 제안한다. 이 기법은 다방향 선형 스캐닝을 이용하여 오목과 볼록의 특징점 후보군을 형성하고 이 후보군에서 특징점을 결정한 후 추출된 특징점에 컨벡스 헐 알고리즘을 적용하여 오목 특징과 볼록 특징을 구분한다. 제안한 기법은 불가사리의 주요 특징인 오목 특징과 볼록 특징을 구분하여 효과적으로 추출한다. 따라서 향후 불가사리 인식을 위한 연구에 기여할 것으로 기대한다.
수산 해적생물인 불가사리의 주정추출물을 농도별로 사료에 첨가하여 조피볼락의 성장, 혈액성상 및 식세포 활성산소 생산에 미치는 효과를 조사하였다. 불가사리추출물의 함량이 다른 사료를 급여한 조피볼락의 1일성장율(SGR), 사료계수(FCR), 비만도(CF), 생존율(SR)을 비교한 결과, 실험군별 유의적인 차이를 보이지 않았다. 이는 불가사리추출물에 의한 먹이 기피현상을 방지하기 위해 사료를 코팅을 하였기 때문인 것으로 사료된다. 불가사리추출물이 조피볼락의 체내 대사계에 미치는 영향을 알아보기 위해 혈청 중의 glucose, total cholesterol의 함량과 GOT를 측정한 결과, glucose와 total cholesterol의 함량은 불가사리추출물의 함량이 증가할수록 높아지는 경향을 보였다. 한편, 혈청 GOT는 실험군별로 유의적인 차이를 보이지 않았으며 이는 불가사리추출물이 독성이 거의 없기 때문인 것으로 추정된다. 불가사리추출물 첨가사료의 급이가 조피볼락의 면역계에 미치는 영향을 알아보기 위하여 식세포의 활성산소 생산량을 측정한 결과, 불가사리추출물의 첨가에 따라 농도 의존적으로 증가하였다. 이상의 실험결과에서 불가사리추출물은 면역증강 기능성 사료첨가제로 활용할 수 있을 것으로 사료되었다.
This study was carried out to evaluate off-shore water quality and to draw marine pollution map to Southern sea in Korea by bioassay using gametes and early development system of Asterias amurensis. From the bioassay, it was determined that the water qualites of Southern sea maintained the grade II, because development of A. amurensis were moderately inhibited. However, Sunchon, Kwangyang, and Jinhae Bay belonged to grade III, because development of A. amurensis were strongly inhibited. This could be due to the inflows of several river systems and poor water circulation. The water quality at Yosu island was the grade I, because A. amurensis were not inhibited in its early development.
Asterias amurensis (starfish) is a marine organism that is harmful to the fishing industry, but is also a potential source of functional materials. The present study was conducted to analyze the profiles of fatty acids extracted from A. amurensis tissues and their anti-inflammatory effects on RAW264.7 macrophage cells. In different tissues, the component ratios of saturated fatty acids, monounsaturated fatty acids, and polyunsaturated fatty acids differed; particularly, polyunsaturated fatty acids such as dihomo-gamma-linolenic acid (20:3n-6) and eicosapentaenoic acid (20:5n-3) were considerably different. In lipopolysaccharide-stimulated RAW264.7 cells, fatty acids from A. amurensis skin, gonads, and digestive glands exhibited anti-inflammatory activities by reducing nitric oxide production and inducing nitric oxide synthase gene expression. Asterias amurensis fatty acids effectively suppressed the expression of inflammatory cytokines such as tumor necrosis $factor-{\alpha}$, interleukin-$1{\beta}$, and interleukin-6 in lipopolysaccharide-stimulated cells. Cyclooxygenase-2 and prostaglandin $E_2$, which are critical inflammation biomarkers, were also significantly suppressed. Furthermore, A. amurensis fatty acids reduced the phosphorylation of nuclear $factor-{\kappa}B$ p-65, p38, extracellular signal-related kinase 1/2, and c-Jun N-terminal kinase, indicating that these fatty acids ameliorated inflammation through the nuclear $factor-{\kappa}B$ and mitogen-activated protein kinase pathways. These results provide insight into the anti-inflammatory mechanism of A. amurensis fatty acids on immune cells and suggest that the species is a potential source of anti-inflammatory molecules.
Two neuropeptides were purified from the acidified tube feet extract of the starfish Asterias amurensis by C18 reversed phase and size-exclusion high-performance liquid chromatography (HPLC). The tube feet extract and the purified peptides (AST-I and AST-II) showed potent contractile activity on dorsal retractor muscle (DRM) of the starfish Asterina pectinifera and intestine (smooth muscle) of the panther puffer Takifugu pardalis. Treatment of the purified peptides with dithiothreitol (DTT) for 60 min at $37^{\circ}C$ significantly altered their retention times, suggesting that these compounds contained disulfide bonds. The molecular weights of AST-I and AST-II were determined to be 2064.2 Da and 6137.2 Da, respectively, by MALDI-TOF mass spectrometry.
Bioassays using gametes of sea urchins are widely used in ecotoxicological assessments of marine environments. Since most of sea urchin species in Korean coastal water spawn from spring to autumn, bioassay with them during the winter is impossible. In the course of developing standard methods for bioassays with Korean species, we found a winter-spawning starfish, Asterias amurensis, Since reproductive mode of asteroids is similar to echinoids, the bioassay protocol for sea urchins could be applied similarly to the starfish. Here, we tested and determined several conditions for the acceptability of bioassay with A. amurensis. The least required time for formation of fertilization membrane of fertilized eggs to be easily distinguished from unfertilized ones was 60 min. The threshold of sperm to egg ratio that could make acceptable fertilization rates in controls was 3000. The allowed time for manipulation of sperm after dilution in seawater was at most 3 hr. The optimal exposure time of sperms when the response against toxicant solution was relatively stable was in the range of 20-60 min. The tolerance range of sperms to the salinity of test solution was 26-38 psu. The sensitivity of A. amurensis sperm was intermediate among marine organisms commonly used in aquatic toxicity tests. The sperm bioassay with A. amurensis can be satisfactorily applied to toxicity assessments of marine environments.
This study examined the antimicrobial, antioxidant, and tyrosinase inhibitory activities of bioactive compounds extracted from two starfish, Asterina pectinifera and Asterias amurensis, using solvent extraction after $Protamex^{TM}$ hydrolysis. Methanol and acetone fractions collected by stepwise extraction from specimens were subjected to silica gel column chromatography (SGCC) (200 mesh and 400 mesh), followed by high-performance liquid chromatography (HPLC). Two fractions (7:3 and 5:5 chloroform : methanol ratio, v/v) eluted using silica gel column chromatography from the two starfishes showed higher antimicrobial activity against Propionibacterium acnes and dermatophyte fungi (Epidermophyton floccosum, Microsporum audouinii, Trichophyton ferrugineum, Trichophyton mentagrophytes, and Trichophyton rubrum), antioxidant activity ($EDA_{50}$, mg/mL), and tyrosinase inhibitory activity compared to the other fractions. The final fractions obtained from Asterina pectinifera (RT 7.53, 8.93, and 10.48 min) and Asterias amurensis (RT 5.02 min) by SGCC (400 mesh) and HPLC from two SGCC fractions (200 mesh) showed 8.94 and 15.59 mg/mL antioxidant activity ($EDA_{50}$) and 46.89 and 40.19 % tyrosinase inhibitory activity, respectively. Extracts from starfishes are potential cosmetic basic material.
우리나라는 삼면이 바다인 환경적 특성으로 인해 연안 양식 산업이 발전해 왔다. 번식력과 포식성이 매우 강한 아무르불가사리에 의해 우리나라 근해의 양식 산업이 받는 피해가 매년 급증하고 있다. 더욱이 아무르불가사리는 살아있는 어패류를 잡아먹기 때문에 양식 어민들의 피해가 매우 크다. 이 논문에서는 수중에서 획득한 아무르불가사리 영상에서 효과적으로 특징을 추출하는 방법을 제안하였다. 제안한 방법에서는 6-방향 스캐닝을 이용한 볼록 특징 추출을 사용하여 기존 방법에 비해서 적은 수의 특징 후보를 산출한다. 또한, 추출된 볼록 특징과 영역 중심점을 이용하여 후보 오목점을 선정한 후 최종 오목 특징을 추출한다. 군집 생활을 하는 불가사리의 특성 때문에 입력 영상의 불가사리 개체도 밀집되어 있다. 따라서 입력 영상에서 추출되는 특징 후보의 수를 최소화하는 것은 중요한 의미를 갖는다. 실험결과, 제안한 특징 추출 방법은 특징 후보수 대비 특징 추출률이 약 88%로 기존 방법에 비해 개선되었다.
전라남도 장흥군 인근 해안에서 채집한 불가사리(Asterias amurensis)를 대상으로 불가사리의 장내에 분포하는 종속영양세균의 균체수를 측정하였고, 분리동정된 균주들을 대상으로 탄소원 이용 특성을 조사하였다. 불가사리의 장내에 분포하는 종속영양세균의 균체수는8.65$\pm$0.65 $\times$$10^3$ cfu . g$^{-1}$이 였으며, 분리 동정된 24균주(그람음성균 11균주, 그람양성균이 13균주)중에서 Vibrio속,Staphylococcus속, Corynebacterium속이 높게 나타났다. 불가사리 장내에서 분리 동정된 세균들은 그람양성의 간균이 높은 비율을차지하였고, catalase와 oxidase의 활성은 각각 54.2%,20.8%로 나타났다. 그람음성균의 경우, 탄소원으로 glucose의 이용률이 가장 높았고, 그 다음으로 sucrose, mannose, maltose,그리고 trehalose의 순으로 높은 이용률을 나타냈으며, adipate, phenol acetate는 이용하지 못하였다. 그람양성균의 경우, glucose가 가장 높은 이용률을 나타냈고, 그 다음은 fructose, trehalose, maltose 순으로 높은 이용률을 보였다.
