Formation and Fusion of Protoplasts from the Cellulolytic Fungi, Aspergillus niger MAN-831 and Aspergillus wentii MAW-538 (Cellulase를 생산하는 Aspergillus niger MAN-831과 Aspergillus wentii MAW-538의 원형질체 형성 및 융합)

  • For the effective utilization of cellulosic biomass, conidial protoplast fusion between Aspergillus niger MAN-831(${\beta}-glucosidase$) and A. wentii MAW-538(CMCase and avicelase), which produced potently cellulolytic enzymes was carried out. Optimal conditions for formation and regeneration of protoplast were conidiospore age-5 dyuas. $2-DG-30\mu\textrm{g}/ml$, preincubation time-4 hours, osmotic stabilizer-0.7M KCl, novozyme(7mg/ml)+driselase(2.5mg/ml) and reaction time of enzyme-5 hours. Optimal conditions for protoplast fusion were obtained by treatment of protoplasts with 15mM CaCl2 and 25% polyethylene glycol 4000(pH 6~7) as fusogenic agent at $36^{\circ}C$ for 25~30 minutes. The frequency was then $7.94{\times}10^{-4}$. CMCase, avicelase and ${\beta}-glucosidase$ activity of fusant F-208 strain was 1.5, 1.3, 1.2 times higher than those of parental strains, respectively.

  • Regeneration of protoplast was effective by preincubating spore suspension containing 30$\mu g$/ml of 2-DG for 4 hours, and CBE medium containing casamino acid, bovine serum albumin, ergosterol and myoinositol was found to be more efficient than any other regeneration medium tested in this experiment. The regeneration frequency was about 30%. Optimal conditions for conidial protoplast fusion were obtained by treatment of protoplasts with 10 mM $CaCl_2$ and 30% polyethylene glycol 4000 (pH 7.5) as fusogenic agent at $37^{\circ}C$ for 10 minutes. The fusion frequency was $8.2\times 10^{-4}$. The higher productivity of enzyme of fusant FWN-56 was achived: 2.3-fold for CMCase, 1.5-fold for avicelase, 1.8-fold for $\beta$-glucosidase and 2.5-fold for xylanase compared to that obtained in two parental strains. The genetic stability of fusant after maintenance on minimal medium for more than 4 weeks was high because segregant rate was below 1%. The conidial DNA content of fusant was 1.4-1.6 times higher than that of the parental strains, The nucleus size of fusants were also higher than that of each parental strains.

  • Through the study of identification of filamentous molds isolated from 12 traditional Nuruk, collected from several regions in Korea, 78 strains of Absidia spp., including Absidia corymbifera, Absidia ramosa, and Absidia sp. of which specific names were not identified, 19 strains of Rhizopus spp. including Rhizopus cohnii, Rhizopus arrhizus, and Rhizopus oryzae, 8 strains of Circinella spp., 1 strain of Actinomucor sp., 49 strains of Aspergillus spp., including Aspergillus oryzae, Aspergillus flavus, Aspergillus flavus var. columnaris, Aspergillus wentii, Aspergillus candidus, Aspergillus fumigatus, Aspergillus niger, and Aspergillus foetidus, 2 strains of Cladospoyium spp. and 2 strains Botryotrichum spp., etc. total 159 kinds of filamentous molds were isolated and identified. There were many differences in numbers and distributions of filamentous molds from each Nuruk according to their collected region. Absidia spp. were most frequently isolated from every Nuruk sample. Aspergillus oryzae, Aspergillus candidus, and Rhizopus cohnii showed even distribution. Penicillium sp. and Mucor sp. were not detected. Actinomucor sp., Aspergillus foetidus, Botryotrichum sp., and Cladosporium spp., which have not been reported by far, were found. Amylase activities, pH stability of amylase, and acid productivity of isolated strains were compared.

  • Dextrinogenic and glucoamylase activities of Aspergillus isolated from various habitat-substrates collected through South Korea are measured, and their amylase activities are surveyed in taxonomical and ecological viewpoints. 1. A. flavous group and A.wentii group exhibited higher activities for both amylases than others. 2. In the relations between amylase activity of Asperguillus and their habitat-sub-strates, the strains isolated from meju and cereals exhibited predominant dextrinogenic amylase activity. 3. Dextrinogenic amylase activity of Aspergillus is higher in the strains isolated from southern coast than the other regions. 4. Among the 601 strains of Aspergillus surveryed, strain No.74 and strain No.421 exhibited the most predominant activity for dextrinogenic amylase and glucoamylase, respectively.

Occurrence of Fungal Species and Mycotoxins from Decayed Sugarcane (Saccharrum officinarum) in Egypt

  • Seventy-three fungal species belonging to forty-three genera were isolated from 40 samples of Saccharrum officinarum (collected from Naage-Hamadi canal in Qena Governorate, Egypt). Aspergillus, Trichoderma, Mucor and Pythium were the most common genera on the two isolation media. The dominant species of Aspergillus were A. niger, A. flavus, A. ustus, A. terreus and A. wentii. Some species were dominant on 40 g/l sucrose such as Aspergillus niger, A. flavus, Emericella nidulans, Trichoderma viride, Torula herbarum and Mamaria echinoeotryoides, while the dominant species on 10 g/l glucose were Mucor circinelloides, Aspergillus niger, Torula herbarum and Trichoderma viride. Mycotoxins including aflatoxins $B_1,\;B_2,\;G_1\;and\;G_2$, zearalenone and diacetoxyscirpenol were detected in the examined samples of Saccharrum officinarum. The mycelial growth of A. flavus, A. niger, Fusarium moniliforme and Torula herbarum decreased with the increase in Dimethoate concentrations, although 25 ppm was less effective than the higher levels of the insecticide ($75{\sim}200\;ppm$). Dimethoate stimulated the activity of Go-Tin A. niger, F. moniliforme and T. harbarum, while the Go-T activity was inhibited in A. flavus with the Dimethoate treatments.

  • From Nuruks, a traditional Korean starter for rice wine, collected from 42 different areas in Korea, 111 fungal strains were isolated. These isolates were identified as 25 species belonging to seven genera of Rhizopus oryzae(14 strains), R. oligosporus(8 strains), R. nigricans(5 strains), R. arrhizus (5 strains), Aspergillus oryzae(12 strains),Asp. parasiticus(8 strains), Asp. fumigatus(3 strains), Asp. ochraceus(7 strains), Asp. wentii(5 strains), Asp. parasiticus(8 strains), Asp. penicilloides(3 strains), Asp. clavatus(4 strains), Penicillium purpurogenum(2 strains), Pen. glabrum(1 strain), Pen. granulaturm (1 strsin), Pen. fellutanum(1 strain), Geotrichum candidum(2 strains), Absidia corymbifera(12 strains), Mucor racemosus(2 strains), M. plumbeus(2 strains) and Curvularia lunatus(3 strains).

  • Nowadays, the damages caused by molds in cotton textile goods becomes influential. In our country, however, the relations between cotton goods and molds are not investigated and studied in detail. Two hundred and fifty seven kind of mold's samples were collected in ninety places through the whole country. The molds samples are mainly gathered according to each regions and seasons from molded cotton textiles. Out of this samples, we isolated six hundred and seventy two strains of molds and the results of isolation are following. 1. The distributed molds were Aspergillus sp., Rhizopus sp., Penicillium sp., etc. among them Aspergillus sp. were most widely distributed, and next were Rhizopus sp., Penicillium sp. etc. 2. The distribution of Aspergillus sp. abounded peculiarly in the dry season, while Rhizopus sp. in the rainy season. 3. The C.M.C, descomposing enzymes forming activity on molds were greatly concerned with intensity damage of cotton textile goods. 4. The formation of C.M.C. decomposing enzyme was only influenced by physiology of each strains. 5. Regarding to the growth. a. The molds which were saprophyting on the cotton textile goods were indicated vigorous growing. b. Among isolated six hundred and seventy two strains, there were above a hundred strains which produced pigment and nearly half of them fifty nine strains were Aspergillus sp. 6. Twenty one strains in isolated six hundred and seventy two strains were indentified which can heavily damage upon cotton textile. As a results of indentification of the selected strains, the following species was abtained, Aspergillus sydowi, wentii, niger, luchuensis, flavus, fumigatus, nidulans, Penicillium frequentants, roqueforti, chrysogenum, albicans, Rhizopus oligosporus, delemar, Mucor rouxii, mucedo, Neurospora sitophila, Monilia variabilis, fructigena, Cladsporium hurbarum and Aspergillus spp. Mucor spp.

  • The Investigation of organisms damaged to papers and cellulose materials of Cultural Property in the Ch'ang Dok Palace The investigation of the airborne fungi, the attached fungi to the papers and cellulose materials of Cultural Properties in addition to the insects inhabiting at the Ku Sonwon Jon, Shin Sonwon Jon and Yonwa ch'anggo in the Ch'ang Dok Palace carried out from Jul. 10 to Jul. 21,1984.The results are summarized as follows ;1. Isolation and identification of the airborne fungi from the three storages were Cladosporium sp., Alternaria alternata, As pergillus cervinus, A. flavus, A. nidulance, A. oryzae, A. terreus, A. versicolor, A. wentii, Penicillium adametzii, P.albicans, P.canescens, P. citreo-viride, P. citrinums, P. decumbens, P. frequentans, P. funiculosum, P.herquei, P.implicatum, P.multicolor, P.nigricans, P.nonatum,P.purpurogenum, P.roqueforti, P.viridicatum, Trichodema viride, Geotrichumcandidum, Curvvlaria lunata, Torula hebarum, T.thermophila, Itersoniliasalmonicolor, Drechsclera avenue, Candida sp., Acremonium sp., and Botrytis sp., It was found that thirty five species in thirteen genera was isolated. Among them, the dominant species was Cladosporium sp., and the order was Penicillium, Aspergillus, Alternaria and so on.2. The attached fungi directly isolated from the papers and cellulose materials of Cultural Properties were twenty-nine species in fourteen genera, namely, Acremonium sp., Albertiniella sp., Alternaria alternata, Aspergillus clavatus,A.niger, A.ornatus, A.versicolor, Botrytis sp., Bysochlamys sp., Carpenteles sp.,Chaetomium globosum, Cladosporium sp., Eurotium sp., Mucor sp., Penicilliumcanescens, P.chermesium, P.citrinum, P.frequentans, P.funiculosum, P.herqueiP .implicatum, P.javanicum, P.luteum, P.purpurogenum, P.thomii, P.viridicatum, Torula thermophila, Trichoderma koningi and T.viride. Among them, the mostfungi distributed on the surface of the papers and cellulose materials was Penicillium and the order was Aspergillus, Alternaria, Cladosporium, Trichodermaand so on.3. The insects collected the three storages were ten genera and ten species including 916 specimens. By classifying the insects collected, the most species of the insects was Stenoscelodes hayashii of 857 specimens occupied about 93% of the total insect. And the other insects were collected as Microgamme costipennisAnobium pertinax, Xenomimetes alni, Anthrenus verbasci, Holoparmecus signatus,Thermobia domestica, Halyomorpha brevis, Drosophila coracine and Brattaorientalis. As described above, it could be known that the most airborne fungi was Cladosporium and the order was Penicillium, Aspergillus, Alternaria in the three storages. And the most attached fungi distributed on the surface of papers and cellulose materials was Penicillium and the other fungi were Aspergillus, Alternaria Cladosporium, Trichoderma and so on. Accordingly, from the results, itwas assumed that the major part of airborne fungi were attached to the papers and cellulose materials of Cultural Properties. The paper and cellulose materials of Cultural Properties in Ch'ang Dok Palace were chiefly damaged by S.hayashii in Coleoptera.

The Study on the Detection of Aflatoxins in the Fermentation Products and Cereals (TLC법에 의한 장류 및 공류중의 Aflatoxin검출에 관한 연구)

  • Aflatoxin, a mixture of the at least four toxic and carcinogenic metabolites, is known to be produced by only a few fungi. The toxins were designated aflatoxins because they were produced by the mold Aspergillus flavus(A. flavus). However, at least four other toxins and other species of the genus A. niger, A. parasiticus A. ruber and wentii have been reported to produce aflatoxins. And also the identical compounds may also be produced by molds, the Pencillium. At least four different species of Penicilliurn have been reported to produce aflatoxins (P. citrinurn, P. frequentans, P. puberulurn. and P. variable). So it is now known that the problem of Aflatoxin is not restricted to the single species A. flavus, even though that is a very common mold. Also additional aflatoxins have been discorvered. For sereral years, only four aflatoxins were known: $B_1, B_2, G_1$ and $G_2$, so designated by reason of their fluorescence and chromatographic charateristics. It is now known that there are really two new toxic materials in the milk. During the past year(1966) they were christened aflatoxin $M_1$ and $M_2$, since they were first found in milk. The two other and most recently discorvered aflatoxins were isolated late in 1966 from cultures of A. flavus, and were designated aflatoxin $B_2a$ and aflatoxin $G_2a$. In order to obtain a breaf information about extent of contamination of foodstuffs by aflatoxin which is known to produce eight different mold, aflatoxin detection of cereals and fermented foods on sale, such as polished rice, barley, wheat, wheat flour, lentil, red bean, soy bean, noodle, kochuj ang and Dwenjang (fermented soy bean paste) and chong Kuk, were carried out. The results of this investigation were summarized as follows: The hexane:$CHCl_3$ extracts of polished rice, barley wheat, wheat flour, lentil, red bean, noodle and kochujang yielded fluorescent spots on thin layer plates. However their Rfvalues were different from those of authentic aflatoxins. The fluorescent substances of the extract from soy bean, Dwenjang and chong kuk showed very similar Rf values to those of the standard aflatoxins. By two dimensional thin layer chromatography and comparison of ultra violet absorption spectra, it was found that these fluorescent substances were not aflatoxins. To conclude, aflatoxins themselves were not detected directly in those samples tested.

  • Mutant strans of Salmonella typhimurium which require histidine for their growth sensitively, were easily revertant and lost the histidine requirement, when the strains contacted with some new mutagen. This work was carried out to determine the mutagenicity of kojic acid and emodin for the mutant strains of Salmonella typhimurium TA 98, TA 100, TA 1535, TA 1537, and TA 1538. Through the metabolic activation with liver microsome enzyme system of rat (S-9), kojic acid was recognized as a strong mutagen for the strain of TA 98, while it responsed weakly for the strain of TA 100. Without S-9 metabolic activation, kojic acid could not induce the mutation for the both strains of TA 98 and TA 100. Emodin was also recogniged as a strong mutagen for the strain of TA 1537 through the metabolic activation with S-9 mix.
