Improvement in the Quality of Takju by the Combined Use of Aspergillus kawachii and Aspergillus oryzae (Aspergillus kawachii와 Aspergillus oryzae의 병용에 의한 탁주의 품질개선)

  • In order to improve the quality of Takju, the effect of combined use of Aspergillus oryzae and Aspergillus kawachii in brewing was investigated. The quality of Takju which was brewed by the combined use of Aspergillus oryzae CF7-koji and Aspergillus kawachii CF1-koji in equal amount was better as cmapared with that brewed by existing method using Aspergillus kawachii-koji only as koji. But the good result did not obtained when the koji was made by mixed culture of Aspergillus oryzae CF7 and Aspergillus kawachii CF1.

  • The objective of this experiment was to improve the quality of Kochujang and to shorten the aging time of it by using the Kojis of various Aspergillus strains. During the autumn season Kochujangs were mashed with Koji of Aspergillus oryzae, Aspergillus kawachii and Aspergillus shirousamii. Chemical composition of the Kochujang was determined during the aging period. There was no significant difference in quality and component between the Kochujang made with Aspergillus oryzae cultured Koji and Aspergillus kawachii cultured Koji. But the Kochujang with Aspergillus shirousamii cultured Koji was better in flavor, color and amino nitrogen contents than the Kochujang with Aspergillus oryzae cultured Koji, though two Kochuangs were almost the same in general components. The significant difference between the two tested Kochuangs was observed in pH, and the pH of the Kochujong with Aspergillus shirousamii cultured Koji was extremely low during the aging period. Thus it is presumed that Aspergillus shirousamii Koji can be used for the improvement of quality of Kochujang if a suitable method is applied to prevent pH from lowering.

  • To substitute the soy-sauce 'koji' used in fish sauce processing with molded sardine meal(MSM) 'koji', the culture conditions of MSM be inoculated with Aspergillus spp. were investigated. To prepare the MSM, Asp. awamori(KFCC. 11439), Asp. guercinus (KFCC. l1595), Asp. niger(KFCC. 11239), Asp. oryzae(KFCC. 32343), and Asp. sojae(KFCC. 11559) were inoculated upon the chopped sardine with $20\%$ (w/w) corn starch after steriling it at $121^{\circ}C$ for 15min. Sporulation time cultured with Asp. spp. at $30^{\circ}C$ was 48hrs and color of mycelium on the surface of MSM inoculated with Asp. awamari and Asp. oryzae were black, that of MSM inoculated with the other strains were yellowish brown. The activity of protease and lipase from the MSM were increased till the 72hrs of culture at $30^{\circ}C$, while the content of trimethylamine was decreased after 96hrs of culture period at same condition with exception of Asp. niger. Asp. oryzae and Asp. sojae showed superior in pro-tease and lipase activity in comparison with the other strains, and maximum activity of protease and lipase of MSM was observed after 72hrs of culture period. The optimum pH and temperature for the activity of MSM inoculated with Asp. oryzae and Asp. sojae were pH 9.0, $30\~35^{\circ}C$ and pH $6\~7$, $35^{\circ}C$, respectively.

  • The study was conducted to investigate the improvement nail health in middle-aged women aged 40~60 years who intake soybean koji by incubating Aspergillus oryzae(황국균), and utilized non-fermented soybeans as a control group and soybean koji as an experiment group for eight weeks. After clinical trials, the lifting and cracking conditions of the nails in the experiment group were improved, The moisture content increased 5.1 times compared to the control group, The water loss was 1.4 times less the reduction to the control group. and nail thickness increased by 1.9 times compared to controls. Wrinkles on the surface improved by about 2.4 times compared to the control group. In conclusion, consumption of this soybean koji of A. oryzae's has been shown to have an impact on improving nail is expected to be used Therefore, I propose this soybean koji of A. oryzae's as an inner beauty for improving nail is in middle-aged women aged 40~60 years.

Evaluating the Headspace Volatolome, Primary Metabolites, and Aroma Characteristics of Koji Fermented with Bacillus amyloliquefaciens and Aspergillus oryzae

  • Production of good Koji primarily depends upon the selection of substrate materials and fermentative microflora, which together influence the characteristic flavor and aroma. Herein, we performed comparative metabolomic analyses of volatile organic compounds (VOCs) and primary metabolites for Koji samples fermented individually with Bacillus amyloliquefaciens and Aspergillus oryzae. The VOCs and primary metabolites were analyzed using headspace solid phase microextraction (HS-SPME) followed by gas chromatography time-of-flight mass spectrometry (GC-TOF-MS). In particular, alcohols, ketones, and furans were mainly detected in Bacillus-fermented Koji (Bacillus Koji, BK), potentially due to the increased levels of lipid oxidation. A cheesy and rancid flavor was characteristic of Bacillus Koji, which is attributable to high content of typical 'off-flavor' compounds. Furthermore, the umami taste engendered by 2-methoxyphenol, (E,E)-2,4-decadienal, and glutamic acid was primarily detected in Bacillus Koji. Alternatively, malty flavor compounds (2-methylpropanal, 2-methylbutanal, 3-methylbutanal) and sweet flavor compounds (monosaccharides and maltol) were relatively abundant in Aspergillus-fermented Koji (Aspergillus Koji, AK). Hence, we argue that the VOC profile of Koji is largely determined by the rational choice of inocula, which modifies the primary metabolomes in Koji substrates, potentially shaping its volatolome as well as the aroma characteristics.

  • This study was carried out to investigate the influences of cultural conditions of koji on the production of saccharogenic amylase during rice-koji making by Aspergillus awamori var. kawachii which is now widely used as koji-mold in brewing Tikju and Yakju in Korea. The optimum cultural temperature for the production of saccharogenic amylase by this mold was 36$^{\circ}C$, and at this temperature it needed 40 hours of cultivation for maximum production of this enzyme. It was favorable for high production of both organic acid and saccharogenic amylase to shift the cultural temperature form initial 36$^{\circ}C$ to 32$^{\circ}C$ after 20~25 hours of cultivation. The production of saccharogenic amylase was low when the water content of steamed rice was below 35%, but its production was high at 40~60% of water content. When the quantity of conidial inoculation was too small, the production of saccharogenic amylase was low in initial phase, but it was retrived after 40 hours of cultivation. When koji-thickness was over 3cm, the production of saccharogenic amylase was markedly restricted. The saccharogenic amylase of this koji was stable at pH 2~7, and showed high activity at pH 2~5.

Comparison of Characteristics of Koji Manufactured with Bacillus subtilis B-4 and Aspergillus oryzae F-5 (Bacillus subtilis B-4와 Aspergillus oryzae F-5로 제조한 코오지의 특성 비교)

  • In order to industrialize traditional Meju-tasting Koji, the characteristics of Koji manufactured with bacteria and fungi found in traditional Meju were investigated. Bacillus subtilis B-4 and Aspergillus oryzae F-5 showing high enzyme activities including those of amylase and protease were used. L-value of Koji manufactured with B. subtilis B-4 had darker color and higher enzyme production than A. oryzae F-5 made one. B. subtilis B-4 made Koji showed higher enzyme production and sensony evaluation score than A. oryzae F-5 Koji. A. oryzae F-5 Koji showed superior color to B. subtilis B-4 Koji. Activity in color, capacity of enzyme production, viable cell count, and sensory evaluation of water activity controlled Koji was superior to the uncontrolled one.

Physiological Function and Enzyme Activity of Koji Cultured by Aspergillus oryzae CNU O4-5 in the Various Grain Materials (원료(原料)의 종류(種類)와 형태(形態)에 따른 Aspergillus oryzae 국(麴)의 생리활성(生理活性) 및 효소력(酵素力))

  • CNU O4-5 isolated from conventional Meju, which is used as raw material for making a soybean fermentation food, identified as an Aspergillus oryzae. To make koji, Aspergillus oryzae CNU O4-5 was cultured for 3-4 days at $30^{\circ}C$ with various grain materials such as flour, soybean powder, flour+soybean powder(1:1), soaked soybean and rice. The koji was evaluated for analyze the angiotesin converting enzyne(ACE) inhibition, antioxidative activity, superoxide dismutase(SOD) activity, amylase and protease activity. $\alpha$-amylase and glucoamylase activities of flour koji were higher than those of the koji soybean powder. However neutral and alkaline protease activities of flour koji were lower than those of flour+soybean powder and soybean koji. Amylase and protease activities of kojies of soaked soybean and rice showed very low level. The range of the ACE inhibition rate by hot water fraction of the kojies, which are cultured with various gain materials, were from 45% to 54%. The anti oxidative activity of ethanol-fraction of koji, which is made by using the soybean powder or soaked soybean, prolonged for 6 days in lard at $60^{\circ}C$. The SOD activity of grinded fraction of koji, which is made by using the flour or soybean powder, was same as 2,000 units per g of each koji.

  • In this study, investigated the effects of four strains belonging to Aspergillus oryzae on the quality of Kochujang. In the Koji and Kochujang making, investigated the difference of enzyme production of each strain, the change of each component and color during the aging, and tested the sensory taste. The results obtained were as follows; 1. Protease activity (acid, neutral) in wheat flour Koji was high in the case of Aspergillus $oryzae-S_1$ and Aspergillus oryzae - M of short stalked type. The strain Aspergillus $oryzae-S_1$ showed maximum activity after two days of Koji making, while the strain Aspergillus oryzae - M showed low activity till two days, but showed maximum activity after three days-four days of Koji making. 2. In $\alpha$-amylase activity, strain Aspergillus $oryzae-S_1$, Aspergillus $oryzae-S_2$ and Aspergillus oryzae - M showed high activity after two days of Koji making. Aspergillus oryzae-NB strain showed slower ${\alpha}$ - amylase activity than that strains. 3. In glucoamylase activity, all strain tested showed high activity after three days of Koji making, but st rain Aspergillus oryzae - NB showed slower activity than ot - hers. 4. In protease activity (acid, neutral) during the aging of Kochujang, strain Aspergillus $oryzae-S_1$ and Aspergillus oryzae - M of short stalked type showed higher activity than that of long stalked type. 5. Amino type nitrogen contents during the aging of Kochujang was very higher in the case of strains Aspergillus $oryzae-S_1$ and Aspergillus oryzae - M of short stalked type than other strains, and each contents was 315mg% and 337mg% after aged for ninty days. 6. The results that analysed free sugar of Kochujang aged for ninty days with HPLC were; glucose 5.84~7.13%, fructose 4.13~5.00%, rhamnose 0.91~1.04%, maltose 0.72~0.92% and presence of xylose was recognized. 7. The results that analysed alcohols of Kochujang aged for ninty days with gas chromatography were; ethanol 1.51~1.78%, n-propyl alcohol 1.13~1.20mg%, iso-amyl alcohol 3.5~4.4mg%. 8. In the sensory test of Kochujang aged for sixty days and for ninty days, the case of strains Aspergillus oryzae-M and Aspergillus $oryzae-S_1$ of short stalked type showed good taste, while the case of strains Aspergillus $oryzae-S_2$ and Aspergillus oryzae-NB of long stalked type showed good flavor and color.

The Chemical Properties of Doenjang Prepared by Monascus Koji (홍국첨가가 된장의 이화학적 특성에 미치는 영향)

  • Monascus koji have been used as a natural food colorant and preservative in Asia countries for centuries. This study was investigated to the chemical properties of Doenjang prepared with Monascus koji. Four kinds of Doenjang were fermented with Aspergillus koji only and Aspergillus koji plus 10%, 20% and 30% Monascus koji of its amount, respectively, and the changes of characteristics were investigated during fermentation. All the Monascus koji substituted Doenjang showed lower protease activities than Aspergillus koji Doenjang over all periods of fermentation. No differences of amino nitrogen and free amino acids were found after 30 days of fermentation and reducing sugars and peptides were found during over all periods of fermentation in each other. The Monascus koji gave higher a-, L- and C-values and lower h-values than Aspergillus koji. Sensory profiles of color, flavor, taste and overall quality on the final products which fermented during 60 days, indicated that there were no differences each other. From these results Monascus koji suggested as 30% substitutes for Aspergillus koji in Doenjang preparation.
