The effects of 3 different strains of Japanese koji (Aspergillus oryzae, A. sojae and A. awamori) in the solid-state fermentation (SSF) of sugarcane bagasse mixed with wheat bran on chemical composition, energy, in vivo digestibility and preference of the fermented bagasse feeds (FBF) in goats were investigated. Diets consisted of lucerne hay cube (basal diet) and unfermented bagasse feed (control), FBF with A. oryzae (O), FBF with A. sojae (S) or FBF with A. awamori (A), which were mixed in a total ration of 7:3 (w/w DM). Three Nubian does were fed each of the diets, i.e. control, O, S and A in the 4 consecutive periods for digestion trials (21-day each). The goats were also used for preference trials (30-min each) of O, S and A. The O was significantly higher in CP content than others (p<0.05). The crude fiber (CF), ADF and cellulose contents of control were significantly lower than those of other diets (p<0.05). The S had significantly higher CF digestibility than control (p<0.05), and it revealed the largest value of all. Digestibilities of NDF, ADF and cellulose in S were significantly higher than those of control (about 10, 18 and 18%, respectively, p<0.05). The DE of S was significantly higher than that of others (p<0.05), though there were no significant differences in DCP and TDN between control and S. The results of preference trials demonstrated that the average intake rate was not significantly different among diets, but O and S are likely to be preferable to A (p<0.1). It was concluded that the SSF of bagasse feeds by Japanese koji can improve the fiber digestion, especially NDF, ADF or cellulose in goats, and there is a marked effect in the feed containing A. sojae, which may lead to the improvement of DE.
엽연초인공숙성(葉煙草人工熟成)에 관여(關與)하는 미생물(微生物)들을 분리(分離)하고 분리균주(分離菌株)에 대(對)하여 배양(培養), 형태(形態), 생리(生理) 및 회수화물(灰水化物) 발효시험(醱酵試驗)을 행(行)하고 이 결과(結果)들로부터 이들 미생물군(微生物群)을 각각(各各) Asperfillus gavus, A. restrictus, A. nidulans, A. awamori, Oidium sp. Edmundmasonia Sp. spicaria Sp. Bacillus subtilis, B. subtilis var aterrimus, B. licheniformis Flaavobacterium harrisonii, Aerobacter aerogenes으로 동정(同定)하였다. 그리고 이들 균주중(菌株中)에서 발효전기간(醱酵全期間)에 걸쳐, 관여(關與)한 것으는 Aspergillus flavus, A. restriclus.,와 Bacillus subtilis, B. licheniformis이었고 이 곰팡과, 세균(細菌)은 각각(各各) 14, 7 일간(日間)에 최고분포(最高分布)를 이루었으며 기후(其後)에는 뚜렷한 분포(分布)의 변화(變化)를 보이지 않았다.
생전분으로부터 에탄올을 효율적으로 생산하는 전분 분해성 산업용 Saccharomyces cerevisiae를 제조하기 위해 생전분을 발효하는 모균주(ATCC 9763/$YIp{\delta}AGSA{\delta}$)의 염색체내 ribosomal DNA loci에 double 18S rDNA-integration 시스템을 이용하여 Bacillus amyloliquefaciens ${\alpha}$-amylase 유전자(Amy) 혹은 Aspergillus awamori glucoamylase 유전자(GA1)를 다중도입시켰다. 얻어진 형질전환 균주들 중 생전분으로부터 가장 효율적으로 에탄올을 생산하는 균주(ATCC 9763/$YIp{\delta}AGSA{\delta}$/YIpAG2rD)의 발효 3일째 에탄올 생산은 모균주에 비해 1.5배 높았다. 이 새로운 균주는 생옥수수 전분이 20% (w/v) 함유된 배지에서 3일간 발효를 통해 에탄올 9.2% (v/v) (72 g/L)를 생산하였고, 생전분 함유량의 75%를 소비하였다.
우리 나라 전통술인 탁주의 주질 개선을 위한 목적으로 천연성과 기능성을 가진 벌꿀을 사용하였고 저장기간의 연장을 위하여 술액의 침전물 양을 조절할 목적으로 본 연구를 진행하였다. 발효방식은 주모발효 후 3단담금 방식으로 하였고, 입국균은 Aspergillus awamori var. kawachi, 원료는 100% 백미, 알코올 발효 효모는 Saccharomyces cerevisiae를 사용하였다. 제성액의 고형분과 술액을 분리하여 벌꿀을 최종농도 5%(w/v) 첨가하여 5, 10, $15^{\circ}C$에서 23일간 숙성하였을 때 온도에 따른 유의한 차이는 보이지 않았으며 숙성기간 동안 알코올이 미소하게 감소했을 뿐 pH, 산도 및 총당량은 거의 일정하였다. 이와 같이 특징적인 술액의 차이는 없었지만 벌꿀숙성으로 제조한 탁주가 벌꿀 첨가 직후의 미숙성 탁주와 비교할 때 우수한 관능 테스트 결과를 보여주었다. 추가적으로 냉동 보관한 고형분을 100%, 50% 및 25% 비율로 벌꿀첨가 숙성된 술액과 혼합한 후 물을 보충하여 알코올 8%의 탁주를 제조하고 37일간 $10^{\circ}C$에서 날짜 별로 분석하였다. pH는 모든 탁주에서 4.5~4.8 사이를 큰 차이 없이 유지하였으며 고형분 첨가량이 많을수록 효모 세포수, 산도 및 알코올 함량은 상대적으로 증가하였고 총당량은 감소함을 보여 주었다. 즉 고형분의 양이 소량일수록 탁주의 주질이 안정하였다. 결론적으로 발효 후 고액 분리하여 상층액에 벌꿀을 첨가함으로써 저온 숙성기간 동안 주질의 큰 변화를 주지 않고 관능적으로 우수한 술액을 만들 수 있었으며, 고형분의 재혼합 비율을 조절할 경우 저장기간이 증대된 탁주를 제조할 수 있을 것이다.

A novel $\alpha$-amylase ($\alpha$-1,4-$\alpha$-D-glucan glucanohydrolase, E.C. 3.2.1.1), ScAmy43, was found in the culture medium of the phytopathogenic fungus Sclerotinia sclerotiorum grown on oats flour. Purified to homogeneity, ScAmy43 appeared as a 43 kDa monomeric enzyme, as estimated by SDS-PAGE and Superdex 75 gel filtration. The MALDI peptide mass fingerprint of ScAmy43 tryptic digest as well as internal sequence analyses indicate that the enzyme has an original primary structure when compared with other fungal a-amylases. However, the sequence of the 12 N-terminal residues is homologous with those of Aspergillus awamori and Aspergillus kawachii amylases, suggesting that the new enzyme belongs to the same GH13 glycosyl hydrolase family. Assayed with soluble starch as substrate, this enzyme displayed optimal activity at pH 4 and $55^{\circ}C$ with an apparent $K_m$ value of 1.66 mg/ml and $V_{max}$ of 0.1${\mu}mol$glucose $min^{-1}$$ml^{-1}$. ScAmy43 activity was strongly inhibited by $Cu^{2+}$, $Mn^{2+}$, and $Ba^{2+}$, moderately by $Fe^{2+}$, and was only weakly affected by $Ca^{2+}$ addition. However, since EDTA and EGTA did not inhibit ScAmy43 activity, this enzyme is probably not a metalloprotein. DTT and $\beta$-mercaptoethanol strongly increased the enzyme activity. Starting with soluble starch as substrate, the end products were mainly maltotriose, suggesting for this enzyme an endo action.
A comparison of dextrinogenic amylase activities in the Asp. niger group was made with their crude and ethanol dialized enzymes before and after heating at high temperature (60-$65^{\circ}C$). The results obtained are as follows ; 1. THe dextrinogenic amylase activity of crude enzymes of Asp. kawachii and Asp. foetidus was strong, but Asp. phoernicis, Asp. carbonarius and Asp.japonicus showed weak activity. The others showed medial grades of activity. 2. The ethanol dialized enzymes of Asp. kawachii, Asp. foetidus and Asp. japonicus was very sesitive to high temperature (60 or $65^{\circ}C$) and their enzymatic activities were diminished greatly. The others did not show diminution of enzymatic activity at 60 or 65.deg.C, but diminished greatly at 70 or $75^{\circ}C$. 3. The ethanol dialized enzymes of the Asp.niger group heated to 65.deg.C was more sensitive at pH 6.0 and 6.5 than at pH 4.5, 5.0 and 5.5. 4. Tested strains in the Asp.niger group were subdivided into 4 subgroups by their dextrinogenic amylase activities before and after heating at 60 or $65^{\circ}C$. The first group showed a medial grade of activity before heating and no diminution of their enzymatic activities after heating. Asp. niger, Asp.pulverulentus, Asp. awamori and Asp. usmii were included in this group. The second group had strong enzymatic activity before heating, but diminished greatly after heating. Asp rawachii and Asp. phoenicis were included in this group. The fourth group showed very weak enzymatic activity before heating, and was inactivated easily by heating. Asp.oryzae of the Asp. flavus group showed a very strong dextrinogenic amylase activity before heating. After the heat treatment, however, its enzymatic activity was diminished greatly.

A field experiment to evaluate the effect of application of different organic manures and inorganic fertilizers on growth, yield and quality of leaf was studied during 2004-05 has showed that, the application of 10 kg each of Azospirillum brasilense and Aspergillus awamori+20% each of recommended N through compost+green manure (Glyricidia maculata)+castor cake+vermicompost+Urea and remaining P and K through fertilizers ($T_{11}$) has recorded significantly higher leaf yield (250 g/ plant and 34.70 tonnes/ha/yr, respectively) with improvement in growth characters as compared to control. Leaf quality status was also improved in terms of N (3.19%), P (1.97%), K (1.28%), total soluble protein (8.39 mg/ml), total soluble sugars (14.40 mg/ml), secondary nutrients viz., Ca (3.00%), Mg (0.60%), S (0.35%) and micronutrients viz., Cu (0.410 ppm), Mn (0.454 ppm) and Zn (0.112 ppm) contents. The mulberry grown with 20 tonnes of compost+300:120:120 Kg of NPK/ha/year through fertilizer ranked second for growth and foliar constituents.
To develop a new traditional Gugija tagju, various koji were prepared using several starch sources along with Aspergillus awamori var. kawachi, and then their effects on the quality of the tagju were investigated. Ethanol contents(11.1${\sim}$13.8%) were not significantly different among the Gugija tagju made from the various types of koji, whereas the tagju made using non-glutinous rice koji had the best total acceptability. The addition of 1% Gugija extracts into the mash increased its total acceptability and anti-hypertensive angiotensin I-converting inhibitory activity, and the optimal fermentation period for brewing the Gugija tagju was 10 days.
식혜에 함유된 이소말토올리고당과 밥알에 30unit/ml의 $\alpha$-아밀라아제, 글루코아밀라아제, $\alpha$-글루코시다아제, $\beta$-아밀라아제를 가하여 작용시킨 결과 글루코아밀라아제 외에는 일부밖에 가수분해하지 못하였다. 그리고, 인체의 효소인 $\alpha$-아밀라아제와 $\alpha$-글루코시다아제를 함께 작용시켜도 25% 이상 가수분해하지 못하므로 비피두스균의 활성인자로 작용할 것으로 보인다. 밥알은 물에 녹지 않기 때문에 식이섬유로도 작용할 것으로 보인다.
DFA (Difructose anhydride)는 특유의 구조적인 안정성 때문에 당뇨병 환자를 위한 당원으로써 적합하다는 연구가 보고 되어 있다. DFA에는 4가지 type이 있는데 inulin에 의한 DFA I DFA III DFAV가 있고 levan에 의한 DFA IV가 있는 것으로 알려져 있다. 특히 DFA IV는 당뇨병 환자를 위한 당원 뿐 만 아니라 rat을 이용한 연구에서 칼슘의 흡수를 도와 준다는 보고가 있었다. 이러한 DFAIV를 생성하는 데 쓰이는 Microbacterium sp. AL-210에서 유래한 LFTase (Levan fructotransferase)의 wild-type과 mutants (D63A, D195N, N85S)의 구조적 특성을 밝히기 위해 정제하였다. LFTase의 wild-type과 mutants들을 대량 발현시킨 후 흡착 크로마토그래피, 이온교환 크로마토그래피 그리고 젤 여과 크로마토그래피를 이용하여 고순도로 분리 정제하였으며 이를 SDS-PAGE를 통하여 확인하였다. 분리 정제된 단백질을 JNET 이차 구조 예측 프로그램, solubility 측정, CD (원 편광 이색성 분광편광계), fluorescence spectroscopy (형광분석법), DSC (시차주사열량계)를 이용하여 분석하였다. 또한 다중 정렬과 2차 구조 예측 프로그램을 이용하여 wild-type의 2차 구조를 분석하였다. Solubility 측정에서 가장 적합한 온도는 $55^{\circ}C$, 최상의 pH는 7.5로 나타났다. CD 분석에서 wild-type과 비교한 결과 다른 mutant에 비해 N85S의 $\alpha$-helix가 많이 감소한 것과 $\beta$ strand와 random coil이 증가한 것을 확인하였다. 또한 DSC 분석을 통해 wild-type이 다른 mutants에 비해 안정적인 구조를 지닌 것을 확인하였다. 형광분석에서 N85S가 wild-type과 가장 유사하게 나타났으며 D63A와 D195N은 wild-type에 비해 높은 강도를 나타내었다. 또한 wild-type의 sequence를 Exo-inulinase from Aspegillus awamori, a plant fructan 1-exohydrolase from Cichorium intybus 그리고 invertase from Thermotogo maritime (Tm)의 sequence와 다중 정렬한 결과 Exo-inulinase와 높은 identity를 보였다.
