Background There are currently no guidelines for the postoperative wound management of the hard-palate donor site in cases involving mucosal harvesting. This study describes our experiences with the use of an artificial dermis for early epithelialization and transparent plate fixation in cases involving hard-palate mucosal harvesting. Methods A transparent palatal plate was custom-fabricated using a thermoplastic resin board. After mucosal harvesting, an alginic acid-containing wound dressing (Sorbsan) was applied to the donor site, which was then covered with the plate. After confirming hemostasis, the dressing was changed to artificial dermis a few days later, and the plate was fixed to the artificial dermis. The size of the mucosal defect ranged from 8×25 to 20×40 mm. Results Plate fixation was adequate, with no postoperative slippage or infection of the artificial dermis. There was no pain at the harvest site, but a slight sense of incongruity during eating was reported. Although the fabrication and application of the palatal plate required extra steps before and after harvesting, the combination of the artificial dermis and palatal plate was found to be very useful for protecting the mucosal harvest site, and resulted in decreased pain and earlier epithelialization. Conclusions The combination of artificial dermis and a transparent palatal plate for wound management at the hard-palate mucosal donor site resolved some of the limitations of conventional methods.
This study was carried out to investigate the histological changes after the infection of nuclear and cytoplasmic polyhedrosis viruses(NPV, CPV) and the resistance to the viruses in various varieties of the silkworm fed on artificial diet. The results obtained were as follows; Among four varieties of silkworm tested, Jam 107$\times$Jam 108 was more resistant than the other varieties tested and Jam 119$\times$Jam 120 was the most resistant to CPV. In case of peroral infection with NPV, Jam 107$\times$Jam 108 showed lower mortality than the remained varieties in low concentration (104/ml). However, all varieties showed high mortality as the concentration of viruses was increased. With infection of CPV, the varieties showed high mortality at the concentration of 107 and 108/ml, while Jam 119$\times$Jam 120 showed the lowest mortality at virus concentration of 104/ml. The fat bodies, epidermal cells and tracheal epithelial cells showed high susceptibility to NPV to break the cells completely and liberate the debris to the body cavity. The CPV infected only the cylindrical cells of mid-gut and formed polyhedrons. In some cells, CPV was liberated to gastral cavity. In the electrophoretic pattern of hemolymph protein of silkworm larvae infected with NPV, bands were dimmed and disappeared as symptom aggravated after infection. Electrophoretic pattern of homolymph proteins of silkworm larvae infected with CPV showed no numerical difference at the later stage of infection, and one or two bands was observed along with lowering the concentrations.
This study was carried out to investigate the effects of vitamin E on the prevention and treatment of Babesia gibsoni. Fifteen mongrel dogs, uninfected with Babesia spp, were assigned to three groups according to vitamin E(${\alpha}-tocopherol$) concentrations in the RBC. The concentrations in each of the three groups were, respectively : ${\alpha}-tocopherol$ in RBC less than $30{\mu}g/{\mu}l$(Group I), $30{\mu}g/{\mu}l{\sim}60{\mu}g/{\mu}l$(Group II), more than $60{\mu}g/{\mu}l$(Group III). Artificial infection was accomplished by injecting $2{\times}10^7{\sim}2{\times}10^8$ erythrocyte of Babesia gibsoni-infected dog into the cephalic vein. We investigated the clinical signs, vitamin E concentrations in RBC and serum, Vitamin A concentrations in serum, hematological values, white blood cell(WBC) viability and RBC membrane osmotic fragility after infection of Babesia gibsoni for a period of 20 days at 5 day intervals. The results obtained are summarized as follows : 1. After infection by Babesia gibsoni, clinical examination revealed depression, anorexia, pale mucous membranes, dark brown urine and diarrhea in proportion as time went on. After 10 days of infection, one dog each of Groups I, II and III revealed depression and anorexia. Two dogs in Group I and one dog each of Groups II and III showed dark brown urine after 15 days. Diarrhea was observed in one dog in each of the 3 groups after 20 days of infection. 2. After 5 days of infection, two dogs in each of Groups I, II and III showed Babesia gibsoni in RBC of blood smear stained with Giemsa. At the 15th day after infection with Babesia gibsoni, they were observed in all experimental animals. After both 5 days and 10 days of infection, the rate of Babesia gibsoni parasitized RBC(permillage, ‰) was 1‰, and increased as time went on. 3. After 5 days of infection by Babesia gibsoni, Group I, which had the lowest vitamin E concentration, showed significantly decreased RBC and PCV levels(p < 0.01). Group II and group III also showed significantly decreased RBC and PCV levels after 15 days of infection(p < 0.05). Particularly after 10 days of infection, Group I showed lower values in RBC and PCV levels compared to Groups II and III. WBC, RBC, fibrinogen and total protein levels between the groups did not differ during experimental periods. 4. According to the WBC differential counts, the ratios of neutrophil to lymphocyte showed a tendency to be slightly higher in Group III (more than $60{\mu}g/{\mu}l$) than in Groups I and II. 5. WBC viability did not differ between the groups. 6. RBC membrane osmotic fragility did not differ between the groups.

Understanding and monitoring virus-mediated infections has gained importance since the global outbreak of the coronavirus disease 2019 (COVID-19) pandemic. Studies of high-throughput omics-based immune profiling of COVID-19 patients can help manage the current pandemic and future virus-mediated pandemics. Although COVID-19 is being studied since past 2 years, detailed mechanisms of the initial induction of dynamic immune responses or the molecular mechanisms that characterize disease progression remains unclear. This study involved comprehensively collected biospecimens and longitudinal multi-omics data of 300 COVID-19 patients and 120 healthy controls, including whole genome sequencing (WGS), single-cell RNA sequencing combined with T cell receptor (TCR) and B cell receptor (BCR) sequencing (scRNA(+scTCR/BCR)-seq), bulk BCR and TCR sequencing (bulk TCR/BCR-seq), and cytokine profiling. Clinical data were also collected from hospitalized COVID-19 patients, and HLA typing, laboratory characteristics, and COVID-19 viral genome sequencing were performed during the initial diagnosis. The entire set of biospecimens and multi-omics data generated in this project can be accessed by researchers from the National Biobank of Korea with prior approval. This distribution of large-scale multi-omics data of COVID-19 patients can facilitate the understanding of biological crosstalk involved in COVID-19 infection and contribute to the development of potential methodologies for its diagnosis and treatment.

Purpose: After damage control surgery, abdominal wall closure may be impossible due to increased intra-abdominal pressure (IAP), and primary closure may induce abdominal compartment syndrome. The purpose of this study was to investigate changes in the IAP and the feasibility of abdominal wall closure using artificial mesh. Methods: From July 2010 to July 2011, 8 patients with intra-abdominal hypertension underwent abdominal wall closure using artificial mesh. Medical data such as demographics, diagnosis, operation, IAP, postoperative complications, mortality and length of hospital stays were collected and reviewed, retrospectively. One patient was excluded because of inadequate measurement of the IAP. Results: Seven patients, 4 males and 3 females, were enrolled, and the mean age was 54.1 years old. Causes of operations were six traumatic abdominal injuries and one intra-abdominal infection. The IAP was reduced from $21.9{\pm}6.6mmHg$ before opening the abdomen to $15.1{\pm}7.1mmHg$ after fascial closure. Fascial closure was done on $14.9{\pm}17.5$ days after the first operation. The mean lengths of the hospital and the intensive care unit (ICU) stays were 49.6 days and 29.7 days respectively. Operations were performed $3.1{\pm}1.5$ times in all patients. Two patients expired, and one was transferred in a moribund state. Three patients suffered from complications, such as retroperitoneal abscesses, enterocutaneous fistulas, and bleeding that was related to the negative pressure wound therapy. Conclusion: After abdominal wall closure using artificial mesh, intra-abdominal pressure was well controlled, and abdominal compartment syndrome does not occur. When the abdominal wall in patients who have intra-abdominal hypertension is closed, artificial mesh may be useful for maintaining a lower abdominal pressure. However, when negative pressure wound therapy is used, the possibility of serious complications must be kept in mind.

Purpose: The forehead flap is the workhorse in nasal reconstruction, which provides a similar skin color, texture, structure, and reliability. There are some disadvantages, including donor site morbidities, 2- or 3-stage operations, and postoperative management after initial flap transfer. Furthermore, there has been little attention to the exposed raw surface wound, after the first stage of an operation. This article describes the authors' modification to overcome this problem, using artificial collagen membrane. Methods: An Artificial collagen membrane is composed of an outer silicone membrane and an inner collagen layer. After a forehead flap elevation, the expected raw surface was covered by an artificial collagen membrane with 5-0 nylon suture. A simple dressing, which had been applied to the site, was changed every 2 or 3 days in an outpatient unit. At 3 weeks postprocedure, a second stage operation was performed. Results: With biosynthetic protection of the raw surface, there were no wound problems, such as infection or flap loss. Thus, the patient was satisfied due to an effortless management of the wound and a reduction in pain. Conclusion: The application of an artificial collagen membrane to the raw under-surface of the flap could be a comfortable and a protective choice for this procedure.
To confirm the possibility of a wild marine crab, Gaetice depressus, as a carrier for white spot syndrome virus (WSSV) and to develop an alternative experimental model for WSSV in winter season, the susceptibility of the crab to WSSV was assessed by artificial challenge and subsequently tested for infection by PCR assay. The results revealed that the crabs were as highly susceptible as penaeid shrimps. WSSV caused 100% mortality in G. depressus within 16 days after intramuscular injection. The presence of WSSV in the moribund crabs was confirmed by PCR and was found in gills and muscle tissue. These results suggest that G. depressus can be naturally infected by WSSV via moribund shrimps, and can act as a potential carrier of WSSV. In addition, G. depressus can be used as an alternative experimental animal for WSSV.
Viral coat protein (CP) encoding cDNA with artificial start and stop codons was synthesized by reverse-transcriptase polymerase chain reaction (RT-PCR) from the Korean isolate of potato virus Y-vein nectrosis strain (pVY-VN). To make PVY CP cDNA to untranslatable form, three stop codons were inserted near the start codon by "megaprimer-PCR" method. The untranslatable CP cDNA was subcloned to plant expression vector and transferred to N. tabacum cv. NC82 by Agrobacterium-mediated transformation. Highly resistant plants to PVY infection were screened, based on symptom development after mechanical virus inoculation. By genomic PCR and Southern blot analysis, one or more copies of the untranslatable CP gene were found in all transformants. From northern blot analysis, highly resistant transgenic lines had very low level of CP transcript but susceptible lines had high level, suggesting resistance to PVY infection should be related to RNA-mediated mechanism.mechanism.
India has maximum genetic materials in early cauliflower, which grow in subtropical conditions. Different disease parameters like linear growth, maximum growth rate per day, AUDPC, apparent infection rate and percent diseased area were calculated in artificially inoculated plants. Apparent infection rate is not co-related with the black rot disease incidence and should never be considered during characterization of disease resistance and varietal screening. Based on the above disease parameters Kunwari-18, Phool Gobhi Kunwari, Kataki-7 and BT-10-2 were selected as moderately resistance to black rot in early cauliflower. These lines can be used for black rot prone area and also for black rot disease improvement programme. Considering the qualitative and quantitative parameters, slow rotting resistance cauliflower lines are selected as such for cultivation and would be best suited in integrated disease programme.
This study is aimed to clarify on the influence of cultivated conditions and fungicides on development of Rhizoetonia diseases of zoysiagrass and creeping bentgrass and results are as follows. Infection rates of disease were observed more serious when inoculum amounts were higher than low. Effects of the watering intervals on the severity of disease with artificial inoculating conditions, infection rates were higher when two days watering interval for creeping bentgrass and one day interval for zoysiagrass. Disease severity of creeping bentgrass incited by Rhizoetonia solani AG 1 (1A) was grater under conditions of double amounts of nitrogen fertilization. A posi- tive correlation was founded between the pH and disease development. The pots with pH 5 and 8 were significantly more diseased grass than those in which the pH was in 6.5. Mepronil and myclobutanil were extremely effective against brown patch of creeping bentgrass and large patch of zoysiagrass and followed by capro, and tebeonazole. Fenari was inferior effect to both diseases.
