Background: Recently, we identified a novel ginseng-derived lysophosphatidic acid receptor ligand, called gintonin. We showed that gintonin induces $[Ca^{2+}]i$ transient-mediated morphological changes, proliferation, and migration in cells expressing lysophosphatidic acid receptors and that oral administration of gintonin exhibits anti-Alzheimer disease effects in model mice. However, little is known about the intestinal absorption of gintonin. The aim of this study was to investigate gintonin absorption using two model systems. Methods: Gintonin membrane permeation was examined using a parallel artificial membrane permeation assay, and gintonin absorption was evaluated in a mouse everted intestinal sac model. Results: The parallel artificial membrane permeation assay showed that gintonin could permeate an artificial membrane in a dose-dependent manner. In the everted sac model, gintonin absorption increased with incubation time (from 0 min to 60 min), followed by a decrease in absorption. Gintonin absorption into everted sacs was also dose dependent, with a nonlinear correlation between gintonin absorption and concentration at 0.1-3 mg/mL and saturation at 3-5 mg/mL. Gintonin absorption was inhibited by the Rho kinase inhibitor Y-27632 and the sodiumeglucose transporter inhibitor phloridzin. Moreover, lipid extraction with methanol also attenuated gintonin absorption, suggesting the importance of the lipid portion of gintonin in absorption. This result shows that gintonin might be absorbed through passive diffusion, paracellular, and active transport pathways. Conclusion: The present study shows that gintonin could be absorbed in the intestine through transcellular and paracellular diffusion, and active transport. In addition, the lipid component of gintonin might play a key role in its intestinal absorption.

Presently, there is an increased demand for livestock products all over the world which has led to more devotion on improving livestock population. Although goats have been bred for a long time in Korea, but there is not much research conducted on traditional Korean black goat (Capra hircus coreanae) compared to other livestock populations. Mutton consumption has been dramatically changing from medicinal use to edible meat and this trend directs the black goat populations declining and also mutton import quantities are increasing consistently. The present study introduced a new estrus synchronizing technique with subsequent artificial insemination (AI) for Korean black goats to enable crossbreeding with non-native breeds for the small or subsistent farmers. Our data highlighted that, the percentage of motile sperm from the electro-ejaculated samples declined significantly after freezing and melting. In addition, the sperm motility significantly declined with regard to sperm incubation period (0, 5, 60, and 120 min at 37℃) and was negatively correlated (64.2 ± 7.9%, 63.3 ± 5.8%, 49.9 ± 6.3%, and 35.9 ± 7.6%, respectively) in frozen-thawed sperm samples. Moreover, the E2 levels were unchanged even 24 h after controlled internal drug releas (CIDR) withdrawal. But, 48 h and 72 h after CIDR removal, E2 levels increased significantly. These data helps us to consider the two time points for AI; CIDR removal after 24 h, at which E2 decreases, and after 48 h, as the time at which progesterone increases. Additionally, the AI after 48 h of CIDR removal group exhibited significantly higher pregnancy and parturition rates (42.9%) compared to AI after 24 h after CIDR removal 28.6% group. In conclusion, these studies will propose an optimal estrus synchronisation process with subsequent timing of AI and also will promote the Korean black goat breeding industry.
Transcription activator like effector nucleases (TALENs) are artificial restriction enzymes generated by fusing a TALE DNA binding domain to a DNA cleavage domain which remove and introduce specific genes to produce transgenic animals. To investigate the efficient laboratory techniques for the injection of TALEN mRNA, pEGFP-N1 commercial plasmid were microinjected into porcine parthenogenetic and in vitro fertilization (IVF). In Experiment 1, to investigate injection time, compared 4 different time durations (2 hr, 4 hrs, 6 hrs & 8 hrs) after post activation of parthenogenetic embryos and after 6 hrs of co-incubation with sperms in IVF embryos. There were significant difference (P<0.05) in development to the blastocysts (4.4, 8.9, 3.9, 0.6%), GFP expression in blastocysts (1.3, 5.7, 2.3, 0.0%) which injected after post activation of 4 hrs compared with other 3 groups. IVF embryos after 2 hrs and 4 hrs injected were expressed GFP significantly higher than rest of two groups (P<0.05). In Experiment 2, compared development of 2 different concentrations ($20ng/{\mu}l$ and $50ng/{\mu}l$) of EGFP injection. There were significant difference (P<0.05) between two treatments which has higher cleavage (58.8 vs 41.9%), blastocysts development rate (13.0 vs 11.1%) and GFP expressed blastocysts (5.7 vs 0.0%) in $20ng/{\mu}l$ than the $50ng/{\mu}l$ in parthenogenetic embryos. In IVF embryos, only $20ng/{\mu}l$ injected embryos were expressed GFP (4.2%) after 7 days of incubation and 77.3 vs 64.7% of cleavage, 26.4 vs 23.5% development to blastocysts. In Experiment 3, three different volumes (5, 10 and 20 pl) were microinjected into porcine embryos to determine the most appropriate volume. Out of 3 groups, significantly higher development rates of cleavage (68.3, 58.0, 29.3%), blastocysts (11.7, 12.7, 0.5%) and GFP expressed blastocysts (2.9, 7.8, 0.0%) were shown in the 10 pl group (P<0.05). In conclusion, these results imply that $20ng/{\mu}l$ concentration, 10 pl of volume and injection at 4 hrs after post activation for parthenogenetic and 2~4 hrs after IVF, $20ng/{\mu}l$ concentration and 10 pl volume for IVF embryos were more effective microinjection conditions.
본 연구에서는 ACE 저해능 및 항산화능이 있는 식물성 젖산균을 다양한 식물체로부터 분리한 후, 그 특성을 조사하였다. 김치, 부추, 포도 및 동동주에서 K-1, K-21, L-5, G-3 및 D-3 균주가 분리되었으며, 16S rRNA gene 염기서열 분석을 통하여 이들은 각각 Pediococcus pentosaceus, Lactobacillus plantarum, Weissella cibaria, L. plantarum 및 L. brevis로 동정되었다. 분리균주들은 MRS broth에서 44.3-71.9%의 ACE 저해능을 나타내었으며, 특히 G-3, L-5, K-1 균주는 skim milk가 함유된 MRS broth에서 59.0-8-98.6%의 높은 ACE 저해능을 나타내었다. 분리 균주는 42.5-82.7%의 DPPH radical 소거능을 나타내었으며, G-3 및 K-1 균주는 pH 2.5의 인공위액에서 42.2-88.1%의 높은 생존율을 나타내었다. 분리균주는 0.3% oxgall에서 24시간 배양시 55.4-112.8%의 내성을 나타내었다. 또한 분리균주는 유기산 생성에 따른 pH 감소 효과로 인하여 Pseudomonas aeruginosa를 포함한 일부 병원성 세균의 생육을 억제할 수 있었다.

BACKGROUND: Biochar (BC) has a great potential for enhancing soil fertility and carbon sequestration while facilitating beneficial waste disposition. Therefore, it is essential to assess and mitigate any inadvertent consequences associated with soil biochar amendment. Earthworm activity is very vital in the soil system, yet there are a limited number of studies that have examined their impact resulting from biochar application to soil. METHODS AND RESULTS: In this study, the survival, growth, reproductive tests, and oxidative DNA damage tests (measured by 8-hydroxydeoxyguanosine (8-OHdG) and catalase (CAT) activities) to assess the potential toxicity to earthworm Eisenia fetida in artificial soil amended with BCs were investigated. The BCs derived from perilla meal, sesame meal, and pumpkin seed were pyrolyzed at 300 and $550^{\circ}C$, and then amended with soil at a rate of 5%. All the earthworms survived, but lost weight compared to control soil after 28 day incubation period. Moreover, the BC-amended soils did not significantly affect the cocoon numbers of earthworms. Slightly higher concentrations of 8-OHdG and CAT were observed in earthworms present in BC-treated soil than those in control soil. Furthermore, the 8-OHdG concentrations in the soil amended with BC produced at $550^{\circ}C$ were greater than those at $300^{\circ}C$, and it slightly decreased as the incubation time increased. CONCLUSION: These observations could be due to higher contents of toxic metal(loid)s and also higher pH in BCs pyrolyzed at $550^{\circ}C$ than $300^{\circ}C$. While BC is efficiently being used in agricultural fields, this study suggests that it is required to assess the unintended negative impacts of BC on soil ecosystems.
본 연구는 돼지 액상정액을 인공수정용 100ml 플라스틱 병에 보존하면서 Lactose-Egg yolk 희석액과 B tschwiler 희석액 간에 보존 온도별 차이를 조사하고, Lactose-Egg yolk 희석액에서의 글리세롤 농도의 효과를 조사하여 돼지 액상정액을 좀더 장기간 사용할 수 있는 방법을 찾고자 실시하였다. 돼지 액상정액을 5$^{\circ}C$ 냉장고에 보존하면서조사한 바에 의하면, 37$^{\circ}C$에서 0.5 및 2시간 배양 후의 정자 운동성은 전체 보존기간동안 Lactose-Egg yolk 희석액이 B tschwiler 희석액보다 유의하게 (P,0.05) 높게 나타났고, 정상첨체비율은 두 희석액간에 차이가 없었다. 돼지 액상정액을 15$^{\circ}C$에 보존하면서 조사한 바에 의하면, 3일부터 7일 보존시까지 정자 운동성과 정상첨체비율에 있어서 B tschwiler 희석액이 Lactose-Egg yolk 희석액보다 유의하데 높게 나타났다. Lactose-Egg yolk 희석액을 이용한 돼지 액상정액의 글리세롤 농도의 효과에 있어서는 최종 글리세롤 농도가 0, 1, 3 및 5%보다 2%일 때 가장 높은 정자 운동성과 정상첨체비유을 나타내었다. 분만율, 복당 생존자돈수 그리고 출생시 평균 생시체중은 Lactose-Egg yolk 희석액과 B tschwiler 희석액간에 차이가 없었다. 이상의 연구 결과를 종합해 볼 때 Lactose-Egg yolk 희석액은 5$^{\circ}C$에서 B tschwiler 희석액은 15$^{\circ}C$에서 6~7일 동안 돼지 액상정액을 보존할 수 있었다.
본 연구에서는 식품용 프로바이토틱 생균인 비스루트균의 인공위액과 인공담즙산에 대한 내성, 효소활성, 항생물질과 알콜에 대한 내성, 그리고 병원성 세균에 대한 항균기작 등 주요 생균제 특성에 대하여 검토하였다. 인공위액 내성 실험에서는 pH 2.5로 조성된 인공위액을 통과한 생존율이 매우 높았으며 균수가 $2.4{\times}10^7\;CFU/mL$의 수준이었다. 인공담즙산에 대한 내성에서는 pH 2.5의 인공위액을 통과한 비스루트균의 인공담즙산의 내성은 oxgall이 함유되지 않은 대조구$(1.6{\times}10^7\;CFU/mL)$에 비해 오히려 균수가 증가되었으며, 최종적인 생존수가 $3.3{\times}10^7\;CFU/mL$ 으로 확인됨으로써 프로바이오틱 생균으로 매우 우수한 특성을 가지고 있었다. 효소활성을 측정한 결과, 비스루트균은 발암효소인 ${\beta}-glucuronidase$ 및 ${\beta}-glucosidase$ 의 활성이 나타나지 않았다. 또한 항생물질에 대한 내성은 rifamycin $10\;{\mu}g/mL$ 이상의 농도에 대해서는 생육 저해를 보였으나, $nisin(100\;{\mu}g/mL)$, $streptomycin(20\;{\mu}g/mL)$, $tetracycline(20\;{\mu}g/mL)$에 대해 내성을 보였다. 그리고 비스루트균의 알콜에 대한 내성은 4%의 농도에서 저해를 받았으며, 8%의 농도에서 더욱 저해를 받으나, $8{\sim}32%$ 범위의 농도에서는 균수가 더욱 크게 감소를 하지는 않았다. Listeria에 대한 향균기작을 측정한 결과, 24시간이내에 L. monocytogenes ATCC 19111을 완전히 저해를 하였으며, bactericidal한 양상을 나타내었다. 본 연구를 통해 밝혀진 비스루트균의 프로바이오틱 특성은 기능성 식품의 개발 등 산업적으로 매우 유용하다고 판단된다.
전통 장류에서 유산균 성상을 보이는 균주를 분리하여, TLC와 PCR을 통하여 HoPS를 우수하게 생성하는 균주 HSB15, JSA22, JSA57, JSB22, JSB66 및 JSB89를 선발하였다. 선발된 6균주의 16S rDNA의 염기서열을 분석한 결과 HSB15는 L. alimentarius, JSA22는 L. plantarum, JSA57은 L. pentosus, JSB22는 L. brevis, JSB66는 L. alimentarius, JSB89는 L. parabrevis로 동정되었다. 항균 활성은 6균주 중에서 JSA22가 4개의 병원성균에 대한 항균을 나타내어 항균 활성이 가장 우수하게 나타났고, 인공위액 저항성은 HSB15, JSA22와 JSA57 균주가 70% 이상의 높은 인공위액 저항성으로 가장 우수했으며, 인공담즙 저항성은 HSB15를 제외한 5균주에서 80% 이상의 생존율을 나타내며 담즙액 저항성이 우수하게 나타났다. 또한 probiotic 특성을 지닌 분리 균주의 prebiotics로써 AOS를 이용한 성장율을 관찰한 결과, 모든 분리 균주가 AOS를 선택적으로 이용하며 성장하는 것을 관찰하였고 특히 JSB22가 AOS 이용 성장율이 가장 우수한 것으로 확인되었다. 본 연구를 통해 장류유래 probiotics를 선발하였고 AOS의 새로운 prebiotics로 활용을 검토한 바, synbiotics로 이용할 수 있는 가치가 있다고 판단되었다.
The effects of seed-associated or free linseed oil on fermentation characteristics and long-chain unsaturated fatty acids composition, especially the formation of conjugated linoleic acid (CLA) and octadecenoic acid (trans-11 $C_{18:1}$, $t-C_{18:1}$) by mixed ruminal bacteria were examined in vitro. Concentrate (1% of culture solution, w/v, as-fed basis) with ground linseed (0.6% of culture solution, w/v, DM basis) or linseed oil as absorbed onto ground alfalfa hay was added to 600 ml mixed solution consisting of strained rumen fluid and artificial saliva at the ratio of 1:1 in a glass culture jar. The culture jar was covered with a glass lid with stirrer, and placed into a water-bath ($39^{\circ}C$) and incubated anaerobically up to 24 h. Seed-associated or free linseed oil did not significantly affect the pH and ammonia concentration in the culture solution. Molar percent of acetate tended to increase while that of propionate decreased with the addition of free oil treatment throughout the incubation. Differences in bacterial number were relatively small, regardless of the form of supplements. Decreasing trends in the compositions of linoleic acid ($C_{18:2}$) and linolenic acid ($C_{18:3}$) but increasing trends of stearic acid ($C_{18:0}$), $t-C_{18:1}$ and CLA compositions were found from culture contents up to 12h incubation when incubated with both ground linseed and linseed oil. The compositions of $C_{18:0}$, $C_{18:2}$ and $C_{18:3}$ were greater but those of oleic acid ($C_{18:1}$), $t-C_{18:1}$ and CLA were smaller in a culture solution containing ground linseed than those containing linseed oil. The ratio of $t-C_{18:1}$ to CLA was lower in the culture solutions containing linseed oil up to 12h incubations as compared to those containing ground linseed.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
