Attribution of Cortical Granules to Formation of Fertilization Envelopes and Polyspermy Block in Urechis unicinctus

  • Cortical reaction and polyspermy block are well defined in most marine invertebrates. In Urechis species, the function of cortical granules (CGs) is not yet known, and there is controversy on whether the cortical reaction occurs, or the fertilization envelope (FE) is attributed to CG releases or functions to prevent polyspermy. This study was carried out to determine the cortical reactions and functions of the FE in Urechis unicinctus. Artificial insemination of the eggs revealed that CG release occurred to give rise to perivitelline space (PS) into the final FE. Both PS and final FE effectively blocked polyspermy. The final FE was accomplished within 10 min after sperm-egg initial binding. No massive release of CGs occurred within the early phase of 5 min after the initial binding, initially and the PS seemed to playa role to prevent polyspermy. The CG massively released its content into the PS in late phase of FE formation, and differentiated PS into five intermediate layers. The layers opened into each other by anastomosis, so that the final FE consisted of two layers, the inner layer ($15{\mu}m$ in thickness) and the outer layer ($1{\mu}m$ in thickness). The outer layer derived from vitelline layer and the inner layer consisted of PS and CG secretions. Immunofluorescence and confocal laser microscopy revealed that the spermatozoon took up residence in the egg cortex during FE formation and successive meioses of the fertilized egg. These results suggest that both PS and final FE of U. unicinctus were equivalent to the early and late block, respectively, of other marine animals.

Influence of Food and Density on the Growth and Survival of Sea Cucumber, Stichopus japonicus (해삼, Stichopus japonicus 유생의 성장과 생존에 미치는 먹이 및 수용밀도의 영향)

  • In order to obtain the basic biological data for artificial seedling production of sea cucumber, Stichopus japonicus, the influence of water temperature and air dry for spawning induction, egg development and larvae rearing was investigated during the period from April, 1995 through September, 1995. Spawning induction rate by the water temperature was 6.0~17.5% and air dry was responsed 1.4~4.0%. Number of eggs spawned of Stichopus japonicus were $50~500\times10^4$ individuals, the fertilization and hatching rate were ranged 84.0~96.0%, 71.4~84.6% respectively. The fertilized egg of Stichopus japonicus appeared mean diameter of $154{\mu}m$. At a constant water temperature of $23^{circ)C$, it become 4 tell stage from fertilization after 2 hours 10 minutes, hatching larvae after 14 hours half, auricularia larvae after 3 days, doliolaria larvare after 11 days and pentactula larvae after 15 days ready for settlement. The suitable food in the larvae reared for 17 days after fertilization were shown the best growth and survival in the larvae food of Chaetoceros calcitrans. Optimum density for larvae rearing were maintained of the larval density lower than 2 individuals/ml.

STUDIES ON THE ARTIFICIAL FERTILIZATION AND DEVELOPMENT OF MERETRIX LUSORIA (대합 Meretrix lusoria의 인공수정 및 발생에 관한 연구)

  • Meretrix lusoria is one of the most favorite edible bivalves inhabiting wide inter-tidal flats along the western coast of Korea. Over the period of July to September 1973, some specimens from a tidal flat near AnMyun Island were submerged in sea water with various concentrations of ammo-mum hydroxide added and careful observations were made on their fertilization, early development, and metamorphosis of the larvae. The highest rate of fertilization was demonstrated by individuals treated with 1/1000 normal solution of ammonium hydroxide, and their fertilized eggs followed normal development, i. e., two cell stage in 1.2 hours after fertilization, gastrula stage after 4.7 hours, and trochophore stage after 5.6 hours. Within 24 hours after fertilization M. lusoria larvae have acquired the form of early straight-hinge veliger with the mean prodissoconch I length of $112\mu$. It takes seven days to get the umbo stage with the mean shell length of $172\mu$ and twenty days to get the metamorphosing stage with the mean shell length of $232\mu$. The larvae were cultured to the metamorphosing stage with the shell length of $272\mu$ in the laboratory condition. The relationship between the shell length (L) and the shell height (H) in veliger stage is shown as H=1.02325L-24.46425 with a significant difference.

  • Cyclina sinensis is an edible bivalve inhabiting wide tidal flats which are exposed to the air at ebb tide along the western coast of Korea. Over the period of June to September 1971, some specimens from a tidal flat near Inchon were submerged in sea water with various concentrations of ammonium hydroxide added and careful observations were made on their fertilization, early development, and metamorphosis of the larvae. The highest rate of fertilization was demonstrated by individuals treated with 1/1000 normal solution of ammonium hydroxide and their fertilized eggs followed normal development, i.e., two cell stage 1.5 hours after fertilization, blastular stage after 4 hours, and trochophore stage after 6 hours. It takes seven days to get the umbo stage with the mean shell length of $190\mu$ and twenty days to get the morphosing stage with the mean shell length of $260\mu$. The larvae were cultured to the metamorphosing stage with the shell length of $270\mu$ in the laboratory condition.

  • The purposes of this research are to select the appropriate kinds of landscape woody plants for afforestation artificial ground and to find out the way of developing planting-tree area of artificial ground through test planting and case study. The summary of the research is as follows; As a result of observing the state of trees' growth, trees on artificial grounds grow as well as, or even better than those on natural grounds. The kinds of trees which represent better growth states on artificial soil of rooftop test area are Thuja occidentalis, Prunus armericana var. ausu, Acer palmatum, Viburnum sargentii for. sterile, Buxus microphylla var. koreana, etc. In addition, soils than on natural soils. As a result of investigating the tree's growth state according to soil beds of artificial soils, shrubs don't represent distinct differences. They show that shrubs can be planted on 30cm soil bed. The rest of trees no differences according to the variation of soil beds, and they are in normal condition or have slight damage. This means that though arbor planting area of artificial ground is made 45~60cm, it is possible for trees to grow on it. In consequence of analyzing the cases on areas of landscaping artificial grounds, as the appropriate kinds of trees for afforestation artificial ground, Diospyros kaki, Hibiscus syriacus, Syringa dilatata, Magnolia kobus, Acer ginnala and cornus alba are selected in Capital region, and Nandina domestica, Taxus cuspidata, Ilex crenata, Viburnum awabuki, Aucuba japonica and Euonymus japonica are selected in southern region. In addition, Juniperus chinensis, Zelkova serrata, Juniperus chinensis var. kaizuka, etc. are selected regardless of regional character. In accordance with the state of soil it shows great difference, namely, fertilization once or more a year and irrigation for drought is advantageous to tree's growth. This research represents that it is necessary that standards related to landscaping artificial grounds are subdivided. In addition to it, the plan for revising related to regulation is drawn up.

Breeding of New Varieties by Ovule Culture of Intergeneric Hybrid in the Aurantioideae (속간교잡 후 배주배양에 의한 감귤류 신품종 육성)

  • This study was carried out to develop new varieties which are dwarf and tolerant to winter cold in the Aurantioideae by intergeneric crossing. to do that, the reciprocal crosses of Hwanggeumyooza and trifoliate orange, yooza and trifoliate orange were done and in vitro immature ovule culture of their hybrid was carried out .The callus formation from immature ovule was good in order of Hwanggeumyooza, Hwanggeumyooza $\times$ tifoliate orange, yooza, and trifoliate orange and best at 1 to 3 mg/L NAA+0.5mg/L zeatin on MT medium. In vitro germination percentage of 20week old hybrid of Hwanggeumyooza $\times$ tifoliate orange and trifoliate orange $\times$ Hwanggeumyooza were 41.3% and 37.7, respectively. The phenotype of hybrid (95%) of Hwanggeumyooza $\times$ trifoliate orange and that (100%) of trifoliate orange $\times$ Hwanggeumyooza were similar to that of trifoliate orange. After Hwanggeumyooza was pollinated by pollens of trifoliate orange, the pollen tubes grew on stigma after 3h of pollination and entered into micropyle after about 24~28 h. One gamete in pollen was fused with polar nuclei after 2 days and other one fused with egg nucleus at 3days after pollination. The fruit set percentage by intergeneric crossing was 14.0% in Hwanggeumyooza $\times$ trtfoliate orange and 17.5% in trifoliate orange $\times$ Hwanggeumyooza. The fruit set percentages of Hwanggeumyooza. and trifoliate orange were 34.2% and 39.5% by artificial self-fertilization, 34.2% and 39.5% by artificial cross fertilization, 3.1% and 1.4% by parthenocarpy and 13.0% and 3.0% by natural fertilization, respectively. The somatic and gametic chromosome numbers of Hwanggeumyooza, yooza, and trifoliate orange were 2n=18 and n=9.

Correlationship between Artificial Maturation Season and Reproduction Coefficient in the Cultured Eel Anguilla japonica (양식산 뱀장어 Anguilla japonica에 있어서 인공성성숙시기와 번식률과의 상관관계)

  • This study investigated the correlationship between artificial maturation season and reproduction coefficient of cultured eel Anguilla japonica from May (spring) to next January (winter). The brood stock, female eels ($400{\sim}600\;g$) were artificially matured by weekly intramuscular injections of salmon pituitary extracts (SPE, 20 mg/fish) to induce a completion of vitellogenesis. After completion of vitellogenesis, final oocyte maturation and ovulation was induced by injection of $17{\alpha}$, $20{\beta}-dihydroxyprogesterone$ (DHP) at about $2\;{\mu}g/g$ body weight. Most fish ovulated $15{\sim}18\;h$ following the DHP injection. The ovulated fish were induced to natural spawning or artificial fertilization by the dry method. Males ($200{\sim}350\;g$) were received weekly intramuscular injections of human chorionic gonadotropin (HCG) at a dosage of 1 IU/g body weight to induce testicular maturation and spermiation. Seasonal reproduction coefficient which includes the rate of ovulation, buoyancy, fertilization and hatching of eggs in the artificially matured eel during spring to summer ($May{\sim}July$) were significantly higher than the other season, while there were no significant difference among spring and summer (P<0.05). Furthermore, the number of eggs spawned and larvae hatched in the artificially maturated eel during spring to summer ($May{\sim}July$) were significantly higher than the other season, while there were no significant difference in spring and summer (P<0.05). These results indicate that artificial maturation by hormone treatment of A. japonica was successful only during spring to summer, which is the maturation period in the wild stock in nature. Consequently, it is possible to determine the period of artificially induced sexual maturity by the reproduction coefficient which includes the rate of ovulation, buoyancy, fertilization and hatching of eggs in the cultured eel A. japonica.

A Study on the Estimation of Zoysia matrella's Evaporation Using Makkink Model (인공지반에서 금잔디의 증발산량 예측에 관한 연구 -퍼라이트 배합토에서 Makkink의 일사법을 이용하여-)

  • The purpose of this study is to find out the difference of Zoysia matrella's evaporation in between 100 percent soil and mixed soil with 50 percent of perlite to create green spaces on the artificial ground. It is believed that the weight against the artificial ground will be reduced, provided the vegetation is possible in the circumstance of the mixed sol with 50 percent of perlite. The study employed a modified Makkink's model by Iwasa who had developed the model for estimating Zoysia matrella's evaporation in the natural ground using the Makkink's formula in 1997 at Chiba University, Japan. The parameter of Makkink's formula is the solar radiation. For that reason, the Makkink's formula is simple and easy to measure the parameter and has a high utility. If the outcomes from mixed soil are close to modified Makkinks formula, the modified Makkink's formula will be applied to estimate in the artificial ground with mixed soil with 50 percent of perlite. Weather observation and actual amount of evaporation of Zoysia matrella have been measured, and the relation between weather condition and actual amount of evaporation had been also investigated. In line with this, we found out that there is a relevant relationship between daily average temperature, the modified Makkink's model by Iwasa, and the actual amount of evaporation. As the results of the experiment, the outcomes from mixed soil with 50 percent of perlite have very high relation to 100 percent soil. In addition, mixed soil has more adhesion with water than natural soil. However, it needs to be adequately maintained in terms of fertilization and damage from disease and harmful insects until the gras fastens its roots into the soil. By using mixed soil with 50 percent of perlite, the load from soil on the artificial ground can be reduced. The study on the growth of the grass throughout the plant vegetation and the actual amount of evaporation in the mixed soil with 50 percent of perlite should be performed in the future.

A Study on Seedling Production of Jicon Scallop, Chlamys farreri 1. Spawning, Development and Rearing of Larvae (비단가리비 종묘생산을 위한 연구 1. 산란, 발생 및 유생사육)

  • Jicon scallop (Chlamys farreri), as one of new culture species, has been taken a growing interest in Korea recently. Artificial seeds are needed since seeds in natural waters are absolutely short. To develop the better methods for the artificial spawning and the rearing larvae on a large scale, experiments were carried out from July 1993 to May 1994. Mother shells were collected from the area near Tong-yeong and Geoje-do, the southern part of the Korean Penninsula. It was estimated that a period of 2-years was needed to be mother shells after the fertilization. Generally, spawning occourred intermittently from March to August and the maximum occurred in July. A best way for the artificial spawning was a chemical incentive, a 'serotonin injection' in parallel with the exposure and the temperature stimulation. Densities of the fertilized eggs for the normal development was from 30 to 40 per mililiter. It took 27-days from the fertilization to the settling larva averaging $155{\mu}m$ in size with water temperature 11.5\~13.0^{\circ}C$. Isochrysis galvana and Chaetoceros calcitrans with a density of $1,500\times10^4$ cells per ililiter were supplied for rearing the larvae. A proper density of larvae was 10 individuals per mililiter, and the survival rate of larvae from the flowing water system was much better than that of larvae from the still water.

Effects of Soil Amendments on Survival Rate and Growth of Populus sibirica and Ulmus pumila Seedlings in a Semi-arid Region, Mongolia (몽골 반건조 지역에서 토양 개량이 백양나무와 비술나무 묘목의 활착 및 생장에 미치는 영향)

  • This study was conducted to investigate effects of soil amendments on survival rate and growth of seedlings in a semi-arid region, Mongolia. 2-year-old Populus sibirica and Ulmus pumila seedlings were planted in alkaline sandy soils and treated with 2 levels of nitrogen, elemental sulfur, artificial moisture retention and converted loess. After 4 months, the seedling survival rate of both species decreased as the amount of nitrogen increased. Nitrogen has been generally known to increase seedling survival rate and growth by supplying nutrients, however, reduced survival rate in this study might be affected by consequential increase in soil osmotic pressure, which was caused by excessive nitrogen fertilization. The root collar diameter (RCD) growth of both species increased significantly by the treatment of converted loess, and only RCD growth of P. sibirica increased by the artificial moisture retention treatment. Although P. sibirica is drought-tolerant, it is in the group of Populus spp. which requires a high capacity of available water. Conversely, the elemental sulfur treatment showed no effect on survival rate and RCD growth for both species due to the low oxidation of sulfur in arid soils. The extended monitoring of seedling growth and soil characteristics is required to elucidate the long-term effects of soil amendments in the semi-arid region, and the further studies are also needed to examine the appropriate amount of fertilizers for both species.
