Evaluation of Antioxidant Activity and Cytotoxicity in Mixture Extract of Artemisia asiatica Nakai and Moringa oleifera Lam

  • The purpose of this study was performed to evaluate antioxidant activity of the Artemisia asiatica Nakai and Moringa oleifera Lam mixture extract. Mixture extracts were manufactured by concentration and compared with a single extract (only the Artemisia asiatica Nakai mixture and only the Moringa oleifera Lam mixture). The experiments conducted Total polyphenol measurements, Total flavonoid measurements, DPPH radical scavenging activty, ABTS radical scavenging activty and LDH assay. The LDH assay assessment shows that all extracts are cells compared to controls. The toxicity was weak. Finally, The antioxidant capacity was rated higher than mixture extract of a single extract. Also, the optimized mixture was determined AM5 (Artemisia asiatica Nakai mixture: Moringa oleifera Lam mixture = 3:1). For extracts of AM5, Total phenol and flavonoid contents were 271.769±18.087 mg/g and 45.384±5.026 mg/g. and DPPH and ABTS scavenging activity were 70.8±6.496% and 77.1±9.634%. Therefore, it is expected that the value of the extract will increase as it increases its antioxidant activity if it is manufactured according to the appropriate ratio.

  • Ethanol ($70\%$) extract of Caesalpinia sappan L. and Forsythiae suspensa Vahl. showed $84\%$ and $72\%$ of anti-urease activity against Helicobacter pylori, respectively. Among the fraction of Artemisia asiatica Nakai extract using methanol ($80\%$), water, ethyl acetate and butanol ethyl acetate fraction showed $90\%$ of the anti-urease activity.

  • It has long teen known in herbal medicine that the mugwort not only exerts a strong hemostatic and parasiticidal actions but also has therapeutic effects for stomachache and diarrhea. In recent pharmaceutical botany the mugwort is known to have antipyretic and astringent actions also. Among the major principles which have been found in the leaves and stems of mugwort are inulin, alkaloids, vitamines, and esestial oil. It is well known that santonin, one of the well known parasiticides, is one of the glucosides extracted from the limited species of mugwort. The present study was undertaken to investigate effects of mugwort(Artemisia asiatica Nakai) on the motility of isolated rabbit duodenum. The results obtained are as follows: At does of 0.2%, 0.5% and 1. 0% AAJ(Artemisia asiatica juice) markedly enhanced contractility of isolated duodenum and tonus of the intestine was also augmented with doses of 0.5% and 1.0%. The augmentative effect of AAJ on intestinal motility was not affected by pretreatment with epinephrine and avil while it was completely abolished by atropine. Therefore it is strongly suggested that augmentative action of AAJ on duodenal motility was exerted solely through muscarinic cholinergic receptors.

  • The wormwood is one of the plants which occur widely throughout the world. Though the precise data on the entire chemical composition of mugwort leaves are not available, the major principles which have been found so far include inulin, alkaloid, thujon, sesquiterpene and several vitamins. Santonin, a parasiticide, is one of the glucosides extracted from the limited species of wormwood. It has long been known in herb medicine that the plants of this family has not only strong hemostatic, analgesic and parasiticidal actions but also therapeutic effects for diarrhea, stomachache and asthma. In recent pharmaceutical botany the wormwood is introduced to have antipyretic and astringent actions also. The mugwort(Artemisia asiatica Nakai) is the most common species of wormwood that occurs in Korea. The usage of this edible leaves of mugwort is rather various. It is used not only for wormwood bath but also as forage, moxa and medicinal agents. Recently Kim et al reported from their study on the effect of mugwort on the motility of isolated intestine of rabbits that tonus and motility were markedly enhanced by mugwort but this effect of mugwort on intestinal motility was almost completely blocked by atropine suggesting that activity of mugwort was exerted through its cholinergic effect. It was the findings of Kim et al that prompted the authors to do the present experiment. The present study was undertaken to investigate effects of mugwort(Artemisia asiatica Nakai) juice on the respiration and blood pressure in cats. And also studied was the mechanism of depressor action of Artemisia asiatica Nakai Juice (AAJ). The results obtained are as follows; 1) It was observed that mean arterial blood pressure and heart rate were decreased markedly by AAJ. Following administration of 0.15 ml/kg and 0.3 ml/kg AAJ into cats the maximum depressor responses observed were $77.5{\pm}2.2\;mmHg$ and $94.0{\pm}3.7\;mmHg$ respectively. 2) Depressor responses to AAJ were blocked markedly by atropine whereas the responses were not affected by propranolol and dibenamine. Therefore it is strongly inferred that depressor action of AAJ results mainly from its cholinergic effect. This inference was further substantiated by the fact that heart rate change which invariably accompanies depressor responses to AAJ was almost completely abolished by atropinization. 3) After administration of AAJ into cats frequency of respiration was markedly increased while depth of respiration decreased during first 2-3 seconds.

Study on the Manufacturing Process of Artemisia asiatica Carbonized Organic Solution(Wormwood Acetic Acid) by Development of Carbonize Equipment (탄화장치 개발에 의한 쑥(Artemisia asiatica) 탄화 유기용액(쑥초액) 제조에 관한 연구)

  • In this study, liquid with carbonized artemisia was developed for the first time that can be used as food additives and outside medicinal agents by burning artemisia species such as Artemisia dubia wall, Artemisia capillaris Thunb, Artemisia montana (Nakai) Pamp), Artemisia keiskeana Miq, Leonurus Japonicus Houtt. etc. In addition, edibility of recovered liquid with carbonized artemisia was verified by comparing the contents according to each ingredient with common refined pyroligneous liquid through the analysis of 9 kinds of organic acid by the species of artemisia, methanol and ethanol. Besides, the result of antimicrobial activities on six kinds of food spoilage and food poisoning bacteria observed by paper disc method for liquid with carbonized artemisia indicated that those two liquids showed strong antimicrobial. At the concentration of 1.5 mg/disc, clear zone for S. sonnei and S. aureus, L. monocytogenes was 13~16 mm, and they showed stronger antimicrobial activities than other strain.

Induction of Phase II Enzyme Activity by Artemisia asiatica Nakai Aqua-acupuncture Solution (애엽(艾葉) 약침액(藥鍼液)에 의한 Phase II 효소 활성 유도)

  • Artemisia asiatica Nakai aqua-acupuncture solution (ANAS) was administered once daily for 10 days before the tumor implantation ($1{\times}10^6\;cells$). Body weight, spleen weight and the number of ascitic tumor cells were measured at 6 days after tumor implantation. The change of body weight and the survival rate of mice were observed for 21 days. It was used three biomarkers (quinone reductase, glutathione, glutathione S-transferase) to test chemopreventive potentials of ANAS. ANAS exerted antitumor activity by inhibiting the growth of Ehrlich ascites tumor cells in vivo. Mice given Ehrlich cells and ANAS at $CV_{12}$ and $BL_{18}$ had 57.1% to 49.2% survival after 21 days. Quinone reductase activity and glutathione levels were increased with ANAS. However, glutathione S-transferase level was 1.1-fold with ANAS. These results suggest that ANAS has chemopreventive potential by inducing QR activity and increasing GSH level.

  • The purpose of this study was to examine the potential for the development of radioprotective agent in extracts manufactured by mixing Artemisia asiatica Nakai and Moringa oleifera Lam known as antioxidant food with the appropriate ratio. Their whole body were irradiated 7Gy radiation after oral administration of a mixed extract of Artemisia asiatica Nakai and Moringa oleifera Lam to SD Rat for two weeks. And 1 day, 7days, 21days later, changes in blood cell components, SOD activations and tissue changes in the uterus were observed.It was confirmed that the AM + IR group had a higher tendency to recover leukocyte (p<0.05) and platelet (p<0.05) levels than the IR group. It was also confirmed that SOD activity was increased and cell death was decreased in uterine tissue.Based on these results, the mixed extract of A and B is expected to be useful as a radiation protection agent capable of reducing blood cell and uterine damage caused by radiation exposure.
