Artemisia argyi H. has been used for centuries as a traditional medicine and food supplement in Asian countries. The objective of this study was to investigate the physiological activities of Artemisia argyi H. extracts prepared by butanol, chloroform, ethyl acetate, ethyl ether, hexane, and methanol extraction. We evaluated total phenol and flavonoid content, antioxidant activity, nitric oxide (NO) and reactive oxygen species (ROS) release, and osteoclastogenesis inhibition. The total phenolic and flavonoid contents were highest in the methanol extract (49.46 mg GAE/g and 24.32 mg QE/g, respectively). The methanol extracts also had the highest antioxidant activity (DPPH and ABTS radical scavenging ability and ferric reducing antioxidant power), while the hexane extract had the lowest. The release of NO and ROS was dose-dependently decreased by pre-treatment with all solvent extracts. At the same concentrations, the ethyl acetate and butanol extracts showed higher inhibition of NO and ROS production when compared with the other extracts. The butanol extract, at a concentration of $20{\mu}g/ml$, inhibited about 89% of the activity of the osteoclast marker, tartrate resistant acid phosphatase (TRAP). These results suggest that butanol extracts of Artemisia argyi H. may be effective natural medications for the prevention and treatment of osteoporosis.
This study has attempted to examine the effect of Artemisia princeps and A. argyi on liver function-related enzymes in rats with $CCl_4$ adminisration. The activities of serum aspartate aminotransferase(AST), alanine aminotransferase(ALT) and alkaline phosphatase(ALP) from A. princeps were decreased by 33, 23 and $19\%$, respectively, compared to control. The activities of AST, ALT and ALP from A. argyi were decreased by 37, 33 and $26\%$, respectively. Total phenol contents were 10.2 mg/mL and 4.7 mg/mL in A. princeps, and A. argyi, respectively. Also, flavonoid contents were $6.1\;mg\%\;and\;3.6\;mg\%$ in A. princeps, and A. ar효i, respectively. Ethanol extract from A. argyi showed higher electron donating ability toward DPPH than A. princeps. A total of 31 volatile components(3 hydrocarbons, 10 terpenes, 5 carbonyls, 8 alcohols and 5 esters) were indentified in A. princeps, and A. argyi. The major volatile components of A. princeps were $\delta$-3-carene($2.2\%$) in terpenes and nerolidol($0.9\%$) in alcohols. The major volatile components of A. argyi were eugenol($1.4\%$) in alcohols and thyl pentadecanoate($1.1\%$) in esters.

Artemisia capillaris Thunb. (Compositae) is a native herb of East Asian countries and has used for the treatment of jaundice, high liver fever, and digestive diseases for a long time, as well as being developed as the source of herbal preparations until now. The major components from A. capillaris were chlorogenic acid (1) and its derivatives substituted with caffeoyl moieties, such as 3,5-dicaffeoylquinic acid (2) and 4,5-dicaffeoylquinic acid (3), and coumarins, such as scoparone. In the study, four compounds, chlorogenic acid, 3,5-dicaffeoylquinic acid, 4,5-dicaffeoylquinic acid and scoparone (4) in the 70% ethanolic extract of A. capillaris were simultaneously determined by using HPLC-UVD system. This method was validated with the terms of linearity, precious and accuracy according to ICH guidelines. The developed method was successfully applied for the quantitative analysis of Artemisia genus, A. capillaris, A. iwayomogi, A. princeps, and A. argyi, distributed in Korea.

Antioxidant and cancer cell growth inhibition activity of hot water extract from five different varieties of Artemisia (A. Argyi H., A. iwayomogi Kitamura, A. Princeps Var Orien talis HARA, A. princeps Pampanini and A. annua L.) in Korea was studied. We determined the phenol and flavonoid contents and examined antioxidant assay, such as DPPH, NO radical scavenging, activity ferric-reducing antioxidant power (FRAP), and bleaching inhibition activity in the ${\beta}$-carotene linolic acid system. Also, we performed HeLa and MCF-7 cancer cell growth inhibition assay of Artemisia extracts. Total phenol and flavonoid contents were the highest in A. iwayomogi Kitamura followed by A. Argyi H. DPPH radical scavenging activity was the highest in A. Argyi H. at 50 ${\mu}g/ml$ concentration, NO radical scavenging activity was more than 50% in A. Princeps Var Orien talis HARA, A. princeps Pampanini, and A. annua L. at 200 ${\mu}g/ml$ concentration. FRAP was higher in A. Argyi H. and A. iwayomogi Kitamura. Antioxidant activity in the ${\beta}$-carotene linolinolic system was also higher in A. Argyi H. and A. iwayomogi Kitamura by 60.50% and 56.90% at 100 ${\mu}g/ml$ concentration, respectively. In cancer cell growth inhibition activities at 400 ${\mu}g/ml$ concentration, A. iwayomogi Kitamura showed greater than 80% on HeLa cell. A. princeps Pampanini and A. Argyi H. extract had growth inhibition activities greater than 80% on MCF cell. The results of this study suggest that the antioxidant and anticancer activities in various Artemisia are a promising source of functional food ingredients.

The effects of gamma ray or electron beam irradiation on herbaceous medicinal plants were investigated in terms of the composition of volatile constituents using the aerial parts or leaves of Artemisia princeps Pamp. cv. Ganghwayakssuk and Artemisia argyi cvs. Namhaeyakssuk and Hwanghaessuk. The composition of volatile constituents in leaves was clearly distinguishable among the three Artemisia cultivars. However, the relative proportions of the major volatile constituents such as 1,8-cineole, ${\alpha}$-pinene, camphene, santolina triene, and artemesia triene, were similarly changed in two or three cultivars by gamma ray or electron beam irradiation. In particular, the proportion of 1,8-cineole was increased up to 1.29- to 1.71-fold in the three cultivars after irradiation with gamma ray. These results suggest that gamma ray or electron beam irradiation can be applied to modulate the composition of volatile constituents in the leaves of Artemisia plants.

The antioxidant activities of 60% ethanol extracts from different parts (leaf, stem, and root) of Artemisia argyi using an in vitro system were examined to find a possibility as natural antioxidant substances. The highest total phenolics (23.08 mg/g) was obtained by 60% ethanol extract of leaf. The scavenging activity of 60% ethanol extract from different parts of A. argyi on the DPPH and ABTS radical was raised with increasing amount of extracts, and 60% ethanol extract from leaf showed the highest radical scavenging activities. The leaf extract presented the highest reducing power and strong inhibitory effect on autooxidation of linoleic acid. In MDA formation using mouse brain homogenates, the 60% ethanol extracts of A. argyi also exhibited antioxidant activity as inhibition of MDA (71.38% at $200{\mu}g/mL$). Therefore, these results suggest that the 60% ethanol extract of A. argyi leaf possess excellent antioxidant activities and thus it has great potential as a source for natural antioxidant.
The water fraction exhibiting anticancer activity was prepared from 70% methanol extract of Artemisis argyi by stepwise solvent partioning. This water fraction(5 $\mu$g/ml concentration) showed a considerable cytotoxicity against leukemic L1210 cells with a maximal value of 92% for 3 days culture. Contrastingly to such substantial anticancer activities the identical fraction showed far low toxicity against normal lymphocytes than chloroform fraction of Artemisia argyi mitomycine and 5-fluorouracil at every concentration ranging 0.01$\mu$g/ml~10.00$\mu$g/ml. The cytotoxicity displayed against L1210 cells by the water fraction of Artemisia was found to be proportinal to the decrease of viability of L1210 cells. On the other hand, $O_2$ion generation in L1210 cells appeared to be elevated in accordance to cytotoxicity by the water fraction with concurrent increases of superoxide dismuatse (SOD) and glutathione peroxidase (GPx) which are responsible for the conversion of $O_2$ ion and $H_2O$$_2$ respectively These findings taken together indicate that the death of L1210 cells by the water fraction of Auemisia atgyi, may be induced at least in part by the detrimental action of reactive oxygen species (ROS) including $O_2$- in spite of substantial extorts of SOD and GPx to overcome the attack of ROS.
SD-994 was prepared from methanol extract of Artemisia argyi by stepwise purification of solvent partioning and silica gel chromatography. In the course of this purification, fractions obtained at each step were investigated for their cytotoxicities against L1210 cells. Fractions A~G prepared from chloroform fraction showed considerable cytotoxicities raging 40~90% against L1210 cells. Subfractions I~IX obtained from fraction A exhibited various cytotoxicities and subfraction I (SD-994) was found to be the most effective compound. $IC_{50}$ values of SD-994 were measured to be $0.5{\;}{\mu\textrm{g}}/ml and less than $0.05{\;}{\mu\textrm{g}}/ml against L1210 cells and normal lymphocytes, respectively: When SD-994 was added to L1210 cell as cytotoxic agent, significantly increased amount of superoxide ($O_2^-$) and dramatically augmented activities of superoxide dismutase (SOD), specially MnSOD and glutathione peroxidase (GPx) were observed according to the concentration and incubation time. Whereas, in case of normal lymphocytes under the same condition, cytotoxicities were not apparent and the generation of superoxide ($O_2^-$) or the activity changes of SOD and GPx were insignificant. These results together indicate that the cytotoxic action of SD-994 against L1210 cell may be achieved via necrosis and/or apoptosis induced by reaction oxygen species which could not probably be completely abolished even by drastically increased antioxidant enzymes, SOD and GPx activities.

A rat chemokine, cytokine-induced neutrophil chemoattractant-1 (CINC-1) has chemotactic and activating properties to neutrophils. Rat kidney epithelioid NRK-52E cells contained 4 ng/ml of ClNC-1 as a basal level and their CINC-1 production was significantly increased by stimulation with lipopolysaccharide (LPS) of E. coli. Maximal induction of ClNC-1 was 58 ng/ml when 3 ${\mu}g/ml$ of LPS was treated to the NRK-52E cells. Inhibitory effects on CINC-1 induction in LPS-stimulated NRK-52E cells by extracts prepared from herbal medicines and wild plants in Korea were analyzed. At the final concentration of 100 ${\mu}g/ml$ , 9 species out of 304 species of herbal extracts exhibited more than 50% of inhibition on the CINC-1 induction. The active extracts prepared from Artemisia argyi, Lythrum salicaria, Machilus thunbergii, Magnolia sieboldii, Nelumbo nucifera, Prunus persica, Rubus coreanus, Sanguisorba officinalis, and Tripterygium regelii have been sequentially fractionated to obtain methylene chloride, ethyl acetate, butanol, and aqueous layers. Among solvent fractions of the active herbal extracts, methylene chloride fractions of Artemisia argyi and Magnolia sieboldii exhibited the highest inhibitory effects on CINC-1 induction in LPS-stimulated NRK-52E cells.
