Illumina를 이용한16S rRNA 기반 미생물생태분석에서 분변의 동결건조에 의한 인공적인 시퀀스 생성 감소효과 (Freeze-drying feces reduces illumina-derived artefacts on 16S rRNA-based microbial community analysis)

  • PCR 산물을 이용한 시퀀싱방법 중 Illumina 플랫폼으로 시퀀싱을 수행하면 100개 이상의 인위적인 시퀀스가 생겨나며, 그러한 인위적으로 형성되는 시퀀스에 의해 Operational taxonomic units를 기반으로 한 미생물생태 변화 및 네트워크 분석에 영향을 미친다. 이러한 문제점이 있음에도 불구하고 분변미생물생태를 분석하는데 Illumina에서 제공하고 있는 시퀀싱을 주된 방법으로 사용하고 있으며, 또한 그러한 시퀀스 기반의 분변미생물 생태분석 결과는 분변샘플상태(i.e., 분변 보관 기간, 분변양, 분변의 신선도)에 따라 상이하게 나타난다. 본 연구에서는 분변샘플의 동결건조가 시퀀스 데이터의 퀄리티를 향상시키는지 관해 조사하였으며, 이를 통해 분변샘플에 동결건조처리는 전체적인 미생물생태구조를 변화시키지는 않지만 인위적으로 형성되었을 가능성이 있는 시퀀스의 수를 감소시키는 것으로 확인되었다. 따라서, 분변으로부터 DNA를 추출하기 이전에 동결건조처리하는 방법을 Illumina 기반의 분변미생물생태분석에 사용하는 것을 권장한다.

  • The chemical Composition of Chinese red pine (Pinus massoniana) submerged in the Yellow Sea for more than 700 years has been examined. When compared to the recent wood, the marked chemical changes in the waterlogged wood is the higher amount of lignin with lesser amount of holocellulose and abnormally high ash content. In the heavily degraded samples, the degradation of cellulose is more severe than that of hemicellulose. However, hemicellulose is much more attacked than the cellulose at the initial stage of deterioration in the sea water. Chemical analysis suggests that the cellulolytic marine microorganisms, whether they are fungi or bacteria. can be regarded as the primary agents for the destruction of the archaeological woods submerged in the sea water.

  • The pulsatile nature of blood flow makes artefacts in 2D Fourier transform image. Spatial presaturation is known to be effective in eliminating flow artefacts when the spin echo acquisition is employed. However. this method requires additional RF pulse and spoiling gradient for presaturation. In this paper a new flow saturation technique which does not require additional saturation-RF and gradient is proposed. The proposed technique is equivalent to the existing saturation technique but the elimination of the flow component is achieved by a pair of tailored $90^{\circ}-180^{\circ}$ RF pulses in tile spin echo sequence. By use of two tailored RF pulses with opposite phase polarity, a linear phase gradient is generated for those moving materials and consequently all the spins of moving materials become dephased thereby no signal is observable. Computer simulations and experimental results obtained using both a phantom and a human volunteer with a 2.0 T whole body system are also presented.

독립성분 분석기법에 의한 심전도 신호의 왜곡 보정 (Suppressing Artefacts in the ECG by Independent Component Analysis)

  • In this study, Independent Component Analysis (ICA) algorithms are suggested to extract the original ECG part from the mixed signal contaminated with the unwanted frequency components and especially 60Hz power line disturbances. With this aim, we implement a novel method to suppress the baseline-wandering disturbances and power line artefacts contained in patch-electrodes sensory ECG data by separating the unmixed signal with finding the optimal weight W based on Kurtosis value. With applying brutal force and gradient ascent searching algorithm to find W, we can conclude that the unwanted frequency components especially in the ambulatory ECG data can be eliminated by Independent Component Analysis.

  • Radionuclide X-ray fluorescence analysis is a method, which has many advantages for analysing various historic artefacts, as it is relatively cheap, sensitive and non-destructive, and it allows measurements in-situ. However, this analysis has also certain limitations especially concerning sensitivity to chemical elements only, irrespective of the compounds or chemical forms in which these elements have been bonded. In addition, light elements emitting very soft X-rays cannot be measured, and in order to detect a wide range of elements, it is necessary to carry out repeated measurements with different radiation sources. Despite these limitations, valuable information can be obtained about the composition of historic materials and data about the origin and age of these artefacts can be derived. Analyses of wall paintings, ancient metal sculptures or other objects of art provide the basis for historic considerations documented in our results for some objects belonging to the Czech cultural heritage. The results are promising. Thus it is expected that our laboratory will expand its work into more fields of the fine and applied arts.

  • Analyzing adaptive possibilities of agents in multi-agents system, we have discovered new aspects of ambient intelligence in distributed modular systems using intelligent building blocks (I-BLOCKS) [1]. This paper describes early scientific researches related to technical design, applicable experiments and evaluation of adaptive processing and information interaction among I-BLOCKS allowing users to easily develop ambient intelligence applications. The processing technology presented in this paper is embedded inside each DUPLO1 brick by microprocessor as well as selected sensors and actuators in addition. Behaviors of an I-BLOCKS modular structure are defined by the internal processing functionality of each I-Blocks in such structure and communication capacities between I-BLOCKS. Users of the I-BLOCKS system can do 'programming by building' and thereby create specific functionalities of a modular structure of intelligent artefacts without the need to learn and use traditional programming language. From investigating different effects of modem artificial intelligence, I-BLOCKS we have developed might possibly contain potential possibilities for developing applications in ambient intelligence (AmI) environments. To illustrate these possibilities, the paper presents a range of different experimental scenarios in which I-BLOCKS have been used to set-up reconfigurable modular systems. The paper also reports briefly about earlier experiments of I-BLOCKS in different research fields, allowing users to construct AmI applications by a just defined concept of modular artefacts [3].

  • In recent years the importance of the preservation of cultural artefacts like ancient tombs has been widely accepted domestically and internationally as the quality of life improves. However not much technical attention has been paid for the facilities and systems to preserve those artefacts. Even the general understanding of the preservatory environment of the underground space as tombs is poor. As a part of the present study, the temperature and relative humidity inside a selected artefact, Shinkwan-ri tomb, have been monitored for a year round by the present author to improve the understanding of the indoor thermal environment, is pursued to provide a predictive tool of numerical modelling of Shinkwan-ri tomb the opened underground space thermal environment. In this study, predictive numerical modelling of Shinkwan-ri tomb using the Computational Fluid Dynamics, calculate the velocity and temperature distribution and offer basic data which are necessary for the best fitted design of tomb air-conditioning device.

Corrosion Prediction of Metallic Cultural Heritage Assets by EIS

  • Electrochemical Impedance Spectroscopy (EIS) was used to predict corrosion behaviour of metallic Cultural Heritage assets in two monitoring campaigns: 1) an iron bar chain exposed indoor from over 500 years in the Notre Dame Cathedral in Amiens (France); and 2) a large weathering steel sculpture exposed outdoor from tens of years in Ferrara (Italy). The EIS portable instrument employed was battery operated. In situ EIS measurements on the iron chain could be used to investigate the phenomena involved in the electrochemical interfaces among various corrosion products and assess and predict their corrosion behaviour in different areas of the Cathedral. Meanwhile, the sculpture of weathering steel, like most outdoor artefacts, showed rust layers of different chemical composition and colour depending on the orientation of metal plates. The EIS monitoring campaign was carried out on different areas of the artefact surface, allowing assessment of their protective effectiveness. Results of EIS measurements evidenced how employing a simple test that could be performed in situ without damaging the artefacts surface is possible to quickly gain knowledge of the conservation state of an artefact and highlight potential danger conditions.

조선시대 철비(鐵碑)의 보존처리와 탈염방법 비교 연구 (A Study of Conservation and Desalination Methods for an Iron Stele of the Joseon Dynasty)

  • 외부 환경에 노출된 철제문화재는 환경오염요인에 직접적으로 영향을 받아 부식에 취약하다. 특히 철제유물에 있어서 활성부식을 일으키는 염화이온($Cl^-$)은 반드시 제거되어야 하지만 옥외 철제문화재는 유물의 특성상 탈염의 대상에서 제외되고 있다. 그러므로 본 연구에서는 조선시대 철비를 대상으로 보존처리 및 탈염실험을 실시하였으며, 탈염은 흡습지를 이용한 탈염방법과 기존의 침적탈염방법을 함께 실시하여 그 효율성을 비교하였다. 탈염의 평가는 실체 금속 현미경, SEM-EDS, pH측정과 이온크로마토그래피(IC), XRD분석을 통하여 알아보았다. 실체 금속현미경으로 조사된 결과 부식은 층을 이루고 있으며 금속조직은 백주철로 확인된다. 또한 탈염 전 부식물에 대한 SEM-EDS분석결과 Fe와 O가 주로 검출되었으며 특히 Cl이 2.48wt%까지 측정되었다. 탈염용액에 대한 pH와 음이온을 분석한 결과에서는 흡습지를 이용한 탈염방법도 침적탈염방법과 유사한 탈염효과를 볼 수 있었다. 마지막으로 탈염 전 후 부식물에 대한 XRD분석 결과 탈염전에는 goethite, magnetite, lepidocrocite와 akaganeite가 검출되었으나 탈염 후에는 akaganeite가 검출되지 않아 흡습지를 이용한 탈염의 효과를 확인할 수 있었다. 그러므로 본 연구를 통하여 흡습지를 이용한 탈염방법에서도 기존의 침적탈염방법과 유사한 탈염효과를 확인할 수 있었다.
