Effect of Chitosan on Quality of Korean Wheat Bread and Imported Wheat Bread (키토산의 첨가가 우리밀빵과 수입밀빵의 품질에 미치는 영향)

  • This study is to find out the effects of the qualify of the breads made from Korean and imported wheat flour respectively after adding chitosan, by examining the dough making, the bread making properties, the Physicochemical properties of the breads. The low polymerized chitosan (M. W. : 37,000) was added respectively 1, 3 and 5% in the form of 10% lactic acid aqueous solution. The Korean wheat bread had 4% more water, 3% more sugar, 0.5% more salt, 3% more margarine and 1.5% more yeast per 100% to wheat flour added than the imported wheat bread. The bread volume for the two groups stayed the same. The water absorption in the chitosan added group was increased more greatly than in the control group, however, pH was reduced. In case of color among all wheat bread groups the ‘L’ value appeared to be higher in the imported wheat bread groups, and the ‘b’ value appeared to be higher in the chitosan added group.

A New Method for Sclerotial Isolation of Two Species of Sclerotium from Infested Soils

  • White rot on Allium crops recently had a high incidence with incrensed cultivating areas of tropical garlic types in Korea. Two types of Sclerotium have known as causal agents that produce different size and shapes of sclerotia in infested fields. Therefore, we developed a new method for isolation of sclerotia from infested field soils that can be used for ecological study of Sclerotium spp. and establishment of control strategy. Soil samples collected from heavily infested fields were evenly mixed and placed on a automatic sieve shaker connected with tap water, After 10 min of shaking, residues on 0.5 mm and 0.25 mm sieves were separately collected and suspended with 70% sugar solution, which method floats sclerotia in aqueous layer, Then, floated fraction was carefully separated and mixed with a same volume of 1% sodium hypochlorite solution to differentiate with organic materials. This method provides a direct count of sclerotia under a dissecting microscopy.

Rheological Properties of Fucoidans from Laminaria religiosa, Sporophylls of Undaria pinnatifida, Hizikia fusiforme and Sagassum fulvellum in Korea (국내산 다시마, 미역포자엽, 톳, 모자반 fucoidan의 리올리지 특성)

  • Rheological properties of partially purified fucoidans of Laminaria religiosa, sporophylls of Undaria pinnatifida, Hizikia fusiforme, and Sargassum fulvellum in Korea were investigated, The fucoidans produced aqueous solutions of low apparent viscosity with pseudoplastic flow behavior. The viscosity of $5\%$ solution of the fucoidan from sporophylls of U. pinnatifida was 2.2 mPa.s. The activaton energies of L. religiosa, sporophylls of U. pinnatifida, H. fusiforme and S. fulvellum were $8.269\times10^6,\;7.498\times10^6,\;7.141\times10^6,\;and\;6.677\times10^6\;J/Kg.mol$ at 1,500 1/s of shear rate, respectively. The viscosity of the fucoidan solution was increased by the addition of sugar, but that was not changed by addition of NaCl.

  • Free sugars were isolated from ginseng root and its products and analyzed by using high performance liquid chromatogrphy. To isolate free sugars from aqueous sample solution fat-soluble components, crude saponin and protein were removed from the solution by extracting with benzene, water-saturated butanol and 80% ethanol, respectively. Free sugars found from both ginseng root and its products were fructose, glucose, sucrose and maltose, and the only sugar detected from red ginseng root and its products was rhamnose. Major sugar detected from fresh ginseng and white ginseng roots was sucrose, while sucrose and maltose were major sugars of red ginseng root.

Analysis of ${\beta}-(1{\to}3)(1{\to}6)-Glucan$ Produced by Aureobasidium pullulans IMS-822 (Aureobasidium pullulans IMS-822가 생산하는 ${\beta}-(1{\to}3)(1{\to}6)-Glucan$의 특성 분석)

  • The exo-polysaccharide producing microorganism, Aureobasidium pullulans IMS-822, was isolated and identified from soil. The viscosity-average molecular weight of exo-polysaccharide was calculated as $8.9{\times}10^5$ by Mark-Houwink equation. The sugar component of exo-polysaccharide was determined as glucose by HPLC analysis. The IR spectra indicated that the exo-polysaccharide has an absorption peak at 890 $cm^{-1}$ for the ${\beta}-configuration$ of D-glucan. The $^{13}C$ NMR signal at ${\delta}$ 86.62 ppm arose from the substituted C-3 of glucose. The signal at ${\delta}$ 72.11 ppm was assigned to C-6 of branched ${\beta}-(1{\to}3)-D-glucosyl$ residues. Viscosity and Congo red reaction indicated that {\beta}-(1{\to}3)(1{\to}6)-glucan$ produced by A. pullulans IMS-822 has a highly ordered hydrogen-bond dependent conformation in aqueous solution, which collapses in strong alkaline solution.

A Study on Flavorous Taste Components in Kimchis -On Free Amino Acids- (김치의 맛 성분(成分)에 관(關)한 연구(硏究) -유리(遊離) 아미노산(酸)에 관(關)하여 -)

  • Free amino acids and total free sugar of kimchi were identified. Free amino acids of kimchis were extracted by 80% ethanol and isolated by ion exchange chromatography. Identification and quantitative determination of individual free amino acids were performed by amino acid autoanalyzer. Free sugar of kimchis was extracted by aqueous ethanol and isolated by ion exchange chromatography. Quantitative determination of it was perfermed by spectrophotometer. The results are summarized as follows: 1. Lysine, histidine, arginine, tryptophan, aspartic acid, threonine, glutamic acid, proline, glycine, alanine, cysteine, valine, methionine, leucine, isoleucine, tyrosine and phenylalanine were found in all kimchis. 2. The change of free amino acid composition during fermentation of kimchis was not observed, but the amount of total free amino acids of fermented kimchi decreased as compared with those of raw kimchi. 3. In kimchi containing 10 ml of fermented anchovy solution/100 g of chinese cabbage, the amount of total free amino acids was more than that of fermented salt kimchi and the characteristic flavor of it was attributed to such amino acids as lysine, aspartic acid, glutamic acid, valine, methionine, isoleucine and leucine. 4. Large amount of free sugar in raw salt kimchi decreased during fermentation, but, after fermentation, significant difference of free sugar content between salt kimchi and kimchi containing fermented anchovy solution was not observed.

Effect of Fluid Viscosity on Centrifugal Pump Performance (유체의 점성이 원심펌프 성능에 미치는 영향)

  • The characteristics of centrifugal pump performance according to fluid viscosity change were studied experimentally. A small volute pump with low specific speed was tested by changing the viscosity of an aqueous solution of sugar and glycerin, which is considered a Newtonian fluid. After finishing the test, the total head, shaft horsepower, and pump efficiency were compared with those of a water pump. The results are summarized as follows: (1) when the fluid viscosity is increased, the shut-off head shows very little change but the total head decreases gradually as the flow increases, and this makes the H-Q curve leaning rapidly, and (2) when the fluid viscosity is increased, the shaft horsepower shows very little change at the shutoff condition; however, the shaft horsepower increases more rapidly with an increase in the flow and viscosity.

Preparation and Physicochemical Properties of Soluble Dietary Fiber Extracts from Soymilk Residue at High Temperature (두유박 수용성 식이섬유의 고온 추출물 제조와 이화학적 특성구명)

  • Thermal treatment of soymilk residue was carried out at 140, 150, and $160^{\circ}C$ for possible use as a raw material for dietary fiber, and some physicochemical properties of the extracts were investigated. Soluble dietary fiber(SDF) content of the extracts prepared under optimal conditions was more than 30% suggesting the conversion of insoluble dietary fiber to SDF. The main sugar components of the extracts were glucose, galactose, and arabinose. Analysis of the moelcular weight distribution by high performance size exclusion chromatography revealed that the proportion of high molecular weight fraction decreased and that of middle-sized polymer increased as the extraction temperature increased. The viscosity of aqueous solution of the extracts decreased with an increase in extraction temperature, but showed no trend as pH changed. The solubility increased with extraction temperature showing the highest at $160^{\circ}C$. The extract at $140^{\circ}C$ had the biggest calcium-binding capacity, which correlated with the changes in viscosity.

  • Biorefineries, in which renewable resources are utilized, are an eco-friendly alternative based on biomass feedstocks. Alginic acid, a major component of brown algae, which is a type of marine biomass, is widely used in various industries and can be converted into value-added chemicals such as sugars, sugar alcohols, furans, and organic acids via catalytic hydrothermal decomposition under certain conditions. In this study, ruthenium-supported activated carbon and magnesium oxide were mixed and applied to the depolymerization of alginic acid in a batch reactor. The addition of magnesium oxide as a basic promoter had a strong influence on product distribution. In this heterogeneous catalytic system, the separation and purification processes are also simplified. After the reaction, low molecular weight alcohols and organic acids with 5 or fewer carbons were produced. Specifically, under the optimal reaction conditions of 30 mL of 1 wt% alginic acid aqueous solution, 100 mg of ruthenium-supported activated carbon, 100 mg of magnesium oxide, 210 ℃ of reaction temperature, and 1 h of reaction time, total carbon yields of 29.8% for alcohols and 43.8% for a liquid product were obtained. Hence, it is suggested that this catalytic system results in the enhanced hydrogenolysis of alginic acid to value-added chemicals.

Quality Characteristics of Smoked Dombaeki (Shark Meat) (돔배기(상어육)의 훈연처리에 따른 품질 특성에 관한 연구)

  • We explored the effects of curing and smoking conditions on the shelf life of Dombaeki (shark meat). Dombaeki cured for 12 h in an aqueous solution containing (per 100 ml) salt 5.6 g, sugar 14 g, and garlic powder 0.6 g, showed the best sensory quality among various samples cured for 0, 3, 6, 9, 12 or 24 hours. The optimum conditions for preparation of smoked Dombaeki (SD) were drying at $60^{\circ}C$ for 30 min, followed by cooking at $80^{\circ}C$ for 30 min and smoking at $65^{\circ}C$ for 40 min, as judged by sensory evaluation of taste, color, flavor, texture, and overall acceptability. The volatile basic nitrogen content of air- or vacuum-packed unsmoked Dombaeki (NSD) was above 20 mg% after storage for either 12 days or 5 weeks. However, the nitrogen contents of air- and vacuum-packed SD were less than 20 mg% after either 21 days or 10 weeks of storage at $10^{\circ}C$. The DPPH free radical-scavenging ability of SD (73.9%) was significantly higher than that of unsmoked meat (4.54%). The total polyphenol content of SD (745.6 g/g) was about 4-fold greater than that of unsmoked meat (179.5 g/g).The viable bacterial count of air- or vacuum-packed unsmoked meat was over $10^6\;CFU/g$ after storage for either 12 days or 5 weeks. However, air- or vacuum-packed SD had counts under $10^4\;CFU/g$ at all storage times tested. Changes in coliform bacterial levels paralleled those of total viable cells. The sensory quality (taste, color, flavor, appearance, texture, and overall acceptability) of SD was significantly better than that of NSD.
