소형 순환여과양식시스템에서 어린 해삼(Apostichopus japonicus) 사육 (A Laboratory-scale Recirculating Aquaculture System for Sea Cucumber Apostichopus japonicus)

  • The sea cucumber Apostichopus japonicus is a commercially valuable aquaculture species in Korea. Aquaculture species require specific nursery culture conditions to increase survival and growth rates. Sea cucumbers hibernate during the high temperatures of summer and during the low temperatures of winter, and suboptimal temperature conditions decrease sea cucumber growth and survival rates. The natural South Korean environment is very unfavorable for culturing sea cucumber; therefore, developing a recirculating aquaculture system (RAS) capable of breeding and growing sea cucumber year-round is necessary. The aim of this study was to investigate growth performance of juvenile sea cucumber in a RAS. Growth and survival rates of juvenile sea cucumber were high during our 24-week experiment. Sea cucumber survival rates were 87.8-93.3%, and specific growth rates were 0.4689-0.7846.

Identification of bacteria isolated from rockworm viscera and application of isolated bacteria to shrimp aquaculture wastewater treatment

  • Large amounts of waste and wastewater from aquaculture have negatively impacted ecosystems. Among them, shrimp aquaculture wastewater contains large amounts of nitrogen contaminants derived from feed residues in an aerobic environment. This study isolated candidate strains from adult rockworms to treat shrimp aquaculture wastewater (SAW) in an aerobic environment. Among 87 strains isolated, 25 grew well at the same temperature as the shrimp aquaculture with excellent polymer degradation ability (>0.5 cm clear zone). Six isolates (strains AL1, AL4, AL5, AL6, LA10, and PR15) were finally selected after combining strains with excellent polymer degradation ability without antagonism. 16S rRNA sequencing analysis revealed that strains AL1, AL4, AL5, AL6, LA10, and PR15 were closely related to Bacillus paramycoides, Bacillus pumilus, Stenotrophomonas rhizophila, Bacillus paranthracis, Bacillus paranthracis, and Micrococcus luteus, respectively. When these six isolates were applied to SAW, they reached a maximum cell viability of 2.06×105 CFU mL-1. Their chemical oxygen demand (CODCr) and total nitrogen(TN) removal rates for 12h were 51.0% and 44.6%, respectively, when the CODCr/TN ratio was approximately 10.0. Considering these removal rates achieved in this study under batch conditions, these six isolates could be used for aerobic denitrification. Consequently, these six isolates from rockworms are good candidates that can be applied to the field of aquaculture wastewater treatment.

양식장 배출수중의 항생제 내성균 분포 및 전자빔 살균처리 (Distribution of Antibiotic Resistant Microbes in Aquaculture Effluent and Disinfection by Electron Beam Irradiation)

  • 육상양식장에서 항생제에 대한 내성을 가진 균을 분리하여 다양한 항생제에 대한 내성특성을 확인하고, 전자빔의 조사에 따른 내성균의 항생제 내성 특성변화와 살균효능에 관한 연구를 수행하였다. 메기, 송어, 뱀장어, 미꾸라지양식장의 배출수에서 분리된 균주 중 Aeromonas sp., Citrobacter sp., Bacillus sp., Marinobacter sp., Pantoea sp., Pseudomonas sp., Enterobacter sp.가 다제내성균으로 확인되었다. 양식장 배출수에서 분리한 균주 중 한가지의 항생제 내성을 가지고 있는 균은 41.7%이며, 2가지 이상의 항생제 내성을 가지고 있는 균은 58.3%로 나타났다. Quantitative Structure Analysis Relationship (QSAR) model에 의한 평가는 실험값과 매우 유사하여 독성 평가의 간접적인 지표로 이용될 수 있음을 알 수 있었다. 항생제 내성균에 대해 전자빔을 조사한 결과 1 kGy 이내의 선량에서 살균효율이 99.9%로 강한 살균력을 확인할 수 있었다.

저염분 노출에 따른 감성돔(Acanthopagrus schlegelii) 아가미의 Na+/K+-ATPase 활성 및 발현 (Gill Na+/K+-ATPase Activity and Expression in Black Sea Bream Acanthopagrus schlegelii Exposed to a Hyposaline Environment)

  • We investigated the branchial osmoregulatory response of black sea bream Acanthopagrus schlegelii to short-term (3-48 h) exposure to a hyposaline environment (5 psu). Gill $Na^+/K^+$-ATPase (NKA) activity was decreased after 3 h in fish transferred to 5 psu compared to salt water-acclimated (control) fish, but the level of activity returned to that observed in the control fish at 6 h after transfer. NKA activity increased significantly at 24 h after transfer, but it returned to the level observed in the control fish at 48 h after transfer. Immunohistochemical staining revealed that gill NKA was localized to chloride cells. The number of chloride cells tended to change in parallel with NKA activity. Substantial decreases in plasma $Na^+$, $Cl^-$, and osmolality were observed after 12 h of exposure to 5 psu; however, these parameters began to recover to the values detected in the controls at 24 h after transfer. In conclusion, our results suggest that black sea bream are able to adjust their osmoregulatory mechanisms to shift from hypo- to hyperosmoregulation within 6 h of exposure to a hypoosmotic environment.

Survival and growth of the red tide organism Cochlodinium polykrikoides after the addition of yellow loess

  • At least 15% of the C polykrikoides cells that precipitated to the bottom layer either by the addition of loess or no addition survived for 1 week at all growth phases, rather than disappearing immediately after precipitating. However, no live cells were observed after 20 days, regardless of phase or loess addition. In the exponential phase, the number of C polykrikoides cells increased to >2886 cells/ml after loess was added. However, in the stationary phase, the number of cells did not increase until 18 days. In the exponential phase, those C polykrikoides that survived precipitation caused by scattering loess on cultures did not appear to have the ability to cause red tides again because of the short red tide periods in the field, long lag time after loess addition, and low survival rate after loess addition.

  • 디지털양식은 경험과 노동력, 자연환경 의존적인 전통 양식산업이 미래 지능형 스마트양식으로 전환하기 위한 ICT 기자재 기반의 양식산업의 디지털화와 디지털 양식데이터의 분석 및 활용을 통한 양식 재현성 향상 및 생산과정의 효율성 향상을 통한 어가의 수용성을 높이기 위해 추진되고 있다. 유럽의 선진 수산국은 양식장에 정보통신기술을 접목한 디지털화를 통한 규모화와 첨단화로 양식 기술 뿐만 아니라 양식기자재산업의 비약적인 성장을 이루고 있는 반면, 국내에는 다품종·소량생산 구조 및 어가수용성 미확보에 따른 양식생산정보의 수집조차 한계에 부딪치고 있다. 이에 본 연구에서는 스마트양식 관련 동향과 기술 요소를 분석하여 향후 지능형 스마트양식으로 전환하기 위한 한국형 디지털양식의 발전 방향을 제시하고자 한다.

일본 어류 양식업의 발전과정과 산지교체에 관한 연구 : 참돔양식업을 사례로 (A study on Development Process of Fish Aquaculture in Japan - Case by Seabream Aquaculture -)

  • When we think of fundamental problems of the aquaculture industry, there are several strict conditions, and consequently the aquaculture industry is forced to change. Fish aquaculture has a structural supply surplus in production, aggravation of fishing grounds, stagnant low price due to recent recession, and drastic change of distribution circumstances. It is requested for us to initiate discussion on such issue as “how fish aquaculture establishes its status in the coastal fishery\ulcorner, will fish aquaculture grow in the future\ulcorner, and if so “how it will be restructured\ulcorner” The above issues can be observed in the mariculture of yellow tail, sea scallop and eel. But there have not been studied concerning seabream even though the production is over 30% of the total production of fish aquaculture in resent and it occupied an important status in the fish aquaculture. The objectives of this study is to forecast the future movement of sea bream aquaculture. The first goal of the study is to contribute to managerial and economic studies on the aquaculture industry. The second goal is to identify the factors influencing the competition between production areas and to identify the mechanisms involved. This study will examine the competitive power in individual producing area, its behavior, and its compulsory factors based on case study. Producing areas will be categorized according to following parameters : distance to market and availability of transportation, natural environment, the time of formation of producing areas (leaderㆍfollower), major production items, scale of business and producing areas, degree of organization in production and sales. As a factor in shaping the production area of sea bream aquaculture, natural conditions especially the water temperature is very important. Sea bream shows more active feeding and faster growth in areas located where the water temperature does not go below 13∼14$^{\circ}C$ during the winter. Also fish aquaculture is constrained by the transporting distance. Aquacultured yellowtail is a mass-produced and a mass-distributed item. It is sold a unit of cage and transported by ship. On the other hand, sea bream is sold in small amount in markets and transported by truck; so, the transportation cost is higher than yellow tail. Aquacultured sea bream has different product characteristics due to transport distance. We need to study live fish and fresh fish markets separately. Live fish was the original product form of aquacultured sea bream. Transportation of live fish has more constraints than the transportation of fresh fish. Death rate and distance are highly correlated. In addition, loading capacity of live fish is less than fresh fish. In the case of a 10 ton truck, live fish can only be loaded up to 1.5 tons. But, fresh fish which can be placed in a box can be loaded up to 5 to 6 tons. Because of this characteristics, live fish requires closer location to consumption area than fresh fish. In the consumption markets, the size of fresh fish is mainly 0.8 to 2kg.Live fish usually goes through auction, and quality is graded. Main purchaser comes from many small-sized restaurants, so a relatively small farmer and distributer can sell it. Aquacultured sea bream has been transacted as a fresh fish in GMS ,since 1993 when the price plummeted. Economies of scale works in case of fresh fish. The characteristics of fresh fish is as follows : As a large scale demander, General Merchandise Stores are the main purchasers of sea bream and the size of the fish is around 1.3kg. It mainly goes through negotiation. Aquacultured sea bream has been established as a representative food in General Merchandise Stores. GMS require stable and mass supply, consistent size, and low price. And Distribution of fresh fish is undertook by the large scale distributers, which can satisfy requirements of GMS. The market share in Tokyo Central Wholesale Market shows Mie Pref. is dominating in live fish. And Ehime Pref. is dominating in fresh fish. Ehime Pref. showed remarkable growth in 1990s. At present, the dealings of live fish is decreasing. However, the dealings of fresh fish is increasing in Tokyo Central Wholesale Market. The price of live fish is decreasing more than one of fresh fish. Even though Ehime Pref. has an ideal natural environment for sea bream aquaculture, its entry into sea bream aquaculture was late, because it was located at a further distance to consumers than the competing producing areas. However, Ehime Pref. became the number one producing areas through the sales of fresh fish in the 1990s. The production volume is almost 3 times the production volume of Mie Pref. which is the number two production area. More conversion from yellow tail aquaculture to sea bream aquaculture is taking place in Ehime Pref., because Kagosima Pref. has a better natural environment for yellow tail aquaculture. Transportation is worse than Mie Pref., but this region as a far-flung producing area makes up by increasing the business scale. Ehime Pref. increases the market share for fresh fish by creating demand from GMS. Ehime Pref. has developed market strategies such as a quick return at a small profit, a stable and mass supply and standardization in size. Ehime Pref. increases the market power by the capital of a large scale commission agent. Secondly Mie Pref. is close to markets and composed of small scale farmers. Mie Pref. switched to sea bream aquaculture early, because of the price decrease in aquacultured yellou tail and natural environmental problems. Mie Pref. had not changed until 1993 when the price of the sea bream plummeted. Because it had better natural environment and transportation. Mie Pref. has a suitable water temperature range required for sea bream aquaculture. However, the price of live sea bream continued to decline due to excessive production and economic recession. As a consequence, small scale farmers are faced with a market price below the average production cost in 1993. In such kind of situation, the small-sized and inefficient manager in Mie Pref. was obliged to withdraw from sea bream aquaculture. Kumamoto Pref. is located further from market sites and has an unsuitable nature environmental condition required for sea bream aquaculture. Although Kumamoto Pref. is trying to convert to the puffer fish aquaculture which requires different rearing techniques, aquaculture technique for puffer fish is not established yet.

양식어장보호를 위한 어장탐지 시스템 개발에 관한 연구 (A Study on the Fishery Detection System for Protection of an Aquaculture Farm)

  • 본 논문에서는 양식어장을 보호하기 위한 어장 탐지 시스템의 개발에 대해 논의한다. 어장탐지시스템(FDS)은 어장에 침입하는 도적선박을 실시간으로 식별하고 어장의 위치변동 둥을 파악하는 기능을 갖는다. 본 연구에서는 어장에 접근하는 물체탐지 및 적아식별을 위해 저가이면서 대역의 레이더 응답기능을 갖는 어장작업선용 AIS(F-AIS)를 개발하여 소형어선 등의 식별기능에 이용하고자 한다.

무인 어장 도적 감시 시스템 구현에 관한 연구 (Implementation of Unmanned Aquaculture Security System)

  • 본 논문에서는 어장에 침입하는 도적을 감시하기위한 어장보호 시스템의 구현절차와 구현결과를 기술하였다. 이 시스템은 사용자의 요구에 따라 저가부터 고가 및 단순 기능부터 복합 기능 등 다양한 형태로 구성할 수 있도록 설계하였다. 육상으로부터 0.5 마일 이내에 총 50개(가로 10열, 세로 5열)의 케이지가 밀집된 전라남도 진도군 소재 전복 양식장에서 구축한 시스템을 현장 실험하였다. 그 결과, 구축한 시스템이 양식장 전체는 물론 단위 케이지까지 세밀하게 감시할 수 있음을 확인하였다.

사육환경에 따른 이매패류 (Crassostrea gigas, Mytilus galloprovincialis)의 외부형질 성장과 Heat Shock Protein 70 유전자 발현 (Expression of the Heat Shock Protein 70 Gene and External Developmental Traits of Two Bivalvia Species, Crassostrea gigas and Mytilus galloprovincialis, under Aquaculture Environments)

  • 연안의 다양한 환경변화는 서식 생물에 영향을 미치고, 양식장의 생산량 감소와 연결되고 있는 추세이다. 본 연구에서는 가막만의 대표적인 양식종인 패류 C. gigas와 M. galloprovincialis의 서식환경에 따른 스트레스 정도를 파악하고자 하였다. 이를 위해, 각 종의 체중량, 각장과 각고, 양식장 사육기간을 조사하고, 각 종의 계통학적 HSP 70 sequence를 비교한 후, 각 종의 HSP 70 유전자 발현을 분석하였다. 그 결과, C. gigas의 체중량, 각장과 각고는 C2 양식장이 높게 나타났으나, 양식장 환경 사육기간과 HSP 70 유전자 발현은 C3 양식장이 가장 높았다. M. galloprovincialis는 M1 양식장의 체중량이 높게 나타났으며 각장과 각고, 사육기간은 M2와 유사하였으나, HSP 70 유전자 발현은 M2 양식장이 통계적으로 유의한 수준으로 높게 나타났다. 그리고 C. gigas와 M. galloprovincialis의 HSP 70 sequence 분석을 통해서 다른 해양 종들과 높은 유사성이 있음을 확인하였다. 이 결과는 서식환경에 따라 생물의 외부적 형질뿐만 아니라 내부적 스트레스를 HSP 70 유전자 발현을 통하여 파악할 수 있으며 HSP 70은 외부환경 스트레스를 평가하는 지표 유전자로서 활용할 수 있을 것이다.
