In the present investigation, bacterial isolates from infected apple trees causing apple canker during winter were studied in the northern Gyeongbuk Province, Korea. The pathogen was identified as Pseudomonas syringae pv. syringae (Pss) through various physiological and biochemical characterization assays such as BIOLOG, gas chromatography of fatty acid methyl esters, and 16S rRNA. Bioassays for the production of phytotoxins were positive for syringopeptin and syringomycin against Bacillus megaterium and Geotrichum candidum, respectively. The polymerase chain reaction (PCR) method enabled the detection of toxin-producing genes, syrB1, and sypB in Pss. The differentiation of strains was performed using LOPAT and GATTa tests. Pss further exhibited ice nucleation activity (INA) at a temperature of -0.7℃, indicating an INA+ bacterium. The ice-nucleating temperature was -4.7℃ for a non-treated control (sterilized distilled water), whereas it was -9.6℃ for an INA- bacterium Escherichia coli TOP10. These methods detected pathogenic strains from apple orchards. Pss might exist in an apple tree during ice injury, and it secretes a toxin that makes leaves yellow and cause canker symptoms. Until now, Korea has not developed antibiotics targeting Pss. Therefore, it is necessary to develop effective disease control to combat Pss in apple orchards. Pathogenicity test on apple leaves and stems showed canker symptoms. The pathogenic bacterium was re-isolated from symptomatic plant tissue and confirmed as original isolates by 16S rRNA. Repetitive element sequence-based PCR and enterobacterial repetitive intergenic consensus PCR primers revealed different genetic profiles within P. syringae pathovars. High antibiotic susceptibility results showed the misreading of mRNA caused by streptomycin and oxytetracycline.
Our study was carried out to determine the control efficacy of sodium hypochlorite (NaOCl) for violet root rot caused by Helicobasidium mompa in apple. The experiment was conducted in the farm located at Chungbuk province in South Korea from 2014 to 2016. When infected apple trees were treated at least two or three times with 31.25 and 62.5 ml/l available chlorine content in NaOCl, it greatly increased the rooting of rootstock, and restored the tree crown density by 44.4-60.5%. In addition, the number of commercial fruit setting was increased by 54.3-64.5%, and the total starch content in shoots was significantly higher than other non-treated apple trees. However, the untreated disease control and thiophanate-methyl WP treated trees showed the symptom of dieback. Therefore, our results indicate that the drenching treatment of NaOCl with 31.25-62.5 ml/l available chlorine content more than two times from late fall to early spring could effectively control the violet root rot and recover tree vigor up to 60%.
K-Maryblyt has been developed for the effective control of secondary fire blight infections on blossoms and the elimination of primary inoculum sources from cankers and newly emerged shoots early in the season for both apple and pear trees. This model facilitates the precise determination of the blossom infection timing and identification of primary inoculum sources, akin to Maryblyt, predicting flower infections and the appearance of symptoms on various plant parts, including cankers, blossoms, and shoots. Nevertheless, K-Maryblyt has undergone significant improvements: Integration of Phenology Models for both apple and pear trees, Adoption of observed or predicted hourly temperatures for Epiphytic Infection Potential (EIP) calculation, incorporation of adjusted equations resulting in reduced mean error with 10.08 degree-hours (DH) for apple and 9.28 DH for pear, introduction of a relative humidity variable for pear EIP calculation, and adaptation of modified degree-day calculation methods for expected symptoms. Since the transition to a model-based control policy in 2022, the system has disseminated 158,440 messages related to blossom control and symptom prediction to farmers and professional managers in its inaugural year. Furthermore, the system has been refined to include control messages that account for the mechanism of action of pesticides distributed to farmers in specific counties, considering flower opening conditions and weather suitability for spraying. Operating as a pivotal module within the Fire Blight Forecasting Information System (FBcastS), K-Maryblyt plays a crucial role in providing essential fire blight information to farmers, professional managers, and policymakers.

The effect of Acibenzolar-S-methyl (ASM) and Rahnella aquatilis Ra39 against apple fire blight disease caused by Erwinia amylovora were tested as a possible alternative to streptomycin. In vitro studies, no inhibition effect against the pathogen was found when ASM was tested. Under greenhouse conditions, application of R. aquatilis Ra39 with the highly susceptible M26 rootstock resulted in a marked disease suppression. Application of ASM and strain Ra39 caused a high decrease of the disease, 82% and 58% respectively; this was correlated with a reduction of the growth of the pathogen within host plants up to 64% and 49.5% respectively. Further studies in the field under artificial infection condition during full bloom revealed that application of ASM and R. aquatilis Ra39 with Gala variety resulted in a control effect up to 21 and 29% respectively. In physiological studies, enhanced activities of PR-proteins (chitinase and $\beta$-1, 3-glucanase) were detected, which are well known as biochemical markers for systemic acquired resistance. Application of ASM to apple shoots caused the highest chitinase activity followed by strain Ra39. The enzyme activity was increased after 2, 4 and 6 days from application. In addition, ASM-treatment caused the higher $\beta$-1, 3-glucanase activity than strain Ra39. Maximum enzyme activity was recorded after 6 days from application and then decreased after 8 and 10 days from application.
This study was carried out to assess the antifungal potential of R. oligosporus and its ethyl acetate (EtOAc) extract against the fungal pathogens causing anthracnose disease in apple fruits using disc diffusion, antagonistic effect and morphological abnormalities in fungal mycelia. The percentage of inhibition of antifungal effect of the ethyl acetate extract (5 ${\mu}l$$disc^{-1}$) of the R. oligosporus against C. acutatum KACC 40848, C. gloeosporioides KACC 40897, C. higginsianum KACC 40806, C. orbiculare KACC 40808, C. coccodes KACC 40008, C. musae KACC 40947, C. boninense KACC 40893, C. liliacearum KACC 40981, C. caudatum KACC 41028 and Colletotrichum sp. KACC 40811 was found to be 44.4, 35.5, 40, 31.1, 33.3, 37.7, 40, 51.1, 28.8 and 28.8%, respectively. Also the fungus R. oligosporus showed potential antagonistic effect of antifungal activity against the tested pathogens of Colletotrichum spp. Further, R. oligosporus had a potential detrimental effect on the morphology of the tested fungi of Colletotrichum spp. such as wrinkle abnormalities, abnormal cell formation, lysis of mycelium, empty cell formation, distorted cell formation and breakage of the mycelium. These findings strongly support the role of R. oligosporus to serve as a potential antifungal agent to control plant pathogenic fungi causing anthracnose disease in apple fruits.

In order to develop an effective spray program for control of apple white rot with reduced use of fungicides, the control efficacy of several fungicides that has been intensively used for control of the disease was assessed. They were sprayed on the same tree with 15 day interval from late May to early August. Just prior to and after each spray, 100 fruits were bagged with two layered fruit bag to limit the chemical application in only one time, and the disease incidence and latent infection frequency on the bagged apples were examined. Some fungicides such as folpet, iminoctadine-triacetate and azoxystrobin showed a high post-infectional activity even though the former two are non- systemic. Folpet suppressed symptom development, iminoctadine-triacetate reduced infection frequency and azoxystrobin acted in both ways. When those fungicides were !! adopted in a spray program, once in a cropping season, their post-infectional activity became much greater. This activity shown by the non-systemic fungicides was supposed to be derived from the peculiar infection process of the white rot fungus of which the pathogen is usually remain latent in the corked cells of lenticel until the apple reach mature stage.

The strains of Colletotrichum gloeosporioides, C. coccodes, C. acutatum isolated from black raspberry were pathogenic to apple and strawberry after dropping inoculation, but showed weak pathogenicity in hot-pepper and tomato. The anthracnose pathogens of C. gloeosporioides, C. orbiculare, C. acutatum isolated from apple, hot-pepper and pumpkin showed pathogenicity in black raspberry. Moreover, the anthracnose pathogens isolated from apple caused disease symptoms in non-wounded inoculation.
Soft rot in apple caused by Rhizopus oryzae was found for the first time in Korea. A detailed description of the specimen is given along with its internal transcribed spacer rDNA sequence. The fungus was identified as Rhizopus oryzae based on the mycological characteristics, molecular data, and pathogenicity testing.
The 4 viruses, the Apple chlorotic leaf spot virus (ACLSV), Apple stem pitting virus (ASPV), Apple stem grooving virus (ASGV), and Apple mosaic virus (ApMV) and 1 viroid, Apple scar skin viroid (ASSVd) are known major viral pathogens of apple trees in Korea. Infection degree of the 5 viral pathogens in the commercial nursery trees of major apple cultivars, 'Hongro', 'Fuji' and bud mutation of 'Fuji' was investigated. Infection ratio of the ACLSV, ASPV and ASGV for scion of an apple cultivar 'Hongro' were 100%, 81.3% and 100%, respectively. On the other hand, no infection for either ApMV and ASSVd detected. For the root stock of the cultivar, infection ratio of ACLSV, ASPV and ASGV showed 87.5%, 81.3% and 100% as well as ApMV and ASSVd were 12.5% and 6.3%, respectively. From the scion of apple cultivars 'Fuji' and bud mutation of 'Fuji', infection ratio of ACLSV, ASPV and ASGV showed 86.7%, 86.7% and 100%, respectively. Whereas, no infection for either ApMV or ASSVd detected. From the root stock of the cultivars, infection ratio of ACLSV, ASPV and ASGV showed 86.7%, 93.3% and 93.3% as well as ApMV and ASSVd were 12.5% and 6.3%, respectively. Result of our study indicates that most of commercial nursery apple trees were supplied with multiple infections by apple viruses causing potential losses for apple growers and, henceforth, agricultural policy for supply of the virus-free trees should be employed as soon as possible.
This study was carried out to select effective fungicides against white and violet root rot caused by Rosellinia necatrix and Helicobasidium mompa with nine fungicides including thiophanate-methyl from 1993 to 1994. Through laboratory, greenhouse and field trials on inhibitory effect of mycelial growth and disease incidence against the two fungal pathogens, 5 fungicides have been selected finally. Thiopanate-methyl, benomyl, iminoctadine-triacetate and isoprothiolane were proven to have high control effect against R. necatrix. In addition to thiopanate-methyl and benomyl, tolclofos-methyl has been selected for effective control of H. mompa, since it showed prominent control effect in field trial than in laboratory or green house test.
