상수 원수에 따른 용존 유기물의 특성 평가 (Characterization of Dissolved Organics Based on Their Origins)

  • This study was carried out to evaluate the characteristics of dissolved organics based on their origins, which were divided into two categories. The first group consisted of river, lake and secondary sewage treatment effluent, which were chosen as representative of their origins. The second group were artificial samples which were made of AHA(Aldrich humic acids) and WHA(Wako humic acids). Physicochemical characteristics, biological degradability and THMEP(trihalomethane formation potential) of the samples were analysed based on the AMWD(apparent molecular weight distribution). Large portion of dissolved organic carbon(DOC) in the river and lake samples was comprised of LMW(low molecular weight), which that of AHA and WHA was HMW(high molecular weight). The DOC of the lake was evenly distributed in the all range of molecular weight. The river, lake and secondary treated effluent have lower ultraviolet(UV) absorbance at 254nm, and have a higher amount of humic acids. Higher absorbance of humic acids means that aliphatic bond and benzenoid type components that absorb UV light were contained in these kind of humic acids. It was expected that lake sample was the most biodegradable in the different samples investigated, and in order of secondary sewage treatment effluent, river, WHA and AHA based on the result of determination of specific ultraviolet absorbance(SUVA). Biodegradability showed similar result except for AHA, while dissolved organics in the range of LMW decreased during the biodegradability test, and on the contrary those of HMW increased. Production of the SMPs(soluble micobial products) was observed during humicfication of dissolved organics and the SMPs were higher production of the SMPs. THM formation was high in the samples containing HMW and similar tendency was shown in the THMEP(trihalomethane formation potential), except for WHA.

Effects of Amylose Contents and Degree of Gelatinization of Rice Flour on In Vitro Starch Digestibility, Physical Characteristics, and Morphological Properties

  • The relationship of in vitro starch digestibility and gel strength was investigated at various concentrations (10-30%) of rice cultivars with different amylose contents (27.9, 17.9, and 5.2%). As the rice flour concentration increased, predicted glycemic index decreased, but gel strength increased regardless of amylose contents. Gel strength correlated strongly with amylose content, whereas in vitro starch digestibility was more highly affected by rice flour concentration than by amylose contents. Moreover, the impact of degree of gelatinization on in vitro starch digestibility of high amylose rice was also examined in terms of structural features and rheological properties. The digestion rate of fully gelatinized flour was 1.7 times higher than that of native flour, while the disrupted structure with a different gelatinization degree during starch digestion was visually demonstrated through the X-ray diffraction and molecular distribution analysis. The rice flour changed from an A-type to a V-type pattern and showed difference in crystalline melting. The low molecular weight distribution increased with increasing degree of gelatinization during starch digestion. The apparent viscosity also increased with degree of gelatinization. These results demonstrated that the starch digestibility of rice was more affected by concentration than by amylose content, as well as by the degree of gelatinization due to structural difference.

실크 피브로인의 대규모 투석 공정의 단위 모듈의 개발 (Development of a Unit Module for the Process of Large Scale Dialysis for Silk fibroin)

  • For the development of a large-scale dialysis process for silk fibroin solution, a batch and continuous dialysis chamber was designed and built, which are to be used as a unit module for the process. A series of dialysis experiments were carried out to estimate the kinetic parameters for dialysis using the solutions of salt-solubilized silk fibroin. The solution was dialyzed by distilled water either batch-wise or continuously. Dialysis kinetics was monitored by measuring refractometery at high salt concentration and conductivity at lower salt concentration. The apparent dialysis coefficients were $5.4g/m^2-hr$ and $1.8g/m^2-hr$, for well mixed batch and continuous dialysis, respectively. The alteration of molecular weight distribution of dialyzed silk fibroin solution was monitored.

  • Serratia marcescens가 세포외로 분비하는 nuclease의 유전자가 발현된 Escherichia coli JM107을 배양하여 다량의 효소를 정제하였다. Matrex green gel과 heparin agarose gel column chromatography법으로 약 50배 정제한 효소는 분자량이 29KDa였으며, 전기영동 상에서 단일 띠를 보였다. 이 단백질을 이용하여 polyclonal antibody를 만들고, 면역조직화학법으로 세포내의 분포를 조사하였다. Nuclease는 주로 세포막에 존재하였고, 이를 토대로 효소가 세포질에서 합성된 후 세포막으로 빠르게 이동함을 알 수 있었다. 이 결과는 세포의 막분획에서 효소의 활성의 대부분이 회수되며, 면역블럿 방법으로 효소의 대부분이 세포막에서 검출된다는 결과와 일치하였다.

효소처리에 의한 분리대두 단백질의 부분 가수분해에 관한 연구 (Studies on the Enzymatic Partial Hydrolysis of Soybean Protein Isolates)

  • 탈지 대두박으로 부터 추출 분리하여 얻어진 분리대두 단백질에 효소(pepsin 및 trypsin)를 처리하여 부분적 단백질 가수분해물을 만들고 이들 제품의 가수분해 정도를 고유점도 측정, 수용성 단백질 및 유리아미노산 농도 측정과 전기영동등으로 평가하였으며 가수분해물의 유체변형성과 식품기능성외 변화를 평가하였다. 가수분해물의 고유점도는 사용된 효소의 종류와 처리시간에 따라 변화하였으며 가수분해에 의한 수용성 단백질의 농도 변화와 일반적으로 역비례 하였다. 0.03 M$CaCl_2$, 용액에서의 용해도와 ninhydrin test에 의한 TCA가용성 단백질 함량은 고유점도의 변화와 밀접한 상관관계를 나타내었으므로 가수분해 정토를 나타내는 지표로 사용될 수 있었다. S. D. S. - P. A. G. 전기영동 패턴은 사용한 효소와 처리시간에 따라 다른 형태를 나타냈으며 pepsin처리에 있어서는 가수분해 정도가 증가함에 따라 주로 11S분획이 선택적으로 분해되었으며 trypsin처리에 있어서는 7S와 11S모두 영향을 받았다. 가수분해물의 흐름형태는 4%용액에서 거의 New tonian 유체형태를 나타내었으며 apparent viscosity는 가수분해 정도가 증가함에 따라 감소하였다. 유화용량 및 유화안정도는 SPI보다 모두 낮은 값을 나타냈으며 같은 효소처리인 경우 가수분해 정도가 증가함에 따라 감소하는 경향을 나타내었다. 기포형성은 SPI보다 현저하게 증가하였으나 기포안정도는 가수분해 정도가 증가함에 따라 급격하게 감소하는 경향을 나타내었다.
