간질성 폐질환에서 TUNEL 아포프토시스 지수와 임상적, 방사선학적, 및 병리학적 지표와의 상관관계 (The Correlation of TUNEL Apoptotic Index with Clinicoradiologicopathologic Scores in Interstitial Lung Disease)

  • 연구배경 : 간질성 폐질환은 병리소견, 임상경과 및 치료에 대한 반응에 있어서 다양한 양상을 보이지만 궁극적으로는 폐섬유화로 진행한다. 최근 간질성 폐질환의 병태생리에 있어서 폐상피세포의 아포프토시스가 폐섬유화로의 진행에 관여한다는 것이 보고되고 있다. 이에 저자들은 간질성 폐질환 환자에서 아포프토시스가 갖는 임상 및 병리학적 의미를 확인하기 위하여 TUNEL을 이용한 아포프토시스의 지수와 임상양상, 병리소견, HRCT소견, 그리고 치료반응 등과의 상관관계를 분석하였다. 방 법 : 개흉폐생검을 통해서 확진된 간질성 폐질환 환자 20명을 대상으로 하였다. 아포프토시스의 지수는 TUNEL 양성세포의 분율에 따라서 0-2로 구분하였다. 임상양상은 CRP 점수체계를 응용하여 이용하였다. 병리소견은 섬유화, 세포충실도, 탈락화, 육아조직성 정도에 따라 점수화하였다. HRCT소견은 간유리음영과 봉와양음영을 각 엽별로 침범한 정도에 따라서 점수화하였다. 치료의 반응은 치료전과 치료후 3개월째에 시행한 CRP 점수체계의 변화율이 10%이상 증가하였을 때 치료효과가 있는 것으로 판정하였다. 결 과 : 대상 환자는 20명(남 14명, 여 6명)으로 조직학적 아형으로는 AIP 3명, NIP 2 명, BOOP 8명, UIP 7 명이었다. 아포프토시스 지수와 CRP 점수체계의 요소들과는 유의한 상관관계가 없었으며 또한 HRCT소견인 간유리음영점수와 섬유화점수는 역시 유의한 상관관계가 없었다. 조직학적소견에서는 섬유화 점수, 세포충실도 점수, 그리고 탈락화 점수에서 아포프토시스 지수와 유의한 상관관계를 보였다. 부신피질호르몬 치료를 받은 16명의 환자중에서 9명 (56.3%)에서 치료효과가 있었는데 부신 피질호르몬 치료효과 유무와 아포프토시스 지수사이에는 상관관계는 없었다. 급, 아급성 간질성폐렴에서는 세포충실도 점수와 탈락화 점수가 아포프토시스 지수와 유의한 상관관계를 보였으며 만성 간질성 폐렴에서는 섬유화 점수와 세포충실도 유의한 상관관계를 보였다. 결 론 : 간질성 폐질환에서 아포프토시스 지수와 병리학적 지표들과의 유의한 상관관계를 관찰한 본 연구를 통하여 간질성 폐질환의 병태생리에 아포프토시스가 관여한다는 사실을 확인할 수 있었다.

Mycoplasma가 오염된 배양 각막상피 세포의 FAS 유도 세포고사의 민감성 (Susceptibility to FAS induced apoptosis in mycoplasma contaminated HCE cells)

  • 본 연구는 mycoplasma가 오염된 배양 각막 상피세포에 anti-FAS and anti-FAS ligand antibody를 노출시킨 후 세포고사 메커니즘을 조사하기 위해 시행하였다. 배양각막 상피세포에 anti-FAS antibody 와 anti-FAS ligand antibody를 2일과 4일 동안 처리하여 주어진 기간 동안 배양하였다. 배양 중인 각막상피세포가 mycoplasma sp.에 의해 오염된 것이 밝혀졌다. mycoplasma removal agent(MRA)가 세포주로부터 세균을 제거하기 위해 이용되었다. MRA를 세포주에 첨가하여 1주일 동안 배양하였다. 그 후 각막상피 세포주는 MRA가 포함되지 않는 배양액에서 수세대 동안 배양되어 커버글라스에 계대배양하여 50-80% 채워졌을 때 MRA 제거여부를 확인하기 위한 검사 키트에 제공된 Hoechst staining을 이용하여 염색하였다. 세포고사 실험은 MRA 제거 전과 제거 후에 시행되었다. mycoplasma가 오염된 배양 각막상피세포에 대한 anti-FAS and anti-FAS ligand antibody의 영향을 알아보기 위해 Hoechst 33342 staining과 Annexin V-FITC and Propidium Iodide Staining을 이용하여 세포고사 유도를 확인하였다. 본 연구는 anti-FAS antibody가 배양각막 상피세포에서 시간과 농도에 비례하는 메커니즘으로 세포고사를 유도한다. mycoplasma가 오염된 세포주는 FAS 유도 세포고사의 민감성을 증가시키는 것으로 나타났다.

실험적 급성 신우신염이 유발된 이유기 백서에서 methylprednisolone이 TGF-${\beta}1$, 세포고사 및 신반흔에 미치는 영향 (Effects of Methylprednisolone on TGF-${\beta}1$, Apoptosis and Renal Scarring in Experimental Acute Pyelonephritic Weaning Rats)

  • 목 적 :성장기 신장에서의 급성신우신염은 신반흔으로 진행된다. 신반흔의 형성에는 세균자체보다도 숙주의 염증반응과 면역반응의 산물인 TGF-${\beta}1$이 세포 고사를 증가시키고 세포증식을 억제함으로서 섬유화를 촉진한다고 하였다. 이에 저자는 항염증제인 methyl-prednisolone (MP)이 실험적으로 급성신우신염을 일으킨 이유기 백서에서 신반흔 형성에 미치는 영향을 관찰하고자 하였다. 대상 및 방법 : 생후 3주(체중 50-60g)된 이유기 Sprague-Dawley 백서의 방광에 삽입된 16 guage의 실리콘 도관내로 107/mL 농도의 E coli (ATCC No. 25922, pili형)를 5 mL씩 주입하여 급성신우신염을 유발하였다. 실험군은 1군 (ceftriaxone 단독투여, n=31)과 2군 (MP와 ceftriaxone투여, n=28)으로 나누었고 대조군 (n=43)에는 약제를 투여하지 않았다. 실험 1주와 3주에 실험동물을 희생하여 병리 조직학적 소견상 염증점수, 세포고사 지수와 TGF-${\beta}1$ 발현점수 및 섬유화 점수를 관찰하였다. 결 과 : 사망률은 II군이 $21.4\%$였으나 대조군 $41.9\%$, I군 $32.3\%$와 유의한 차이는 없었다. 염증 점수는 실험 1주에 II군에서 $0.8{\pm}0.87$로 대조군의 $2.3{\pm}0.87$, I군의 $1.7{\pm}0.79$에 비하여 유의하게 낮았다 (P<0.05. 세포고사 지수는 실험 1주에 II군에서 $2.9{\pm}2.15$로 대조군의 $10.0{\pm}1.95$, I군의 $8.3{\pm}2.53$에 비하여 유의 하게 낮았다 (P<0.05). TGF-${\beta}1$발현도 실험 1주에 II군에서 $0.8{\pm}0.72$로 대조군의 $1.90{\pm}67$, I군의 $1.8{\pm}0.60$에 비하여 유의하게 낮았다 (P<0.05). 섬유화 지수는 실험 3주에 II군에서 $0.8{\pm}0.63$로 대조군의 $1.8{\pm}0.83$에 비하여 유의하게 낮았다 (P<0.05) 결 론 : 성장기 백서의 실험적 급성 신우신염에서 MP는 ceftriaxone단독 투여에 비하여 염증 반응, 세포고사, TGF-${\beta}1$발현, 섬유화를 모두 감소 시켰다. 즉 항생제 외에 항염증제의 병용투여가 신반흔의 정도를 감소시킬 수 있으므로 치료지연등 신반흔의 위험인자가 있는 경우에 고려할 수 있을 것으로 생각된다.

The anti-tumor effect of combined treatment with arsenic trioxide and interferone-α on transplanted murine Lewis lung carcinoma

  • In the present study, we expected the anti-tumor effect by combined treatment of arsenic trioxide and interferon (IFN)-${\alpha}$ on murine Lewis lung carcinoma (LL2) cells through in vivo study. As a experimental model, LL2 cells ($1{\times}10^{6}$/mouse) were injected subcutaneously into the back region of mice. When the tumor volume reached $100mm^3$, mice were treated with 1 mg/kg arsenic trioxide, 50000 IU IFN-${\alpha}$, or arsenic trioxide and IFN-${\alpha}$. The development of tumor cells was significantly inhibited by combined treatment with arsenic trioxide and IFN-${\alpha}$. In arsenic trioxide and IFN-${\alpha}$ treated group, apoptotic index was reached a peak valve at 48 hr after the treatment and it was restored to approximately the control level at 8 days. Also, positive signals of Bax and Bad were increased at 48 to 96 hr and decreased at 8 day. Whereas, positive cells of Bcl-2 were steadily decreased at 12 to 48 hr and restored to the background level at 8 days. Our data showed that immunoreactivity of Bcl-2 was decreased at 12 to 48 hr, while positive signals of Bax and Bad were increased in accordance with apoptotic index at these times. In conclusion, our results suggest that the combined treatment with arsenic trioxide and IFN-${\alpha}$ significantly inhibited the growth of LL2 tumor cells and induced apoptosis through the up and down-regulation of Bcl-2 gene family.

Sulforaphane Inhibits Growth of Human Breast Cancer Cells and Augments the Therapeutic Index of the Chemotherapeutic Drug, Gemcitabine

  • Phytochemicals are among the natural chemopreventive agents with most potential for delaying, blocking or reversing the initiation and promotional events of carcinogenesis. They therefore offer cancer treatment strategies to reduce cancer related death. One such promising chemopreventive agent which has attracted considerable attention is sulforaphane (SFN), which exhibits anti-cancer, anti-diabetic, and anti-microbial properties. The present study was undertaken to assess effect of SFN alone and in combination with a chemotherapeutic agent, gemcitabine, on the proliferative potential of MCF-7 cells by cell viability assay and authenticated the results by nuclear morphological examination. Further we analyzed the modulation of expression of Bcl-2 and COX-2 on treatment of these cells with SFN by RT-PCR. SFN showed cytotoxic effects on MCF-7 cells in a dose- and time-dependent manner via an apoptotic mode of cell death. In addition, a combinational treatment of SFN and gemcitabine on MCF-7 cells resulted in growth inhibition in a synergistic manner with a combination index (CI)<1. Notably, SFN was found to significantly downregulate the expression of Bcl-2, an anti-apoptotic gene, and COX-2, a gene involved in inflammation, in a time-dependent manner. These results indicate that SFN induces apoptosis and anti-inflammatory effects on MCF-7 cells via downregulation of Bcl-2 and COX-2 respectively. The combination of SFN and gemcitabine may potentiate the efficacy of gemcitabine and minimize the toxicity to normal cells. Taken together, SFN may be a potent anti-cancer agent for breast cancer treatment.

Effect of Two Doses of Different Zinc Sources (Inorganic vs. Chelated form) on the Epithelial Proliferative Activity and the Apoptotic Index of Intestinal Mucosa of Early-weaned Pigs Orally Challenged with E. coli K88

  • The effect of two doses of different sources of zinc, inorganic (zinc oxide) or chelated (zinc glutamate chelate), on morphology and turn-over of the small intestine was assessed in early-weaned pigs orally challenged with enterotoxigenic E. coli K88 (ETEC). Sixty pigs weaned at 21 days were assigned to one of the following 5 diets: control (C); C+Zinc oxide (ZnO), either a 200 or a 2,500 mg Zn/kg dose; or C+zinc chelate with glutamic acid (Glu-Zn), either a 200 or a 2,500 mg Zn/kg dose. On d 2, the pigs were orally inoculated with 1.5 ml of a $10^{10}$ CFU/ml E. coli K88ac O148 suspension. Zinc supplements did not improve the performance of the pigs, but on d 5 faecal excretion of ETEC was reduced, and this was mainly due to high zinc doses (p<0.05). The villous height in the duodenum was improved by the zinc supplements (p<0.01) whatever the source and the level, whereas no effect was seen in the other two tracts of small intestine. The diet did not affect apoptosis and mitosis counts, while ETEC-susceptible pigs had more mitotic cells in the villi than non-susceptible pigs, particularly in the jejunum (p<0.01). The duodenum had fewer mitotic cells in the villi (p<0.05) and in the crypts (p<0.01) and more apoptotic cells in the villi. High dietary doses of ZnO or Zn-Glutamate improve villous height of the duodenum, but not of the jejunum and the ileum, and do not affect the epithelial proliferative activity and apoptotic index of intestinal mucosa of early-weaned pigs orally challenged with ETEC.

An in silico Appraisal to Identify High Affinity Anti-Apoptotic Synthetic Tetrapeptide Inhibitors Targeting the Mammalian Caspase 3 Enzyme

  • Apoptosis is a general phenomenon of all multicellular organisms and caspases form a group of important proteins central to suicide of cells. Pathologies like cancer, Myocardial infarction, Stroke, Sepsis, Alzheimer's, Psoriasis, Parkinson and Huntington diseases are often associated with change in caspase 3 mediated apoptosis and therefore, caspases may serve as potential inhibitory targets for drug development. In the present study, two series of synthetic acetylated tetrapeptides containing aldehyde and fluromethyl keto groups respectively at the C terminus were proposed. All these compounds were evaluated for binding affinity against caspase 3 structure. In series 1 compound Ac-DEHD-CHO demonstrated appreciable and high binding affinity (Rerank Score: -138.899) against caspase 3. While in series 2 it was Ac-WEVD-FMK which showed higher binding affinity (Rerank Score: -139.317). Further these two compounds met ADMET properties and demonstrated to be non-toxic.

Folate-Targeted Nanostructured Lipid Carriers (NLCs) Enhance (Letrozol) Efficacy in MCF-7 Breast Cancer Cells

  • Objective: Targeted-drug-delivery based lipid nanoparticles has emerged as a new and effective approach in cancer chemotherapy. Here, we investigated the ability of folate-modified nanostructured lipid carriers (NLCs) to enhance letrozol (LTZ) efficacy in MCF-7 breast cancer cells. Methods: New formulations were evaluated regarding to particle size and scanning electron microscope (SEM) features. Anti-proliferative effects of LTZ loaded nanoparticles were examined by MTT assay. To understand molecular mechanisms of apoptosis and cell cycle progression, flow cytometric assays were applied. Results: Optimum size of nanoparticles was obtained in mean average of $98{\pm}7nm$ with a poly dispersity index (PDI) of 0.165. The IC50 value was achieved for LTZ was $2.2{\pm}0.2{\mu}M$. Folate-NLC-LTZ increased the percentage of apoptotic cells from 24.6% to 42.2% compared LTZ alone (p<0.05). Furthermore, LTZ loaded folate targeted NLCs caused marked accumulation of cells in the subG1 phase. Conclusion: Taken together, our results concluded that folate targeted LTZ can be considered as potential delivery system which may overcome limitations of clinical application of LTZ and improve drug efficacy in tumor tissue.

2000년 이후 국내 한의학 암 관련 연구 동향 분석 - Latent Dirichlet Allocation 기반 토픽 모델링 및 연관어 네트워크 분석 (Cancer Research Trends in Traditional Korean Medical Journals since 2000 - Topic Modeling Using Latent Dirichlet Allocation and Keyword Network Analysis)

  • Objectives: The aim of this study is to analyze cancer research trends in traditional Korean medical journals indexed in the Korea Citation Index since 2000. Methods: Cancer research papers published in traditional Korean medical journals were searched in databases from inception to October 2022. The numbers of publications by journal and by year were descriptively assessed. After natural language processing, topic modeling (based on Latent Dirichlet allocation) and keyword network analysis were conducted. Results: This research trend analysis involved 1,265 papers. Six topics were identified by topic modeling: case reports on symptom management, literature reviews, experiments on apoptosis, herbal extract treatments of breast carcinoma cell lines, anti-proliferative effects of herbal extracts, and anti-tumor effects. Keyword network analysis found that the effects of herbal medicine were assessed in clinical and experimental studies, while acupuncture was mainly mentioned in clinical reports. Conclusions: Cancer research papers in traditional Korean medical journals have contributed to evidence-based medicine. Further experimental studies are needed to elucidate the effects of on different hallmarks of cancer. Rigorous clinical studies are needed to support clinical guidelines.

Gelam Honey and Ginger Potentiate the Anti Cancer Effect of 5-FU against HCT 116 Colorectal Cancer Cells

  • The development of chemopreventive approaches using a concoction of phytochemicals is potentially viable for combating many types of cancer including colon carcinogenesis. This study evaluated the anti-proliferative effects of ginger and Gelam honey and its efficacy in enhancing the anti-cancer effects of 5-FU (5-fluorouracil) against a colorectal cancer cell line, HCT 116. Cell viability was measured via MTS (3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulphenyl)-2H-tetrazolium) assay showing ginger inhibiting the growth of HCT 116 cells more potently ($IC_{50}$ of 3mg/mL) in comparison to Gelam honey ($IC_{50}$ of 75mg/mL). Combined treatment of the two compounds (3mg/mL ginger+75mg/mL Gelam honey) synergistically lowered the $IC_{50}$ of Gelam honey to 22mg/mL. Combination with 35 mg/mL Gelam honey markedly enhanced 5-FU inhibiting effects on the growth of HCT 116 cells. Subsequent analysis on the induction of cellular apoptosis suggested that individual treatment of ginger and Gelam honey produced higher apoptosis than 5-FU alone. In addition, treatment with the combination of two natural compounds increased the apoptotic rate of HCT 116 cells dose-dependently while treatment of either ginger or Gelam honey combined with 5-FU only showed modest changes. Combination index analysis showed the combination effect of both natural compounds to be synergistic in their inhibitory action against HCT 116 colon cancer cells (CI 0.96 < 1). In conclusion, combined treatment of Gelam honey and ginger extract could potentially enhance the chemotherapeutic effect of 5-FU against colorectal cancer.
