Castration induces the accumulation of body fat and deposition of intramuscular fat in Korean cattle, resulting in improved beef quality. However, little is known about the metabolic adaptations in the liver following castration. To understand changes in lipid metabolism following castration, hepatic expression levels of lipid metabolism genes were compared between Korean bulls and steers. Steers had higher (p<0.001) hepatic lipids contents and higher (p<0.01) mRNA levels of lipogenic acetyl-CoA carboxylase. This differential gene expression may, in part, contribute to increased hepatic lipid content following the castration of bulls. However, we found no differences in the hepatic expression levels of genes related to triglyceride synthesis (mitochondrial glycerol-3-phosphate acyltransferase, diacylglycerol O-acyltransferase 1 and 2) and fatty acid (FA) oxidation (carnitine palmitoyltransferase 1A, C-4 to C-12 straight chain acyl-CoA dehydrogenase, very long chain acyl-CoA dehydrogenase) between bulls and steers. No differences in gene expression for very-low-density lipoprotein (VLDL) secretion, including apolipoprotein B mRNA and microsomal triglyceride transfer protein (MTTP) protein, were observed in the liver although MTTP mRNA levels were higher in steers compared to bulls. In conclusion, FA synthesis may contribute to increased hepatic lipid deposition in steers following castration. However, hepatic lipid metabolism, including triglyceride synthesis, FA oxidation, and VLDL secretion, was not significantly altered by castration. Our results suggest that hepatic lipid metabolism does not significantly contribute to increased body fat deposition in steers following castration.

Recent findings from The Cancer Genome Atlas project have provided a comprehensive map of genomic alterations that occur in hepatocellular carcinoma (HCC), including unexpected mutations in apolipoprotein B (APOB). We aimed to determine the clinical significance of this non-oncogenetic mutation in HCC. An Apob gene signature was derived from genes that differed between control mice and mice treated with siRNA specific for Apob (1.5-fold difference; P < 0.005). Human gene expression data were collected from four independent HCC cohorts (n = 941). A prediction model was constructed using Bayesian compound covariate prediction, and the robustness of the APOB gene signature was validated in HCC cohorts. The correlation of the APOB signature with previously validated gene signatures was performed, and network analysis was conducted using ingenuity pathway analysis. APOB inactivation was associated with poor prognosis when the APOB gene signature was applied in all human HCC cohorts. Poor prognosis with APOB inactivation was consistently observed through cross-validation with previously reported gene signatures (NCIP A, HS, high-recurrence SNUR, and high RS subtypes). Knowledge-based gene network analysis using genes that differed between low-APOB and high-APOB groups in all four cohorts revealed that low-APOB activity was associated with upregulation of oncogenic and metastatic regulators, such as HGF, MTIF, ERBB2, FOXM1, and CD44, and inhibition of tumor suppressors, such as TP53 and PTEN. In conclusion, APOB inactivation is associated with poor outcome in patients with HCC, and APOB may play a role in regulating multiple genes involved in HCC development.
BACKGROUND/OBJECTIVES: Several previous studies have investigated whether regular walnut consumption positively changes heart-health-related parameters. The aim of this study was to investigate the effects of daily walnut intake on metabolic syndrome (MetS) status and other metabolic parameters among subjects with MetS. SUBJECTS/METHODS: This study was a two-arm, randomized, controlled crossover study with 16 weeks of each intervention (45 g of walnuts or iso-caloric white bread) with a 6 week washout period between interventions. Korean adults with MetS (n = 119) were randomly assigned to one of two sequences; 84 subjects completed the trial. At each clinic visit (at 0, 16, 22, and 38 weeks), MetS components, metabolic parameters including lipid profile, hemoglobin A1c (HbA1c), adiponectin, leptin, and apolipoprotein B, as well as anthropometric and bioimpedance data were obtained. RESULTS: Daily walnut consumption for 16 weeks improved MetS status, resulting in 28.6%-52.8% reversion rates for individual MetS components and 51.2% of participants with MetS at baseline reverted to a normal status after the walnut intervention. Significant improvements after walnut intake, compared to control intervention, in high-density lipoprotein cholesterol (HDL-C) (P = 0.028), fasting glucose (P = 0.013), HbA1c (P = 0.021), and adiponectin (P = 0.019) were observed after adjustment for gender, age, body mass index, and sequence using a linear mixed model. CONCLUSION: A dietary supplement of 45 g of walnuts for 16 weeks favorably changed MetS status by increasing the concentration of HDL-C and decreasing fasting glucose level. Furthermore, consuming walnuts on a daily basis changed HbA1c and circulating adiponectin levels among the subjects with MetS. This trial is registered at ClinicalTrials.gov as NCT03267901.
본 연구는 다중절편 방사선단층촬영을 이용하여 관상동맥 석회화와 심혈관질환 위험인자와의 관련성에 관하여 알아보기 위하여 수행되었다. 이를 위하여 일개 대학병원에서 2010년 1월부터 2011년 12월까지 다중절편 방사선단층촬영을 시행한 30~59세의 성인 남성 5,899명을 대상으로 문진, 설문, 신체계측, 혈액검사 등을 시행하였다. 다중절편 방사선단층촬영 영상을 통해 대상자의 관상동맥 칼슘 점수를 확인하고, 기존의 심혈관질환 위험인자에 대해 확인하였다. 대상자 5,899명의 평균 관상동맥 칼슘 점수는 8.20이었고, 773명(13.1%)에서 관상동맥 석회화가 관찰되었다. 관상동맥 석회화 유무와 알려진 심혈관질환 위험인자(나이, 혈압, 공복혈당, 콜레스테롤, 아포지방단백, 비만)는 유의한 관련성을 보였으며, 심혈관질환 발병 위험도평가도구들과도 유의한 관련성이 있었다. 30-39세, 40-49세, 50-59세의 연령 군에서 관상동맥 석회화와 통계적으로 유의한 관련 요인은 각기 다른 양상을 나타냈다. 본 연구를 통하여 관상동맥 석회화와 전통적인 심혈관질환 위험인자와의 관련성 및 심혈관질환 발병위험도 평가도구와의 관련성을 확인할 수 있었다. 또한 연령 군에 따라 각기 다른 양상의 위험인자를 확인할 수 있었다. 따라서 향후 심혈관질환 예방을 위한 관리전략 수립에 연령에 따른 차이를 고려한 접근이 필요할 것으로 사료된다.
CSRP3, APOBEC2, CAV1, CAV2 및 CAV3 유전자들은 포유동물에서 도체와 육질 형질에 중요한 역할을 하는 것으로 보고되고 있다. 따라서, 이 유전자들의 단일염기다형(Single nucleotide poly- morphism; SNP)을 8개의 다른 소의 품종에서 확인한 결과 coding region에서 caveolin family 유전자에서 9개의 SNP, CSRP3유전자에서 1개의 SNP 및 APOBEC2 유전자에서 3개의 SNP가 존재함을 확인하였다. 이 coding region의 SNP들은 PCR-RFLP 방법에 의해 재확인하였으며 이들 유전자의 intronic region에서도 9개의 SNP가 존재함을 확인할 수 있었다. 8개의 다른 품종 소에 각 유전자들의 SNP들을 이용하여 유전자 빈도를 확인한 결과 CAV2, CAV3, CSRP3 및 APOBEC2 유전자의 SNP 중에서 5개가 품종간에서 유의적으로 차이가 있음을 확인할 수 있었다. 이 SNP들은 차후 검증작업을 통하여 육질관련 형질 마커로 이용될 수 있을 것으로 사료된다.
DSG에 몰로키아 분말을 첨가하여 발효시킨 발효물이 콜레스테롤 개선효과를 가지는지 in vitro 및 in vivo에서 살펴보았다. B. subtilis NUC1균주로 발효한 탈지대두 grits 발효물인 FD와 탈지 대두 grit에 몰로키아를 5, 10%로 각각 첨가하여 발효한 FDC군의 콜레스테롤 흡착능을 살펴본 결과, 모든 군에서 70% 이상의 흡착능을 보였다. HepG2 세포를 이용한 apo-AI, apo-CIII의 분비능 측정에서는 FD군에 비해 몰로키아를 첨가한 FDC군에서 더 우수한 개선효과를 보였다. 한편 FDC가 고지방식이를 급여한 흰쥐의체내 콜레스테롤 개선에 미치는 영향을 살펴보고자, 기본식이군(Con), 고지방식이군(HF) 그리고 고지방식이와 FD첨가군(HFFD), 고지방식이와 FDC 첨가군(HF-FDC)으로 나누어 4주간 사육하였다. 그 결과, 혈장 내 총콜레스테롤 및 LDL-콜레스테롤 함량, 그리고 간 조직 내의 총콜레스테롤 및 중성지질 함량은 HF군에서 증가하였다가 HF-FDC 첨가군에서 유의적으로 감소하였고, 분변 내의 콜레스테롤 및 중성지질 배설량은 FDC 투여로 인해서 HF군에 비해 증가하였다. 또한 간내 HMG-CoA reductase 활성은 HF군에서 증가되었다가, HF-FDC첨가군에서 유의성 있게 감소되었다. 따라서 FDC는 콜레스테롤 흡착 및 조절능을 가지며, 그리고 혈중 콜레스테롤 함량, 조직 내의 지질 및 콜레스테롤 축적을 감소시켜 체내 콜레스테롤 개선에 효과적인 것으로 생각된다.
비타민-C를 다량 함유하고 있는 생열귀나무의 자원화를 위한 연구로서 페놀성 화합물의 함량 및 사람의 저밀도지방단백질의 산화에 대한 항산화 활성을 측정하였다. 생열귀나무 추출물의 페놀성 화합물 함량을 측정한 결과, 모든 부위에서 100g당 8.3-10.6g으로 매우 높은 양을 함유하고 있었다. 또한, 생열귀나무 추출물 및 분획물에 대한 항산화 물질의 함량을 측정 한 결과, 생열귀나무 뿌리 메탄올 추출물과 뿌리의 에틸아세테이트 및 부탄을 분획물이 285nm에서 흡수를 나타내는 페놀성 화합물을 다량 함유하고 있음을 확인하였다. TBARS에 의한 LDL산화 억제효과 측정시, 생열귀나무 뿌리의 메탄올 추출물이 가장 좋은 효과를 나타내었다. 생열귀나무 뿌리의 에틸아세테이트 및 부탄을 분회물의 경우, $30\;{\mu}g/mL$ 첨가시 85.3% 및 93.2%까지 LDL 산화를 억제함을 확인하였다. 또한 $CHCl_3$ 분획물, EtOAc 분획물, 그리고 BuOH 분획물은 $30\;{\mu}g/mL$ 분획물 첨가시 $Cu^{2+}$에 의한 LDL 산화를 8시간까지 억제함을 확인하였다. $Cu^{2+}$에 의한 apoB carbonylation과 conjugate diene형성 억제효과 측정시, EtOAc 분획물과 BuOH 분회물이 가장 탁월하게 억제함을 보여주었다. 생열귀나무는 다량의 페놀성 화합물을 함유하고 있으며 항산화 활성이 매우 탁월하므로 새로운 기능성식품 소재로의 활용이 기대되는 약용식물자원이다.
BACKGROUND/OBJECTIVES: Dyslipidemia causes metabolic disorders such as atherosclerosis and fatty liver syndrome due to abnormally high blood lipids. Purple perilla frutescens extract (PPE) possesses various bioactive compounds such as α-asarone, chlorogenic acid and rosmarinic acid. This study examined whether PPE and α-asarone improved dyslipidemia-associated inflammation and inhibited atheroma formation in apolipoprotein E (apoE)-deficient mice, an experimental animal model of atherosclerosis. MATERIALS/METHODS: ApoE-deficient mice were fed on high cholesterol-diet (Paigen's diet) and orally administrated with 10-20 mg/kg PPE and α-asarone for 10 wk. RESULTS: The Paigen's diet reduced body weight gain in apoE-deficient mice, which was not restored by PPE or α-asarone. PPE or α-asarone improved the plasma lipid profiles in Paigen's diet-fed apoE-deficient mice, and despite a small increase in high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein (LDL)-cholesterol, and very LDL were significantly reduced. Paigen's diet-induced systemic inflammation was reduced in PPE or α-asarone-treated apoE-deficient mice. Supplying PPE or α-asarone to mice lacking apoE suppressed aorta atherogenesis induced by atherogenic diet. PPE or α-asarone diminished aorta accumulation of CD68- and/or F4/80-positive macrophages induced by atherogenic diet in apoE-deficient mice. Treatment of apoE-deficient mice with PPE and α-asarone resulted in a significant decrease in plasma cholesteryl ester transfer protein level and an increase in lecithin:cholesterol acyltransferase reduced by supply of Paigen's diet. Supplementation of PPE and α-asarone enhanced the transcription of hepatic apoA1 and SR-B1 reduced by Paigen's diet in apoE-deficient mice. CONCLUSIONS: α-Asarone in PPE inhibited inflammation-associated atheroma formation and promoted hepatic HDL-C trafficking in dyslipidemic mice.
목적 : 말기 신부전 환자에서 심혈관계 질환은 유병율 및 사망율의 중요한 원인중 하나이다. 이들 환자에서 흔히 동반되는 고지질단백 혈증은 심혈관계 질환의 위험인자로 작용하며, 특히 lipoprotein(a)[Lp(a)]는 관상동맥 질환과 혈전증의 위험 인자로 알려져 있다. 소아에서도 만성 신부전시 고지질 혈증이 보고되어 있다. 저자들은 투석 치료중인 만성 신부전 소아를 대상으로 혈중 지질, 지질단백 및 Lp(a)의 농도 변화를 분석하였다. 방법 : 서울대학교 어린이병원 소아과에서 유지 혈액 투석을 받고 있는 환아 10명과 지속성 외래 복막 투석을 받고 있는 환아 14명을 대상으로 정맥 혈청중 지질, 지질단백 및 Lp(a)농도를 측정하여 건강 대조군과 비교하였으며, Lp(a)의 혈중 농도에 따른 다른 지질단백의 혈중 농도를 비교하였다. 혈액투석(hemodialysis, HD)환아군의 평균 연령이 $162{\pm}59$ 개월, 남녀비는 7:3 이었으며 복막투석(peritoneal dialysis, PD)환아군의 평균 연령은 $123{\pm}69$개월, 남녀비는 6:8 이었다. 결과 : 1) 연령, 성별, 비만지수, 투석기간은 HD군과 PD군 간의 차이가 없었으나 혈중 단백과 알부민치는 PD군에서 유의하게 낮았다. 2) HD군과 PD군 모두에서 혈중 콜레스테롤, 중성지방, 저비중 지질단백 콜레스테롤(low density lipoprotein-cholesterol, LDL-콜레스테롤)과 고비중 지질단백 콜레스테롤(high density lipoprotein-cholesterol, HDL-콜레스테롤)의 비는 대조군에 비해 증가되어 있었으며, HDL-콜레스테롤과 콜레스테롤의 비는 대조군에 비해 유의하게 낮았다. 3) 혈중 아포단백 B치는 대조군에 비해 상승되어 있었지만 아포단백 Al과 아포단백 B의 비는 차이가 없었고, 혈중 Lp(a)치는 PD군에서만 대조군보다 상승되어 있었다. 4) 혈중 Lp(a)치가 30 mg/dl 미만인 환아는 13명, 30 mg/dl 이상인 환아는 11명으로 이들 간에 연령, 비만지수, 투석방법, 투석기간은 차이가 없었으나 혈중 알부민은 Lp(a)가 상승된 환아들에서 감소되어 있었다. 5) 혈중 Lp(a)농도와 다른 지질 단백과의 비교에서 아포 단백 B만이 유의한 차이를 보였다. 결론 : 투석 치료중인 만성 신부전 소아에서 혈중 지질단백 이상이 관찰되었으며 특히 PD군에서는 혈중 Lp(a)치가 증가되어 있었다. 따라서 만성 신부전 소아에서 관상 동맥 질환의 위험도가 높을 것으로 사료되며 이에 대한 장기적이고 지속적인 관찰이 필요하다.
과체중과 비만은 당뇨병, 고혈압, 고지혈증과 같은 신진 대사 질환의 발병과 강력하게 연관되어 있다. 비만의 원인은 여러가지가 있겠지만, 고지방식이는 비만의 원인 중 가장 중요한 요소 중 하나이다. 본 연구에서는 C57BL/6 생쥐에게 22주간의 고지방 식이를 주었으며, 이를 통해 비만과 고혈당을 유도하였다. 22주 후에 고지방식이를 한 생쥐들에서 체중과 혈장 포도당 수준이 정상 식이를 한 생쥐들에 비해 크게 증가함을 관찰하였다. 복막 내 당 부하 검사(IPGTT)에서도 고지방 식이를 한 생쥐들은 정상 식이를 한 생쥐들에 비해서 당 내성 이상 반응을 보여주었다. 이러한 결과들은 고지방식이가 C57BL/6 생쥐에서 비만 및 고혈당을 유도한다는 사실을 확인시켜 주었다. 고지방식이군 생쥐들에서는 정상식이군의 생쥐들에 비해 혈장의 중성지방과 총 콜레스테롤의 양이 증가됨이 관찰되었다. 간에서의 중성지방 및 총 콜레스테롤의 수준도 역시 증가하였다. 따라서, 간에서의 지질대사가 어떻게 변하였는지를 알기 위해, 지질대사에 관련된 단백질들의 변화를 관찰하였다. 지방 합성과 관련된 효소들 중 FAS와 GPAT가 고지방식이 군에서 의미있게 증가 되어있었으며, 지방 수송에 관련하는 단백질 중에서도 ApoB 및 MTP의 큰 증가가 고지방식이군에서 관찰되었다. 흥미롭게도, 대사 조절 인자로 알려진 AMPK의 단백질의 양과 인산화 정도는 정상식이군에 비해 고지방식이군에서 의미있게 증가되었음이 관찰되었다. 결론적으로, 본 연구에서 우리는 고지방식이가 지질 합성과 지질 수송과정을 생리학적으로 증가시키지만, 역설적으로 AMPK의 활성화를 유발한다는 것을 확인하였다.

② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는

- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
