볶은 원두커피 갈색추출물의 항산화 효과 (Antioxidative Effect of Brown Materials Extracted from Roasted Coffee Beans)

  • 볶은 원두커피로부터 얻은 갈색 추출물의 항산화력에 대한 볶음시간과 추출온도의 영향을 시험하였다. 볶은 원두커피의 갈색추출물을 돈지에 첨가하였을 때 항산화력을 나타냈으며, 그 항산화력은 원두의 품종에 따라 큰 차이가 없었으나 Colombia 원두가 Robusta 원두 또는 Brazil 원두에 비해 강하였다. Colombia 원두를 $210^{\circ}C$에서 볶음 시간을 달리하였을 때 갈색추출물의 갈색도와 항산화력은 볶음시간 16분까지는 함께 증가하였으나, 그 후는 갈색도는 계속 증가하나 항산화력은 감소하는 경향이었다. 이것은 볶음에 의해 생성된 항산화성 갈색물질이 가열에 의해 분해되기 때문인 것으로 추측되었다. 볶은 원두커피를 $180^{\circ}C$까지 추출온도를 달리하여 갈색도와 항산화력을 측정한 결과 추출온도는 갈색도와 항산화력에 영향을 주지 못하였다.

Hesperidin 과 Naringin 이 흰쥐의 항산화능에 미치는 영향 (Effects of Hesperidin and Naringin on Antioxidative Capacity in the Rat)

  • This study was performed to investigate effects of hesperidin and naringin on linpid peroxide formation and antioxidative enzyme activities in rats. Thiobarbituric acid reactive substance (TBARS) concentrations were measured in plasma and liver. Catalase, superoxide dismutase, and glutathione peroxidase activities were measured in erythrocyte and liver. Forty-nine male Sprague-Dauley rats weighing 275.3$\pm$3.3g were blocked into seven groups according to body weight and were raised fro four weeks on diets containing 0.25, 0.50 or 1.00%(w/w) hesperidin or naringin . Food intake, weight gain , food efficiency ratio, and weights of liver, kidney, spleen ,and epididymal fat pad were not significantly different among groups. In 0.50 and 1.00% naringin groups , plasma TBARS concentrations were significantly decreased with a dose response patter. In 0.25, 0.50 and 1.00% hesperidin groups, liver TBARS concentrations were significantly decreased without a dose dependent patter. Antiosidative enzyme activities in erythrocyte and liver were not significantly affected by type and amountof dietary bioflavonoid, but in the 1.00% hesperidin group, catalase, superoxide dismutase, and glutahione perosidase activities in linver showed a tendency to increase. In conclusion, naringin inhibited lipid peroxide formation with a dose response pattern in plasma without changing the activities of antioxidative enzymes. Hesperidin adminstration, regardless of the level in the diet, inhibited lipid peroxide formation in liver.

Nutritional Compositions of Ray and Effects of Steaming on Antioxidative Activities of Ray Hydrolysates

  • This study investigated the effects of steaming on the antioxidative activities of ray hydrolysates in vitro. Based on the results of 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay, steamed ray hydrolysate possessed significantly higher antioxidative activities than raw ray hydrolysates (p<0.05). The 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity of steamed ray hydrolysate by pancreatin was slightly lower than raw ray hydrolysate at all concentrations. Raw ray hydrolysate displayed moderate hydroxyl radical scavenging activity, while steamed ray hydrolysate showed a greater increase in hydroxyl radical scavenging activity than raw ray hydrolysate at concentrations above 21.33 mg/mL, and it reached 58.21% at 42.67 mg/mL. The results of this study show that ray hydrolysates have potent free radical scavenging activities and reducing power, and the steaming has a partial impact on the antioxidative activity of ray hydrolysates.

울금 분말 첨가가 매작과의 품질특성 및 항산화에 미치는 영향 (Quality Characteristics and Antioxidative Activities of Majakgwa with added Turmeric Powder)

  • This study was carried out for the purpose of inquiring into the effects on shelf-life, quality characteristics and antioxidative activities of Majakgwa with regard to the use turmeric powder at varying concentrations in the recipe. Chemical composition analysis showed that the contents of crude fat, crude protein and carbohydrates are high in turmeric powder. With an increase of the amount of turmeric powder used in Majakgwa recipes, dough pH was higher than that of the control group. Antioxidative activities were the highest in the groups with 3% and 9% turmeric powder content, and the value increased largely after 12 days of storage. Excellent antioxidative activities were observed in the 6% turmeric content group. For recipes with turmeric contents of more than 15%, oxidation was accelerated from early on in storage. The hardness, brittleness and bitterness increased significantly with the increase of the turmeric content. The intensity, chromaticity, crunch and intensity of the surface colors, all increased significantly with the increase of turmeric content. The general preference ratings for Majakgwa with added turmeric was highest for the 9% content recipe. Over all Majakgwa recipes with 6~9% turmeric powder content were found to ideal.

아질산에 노출된 뱀장어(Anguilla japonica)의 혈액학적 반응과 항산화효소의 활성 변화 (Changes in Hematological Responses and Antioxidative Enzyme Activities of Japanese Eel Anguilla japonica Exposed to Elevated Ambient Nitrite)

  • The study was performed to investigate changes in hematological responses and antioxidative enzyme activities (superoxide dismutase, SOD; catalase, CAT) of Japanese eel Anguilla japonica following exposure to 0 (control), 2.33, 4.60, 6,64 and 8.78 mM nitrite-N in fresh water for 48 h. Hematological parameters such as plasma nitrite, electrolytes, cortisol, glucose, glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), hemoglobin (Hb), methemoglobin (metHb) and NADH-methemoglobin reductase (NMR) were measured. Plasma nitrite, cortisol, metHb and NMR increased directly with increasing ambient nitrite concentration, while Hb content showed a progressive decline. Levels of plasma potassium, GOT and GPT of the eel exposed to 6.64 mM ambient nitrite were significantly higher than the control fish. The activity of SOD and CAT in plasma, gill and liver of the eel following exposure to nitrite were augmented by increasing ambient nitrite. Levels of plasma nitrite, metHb, NMR, cortisol, glucose and antioxidative enzyme activities of the eel exposed to 2.33 mM ambient nitrite were significantly higher than the control fish. This study suggested that the eel acutely exposed to elevated ambient nitrite causes nitrite-induced stress responses, changes in antioxidative enzyme activities and hematological parameters.

목단피 추출물의 항균 및 항산화 작용 (Antimicrobial and Antioxidative Activities from Moutan Cortex Extract)

  • 목단피의 메탄을 추출물과 용매 분획물을 이용하여 항균작용, 프리라디칼 소거작용 및 항산화 작용을 검색하고, HPLC와 GC를 이용하여 페놀화합물을 분석하였다. 항균작용은 에칠아세테이트 분획에서 가장 높게 나타났으며, 그람 음성균보다는 그람 양성균에 대하여 더욱 효과적이었다. 에칠아세테이트 분획의 MIC를 측정한 결과, 그람 양성균에 대하여는 156-1250$\mu\textrm{g}$/ml, 그람 음성균에는 2500-5000$\mu\textrm{g}$/ml 이었다. DPPH 라디칼에 대한 프리라디칼 소거활성과 linoleic acid를 이용한 항산화 시험에서도 에칠아세테이트 분획이 가장 높은 활성을 나타내었다. 에칠아세테이트 분획의 항산화 활성은 $\alpha$-tocopherol과 비슷하였다. 에칠아세테이트 분획으로부터 U와 HPLC를 이용하여 3종의 페놀화합물을 확인하였으며, 그 함량은 p-hydroxybenzoic acid 1.35%, methyl gallate 14.61%, gallic acid 4.01% 이었다.

Comparative Analysis on Antioxidative Ability of Muscle between Laiwu Pig and Large White

  • This study was conducted to evaluate effects of storage temperatures ($4^{\circ}C$ and $20^{\circ}C$) and pig breeds (Laiwu pig and Large White pig) on the main antioxidative enzymes (superoxide dismutase, catalase, and glutathione peroxidase) activity and lipid oxidation in porcine Longissimus dorsi muscle. Activities of antioxidative enzymes (AOE) decreased slightly during storage, regardless of storage temperatures. Muscle antioxidative enzymes activities stored at $4^{\circ}C$ were higher than that stored at $20^{\circ}C$. Laiwu pig's enzymes activities were significantly (p<0.01) higher than Large White's. The level of malondialdehyde is a direct expression of the grade of lipid oxidation in meat. In our study, the malondialdehyde contents increased after 6 days storage. However, malondialdehyde contents of Laiwu pig were significantly (p<0.01) lower than Large White's. A lower content of malondialdehyde corresponds to a lower oxidation of lipids. These results indicated the muscle antioxidative ability of Laiwu pig was higher than Large White pig. It also implied that antioxidative enzymes were involved in the essentials and deciding mechanisms of meat quality by quenching oxygen free radicals and inhibiting lipid oxidation in muscle.

밤 귀피의 용매분획별 항산화 활성과 항산화 물질의 분리 (Antioxidative Activity of Solvent Fraction and Isolation of ANtioxidative Compound from Chestnut Husk)

  • 밤 가공공장에서 폐기되고 있는 밤의 귀피를 항산화 기능성 자원으로 활용하고자 밤 귀치의 methanol 추출물 및 용매획분을 benzoic acid 수산화법으로 hydroxyl radical 소거능과 ferric thiocyanate법으로 과산화물생성 억제능을 시험하였다. 대부분의 획분에서 항산화성을 보였으며 특히 ethyl acetate 획분에서 BHA나 $\alpha$-tocopherol과 거의 유사한 rkdgkshydroxyl radical 소거능과 지질과산화물생성 억제능을 보였다. Ethyl acetate 획분으로부터 항산화성 물질을 분리하기 위하여 Sephadex LH-20 column chromatography 및 prep HPLC를 행하여 단일 물질로 정제하였다. 분리 정제된 물질을 $^{1}$H-NMR과 $^{13}$C-NMR로 구조 분석한 결과 gallic acid로 동정하였다. 페놀산을 GC로 분석한 결과 밤 귀피의 주요 페놀산으로 ellagic acid(172.22 mg%) 와 gallic acid(107.39 mg%)가 확인되었다.

Screening of Antioxidative Activity of Bifidobacterium Species Isolated from Korean Infant Feces and Their Identification

  • Among 59 Korean isolated, 20 were confirmed as members of the genus Bifidobaferium species based on gram staining, microscopic examination of cell morphology and the TLC method, The oxygen tolerance and antioxidative activities of these 20 Bifidobacterium strains and 5 standard Bifidobaferium strains were tested. All the strains demonstrated antioxidative activities as regards inhibiting linoleic acid peroxidation. The antioxidative activities of isolated and standard strains were found to range from 10.7-46.4% and from 10.7-22.2%, respectively. In addition, all tested strains exhibited a Scavenging ability on DPPH free radicals, range from 15-41% for the isolated strains and 8.3-22% for the standard strain. Accordingly. the isolated Bifidobarterium strains demonstrated higher antioxidative artivities than the 5 standa rd Bifidobarterium strains. On the base of grades for each test, HJL 7511 was identified 35 the best strain, followed by HJL 7501. 2 strains were identified with Polymerase Chain Reaction (PCR) assay using group-specific primers designed from the nucleotide Sequences of the 16S rRNA and internal transcribed spacer (ITS) regions of the Bifidobacteria. Based on the Sequencing results, HJL 7511 and HJL 7501 were identified as Bifidobacterium infantis.
