Antioxidative Effect of Brown Materials Extracted from Roasted Coffee Beans (볶은 원두커피 갈색추출물의 항산화 효과)

  • Antioxidative activities of brown materials extracted from the three kinds of roasted coffee beans by water were examined. Antioxidative activity was assessed by the AOM at $120^{\circ}C$ and the oven test at $60^{\circ}C$ on lard. The brown materials of them showed the considerable antioxiant activity. The brown materials of Colombian coffee beans were more effective than those of Robusta or Brazil beans. The antioxidative activities of brown materials from Colombian coffee beans roasted at $210^{\circ}C$ increased in proportion to the browning intensity up to 16 minutes of roasting time, but the antioxidant activities of the brown materials upon further roasting time decreased gradually. In changes of extraction temperature from $30^{\circ}C\;to\;180^{\circ}C$ on the coffee beans roasted at $210^{\circ}C$ for 16 minutes, the antioxidative activities of brown materials did not change as the extraction temperature increased to $180^{\circ}C$. The antioxidative activities of the coffee extracts were thought to be caused by browning reaction materials produced during the roasting process only.

Effects of Hesperidin and Naringin on Antioxidative Capacity in the Rat (Hesperidin 과 Naringin 이 흰쥐의 항산화능에 미치는 영향)

  • This study was performed to investigate effects of hesperidin and naringin on linpid peroxide formation and antioxidative enzyme activities in rats. Thiobarbituric acid reactive substance (TBARS) concentrations were measured in plasma and liver. Catalase, superoxide dismutase, and glutathione peroxidase activities were measured in erythrocyte and liver. Forty-nine male Sprague-Dauley rats weighing 275.3$\pm$3.3g were blocked into seven groups according to body weight and were raised fro four weeks on diets containing 0.25, 0.50 or 1.00%(w/w) hesperidin or naringin . Food intake, weight gain , food efficiency ratio, and weights of liver, kidney, spleen ,and epididymal fat pad were not significantly different among groups. In 0.50 and 1.00% naringin groups , plasma TBARS concentrations were significantly decreased with a dose response patter. In 0.25, 0.50 and 1.00% hesperidin groups, liver TBARS concentrations were significantly decreased without a dose dependent patter. Antiosidative enzyme activities in erythrocyte and liver were not significantly affected by type and amountof dietary bioflavonoid, but in the 1.00% hesperidin group, catalase, superoxide dismutase, and glutahione perosidase activities in linver showed a tendency to increase. In conclusion, naringin inhibited lipid peroxide formation with a dose response pattern in plasma without changing the activities of antioxidative enzymes. Hesperidin adminstration, regardless of the level in the diet, inhibited lipid peroxide formation in liver.

Nutritional Compositions of Ray and Effects of Steaming on Antioxidative Activities of Ray Hydrolysates

  • This study investigated the effects of steaming on the antioxidative activities of ray hydrolysates in vitro. Based on the results of 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay, steamed ray hydrolysate possessed significantly higher antioxidative activities than raw ray hydrolysates (p<0.05). The 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity of steamed ray hydrolysate by pancreatin was slightly lower than raw ray hydrolysate at all concentrations. Raw ray hydrolysate displayed moderate hydroxyl radical scavenging activity, while steamed ray hydrolysate showed a greater increase in hydroxyl radical scavenging activity than raw ray hydrolysate at concentrations above 21.33 mg/mL, and it reached 58.21% at 42.67 mg/mL. The results of this study show that ray hydrolysates have potent free radical scavenging activities and reducing power, and the steaming has a partial impact on the antioxidative activity of ray hydrolysates.

Quality Characteristics and Antioxidative Activities of Majakgwa with added Turmeric Powder (울금 분말 첨가가 매작과의 품질특성 및 항산화에 미치는 영향)

  • This study was carried out for the purpose of inquiring into the effects on shelf-life, quality characteristics and antioxidative activities of Majakgwa with regard to the use turmeric powder at varying concentrations in the recipe. Chemical composition analysis showed that the contents of crude fat, crude protein and carbohydrates are high in turmeric powder. With an increase of the amount of turmeric powder used in Majakgwa recipes, dough pH was higher than that of the control group. Antioxidative activities were the highest in the groups with 3% and 9% turmeric powder content, and the value increased largely after 12 days of storage. Excellent antioxidative activities were observed in the 6% turmeric content group. For recipes with turmeric contents of more than 15%, oxidation was accelerated from early on in storage. The hardness, brittleness and bitterness increased significantly with the increase of the turmeric content. The intensity, chromaticity, crunch and intensity of the surface colors, all increased significantly with the increase of turmeric content. The general preference ratings for Majakgwa with added turmeric was highest for the 9% content recipe. Over all Majakgwa recipes with 6~9% turmeric powder content were found to ideal.

Changes in Hematological Responses and Antioxidative Enzyme Activities of Japanese Eel Anguilla japonica Exposed to Elevated Ambient Nitrite (아질산에 노출된 뱀장어(Anguilla japonica)의 혈액학적 반응과 항산화효소의 활성 변화)

  • The study was performed to investigate changes in hematological responses and antioxidative enzyme activities (superoxide dismutase, SOD; catalase, CAT) of Japanese eel Anguilla japonica following exposure to 0 (control), 2.33, 4.60, 6,64 and 8.78 mM nitrite-N in fresh water for 48 h. Hematological parameters such as plasma nitrite, electrolytes, cortisol, glucose, glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), hemoglobin (Hb), methemoglobin (metHb) and NADH-methemoglobin reductase (NMR) were measured. Plasma nitrite, cortisol, metHb and NMR increased directly with increasing ambient nitrite concentration, while Hb content showed a progressive decline. Levels of plasma potassium, GOT and GPT of the eel exposed to 6.64 mM ambient nitrite were significantly higher than the control fish. The activity of SOD and CAT in plasma, gill and liver of the eel following exposure to nitrite were augmented by increasing ambient nitrite. Levels of plasma nitrite, metHb, NMR, cortisol, glucose and antioxidative enzyme activities of the eel exposed to 2.33 mM ambient nitrite were significantly higher than the control fish. This study suggested that the eel acutely exposed to elevated ambient nitrite causes nitrite-induced stress responses, changes in antioxidative enzyme activities and hematological parameters.

Antimicrobial and Antioxidative Activities from Moutan Cortex Extract (목단피 추출물의 항균 및 항산화 작용)

  • Methanol extract and various solvent fractions from Moutan Cortex were tested for their antimicrobial activities, free radical scavenging activities and antioxidative activities, and phenolic compounds in ethylacetate fraction were analyzed by GC and HPLC. In the antimicrobial activities test, the ethylacetate fraction of methanol extract showed stronger than other fractions. The antimirobial activities were more effective against Gram positive bacteria than Gram negative bacteria. Minimum inhibitory concentration(MIC) of ethylacetate fraction showed 156-1250$\mu\textrm{g}$/ml against Cram positive bacteria and 2500-5000$\mu\textrm{g}$/mg against Gram negative bacteria. The free radical scavenging activities and antioxidative activities using linoleic acid were higher in ethylacetate fraction. The antioxidative activity of ethylacetate fraction was similar to ${\alpha}$-tocopherol. The 3 major phenolic compounds were analyzed by GC and HPLC and these content were determined. The content of p-hydroxybenzoic acid, methyl gallate and gallic acid were 1.35%, 14.61% and 4.01%, respectively.

Comparative Analysis on Antioxidative Ability of Muscle between Laiwu Pig and Large White

  • This study was conducted to evaluate effects of storage temperatures ($4^{\circ}C$ and $20^{\circ}C$) and pig breeds (Laiwu pig and Large White pig) on the main antioxidative enzymes (superoxide dismutase, catalase, and glutathione peroxidase) activity and lipid oxidation in porcine Longissimus dorsi muscle. Activities of antioxidative enzymes (AOE) decreased slightly during storage, regardless of storage temperatures. Muscle antioxidative enzymes activities stored at $4^{\circ}C$ were higher than that stored at $20^{\circ}C$. Laiwu pig's enzymes activities were significantly (p<0.01) higher than Large White's. The level of malondialdehyde is a direct expression of the grade of lipid oxidation in meat. In our study, the malondialdehyde contents increased after 6 days storage. However, malondialdehyde contents of Laiwu pig were significantly (p<0.01) lower than Large White's. A lower content of malondialdehyde corresponds to a lower oxidation of lipids. These results indicated the muscle antioxidative ability of Laiwu pig was higher than Large White pig. It also implied that antioxidative enzymes were involved in the essentials and deciding mechanisms of meat quality by quenching oxygen free radicals and inhibiting lipid oxidation in muscle.

Antioxidative Activity of Solvent Fraction and Isolation of ANtioxidative Compound from Chestnut Husk (밤 귀피의 용매분획별 항산화 활성과 항산화 물질의 분리)

  • To enhance the utilization of chestnut husk discarded in the processing company antioxidative activities and compounds were investigated. Antioxidative activities of solvent fractions from chestnut husk were examined by benzoic acid hydroxylation method ferric thiocyanate method and DPPH test. Ethyl acetate fraction showed strong antioxidative activities comparable to BHA. Active compounds were isolated and purified from ethyl acetate fraction by Sephadex LH-20 column chromatography and preparative HPLC. A major active compound gallic acid was identified by $^{1}$H and $^{13}$ C-NMR. The phenolic acid contents was analyzed by GC and ellagic acid (172.22 mg%) and gallic acid (107.39 mg%) were major phenolic acid of chestnut husk.

Screening of Antioxidative Activity of Bifidobacterium Species Isolated from Korean Infant Feces and Their Identification

  • Among 59 Korean isolated, 20 were confirmed as members of the genus Bifidobaferium species based on gram staining, microscopic examination of cell morphology and the TLC method, The oxygen tolerance and antioxidative activities of these 20 Bifidobacterium strains and 5 standard Bifidobaferium strains were tested. All the strains demonstrated antioxidative activities as regards inhibiting linoleic acid peroxidation. The antioxidative activities of isolated and standard strains were found to range from 10.7-46.4% and from 10.7-22.2%, respectively. In addition, all tested strains exhibited a Scavenging ability on DPPH free radicals, range from 15-41% for the isolated strains and 8.3-22% for the standard strain. Accordingly. the isolated Bifidobarterium strains demonstrated higher antioxidative artivities than the 5 standa rd Bifidobarterium strains. On the base of grades for each test, HJL 7511 was identified 35 the best strain, followed by HJL 7501. 2 strains were identified with Polymerase Chain Reaction (PCR) assay using group-specific primers designed from the nucleotide Sequences of the 16S rRNA and internal transcribed spacer (ITS) regions of the Bifidobacteria. Based on the Sequencing results, HJL 7511 and HJL 7501 were identified as Bifidobacterium infantis.
