선씀바귀 추출물의 항산화 및 항당뇨 효과 (Antioxidant and anti-diabetic effects of Ixeris strigosa extract)

  • 본 연구는 선씀바귀의 항산화 및 탄수화물 소화효소 억제 효과를 평가하여 제 2형 당뇨환자에게 중요한 항산화제 및 혈당 상승 억제제로서의 가능성을 타진하고자 하였다. 선씀바귀 추출물의 총 폴리페놀, 플라보노이드 함량과 in vitro 항산화 활성 및 소화효소 저해 활성을 조사한 결과 ISE가 ISW보다 총페놀과 플라보노이드 함량이 높았고 항산화 활성 및 소화효소 저해 활성 또한 높았다. ISE의 계통분획물 중에 에틸아세테이트 분획물과 부탄올 분획물의 소화효소 저해 활성이 가장 우수하고, 추출물의 항산화활성과 탄수화물 소화효소 억제효과가 총 페놀 및 플라보노이드함량과 높은 상관성을 가지는 것으로 볼 때, ISW에 비해 상대적으로 높은 ISE의 항산화 및 소화효소 저해 활성은 페놀 화합물, 특히 플라보노이드 화합물 때문임을 알 수 있었다. 따라서 ISE와 에틸아세테이트 그리고 부탄올 분획물은 우수한 항당뇨 기능성 소재라고 판단된다.

  • Effective synthetic antioxidants such as butylated hydroxyanisole(BHA) and butylated hydroxytoluene(BHT) have been widely used in the food industry, but they are suspected to be toxic and carcinogenic effects. Therefore, the development of safely available natural antioxidants such as ascorbic acid, ${\alpha}$-tocopherol, ${\beta}$-carotene, flavonoids and selenium is essential. In particular, flavonoids, 2-phenyl-benzo-${\alpha}$-pyrones, are polyphenolic compounds that occur ubiquitously in food of plant origin. flavonoids occur in foods generally as O-glycosides with sugars bound usually at the C\ulcorner position. And variations in their heterocyclic ring gibes rise to flavones, flavonols, flavanones, flavanols, catechins, anthocyanidins, chalcone and isoflavones. Vegetables, fruits, and beverages are the main dietary sources of the flavonols, primarily as quercetin, kaempferol, and myricetin and the corresponding flavones, apigenin and luteolin. These flavonoids have biological activity such as antioxidant, anti-inflammatory, antithrombotic, antimutagenic, anticarcimogenic antiallergic and antimicrobial activity effects in vitro and in vivo. Flavonoids posses strong antioxidant activities acting as oxygen radicals scavenger, metal chelators and enzyme inhibitor. The antioxidant activity of flavonoids is determined by their molecular structure and more specially, by the position and degree of hydroxylation of the ring structure. All flavonoids with the 3`, 4`-dihydroxy(ortho-dihydroxy) posses marked antioxidant activity. And antioxidant activity increases with the number of hydroxyl groups substituted on the A-and B-rings. There is as yet no certainty about the effect of the presence of a double bond between C\ulcorner and C\ulcorner on the antioxidant activity of flavonoids.

대추 추출물의 항산화 및 아토피 피부염 관련 항염증 효과 (Anti-inflammatory and Antioxidant Effects of Zizyphus jujuba Miller var. inermis extracts)

  • In the present study, the effect of Z. jujuba Miller var. inermis Rehder extracts on the secretion of atopic dermatitis (AD)-related cytokines and hyaluronidase activity was investigated. We prepared four fractions, butylene glycol (JB), ethanol (JE), and water (JW), with Z. jujuba Miller var. inermis extracts. JW significantly reduced the secretion of interleukin-8 and JE reduced the secretion of tumor necrosis factor-alpha in human keratinocyte HaCaT cells. Also, hyaluronidase activity was measured by enzyme assay and the fractions inhibited the activity in a dose-dependent manner. In addition, the human dermal fibroblast, HDF-n cells were treated with the extracts and antioxidant activities were measured. The results showed that the extracts increased the free radical scavenging activity and the superoxide dismutase activity. Taken together, Z. jujuba Miller var. inermis extracts reduced the secretion of AD-related cytokines and inhibited the hyaluronidase. In addition, the extracts showed antioxidant activity.

비타민 C 보충 및 영양 교육이 흡연 남자 청소년의 영양지식, 식행동과 체내 일부 항산화 관련 효소체계에 미치는 영향 (The Effects of Vit C Supplementation and Nutrition Education on Nutrition Knowledge, Food Habits and Antioxidative Enzyme Activity in Male Adolescent Smokers)

  • The study evaluated the effectiveness of intervention for male adolescent smokers by making an assessment in terms of changes in food habits, nutrition knowledge, plasma catalase, superoxide dismutase(SOD), glutathione peroxidase peroxidase(GSH-px) activities and thiobarbituric acid reactive substance(TBARS) after Vit C supplementation and nutrition education. The subjects, male adolescent smokers, were assigned into four groups : Control group(19 students), education(Educ.) group(19 students), Vit C supplementation (suppl) group(19 students), and Educ. + Vit C suppl. group(19 students). The Educ. group and Educ. + Vit C suppl. group received nutrition education once a week for 2 weeks. The Vit C suppl. group Educ. + Vit C suppl. group received 500 mg ascorbic acid for 35 days. All data were collected before intervention and after intervention. Nutrition knowledge of those who received education increased, and the frequency of fruit and yellow-green vegetable consumption also increased. Plasma antioxidant enzyme activities were not different except for the SOD activity in the Educ. + Vit C suppl. group, which was significantly increased. The plasma ceruloplasmin level of groups that received Vit C supplementation was reduced more than any other groups, and the specific ceruloplasmin ferroxidase activity of groups that received Vit C supplementation was elevated more than other groups. These intervention programs had an impact on food habits, nutrition knowledge, plasma antioxidant enzyme activities, and plasma TBARS in male adolescent smokers. Various nutrition education programs must be implemented for adolescent smokers, and further studied are needed regarding sorts and amount of antioxidant nutrients and supplementation duration.

콩나물 당 침지액의 항산화 효능 및 알코올 분해 효소 활성 연구 (Study of the Antioxidant and Alcohol-degrading Enzyme Activities of Soybean Sprout Sugar Solutions)

  • 콩나물은 숙취해소에 좋은 식품으로 잘 알려져 있으나 콩나물 특유의 비린 맛으로 인해 가공식품 원료로 이용하기에 어려움이 있다. 이에 숙취해소효과로 잘 알려진 설탕 및 올리고당과 같은 당을 이용하여 콩나물의 비린 맛을 없애고 섭취를 용이하게 하며 항산화 효과 증진 및 숙취해소에 긍정적인 효과를 기대하고 실험을 설계하였다. 콩나물에 설탕과 올리고당의 2종류 당액($50^{\circ}Bx$)을 25, 50, 75%(w/w) 비율로 첨가하여 1, 3, 6, 12, 24시간 침지하였으며, 침지한 콩나물의 여액을 여과하여 콩나물 당침용액을 샘플로 사용하였다. 세포내 산화적 스트레스 억제 효과를 제외한 모든 실험 결과에서 올리고당이 대조구나 설탕 침지보다 DPPH radical 소거능, 알코올 분해능이 좋은 것을 확인하였다. 총페놀, 총플라보노이드 함량 및 ADH ALDH 효소 활성 실험 결과, 당액 첨가비율 25%에 비해 50%에서 유의적으로 높은 활성을 띄었고, 50%와 75%에서는 유의적인 차이가 발생하지 않았다. 추출시간에 따른 항산화능과 알코올 분해능은 시간 증가에 따른 일정한 증가 추세는 나타나지 않았다. 추출 시간만을 고려하였을 때, DPPH radical 소거능 및 총페놀, 플라보노이드 함량과 알코올 분해능 실험에서 1-3시간 추출 시 대부분 대조구보다 높은 활성을 보였고 추출 시간이 길어질수록 오히려 활성이 떨어지는 경우가 더 많았다. 이러한 결과를 종합할 때 콩나물을 올리고당 당액($50^{\circ}Bx$) 50% (w/w) 비율 이상의 수준에서 단시간 침지하여 추출하는 것만으로도 항산화능과 알코올 분해능이 좋은 콩나물 추출액을 얻을 수 있을 것으로 판단된다. 본 연구를 통하여 콩나물을 이용한 숙취해소 음료를 제조하기 위하여 과량의 당액을 사용함으로서 발생할 수 있는 비용적인 절감의 근거를 마련할 수 있을 것으로 생각된다.

담쟁이덩굴의 항산화 방어계의 탐색 (Screening of the Antioxidant Defense Systems from Parthenocisuss tricuspidata PLANCH)

  • 담쟁이 덩굴 추출물로부터 항암예방제로서의 물질을 탐색코자 항산화 비효소활성 및 효소활성을 조사해 본 결과, 부위별 항산화력 및 효소활성들은 차이를 나타내었다. 부위별 에틸아세테이트 분획물은 잎에서 DPPH 소거활성이 가장 높았으며, 실리카겔 컬럼크로마토그래피에서 잎, 줄기, 뿔리에서 각각 7.06, 6.99, 12.39($\mu\textrm{g}/m\ell$)이었다. 에틸아세테이트 분획에서 활성이 가장 높은 분획을 Sephadex LH-20 column chromatography로 분리 정제한 결과 잎, 줄기 그리고 뿔리에서 각각 5.5, 6.11, 5.29 ($\mu\textrm{g}/m\ell$)의 DPPH 활성이 높은 분획을 얻었다. Sephadex LH-20에 의해 활성이 가장 높은 분획을 GC/MS로 분석한 결과, Benzoic acid(Gallic acid), 1,2,3-Benzenetriol, 4-amino-Benzenemethanol과 같은 페놀화합물의 항산화물질들이 동정되었다. 줄기와 뿌리에서 POD활서은 잎에서 보다 활성이 높았고, SOD 활성은 잎, 줄기 그리고 뿌리 순으로 높은 활성을 나타내었으며 특히, 잎은 뿌리보다 2배 이상의 높은 활성을 보였다.

Biocide sodium hypochlorite decreases pigment production and induces oxidative damage in the harmful dinoflagellate Cochlodinium polykrikoides

  • The biocide sodium hypochlorite (NaOCl) is widely used for controlling algal growth, and this application can be extended to marine environments as well. This study evaluates the biocidal efficiency and cellular toxicity of NaOCl on the harmful dinoflagellate Cochlodinium polykrikoides, with emphasis on pigment production and antioxidant enzyme activity. The test organism showed dose-dependent decrease in growth rate on exposure to NaOCl, and the 72 h $EC_{50}$ was measured to be $0.584mg\;L^{-1}$. NaOCl significantly decreased pigment levels and chlorophyll autofluorescence intensity, indicating possible detrimental effects on the photosystem of C. polykrikoides. Moreover, it significantly increased the activities of antioxidant enzymes, suggesting the production of reactive oxygen species in the cells. These data indicate that NaOCl exerted deleterious effects on the photosynthetic machinery and induced oxidative damage in the dinoflagellate and this biocide could be effectively used for the control of algal blooms.

Phaleria macrocarpa Suppress Nephropathy by Increasing Renal Antioxidant Enzyme Activity in Alloxan-Induced Diabetic Rats

  • The protective effects of Phaleria macrocarpa (PM) against oxidative stress in diabetic rats were investigated. Diabetes was induced in male Sprague Dawley rats using alloxan (150 mg/kg i.p). After the administration of PM fractions for two weeks the diabetic symptoms, nephropathy and renal antioxidant enzymes were evaluated. The results showed that the oral PM treatments reduced blood glucose levels in diabetic rats. The PM fractions decreased kidney hypertrophy and diminished blood urea nitrogen (BUN) in diabetic rats. Malondialdehyde (MDA), a lipid peroxidation marker, was increased in diabetic animals, but was suppressed by the PM treatments. In addition, the superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) activities, and glutathione (GSH) level in the alloxan-induced diabetic rats were significantly decreased compared with those in the normal rats, but were restored by PM treatments. The PM fractions also suppressed the level of MDA in the kidney. In conclusion, the anti hyperglycemic and anti-nephropathy of P. macrocarpa may be correlated to the increased renal antioxidant enzyme activity in the kidney.

Antioxidant Enzyme Activity in Rat Liver and Kidney Related to Coix Intake

  • The effects of dietary Coix(lacryma-jobi) water extract on the antioxidant enzyme activity in the liver and kidney of Sprague-Dawley rats were studied. Forty-five rats were fed for 3 weeks with either control diet or experimental diets that contain either Coix water extract or Coix water residue. Twenty percent of the carbohydrate was replaced with Coix water residue by dry weight in the water residue diet, while distilled water was replaced by Coix water extract to make a pellet-form diet in the Coix water extract diet. The levels of glutathione, glutathione-peroxidase, and glutathione-S-transferase activities in liver and kidney were measured . It has been found that glutathione, glutathione peroxidase, and glutathione-S-transferase enzyme activities from activities from liver and kidneyof the rats were enhanced in the group fed with Coix water extract.

운동으로 유발된 산화 스트레스와 마늘의 항산화 작용 (Antioxidant Effect of Garlic Supplement against Exercise-Induced Oxidative Stress in Rats)

  • This study was to investigate lipid peroxidation, antioxidant enzyme activity and DNA damage after exercise, and the protective effect of garlic against exercise-induced oxidative stress. Male Sprague-Dawley rats(4 weeks old) were randomly divided into three groups of 6 rats each; control group(Con) without garlic and exercise, Ex group with exercise alone, and Ex-G group with 2% garlic and exercise. For 4 weeks, rats were given diets containing 15% corn oil and 1% cholesterol with or without garlic. The swimming was selected as a model for exercise performance. Rats swam for 40 min a day, for 5 days a week. Group Ex and Ex-G showed significant lowering in body weight gain and fat accumulation compared to control. No significant changes were observed in levels of plasma cholesterol and triglyceride among three groups, demonstrating that exercise and garlic had no effects on changes of blood lipid. This finding of blood lipid seems to be due to higher plant sterol content in corn oil. The DNA tail moment of lymphocytes showed greater tendency in Ex and Ex-G than in control, but garlic supplements failed to suppress DNA damages. Compared to control, Ex had higher plasma TBARS which was lowered to the control's level in Ex-G with 2% garlic supplementation(p<0.05). Ex-G led to a higher hepatic superoxide dismutase(SOD) activity than control and Ex(p<0.05). Activity of hepatic catalase was also increased in Ex-G, while in Ex it was significantly low(p<0.05). It seemed that TBARS levels were related to the activities of SOD and catalase, and that garlic contributed to increasing the enzyme activities and led to decrease of TBARS. These results demonstrate that lipid peroxidation and DNA damage occur as a consequences of oxidative stress after exercise, and that antioxidant defense against oxidative stress could be enhanced by garlic supplementation through the induction of antioxidant enzymes. However, further investigations should be done on the garlic effect on DNA damage.
