Potential antioxidant activities of methanol and water extracts of Cirsium japonicum var. ussuriense (CJ) leaves were examined. The reducing power and hydroxyl raical-scavenging activity assays showed that the methanol extract had a significantly higher activity than the water extract. In addition, the methanol extract showed a concentration-dependent reducing power, ranging from 0.228 to 1.072($0.1{\sim}0.5\;mg/mL$), as well as a high DPPH free radical-scavenging activity($EC_{50}=40.25\;{\mu}/mL$). The total phenolic(as tannic acid) and flavonoid(as quercetin) contents of the extract were 62.41 mg/g and 13.48 mg/g, respectively. The cytotoxic activity indicated that the methanol extract has an inhibition activity in the stomach carcinoma cell (35.40%), suggesting that the methanol extract of CJ leaves could be used as a potential source of pharmaceutical material.

The anti-cardiovascular effect of estrogen replacement therapy in postmenopausal women is known to be associated with its role as an antioxidant, its ability to protect cells from DNA damage. Genistein concentrated polysaccharide (GCP) is a functional food produced by fermentation of soybean isoflavone extracts with Basidiomycetes, containing rich content of genistein aglycones. The aim of this study was to investigate the effect of GCP on oxidative DNA damage and plasma total antioxidant potential, comparing to the effect of estrogen.(omitted)
Coronavirus disease-19 (COVID-19) is caused by the severe acute respiratory syndrome coronavirus 2 (SARS CoV-2). Currently, the management of patients with COVID-19 depends mainly on repurposed drugs which include chloroquine, hydroxychloroquine, lopinavir/ritonavir, ribavirin, remdesivir, favipiravir, umifenovir, interferon-α, interferon-β and others. In this review, the potential of Nigella sativa (black cumin seeds) to treat the patients with COVID-19 analyzed, as it has shown to possess antiviral, antioxidant, anti-inflammatory, anticoagulant, immunomodulatory, bronchodilatory, antihistaminic, antitussive, antipyretic and analgesic activities. Medline/PubMed Central/PubMed, Google Scholar, Science Direct, Directory of open access journals (DOAJ) and reference lists were searched to identify articles associated with antiviral and other properties of N.sativa related to the signs and symptoms of COVID-19. Various randomized controlled trials, pilot studies, case reports and in vitro and in vivo studies confirmed that N.sativa has antiviral, antioxidant, anti-inflammatory, immunomodulatory, bronchodilatory, antihistaminic, antitussive activities related to causative oraganism and signs and symptoms of COVID-19. N. sativa could be used as an adjuvant therapy along with repurposed conventional drugs to manage the patients with COVID-19.
Hydrolysates of Trianthema portulacastrum in acidified methanol were evaluated for their total phenolic (TP) constituents and respective antioxidant activities using in vitro assays (i.e., 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, percent inhibition of linoleic acid peroxidation, and ferric reducing power). The observed results indicate that root, shoot, and leaf fractions of T. portulacastrum contain 50.75~98.09 mg gallic acid equivalents/g dry weight of TP. In addition, these fractions have substantial reducing potentials (0.10~0.59), abilities to inhibit peroxidation (43.26~89.98%), and DPPH radical scavenging capabilities ($6.98{\sim}311.61{\mu}g/mL$$IC_{50}$). The experimental data not only reveal T. portulacastrum as potential source of valuable antioxidants, but also indicate that acidified methanol may be an ideal choice for the enhanced recovery of phenolic compounds with retained biological potential for the food and pharmaceutical industry.
The purpose of this study was to purify antioxidant substances from steamed squid extract (SSE). The yield of SSE was 8% by dry weight. The approximate compositions of SSE proteins, lipids, moisture, carbohydrate and ash were 64.95%, 1.69%, 7.23%, 4.44% and 21.69%, respectively. The major amino acids in SSE were taurine (29.17%), glycine (20.33%), alanine (12.51%), and glutamic acid (9.83%). Antioxidant activities were evaluated using 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity, which was measured as 24.7% at 1.0 mg/mL. Four SSE fractions were isolated by Sephadex G-25 gel chromatography; the F2 fraction showed the highest DPPH radical scavenging activity. The F2 fraction was separated by reverse-phase high performance liquid chromatography (HPLC) using an octadecylsilane (ODS) column, yielding a purified antioxidant substance with a DPPH radical scavenging activity of 64.41% at 1.0 mg/mL, representing a 2.64-fold increase in the scavenging activity of SSE purified by the 3-step procedure. The amino acid compositions showed that purified SSE was rich in taurine, glycine, glutamic acid and alanine. The purified SSE significantly elevated 2',7'-dichlorodihydrofluororescein diacetate (DCFH-DA) fluorescence probe, which confirms its effective radical scavenging potential in cellular ROS. In addition, the SSE significantly inhibited oxidative damage of purified genomic DNA. These results suggest that a purified antioxidant substance from SSE can be used as a potential natural compound-based antioxidant in the functional food and pharmaceutical industries.
The antioxidant properties of 46 lichen species, collected from the highly UV-exposed alpine areas of southwestern China, were evaluated for their potential therapeutic utilization. The anti-linoleic acid peroxidation activity, 1,1-diphenyl-2-picryl-hydrazyl (DPPH) scavenging activity, reducing power, and total phenolic contents were all assessed in vitro in the methanol extract of the lichens. A potent reducing power was detected in a number of the lichen extracts, when compared with butylated hydroxyanisole (BHA). In general, it was found that many of the lichens, with antioxidant properties, contained large quantities of phenolic content. Extracts of Peltigera praetextata and Sticta nylanderiana were found to exhibit the most potent activity in all of the antioxidant tests. In particular, extracts of S. nylanderiana displayed a 1.37 times greater anti-linoleic acid peroxidation activity, when compared with the ascorbic acid used as the positive control. S. nylanderiana also possessed the strongest free radical scavenging activity amongst all the tested species, with an inhibition rate of 90.4% at concentration of $330{\mu}g/ml$. Activity-guided bioautographic TLC and HPLC analyses were used to establish which compounds were responsible for the potent antioxidant activities of the S. nylanderiana extract. These analyses revealed lecanoric acid to be primarily responsible for the effective antioxidant properties of S. nylanderiana. Overall, these results have indicated that several highland lichens have the potential of being utilized as novel bioresources for naturally occurring antioxidant therapies.
Legumes have been important components of the human diet. They contain not only protein, starch, and dietary fiber, but also various phenolic compounds such as flavonoids and phenolic acids. The importance of phenolic compounds to human health is well known due to their antioxidant activities. In this study, three legumes (adzuki beans, common beans, and black soybeans) frequently cultivated in Korea were evaluated for their total phenolic content (TPC) and antioxidant activities using DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2'-azinobis (3-ethylbenzothiazoline 6-sulfonate)), and FRAP (ferric reducing antioxidant potential) assays. In addition, correlations between agricultural traits and antioxidant activities of these three legumes were analyzed. Antioxidant activities assessed by DPPH, ABTS, and FRAP assays and TPC showed wide variations among legumes types and accessions. Among the three legumes, adzuki beans showed higher TPC and antioxidant activity than the other two legumes. In correlation analysis, seed size showed negative correlations with antioxidant activities and TPC. In principal component analysis and hierarchical clustering analysis, each of the three legumes was clearly separate. Results of this study can be used as basic information for developing functional materials for each legume. They can also help us understand the overall antioxidant activity of the three legumes.
The antioxidant activity of hot water extracts of various parts, the leaf, stem, and root of Synurus deltoides was evaluated by various antioxidant assays, including total phenolic content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, hydroxyl radical (${\cdot}OH$) scavenging, superoxide dismutase (SOD), and xanthine oxidase (XOI) activities. The various antioxidant activities were compared with the standard antioxidants such as L-ascorbic acid, $\alpha$-tocopherol, and butylated hydroxyanisole (BHA). Among the different plant parts, stem has been found to possess the highest activity in all tested model systems, the activity decreased in the order stems>roots>leaves. These results indicate that stem extract could be used as potential source of natural antioxidant.
Free radical scavengers or quenching agents for reactive oxygen species (ROS) present in consumable fruits, vegetables, and beverages have received considerable attention as potential antioxidants, and thus uses for treatment of several human diseases. In this study, grapevine shoot extract (GSE) containing high concentration of resveratrol and viniferine was evaluated for antioxidant potential and inhibition of pro-inflam-matory mediator production. Utilizing 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity and xanthine oxidase (XOD) inhibition assay the GSE showed inhibitory effects of DPPH radical scavenging and XOD activity with the $IC_{50}$/ values of 34.5 and 155 $\mu\textrm{g}$/ml, respectively. In addition, GSE also exhibited the inhibition of prostaglandin E$_2$ (PGE$_2$) and nitric oxide (NO) production in lipopolysaccharide (LPS)-induced mouse macrophage RAW264.7 cells with the $IC_{50}$/ value of 6.4 and 14.5 $\mu\textrm{g}$/ml, respectively. This result suggests that grapevine shoot extract has the potential activity as a natural antioxidant or antiinflammatory agent.
Strawberry is widely consumed in diet and has been attracted much attention due to its potential for human health benefits. Strawberry contains a diverse range of phytochemicals but the biological activities with molecular mechanisms are poorly elucidated yet. In this study, the effects of the extracts of strawberry (Maehyang cultivar) on antioxidant, antiinflammatory, and antiproliferative potential against various cancer cells were investigated. The strawberry extracts (SE) of Maehyang cultivar showed 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activities. In addition, SE inhibited the growth of human colon (HCT-116), lung (A549), stomach (SNU-638) and fibrosarcoma (HT-1080) cancer cells. The strawberry extracts also exhibited the inhibitory effect on lipopolysaccharide (LPS)-stimulated nitric oxide (NO) production and suppressed LPS-induced inducible nitric oxide synthase (iNOS) protein and mRNA expression in mouse macrophage RAW 264.7 cells. These findings suggest that the strawberry extracts (Maehyang cultivar) might have antioxidant, antiinflammotry, and anticancer activities.
