Purification and Identification of a Natural Antioxidant Protein from Fertilized Eggs

  • Fertilized hen eggs are rich in a variety of bioactive ingredients. In this study, we aimed to obtain an antioxidant protein from fertilized eggs and the radical scavenging abilities on 1, 1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical (${\bullet}OH$), superoxide anion ($O^{2-}{\bullet}$) were used to evaluate the antioxidant activity of the purified protein. During 20 d of incubation, the radical scavenging ability of protein extracted from fertilized eggs exhibited significantly differences and the protein on day 16 showed higher antioxidant capacity. Based on this, the antioxidant protein of the samples on day 16 were isolated for the follow-up study. With a molecular weight 43.22 kDa, the antioxidant protein was purified by Diethylaminoethyl cellulose -52 (DEAE-52) column and Sephadex G-100. The LC-MS analysis showed that the purified protein molecular weight was 43.22 kDa, named D2-S. The sequence of amino acids was highly similar to ovalbumin and the coverage reached to 84%. The purified protein showed a radical scavenging rate of $52.34{\pm}3.27%$ on DPPH and $63.49{\pm}0.25%$ on ${\bullet}OH$, respectively. Furthermore, the C-terminal amino acid sequence was NAVLFFGRCVSP, which was consistent with the sequence of ovabumin. These results here indicated that purified protein may be a potential resource as a natural antioxidant.

추출용매에 따른 참죽나무 순 분말 추출물의 항산화 활성 (Antioxidant Activities of Cedrela sinensis Tender Leaf Powder Extracts obtained from Different Solvents)

  • 본 연구에서는 전통 식품 소재로의 활용성을 높이기 위해 참죽나무 순을 동결건조하여 일반성분과 무기질 함량을 측정하였고, 추출용매를 달리하여 얻은 열수 추출물과 70% 에탄올 추출물로부터 총 폴리페놀 및 플라보노이드 함량과 항산화능을 살펴보았다. 참죽나무 순의 일반성분 및 무기질 분석 결과, 조단백질, 조섬유, 칼슘과 칼륨 함량이 높게 측정되었다. 총 폴리페놀과 플라보노이드는 참죽나무 순 분말 열수 추출물보다 70% 에탄올 추출물에서 함량이 더 높았으며, DPPH 유리 라디칼 소거능과 FRAP 또한 70% 에탄올 추출물에서 더 높은 항산화력을 나타냈다. 이상의 결과로 볼 때 참죽나무 순 분말은 영양성과 식이섬유소의 기능성이 높아 식품의 재료로 이용가능성이 높으며, 참죽나무 순 분말로부터 유용성분 용출 및 기능성 증대를 위해서는 70% 에탄올로 추출하는 것이 바람직하다고 생각된다.

Antioxidant activities of Erythrina stricta Roxb.using various in vitro and ex vivo models

  • Erythrina stricta, a deciduous tree widely used traditionally in indigenous system of medicine for various ailments such as rheumatism, fever, leprosy, epilepsy etc. The leaves of Erythrina stricta was extracted with ethanol (70%) and used for the evaluation of various in vitro antioxidant assays which includes H - donor activity, nitric oxide scavenging, superoxide anion scavenging, reducing ability, hydroxyl radical, hydrogen peroxide scavenging, total phenolic content, total flavonoid content, total antioxidant activity by thiocyanate and phosphomolybdenum method, metal chelating, $\beta$-carotene bleaching, total peroxy radical assays. The pro-oxidant activity was measured using bleomycin-dependent DNA damage. Ex vivo models like lipid peroxidation and erythrocyte haemolysis were also used to study the antioxidant property of the extract. The various antioxidant activities were compared with suitable standard antioxidants such as ascorbic acid, butylated hydroxyl toluene, $\alpha$-tocopherol, curcumin, quercetin and Trolox. The generation of free radicals viz. $O_2^{{\cdot}-}$, $OH^{\cdot}$, $H_2O_2$, $NO^{\cdot}$ and peroxyl radicals were effectively scavenged by the ethanolic extract of Erythrina stricta. In all the methods, the extract offered strong antioxidant activity in a concentration dependent manner. The total phenolic content, flavonoid content and total antioxidant activity in Erythrina stricta were determined as microgram (g) pyrocatechol, quercetin and $\alpha$-tocopherol equivalent/mg respectively. The extract did not exhibit any prooxidant activity when compared with ascorbic acid. The results obtained in the present study clearly indicates that Erythrina stricta scavenges free radicals and reduces lipid peroxidation, ameliorating the damage imposed by oxidative stress in different disease conditions and serve as a potential source of natural antioxidant.

청심연자탕(淸心蓮子湯)의 항산화(抗酸化) 효과(效果)와 기전(機轉)에 관한 연구(硏究) (Antioxidant effects and its mechanism of Cheongsimyeonjatang in Sasang Constitutional Medicine)

  • To investigate the antioxidant capacity of traditional Korean herb medicines, water extracts from 42 species which has been used for the Taeum constitution of Sasang Medicine, were tested on their antioxidant activity using radical scavenging effects against ABTS. Some Of them showed strong antioxidant capacities at $50{\mu}g$ concentration. And, antioxidant capacities of 2 prescriptions extract of Sasang Medicine, Cheongsimyeonja-tang and Yeoldahanso-tang, in the different concentration ($10{\mu}g$, $50{\mu}g$, and $100{\mu}g$) were determined. At the same time, the antiperoxidation effects of these 2 prescriptions extract were determined. Lipid peroxidation in brain homogenates induced by NADPH and $ADF-Fe^{2+}$ was strongly inhibited by Cheongsimyeonja-tang in vitro. The above-mentioned 2 prescriptions are a potent antioxidant capacity and antiperoxidation activity, further investigation into the in vivo antioxidant therapeutic potential for treatment of human disorders in brain tissue.

Screening of antioxidant and antimicrobial activities of Caesalpinia bonducella Flem., leaves (Caesalpiniaceae)

  • The study was aimed at evaluating the antioxidant and antimicrobial activities of methanol extract of Caesalpinia bonducella leaves (MECB) (Family: Caesalpiniaceae). The effect of MECB on antioxidant activity, reducing power, free radical scavenging (DPPH radical, nitric oxide radical, superoxide anion radical, hydroxyl radical and hydrogen peroxide radical scavenging), total phenolic content and antimicrobial activities were studied. The antioxidant activity of MECB increased in a dose dependent manner. About 50, 100, 250 and 500 g of MECB showed 53.4, 61.2, 69.1 and 76.2 % inhibition respectively on peroxidation of linoleic acid emulsion. Like antioxidant activity, the effect of MECB on reducing power increased in a dose dependent manner. The free radical scavenging activity of MECB was determined by DPPH radical scavenging method. The potency of this activity was increased with increased amount of extract. MECB was found to inhibit the nitric oxide radicals generated from sodium nitroprusside $(IC_{50}\;=\;102.8\;g/ml)$ whereas the $IC_{50}$ value of curcumin was 20.4 g/ml. Moreover, the MECB was found to scavenge the superoxide generated by photoreduction of Riboflavin. MECB was also found to inhibit the hydroxyl radical generated by Fenton reaction, where the $IC_{50}$ value is 104.17 g/ml compared with catechin 5 g/ml, which indicates the antioxidant activity of MECB. The MECB capable of scavenging hydrogen peroxide in a concentration-dependent manner. The amounts of total phenolic compounds were also determined. Antimicrobial activities of MECB were carried out using disc diffusion methods with five Gram positive, four Gram negative and four fungal species. The results obtained in the present study indicate that MECB leaves are potential source of natural antioxidant and antimicrobial agents.

볶음처리에 의한 자색고구마의 항산화 증진 효과 (Enhances Antioxidant Effect of Purple Sweet Potato by Roasting)

  • 자색고구마는 항당뇨, 항염증, 및 항산화 활성 등의 다양한 생리활성 효과가 있다. 본 연구에서는 자색고구마의 볶음 처리 후 DPPH, ABTs 및 FRAP를 분석하여 항산화 활성을 비교하였다. 부가적으로 우리는 볶음 처리에 의한 자색 고구마의 항산화 활성 변화 요인을 확인하기 위해 총 phenolics와 flavonoids 함량, 갈변물질 및 phenolic acids 함량을 측정하였다. 볶은 자색고구마는 볶지 않은 자색고구마보다 상당히 높은 항산화 활성을 나타내었다. 특히, $200^{\circ}C$에서 10분간 볶음처리 한 자색고구마는 다른 처리 조건들보다 높은 항산화 활성을 나타내었다. 볶음 처리 후 총 phenolic와 flavonoid 함량, 갈변물질 및 phenolic acids 함량이 현저하게 증가하였고, 이에 상응하여 항산화 활성이 증가하였다. 이 결과로부터 볶음 처리한 자색고구마는 식품에서 잠재적인 천연 항산화제로 사용할 수 있을 것으로 판단되었다.

수확시기가 참외의 항산화 활성 및 기능성 성분에 미치는 영향 (Effect of Harvest Time on Antioxidant Activity and Functional Constituents of Oriental Melons)

  • 수확시기에 따른 참외 부위별 항산화 활성과 기능성 성분의 변화를 측정하기 위해 본 연구를 실시하였다. 참외를 2년에 걸쳐 주요 수확시기(6월, 7월, 8월)별로 수확하여 껍질, 과육, 태좌 세 부분으로 나눈 다음 에탄올로 추출하여 항산화 활성 및 기능성 성분 함량을 측정하였다. 기능성 성분 함량을 측정한 결과 2008, 2009년 껍질, 과육, 태좌 모든 부위에서 총 페놀, 총 플라보노이드 함량이 6월에 수확한 경우 가장 높았고, 2008년 껍질을 제외한 나머지 부위에서는 7월에 수확한 참외에서 기능성분 함량이 가장 낮았다. ABTS와 FRAP를 이용한 항산화 실험 결과도 마찬가지로 6월에 수확한 참외의 모든 부위가 강한 항산화 활성을 보여주었고, 7월에는 활성이 급격히 떨어지는 것을 확인할 수 있었다. 이상의 연구결과로 6월에 수확하는 참외가 기능성분 함량이 높고 품질이 좋은 건강 참외임을 알 수있었다. 수확시기에 따라 기능성 성분과 항산화 활성이 변화하는 연구결과는 제철 과일의 우수성을 보여주는 기초 자료로 이용될 수 있을 것이다.

Antioxidant activities of brown teff hydrolysates produced by protease treatment

  • Purpose: Various plants, herbal medicines, and marine foodstuffs have been used in kimchi preparation to improve its overall quality. Teff, which is rich in minerals and starches, facilitates stable blood glucose levels and is well-suited for use in gluten-free products; hence, it can be used to reinforce the mineral composition of kimchi. In this study, we probed the antioxidant activities of hydrolysates prepared by treatment of brown teff with three proteases under different conditions. Methods: The mineral composition of brown teff was determined by inductively coupled plasma spectrophotometry-mass spectrometry, and we established optimal hydrolysis conditions by determining the total phenol and flavonoid contents of teff hydrolysates obtained using three different proteases (protamax, flavourzyme, and alcalase), two different protease concentrations (1 and 3 wt%), and three different incubation times (1, 2, and 4 h). The antioxidant activity of the hydrolysates was further investigated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity, total antioxidant capacity (TAC), and ferrous reducing antioxidant power (FRAP) assays. Results: Brown teff was rich in I, K, Mg, and Ca, and the highest total phenol content ($24.16{\mu}g/mL$), total flavonoid content ($69.08{\mu}g/mL$), and TAC were obtained for 1 wt% protamax treatment. However, the highest DPPH scavenging activity and FRAP values were observed for hydrolysates produced by alcalase and flavourzyme treatments, respectively. Conclusion: Treatment of brown teff with proteases affords hydrolysates with significantly increased antioxidant activities and high total phenol and flavonoid contents, and these antioxidant activities of teff hydrolysates have the potential to enhance the quality and functionality of kimchi in future applications.

현미 품종별 에탄올 추출물의 항산화 성분 및 항산화 활성 (Antioxidant Compounds and Antioxidant Activities of Ethanolic Extracts from Brown Rice Cultivars)

  • This study investigated chemical components, antioxidant compounds, and activity of brown rice cultivars, to select good cultivar to be used for processing of mixed-rice in the food industry. Proximate compositions, phytic acid, phenolic compounds, and antioxidant activity of brown rice were significantly different among cultivars. Moisture, crude ash, fat, protein, and carbohydrate contents of brown rice were 9.51~12.82, 1.05~1.93, 1.84~6.24, 5.90~9.60 and 71.75~80.34 g/100 g, respectively. Phytic acid content of brown rice cultivars was 7.39~0.87 mg/g. Total polyphenol content of Joeunheukmi and Geonganghongmi cultivars, were 615.25 and $311.14{\mu}g\;GAE/g$, total flavonoid content was 267.75 and $100.67{\mu}g\;CE/g$, respectively. DPPH radical scavenging activity of Geonganghongmi, Joeunheukmi and Hyeugkwang cultivars was 89.17, 87.94 and 43.17%, ABTS radical scavenging activity was 113.57, 113.34, and $93.53{\mu}mol\;TE/g$, and ferric reducing antioxidant potential was 951.67, 1,075.75, and $508.33{\mu}M/g$, respectively. As a result, phenolic compounds and antioxidant activities of pigmented brown rice were high, and it could be used as a functional material.

Characterization of Mulberry Root Bark Extracts (Morus alba L.) Based on the Extraction Temperature and Solvent

  • Mulberry root bark is one of potential plant sources for antioxidant materials which can be used for the relief of oxidative stress. To explore the effects of solvent type and temperature on the structural characteristics and antioxidant activity of the root bark extracts, we prepared various extracts of mulberry root bark (Morus alba L.) using 0 - 100 % ethanol (EtOH) at RT - 100℃. EtOH concentration and temperature critically affected the extraction yields, the content of bioactive components, and antioxidant activity of the extracts. Use of high content of EtOH solvent and low temperature resulted in the low extraction yield. Meanwhile, it was revealed that the extract prepared using absolute EtOH at room temperature contained polyphenols and flavonoids with the highest contents among other extracts. Interestingly, the temperature differently affected the polyphenol and flavonoid contents according to the solvent types. In the case of 30% EtOH solvent, polyphenol and flavonoid contents increased with an increase in temperature, whereas in the case of 70 and 100 % EtOH, these contents decreased. Using the radical scavenging assay, it was confirmed that the 100% EtOH extracts had higher antioxidant activity compared to distilled water (DW) extracts regardless of temperature. Also, heating might extract more antioxidant components from the root bark. Especially, the extract prepared using 30% EtOH solvent at 100℃ showed the highest antioxidant activity. Taken together, these experimental results imply that the extraction parameters should be designed carefully considering the productivity, the extracted bioactive components, and antioxidant activity.
