The Cytotoxicity and Chemosensitizing Effects of native camellia(Camellia japonica) and nutraceutical camellia teas

  • The present study has been undertaken to characterize availability of camellia(Camellia japonica L.) as a medicinal plant with antineoplastic and chemosensitizing activities. The crude extracts from fresn camellia flower, young leaves and nutraceutical tea of camellia leaf and flower buds were evaluated on their potential activities against various human cancer cells and multidrug resistance to cancer cells in vitro. The range of cytotoxicity displayed from 120$\mu\textrm{g}$/mL to 200$\mu\textrm{g}$/mL. Catemix 1(CT-1) mixed with camellia and green tea showed high toxicity(respectively IC$\sub$50/=l16$\mu\textrm{g}$/mL, 129$\mu\textrm{g}$/mL) against AML-2/WT, acute myelogenous leukemia cell and MCF-7, brest adenocarcinoma pleual effusion cell. Generally camellia tea mixed with green tea showed higher cytotoxicity than the other camellia teas mixed with some herbs(CH). Methanol extract of steamed camellia tea and roasted camellia tea had a chemosensitizing effect to reverse Pgp-mediated MDR. In addition, camellia flower tea of insignificant cytotoxicity, chemosensitizing effect were increased remarkably chemosensitizing effect in mixed flower tea with some herbs.

  • Alcohol extracts of six plant namely Coix agrestis L$_{OUREIRO}$, Chloranthus Japonicus (Siebi) N$_{AKAI}$, Ajuga Spectabilis N$_{AKAI}$, Juglans mandsuria Max., Cirsium Macki var Ussuriense $K_{ITA}$ and Sasamorpha Purpurascense (Hack) N$_{AKAI}$ were subjected to the screening tests for anti-tumor activities against SN-36 Leukemia, Sarcoma 180 and Ehrlich ascites carcinoma. Of the six extracts, Ajuga Spectabilis N$_{AKAI}$ and Coix agrestis L$_{OUREIRO}$ were found to be potential in survival effect of tumor transplanted mice.

  • The present study has been undertaken to detect edible medicinal plants with antineoplastic property on the basic of a number of traditional Korean medical literatures, besides studies on development of anti-cancer medical wild plants growing in Korea and to prove experimentally their efficacy by in vitro and in vivo tests.235 species from 45 family 79 genus were screened primarily as edible sources of antitumor effect. Among those the crude. extracts of 40 spp. showed considerable cytotoxicity in vitro and especially Pegangkuen(Patrinia scabiosaefolia), Deod-eog(Codonopsis lanceolata), Okssusu(Zea may), and Geureong(Eragrositis ferru-ginea) exhibited significant antitumor activity against sarcoma 180 asites mice. However, additional researches should be mode for the confirmation of their availability as antitumor plants.

Effect of Podophyllotoxin Conjugated Stearic Acid Grafted Chitosan Oligosaccharide Micelle on Human Glioma Cells

  • Objective : To study the physiochemical characteristics of podophyllotoxin (PPT) conjugated stearic acid grafted chitosan oligosaccharide micelle (PPT-CSO-SA), and evaluate the ability of the potential antineoplastic effects against glioma cells. Methods : PPT-CSO-SA was prepared by a dialysis method. The quality of PPT-CSO-SA including micellar size, zeta potential, drug encapsulation efficiency and drug release profiles was evaluated. Glioma cells were cultured and treated with PPT and PPT-CSO-SA. The ability of glioma cells to uptake PPT-CSO-SA was observed. The proliferation of glioma cells was determined by 3-[4, 5-dimethyl-2-thiazolyl]-2, 5-diphenyl-2H-tetrazolium bromide (MTT) assay. The apoptosis and morphology of U251 cells were observed by 4',6-Diamidino-2-phenylindole dihydrochloride (DAPI) dye staining. Cell cycle analysis was performed by flow cytometry. The migration ability of U251 cells was determined by wound healing test. Results : PPT-CSO-SA had nano-level particle size and sustained release property. The encapsulation efficiency of drug reached a high level. The cellular uptake percentage of PPT in glioma cells was lower than that of PPT-CSO-SA (p<0.05). The inhibitory effect of PPT-CSO-SA on glioma cells proliferation was significantly stronger than that of PPT (p<0.05). The morphologic change of apoptosis cell such as shrinkage, karyorrhexis and karyopyknosis were observed. The percentage of U251 cells in G2/M phase increased significantly in the PPT-CSO-SA group compared with PPT group (p<0.05). Compared with the PPT group, the cell migration ability of the PPT-CSO-SA group was significantly inhibited after 12 and 24 hours (p<0.05). Conclusion : PPT-CSO-SA can effectively enhance the glioma cellular uptake of drugs, inhibit glioma cells proliferation and migration, induce G2/M phase arrest of them, and promote their apoptosis. It may be a promising anti-glioma nano-drug.

  • Recombinant interleukin-2 (IL-2) has demonstrated as an antineoplastic agent in mice and human, but the relatively low response rates observed in clinical trials. Therefore, the present study was undertaken in order to evaluate therapeutic activities of IL-2 for the establishment of therapeutic applications. At the onset of the experiment, normal C3H/HeN mice were injected with $3{\times}10^6$ RD-995 tumor cells, murine ultraviolet radiation-induced fibrosarcoma, subcutaneously. Beginning on day 25, experimental groups were treated with a 5-day course of IL-2 (subcutaneous injection of 30,000 IU every 12 hours for 5 days). The result of this experiment revealed that RD-995 tumor grew progressively in control mice. Subcutaneous IL-2 therapy decreased tumor growth until day 23, then the tumor grew progressively. No significant difference in the survival of IL-2 therapy decreased tumor growth until day 23, then the tumor grew progressively. 5-HT1A receptors mediate the analgesic effect of rosavin in a mouse model of oxaliplatin-induced peripheral neuropathic pain

  • Oxaliplatin, a third-generation platinum derivative, is the mainstay of current antineoplastic medications for advanced colorectal cancer therapy. However, peripheral neuropathic complications, especially cold allodynia, undermine the life-prolonging outcome of this anti-cancer agent. Rosavin, a phenylpropanoid derived originally from Rhodiola rosea, exhibits a wide range of therapeutic properties. The present study explored whether and how rosavin alleviates oxaliplatin-induced cold hypersensitivity in mice. In the acetone drop test, cold allodynia behavior was observed from days 3 to 5 after a single injection of oxaliplatin (6 mg/kg, i.p.). Cold allodynia was significantly attenuated following rosavin treatment (10 mg/kg, i.p.). Specific endogenous 5-HT depletion by three consecutive pretreatments with parachlorophenylalanine (150 mg/kg/day, i.p.) abolished the analgesic action of rosavin; this effect was not observed following pretreatment with naloxone (opioid receptor antagonist, 10 mg/kg, i.p.). Furthermore, 5-HT1A receptor antagonist WAY-100635 (0.16 mg/kg, i.p.), but not 5-HT3 receptor antagonist MDL-72222 (1 mg/kg, i.p.), blocked rosavin-induced analgesia. These results suggest that rosavin may provide a novel approach to alleviate oxaliplatin-induced cold allodynia by recruiting the activity of 5-HT1A receptors.

Pro-apoptotic Effects of Sanguisorbae Radix Ethanol-Extracts on Two Cholangiocarcinoma Cell Lines, SNU-1079 and SNU-1196 (담관암세포주에서 지유(地楡) 에탄올 추출물의 항암 효과)

  • Objectives : This study was performed to investigate the antineoplastic effect of ethanol extracts from Sanguisorbae Radix on cholangiocarcinoma cells that was established from biliary tract cancer tissue. Materials and Methods : Two cholangiocarcinoma cell lines, SNU-1079 and SNU-1196, were studied. The mRNA expression of Caspase 3, 8, 9, Bcl-2, Bax, P53, and P21 was examined by RT-PCR. Cell viability was determined by MTT assay. The cell cycle was analyzed by flow cytometry and apoptosis by cell death detection ELISA kit. Results : Proliferation of SNU-1079 and SNU-1196 was inhibited by Sanguisorbae Radix treatment in a dose-dependent manner. All cells treated with Sanguisorbae Radix showed increased dose- and time-dependent apoptosis. The expression of caspase 3, 8, 9, p53, and p21 was increased in all cells after the treatment of Sanguisorbae Radix. The expression of Bcl-2 was decreased in SNU-1196 and Bax expression was increased in all cells after the treatment of Sanguisorbae Radix. Conclusions : These results suggest that Sanguisorbae Radix would be beneficial in the treatment of cholangiocarcinoma.

Effect of Gleditsiae Spina on Hep G2 cells cytotoxicity and Apoptosis and No (조각자(皂角刺)의 간암세포주(Hep G2)에 대한 세포독성, Apoptosis 및 NO에 대한 실험)

  • In this study, antineoplastic activity against human hepatocellular carcinoma cell line(Hep G2) was tested in Gleditsiae Spina. Gleditsiae Spina was extracted with water, and the cytotoxic activity was tested using a calorimetric tetrazolium assay(MTT assay), the apoptosis was tested using a DNA electrophoresis and flow cytometry. The nitric oxide production from mouse peritoneal macrophage was tested using a Griess method. Gleditsiae Spina extracts against the proliferation of Hep G2 cells not showed cytotoxicity at the concentration of less than $100{\mu}g/ml$, and Gleditsiae Spina extracts not showed the cytotoxicity of mitomycin C and the cytotoxicity of cisplatin on Hep G2 cells. Gleditsiae Spina extracts aginist the proliperation of BALB/c 3T3 cells not showed cytotoxicity, the proliperation of mouse thymocytes and splenocytes not showed cytotoxicity at the concentration of less than $100{\mu}g/ml$. Gleditsiae Spina extracts not showed nitric oxide production from mouse peritoneal macrophage in vitro. Gleditsiae Spina was administered orally for 7 days at 300mg/kg increased nitric oxide production from mouse peritoneal macrophage.

Impressive effect of cisplatin monotherapy on a patient with heavily pretreated triple-negative breast cancer with poor performance

  • Systemic therapy for metastatic triple-negative breast cancer (TNBC) still remains challenging because there are no targeted agents or endocrine therapies currently available. The present case report documents the successful use of cisplatin monotherapy to manage a heavily pretreated TNBC patient showing poor response to therapy. The patient was a 51-year-old woman who had already undergone several lines of systemic chemotherapy for widespread TNBC. Although the mutation analysis performed on DNA isolated from blood cells and progressed lesion samples confirmed the tumor to be germline BRCA wild-type, cisplatin monotherapy was administered based on the increasing evidence of safety and efficacy of platinum for breast cancer. After three cycles of cisplatin treatment, the patient's metastatic lesions dramatically improved without any major toxicity, and she completed 17 cycles with good response. This case study indicates that patients with heavily pretreated TNBC can potentially achieve a good response to cisplatin monotherapy.

  • Purpose : This study was conducted to determine the relationships between conditioned variables and the quality of life in the gastric cancer patients receiving chemotherapy after surgery. Method : 56 gastric cancer patients receiving chemotherapy included Cisplatin and 5-Fu were recruited from one university hospital located in Seoul. Questionnaires for this study were Quality of life Index developed by Padilla et al. at 1983, Rhode's nausea and vomiting scale and tool for side effects after chemotherapy at home developed by authors. Result : Quality of life in this study was correlated significantly with age, stage of cancer, nausea and vomiting, and level of side effects of chemotherapy. Quality of life of woman was lower than that of man. Conclusion : It is suggested that nurses be reinforce nurse's discharge education for the patients about increasing oral intake and preventing side effect after discharge from hospital and pay more attention to the change of the quality of life and relevant variables among the gastric cancer patients.
