The inhibitory effect of Ramaria botrytis on the mutagenicity in Salmonella assay and cytotoxicity on cancer cell were studied. Ramaria botrytis methanol extracts showed antimutagenic effects of 60~90% on B(a)P and AFB1 in S. typhimurium TA98 and TA100. These extracts showed 73~85% antimutagenicity on TA100 against MNNG. The methanol extracts with strong antimutagenic activities were further fractionated by ethylacetate and water, the ethylacetate fraction were found to be stronger antimutagen icity against MNNG than water fraction. Ramaria botrytis methanol extracts and ethylacetate fraction revealed the highest cytoxicity against HT 29 human colon adenocarcinoma cells in which cell growth was inhibited by 57~88% and 69~94% at 0.25~1.0mg/ml, respectively. These methanol extract and ethyl acetate fraction exhibited 53~79% and 66~87% inhibition against HepG2 human hepatocarcinoma cells, respectively. But water fraction showed only 10~24% inhibition. However, these extract and fractions did not show cytotoxic effect against human chang liver cells. From these results, it is considered that Ramaria botrytis has stronger antimutagenic and anticancer effects in vitro.

Methanol extract of garlic was fractionated to chloroform and aqueous fractions. The cholorform fraction possessed the highest antimutagenic activity in Salmonella typhimurium TA 100 and TA98, and was further fractionaed into four Allium sativum chromatography fractions (ASC F1, 2, 3 and 4) by column and thin layer chromatographies. The ASC F2 exhibited the higher antimutagenic activity and contained 18 chemical compounds tentatively identifed by GC-MS, NMR and FT-IR. Among the 18 compounds, methyl linoleate was a major compound to exhibit the antimutagenicity.
The antimutagenic activity of a methanol extract of roasted Cassia tora seed against aflatoxin $B_1(AFB_1)$ was demonstrated with the Salmonella typhimurium assay. The numbers of revertants per plate decreased significantly when this extract was added to the assay system using Salmonella typhimurium TA100 and or TA98. The MeOH extract was then sequentially partitioned with $CH_2Cl_2$, EtOAc, n-BuOH, and $H_2O$. The $CH_2Cl_2$ and n-BuOH fractions possessed antimutagenic activity, but the EtOAc and $H_2O$ fractions were inactive. Both the MeOH extract and its fractions were capable of inhibiting the indirect-acting mutagen $AFB_1$, suggesting that these fractions may prevent the metabolic activation of $AFB_1$, or scavenge the electrophilic intermediate capable of inducing mutations. Column chromatography using silica gel yielded pure alaternin from the $CH_2Cl_2$ a fraction, and adenosine and isorubrofusarin gentiobioside from the n-BuOH fraction. Alaternin and isorubrofusarin gentiobioside demonstrated significant antimutagenic activities.
Free radicals formed during the metabolism of environmental chemicals are known to induce mutagenicity, while different types of antioxidants suppress this event. The purpose of this study was to determine the antioxidative and antimutagenic effects of soybean saponins, and to examine the relationship between these two effects for the elucidation of mechanisms involved in the anticarcinogenicity of soybean saponins. Also, antioxidative and antimutagenic effects of soybean saponins were compared with those of kinown antioxidants. For the measurement of antioxidative capacity, soybean saponins, L-ascorbic acid, $\alpha$-tocophoerol, and BHT at concentrations between 005 and 1.0mg/ml were tested for their ability to donate hydrogens and to reduce the formation of thiobarbituric substances(TBARS). Antimutagenic activity was examined using the Ames salmonella test system at concentrations of 600, 900 or 1200ug/ml. Study results showed soybean saponins and all of the other antioxidants tested possessed dose-dependent antioxidative activities. The ability of hydrogen-donation to DPPH was in the order of L-ascorbic acid>$\alpha$-tocopherol=>BHT>soybean saponins. TBARS formation was also inhibited by these compounds, in the order of BHT>$\alpha$-tocopherol=L-ascorbic acid>soybean saponins. Soybean saponins and other antioxidants also showed antimutagenicity in a dose-dependent manner. Especially, soybean saponins and BHT were excellent antioxidants compounds, inhibiting near 80% of the mutagenic effects at a concentration of 1200ug/ml. The correlation coefficients between antioxidative capacity and antimutagenicity for each compund was statistically significant at p<0.05. These results indicate that soybean saponins possess antioxidative and antimutagenic capacities. Also, antimutagenicity of saponins and other antioxidats is partly due to their antioxidative activities.

Antimutagenic effects of kale juice on the mutagenicity induced by $B_{1}(AFB_{1})$ N-methyl-N'-N-nitrosoguanidine(MNNG) in Salmonella assay system were studied. The freeze dried kale juice significantly reduced the mutagenicity induced by $AFB_{1}$ in Salmonella typhimurium TA100 and TA98. However, the kale juice exhibited less inhigbitory effect on the mutagenicity induced by MNNG as the concentrations of the juice sample increased. Also, kale juice after dialysis (>12,000, Mw) appeared to have 42.3∼89.5% of inhibitory effects against $AFB_{1}$, however, the dialyzate did not show any inhibitory effect against MNNG. To separate and identify the antimutagenic compounds from the kale juice, the dialyzates were further fractioned by using Sepharose CL-6B-200 gel filtration. Fraction number 13 showed the strong antimutagenic activity against $AFB_{1}$, and the fraction exhibited positive results of a characterized colour reactions of protein, carbohydrate and phenolic compound. Therefore, one of the possible active compounds from the kale juice was supposed to a glycoprotein(Mw. 270,000) which seemed unstable with heating.
Antimutagenic effects of 5 kinds of kochujang(Korean red pepper soybean paste) samples compared with doenjang(Korean soy paste) were studied using the Ames test with Salmonella typhimurium TA100 and the SOS chromotest, with E. Coli PQ37. Th eantimutatenic effects of methanol extracts from red pepper powder and meju(fermented soybean) powder, the major ingredients of the kochujang,were also evaluated for the mutagenicity of aflatoxin B1 (AFB1) in the Ames assay. The methanol extracts from the kochujang samples showed lower antimutagenicities than those of doenjang against AFB1 and N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in the Ames assay. Traditional kochujang I and II exhibited strong antimutagenic activity against AFB1 and MNNG. The traditional kochujang samples against MNNG were aslo observed in the SOS chromotest system with the same fashions as shown in the Ames mutagenicity test. The methanol extracts from meju powder had the strongest inhibitory effects on mutabenicity induced by AFB1, however, those form red pepper powder showed lower inhibition rate than kochujang. These results suggest that traditional kochujang exhibit higher antimutagenic acitivity than the commercial variety, and that meju powder seems to be one of the major antimutagenic components in kochujang.

To enhance the antimutagenic and anticancer activities of chinese cabbage kimchi, 13 kinds of kimchi, which were different kinds and levels of sub-ingredient added kimchi, were prepared and fermented at 15℃ for 1 day and then at 5℃ up to pH 4.3. The antimutagenic effects of the methanol extracts of the kimchi were studied by using Ames mutagenicity test in Salmonella typhimurium TA100 and SOS chromotest in E. coli PQ37. Among the kimchi samples, high ratio of red pepper powder(7%) and garlic(2.8% or 5.2%) added kimchi, 1% chinese pepper powder added kimchi and organic cultivated chinese cabbage kimchi significantly reduced(p<0.05) the mutagenicity induced by aflatoxin B1(AFB1) in Ames test and SOS response against N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in SOS chromotest, and also the kimchi inhibited more effectively(p<0.05) the survival and growth of AGS human gastric adenocarcinoma cells than the standarized kimchi on the SRB assay, MTT assay and growth inhibition test. These results suggest that the antimutagenic and anticancer activites of kimchi can be increased by the sub-ingredients such as organic cultivated chinese cabbage, red pepper powder, garlic and chinese pepper powder.

The inhibitory effect of Coprinus comatus on the mutagenicity in Salmonella assay system and SOS chromotest were studied. In Ames test, the ethanol and water extracts and the cultured mycelia fractions of Coprinus comatus did not show any mutagenicity, but the Coprinus comatus ethanol extracts showed inhibitory effects of 8 0∼90% on the mutagenicity induced by indirect mutagen of benzo(a)pyrene(B(a)P) and aflatoxin B1(AFB1) in Salmonella typhimurium TA98 and TA100. The antimutagenic effect increased with increasing concentration of the ethanol extract toward N methyl N' nitro N nitrosoguanidine(MNNG). However, the water extracts inhibited about 40∼50% against direct and indirect mutagen. The cultured mycelial filtrate of Coprinus comatus, the fractionⅡ, showed antimutagenic effect of 90% on MNNG and 25∼50% on B(a)P and AFB1. In SOS chromotest, the ethanol extracts of Coprinus comatus showed antimutagenic effect of 65∼81% on SOS function induced by 4 NQO, and the cultured mycelia fractionⅡ showed low inhibitory effect of 20∼50%.
The study was carried out to evaluate the antimutagenic and anticancer effects of small water dropwort. The methanol extracts from small water dropwort significantly reduced the mutagenicity induced by aflatoxin $B_1\;(AFB_1)$ and N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) in Salmonella typhimutium TA 100. Also, the methanol extracts inhibited the growth of AZ-521 human gastric cancer cells and HT-29 colon cancer cells. The chloroform fraction from methanol extracts of small water dropwort inhibited $40\;to\;80\%$ of the mutagenicity by $AFB_1$ in Sal. typhimurium TA 100 by the addition of 2.5 to $10\%$. To separate active compounds, the chloroform fraction was subjected to column chromatography on a silica gel and separated into five fractions. Among the five fractions, fraction 4 showed the highest antimutagenic effect against $AFB_1$ and an anticancer effect in the HT-29 colon cancer cell. As the result of the analysis in GC-MS, 1-napthalene carbonitrile, 5,6,7,8-tetrahydrol and benzene, 1,1'-(1,4-pentadiene-1,5-diyl) bis-,(E,E) were identified potentially from fraction 4.

Melanoidins, as brown colored polymers, are formed through the diversified reaction systems of Maillard type and other reactions. Melanoidins are important components in relation to food quality and also are known to have antioxidative, mutagenic and antimutagenic activities. Since these aspects have been extensively reviwed elsewhere, only the recent studies regarding to their antioxidative and antimutagenic activities are discussed in this review. Even though their mechanisms are not clearly identified, melanoidins or specific fractions isolated from their mixtures have shown varied antioxidative activities depending on the reaction systems and reaction conditions. Those activities presumely are derived from the complex functional properties of hydrogen / electron donors and metal chelating power, which are originated from their reductone structure and others. It is considered that pyrolysate and other mutagens are formed by the given conditions in some cases during browning reaction, whereas melanoidins and their fractions have antimutagenic effects on chemical and other mutagens. There are positive correlationship among the color intensity, antioxidative activity and antimutagenicity of melanoidins or their fractions. These suggest that the antimutagenicity of melanoidins could be attributed to their antioxidative properties, however, it might also be due to other factors, because the relevant responses for antimutagenicity are very complicate and not clear. Accordingly, further studies are required to determine the actual acitivities and mechanisms involved in antioxidation and (anti)mutagenicity of melanoidins by reaction systems / conditions and by the isolated fractions. And also, additional studies are needed to evaluate the applications of melanoidins and their relevant effects to food and human health.
