The Antimicrobial Effects of Natural Aromas for Substitution of Parabens (합성 항균제를 대체하기 위한 천연물질의 항균 효과)

  • Aroma oils extracted from the natural material have antibacterial, antivirus, antiinflammatory, and preservative effect. The preserve efficacy testing between aroma oils and parabens as an artificial preservative had been performed and then it had been suggested that aroma oil was possibile to apply to the cosmetics. Aroma oils were pine, rosemary, lemon and eucalyptus, and parabens were methylparaben, blitylparaben. Antiseptic concentrations of aroma oils and parabens having 0.0, 0.1, 0.2, 0.4, 0.8, 1.0wt% were tested respectively. Escherichia coil(ATCC No.8739), Pseudomonas aeruginosa(ATCC No. 9027) which are gram-negative and Staphylococcus aureus (ATCC No. 6538), Bacillus subtilis(ATCC No. 6633) which are gram-positive were used as the test organisms. Disk paper and broth dilution methods were used as the methods of preservative efficacy testing. The antibacterial activity of aroma oils and parabens for gram-positive were better than that for gram-negative. For the antibacterial activity aroma oils were better than parabens. Among the aroma oils, rosemary and pine having superior antibacterial activity were selected and blended to illuminate if there is any synergy, There was synergical effect and optimum ratio of aroma blend is 3 : 1(rosemary pine) in this study.

  • The essential oils were extracted from the needles of several softwood tree species, Pinus densiflora, Pinus koraiensis, Chamaecyparis obtusa and Chamaecyparis pisifera to investigate their biological activities. The antifungal activity by hyphal growth inhibition method was determined by three wood rot fungi, one mouldous fungi, and three tree pathogenic fungi and the antibacterial activity by paper disc method and turbidiometric method by three gram positive bacteria and one gram negative bacteria, Escherichia coli, were also tested. Antifungal activity was the best in the essential oil from C. obtusa, and the following was resulted in order of P. densiflora and P. koraiensis. The essential oil from P. densiflora showed higher antifungal activity against wood rot fungi than that from P. koraiensis while the result was contrary against tree pathogenic fungi. However, the antibacterial activity was the best in the essential oil from P. densiflora, and the following was in order of C. obtusa and P. koraiensis. From these results, the variation of biological activity seemed to be due to the chemical composition of essential oils.

Study on the biological activity of Artemisia iwayomogi KITAMURA (한인진(韓茵蔯)의 생리활성에 관한 연구)

  • This study was carried out to investigate antioxidative, antimicrobial activity and the effect on hepatotoxicity in various extracts of Artemisia iwayomogi. The herb has been used widely for jaundice, hepatitis and liver cirrhosis in chinese medicine. Solid yield by various extraction solvents, 18.1%, was the highest in water extract. To find antioxidative activity in Artemisia iwayomogi was estimated radical scavenging effect by DPPH method in various extracts and change of the POV(peroxide value) of various extracts added in soybean oil during 20 days at $60^{\circ}C$. Radical scavenging effect by DPPH method was the most effective in methanol extract. Added 1,000ppm water extract and methanol extract in soybean oil, the POV of them, 46.8(meq/kg) and 50.8(meq/kg) were lower than that of control, 79.1(meq/kg), during 20 days storage. After antimicrobial activity of various extracts of Artemisia iwayomogi on bacteria was carried out by paper disc method, it found that the ethanol extract was the strongest activity on Vibrio parahaemolyticus. In vivo experiment was to investigate the effect of Artemisia iwayomogi water extract(AIWE) on hepatotoxicity by carbon tetrachloride$(CCl_4)$ in rats. The experiment groups were divided into five groups for recovery(for 3 days) and three groups for protection(for 10 days) in rat liver. The weights and morphological changes of liver and the body weight were examined in each groups. Compared with $CCl_4$ treatment groups$(CCl_4\;only)$, liver and liver/body(%) weights of AIWE pretreatment groups for 3 days and AIWE posttreatment groups for 10 days were declined. In macrography, fibrious exudates and swelling of liver were decreased in AIWE treatment groups. Accumulation of lipid droplets and necrosis of hepatocytes were also decreased in AIWE treatment groups in microscopically. In these results, AIWE seems to enhance hepato-protective and recoverable effect on $CCl_4$ induced hepatotoxicity in rats.

Antimicrobial Activity against Food Hazardous Microorganisms and Antimutagenicity against Salmonella serotype Typhimurium TA100 of an Ethanol Extract from Sanguisorba officinalis L. (지유 에탄올 추출물의 식품 위해성 세균에 대한 항균 활성 및 Salmonella serotype Typhimurium TA100에 대한 항돌연변이 활성 효과)

  • This study was performed to analyze the antibacterial activity against food hazardous microorganisms and antimutagenic effects of Sanguisorba officinalis L. ethanol extracts on Salmonella Typhimurium TA100. The antibacterial activity was evaluated by paper disc diffusion assay, minimum inhibition concentration (MIC), and optical density of the culture with the ethanol extract for 24 hr. Antibacterial activity was tested with seven microorganisms including Escherichia coli, Escherichia coli O157:H7, Pseudomonas aeruginosa, Salmonella Typhimurium, Listeria monocytogenes, Bacillus cereus, and Staphylococcus aureus. The paper disc diffusion assay showed distinct clear inhibition zones around the discs treated with the extract for five microorganisms, except Escherichia coli and Escherichia coli O157:H7. MIC values were 0.625-2.5 mg/mL for these five strains that showed clear zones. The time-kill assay was consistent with the results from the paper disc diffusion assay and MIC test. Additionally, antimutagenicity of the extract was determined using the Ames test. The ethanol extract at 5 mg/plate inhibited 72.42% and 89.85% of mutagenicity induced by 4-nitroquinoline 1-oxide and sodium azide, respectively. These results demonstrate that the ethanol extract from S. officinalis L. has remarkable antibacterial activity and antimutagenicity.

A Study on Antibacterial Effects of Ascorbic Acid against Mycobacterium tuberculosis (Ascorbic acid의 항결핵균 작용에 관한 연구)

  • Vitamin C is known as an integral element for the formation and maintenance of intercellular supporting structures. Ascorbic acid has been used for the treatment of certain poisonings and hypovitaminosis(scurvy) but also known as a powerful reducing agent, and can kill a variety of bacteria and detoxify bacterial exotoxins including various clostridial exotoxins in vitro. For viruses, vitamin C inactivates herpes virus, vaccinia virus and influenza virus and has been used for the prevention and treatment of the common cold. Thus ascorbic acid plays an important role in antimicrobial action. Scurvy also promotes the development of tubercles in experimentally infected guinea pig and the tuberculosis patients require more vitamin C than normal persons. However there is no reports that ascorbic acid could inhibit the growh of M. tuberculosis. In this paper, antibacterial effects of ascorbic acid against M. tuberculosis were studied. The results are as follows: 1. The single use of the ascorbic acid exhibited antibacterial effect in vitro against $5{\times}10^3/ml$ of M. tuberculosis $H_{37}$ Rv at the concentration of ascorbic acid 0.625mg/ml over 3 hours exposure and 0.05mg/ml over 9 hours exposure. 2. In vivo mice administered with ascorbic acid 50mg/day for 5, 10 and 15 days respectively were protected from M. tuberculosis $2LD_{50}$, $3LD_{50}$, $4LD_{50}$ and $5LD_{50}$ given intravenously.

Amino Acid-Based Material for the Complementary Therapy of Decubitus Ulcers

  • Chronic wounds, pressure sores, lesions, and infections of microbial origin in bedridden, paralyzed, or malnutrition patients remain the object of study of many researchers. A variety of factors behind the development of these disorders are related to the patient's immune system, making it unable to respond effectively to the treatment of the wound. These factors can be properly controlled, giving particular importance to the ethiology and stage of the wound, as well as the time periods corresponding to the replacement of the dressings. The present research reports a novel foam/soft material, ${{\small}L}$-Cys-g-PCL, with an application for decubitus/pressure ulcers, especially for wounds with a difficult healing process due to infections and constant oxidation of the soft tissues. During this work, the interactions between S. aureus and ${{\small}L}$-Cys-g-PCL foam were studied under conditions that simulate decubitus ulcers; namely, pH and exudate. The effects of duration of grafting (1 or 8 h) and pH (7.0 and 8.9) on wettability, surface energy, swelling, and porosity were also evaluated. Results showed an effective microbicidal activity exhibiting an inhibition ratio of 99.73% against S. aureus. This new ${{\small}L}$-Cys-g-PCL soft material showed saftey to contact skin, ability to be shaped to fill in sunken holes (craters) - pressure ulcers stage III - and to act as a smart material responsive to pH, which can be tailored to develop better swelling properties at alkaline pH where exudates are normally higher, so as to address exudate self-cleaning and prevention of desiccation.

Bioactivities of Korean Ginkgo (Ginkgo biloba L.) Extract and Its Potential as a Natural Pesticide (은행나무 추출물의 생물활성 및 천연물농약으로 이용 가능성)

  • Bioactivities of Korean ginkgo (Ginkgo biloba L.) extract were investigated against several fungi, general bacteria and insect pests. Crude methanolic extracts of different parts of Korean ginkgo showed different bioactivities depending on the target organisms. The methanolic extract showed in vitro antimicrobial activity at dose of 200 ug per paper disc. The extract of ginkgo stalk was some higher than seed coat and root. The extract also showed a remarkable in vivo antifungal activity against green mold (Trichoderma harzianum) on compost surface of spawn bags and in vivo insecticidal activity to Nilaparvata lugens, Plutella xylostella and Tetranychus urticae. This study suggests that Korean ginkgo extracts have a potential as a natural pesticide.

  • Foodborne diseases occur frequently and have various being related to the intake of contaminated foods. Seafood products are susceptible to contamination due to higher water content and microorganisms, which combine to give them a short shelf-life. Various approaches have been applied to overcome this problem. Edible coatings that are also biodegradable and biocompatible have been discussed as one of the applicable solutions. These coatings can actually help to maintain seafood quality by inhibiting the growth of microorganisms and delaying the loss of moisture. This paper presents the effects of various natural bio-polymers, antimicrobial substances and physical sterilization techniques such as gamma irradiation, ultraviolet (UV) sterilization, and light-emitting diode (LED) sterilization on seafood coatings.

  • The work has been carried out for the development of antifungal antibiotics possessing curative effect in the control of sheath blight disease of rice plant. Soil samples were collected from over 1600 spots throughout the country. More than 1300 specimens which seem to be the genus Streptomyces were isolated from the soil samples. Screening procedures consist of respective processes by four steps. Those are growth inhibition test in liquid culture, paper disk method, dendroid test and green house test. 102 isolates appeared to be active against Pellicularia sasakii when all specimens isolated were examined by the first growth inhibition test. Finally a strain of Streptomyces forming strong antifungal substances against P. sasakii was selected from a soil sample of Mt. Soyo, Kyeongi Province. Antifungal substances formed by the strain were isolated and purified from the culture broth and examined for antimicrobial activities as to be specific against fungi but not active on bacterial growth.

  • 최근 국산 농산물의 수요급증과 더불어 소비자들은 안전한 제품의 요구가 높아지고 있다. 이러한 요구에 맞는 제품 개발의 일환으로 미생물 제어가 된 고춧가루 포장제품의 품질향상을 위한 천연항균 물질 소재를 탐색하여 제품개발에 응용하였다. 천연물과 한방재료 중 고춧가루 제품에 적합한 소재를 스크리닝하여 항균물질을 탐색하였으며 고춧가루 제품 패킹에 응용하였다. 천연물 항균제의 개발 원칙은 산취 등 이취가 없어야 하며, 제품에 응용할 수 있는 용해성이나 믹싱의 효과를 높이도록 공정을 최적화 하였다. 또한 최종 천연물 항균제의 형태는 분말 형태로 개발하였다. 천연물에 대한 항균활성은 paper disk test를 통하여 비교하였으며, 항균력이 가장 우수한 소재는 황금추출물이었다. 또한 이들 추출물과 비타민 $B_1$ 인산염의 영향을 함께 비교하였다. 이 천연물 소재 추출물로 개발한 항균물질에 대한 효과는 일반세균, 대장균 검사를 실시하였다. 제품의 안정성 확보 및 유통기한 연장 가능성을 실험한 결과, 일반세균은 $1.0{\times}10^5$cfu/g, 대장균군은 $1.0{\times}10^2$cfu/g, 대장균은 불검출 되었다. 이들 추출물의 항균제 개발과 더불어 다양한 식품에 천연물 항균제로서의 적용 가능성을 모색하고자 하며, 안전한 천연 항균제 개발에 대한 기초 자료를 제공하는 기초 연구를 하고자 하였다.
