The antibacterial and insecticidal properties of ethanol extracts and volatile components extracted from Eugenia caryophyllata, Boswellia carerii, Agastache rugosa, Aristolochia contorta, and Aquilaria agallocha were evaluated. The ethanol extract and volatile component of E. caryophyllata showed strong antimicrobial effect against all strains (Mucor hiemalis, Aspergillus niger, Penicillium funiculosum, Trichodema viride) and the volatile component of B. carterii showed antimicrobial effect against all strains except T. viride. The ethanol extract of E. caryophyllata and A. contorta showed $100\%\;and\;32\%$ mortality against Reticulitermes spertus kyushuensis Morimoto for 48 hours and 72 hours, respectively. In the case of volatile component, R. aryophyllata showed $100\%\;and\;20\%$ mortality against R. spertus and Lyctus linearis GOZE, respectively. The main constitute, eugenol $(92\%)$ among nine components from volatile component of E. aryophyllata were identified as antibacterial active substance.
Bacillus sp.LPO3 (producing emulsifying substances such as bio-surfactant) was used as a bio-control agent to degrade hydrocarbon (gasoline in oil spilled crop soil). The soil (brought from fertilizer store)was mixed with gasoline-spilled soil (made with Diatomaceous Earth, Sigma.U.S.A). The study was conducted for a period of 13 days, 13 days during which bacterial growth, hydrocarbon degradation and growth parameters of Bacillus sp.LP03 including shoot and root length were studied. We found that the effective of bacterial producing substance might bio-surfactants let the plants survive even more promote the growth of shoot and root length and showed antifungal activity against gray mold. Without the bacteria, they couldn't grow in oil-spilled soil not even survive. According to the results of the above experiments, we can see with following results, hydrocarbon in gasoline was reduced, day by day, then RNA dot blotting was done and it fit the results we had done. Finally, this Bacteria(producing bio-surfactant) were found to have effective bio-control agent for cropping in oil spilled soil and infected by gray mold.
Strain BCNU 2001 was isolated from soil samples collected from Tea-baek Mountain area. The biochemical characteristics and 16S ribosomal RNA gene sequences of the isolate revealed that the strain belonged to the Pseudomonas aeruginosa. The supernatants had an antimicrobial effect on various kind of bacteria and fungi. Especially BCNU 2001 was able to greatly inhibit the growth of Micrococcus luteus, Proteus mirabilis, Proteus vulgaris, and Aspergillus niger, and its inhibition zone was measured as 18.5 mm against Micrococcus luteus, 19.0mm against Proteus mirabilis, 17.0mm against Proteus vulgaris, and 13.5 mm against Aspergillus niger, respectively. Hexane and dichloromethane extracts of BCNU 2001 exhibited significant activity against bacteria, and dichloromethane and ethylacetate extracts showed significant activity against fungi. Pseudomonas strain BCNU 2001 was also determined to have antimicrobial peptide against various microorganisms including Gram positive bacteria, Gram negative bacteria and fungi. The obtained results may provide preliminary support for the usefulness of Pseudomonas strain BCNU 2001.
Insects constitute the largest and most diverse group of animals in the world. They also serve as the hosts or nutrient sources for an immense assemblage of pathogens, parasites, and predators. More than 700 fungal species from 100 genera have adopted an entomopathogenic lifestyle. Although entomopathogenic fungi were studied as only biocontrol agents against a variety of pests in various countries, it has been recently focused their additional roles in nature. They are antagonists to/against plant pathogens, endophytes, and possibly even plant growth promoting agents. The potential antimicrobial effect against fungal plant pathogens by an isolate of entomopathogenic fungi including Beauveria bassiana, Lecanicillium spp., and Isaria fumosorosea have been reported since late 1990s, but wasn't reported pathogenicity of the isolate against pests. Later, a Canadian Lecanicillium sp. isolate and L. longisporium isolated from Vertalec$^{(R)}$ showed simultaneous control effect against both aphid and cucumber powder mildew. Therefore, the antimicrobial activities of 342 fungi isolates collected from various regions and conditions in Korea were evaluated against plant pathogenic fungus Botrytis cinerea using dual culture technique on agar plate. As a result, 186 isolates (54.4%) shown the antifungal activity against B. cinerea. The culture filtrates of selected fungi completely suppressed the growth of the microorganisms, indicating that suppression was due to the presence of antimicrobial substances in the culture filtrate. Mode of action of these fungi against insect involves the attachment of conidia to the insect cuticle, followed by germination, cuticle penetration, and internal dissemination throughout the insect. During infection process, secreted enzymes, proteinous toxins, and/or secondary metabolites secreted by entomopathogenic fungi can be used to overcome the host immune system, modify host behavior, and defend host resources. Recently, secondary metabolites isolated from entomopathogenic fungi have been reported as potential bioactive substances. Generally, most of bioactive substances produced by entomopathogenic fungi have reported low molecular weight (lower than 1,000 g/mol) as peptide and, in contrast the high molecular weight fungal bioactive substances are rare. Most substances based on entomopathogenic fungi were shown antimicrobial activity with narrow control ranges. In our study we analyzed the antimicrobial substances having antagonistic effects to B. cinerea. Antimicrobial substances in our fungal culture filtrates showed high thermostability, high stability to proteolytic enzymes, and hydrophilicity and their molecular weights were differed from substance. In conclusion, entomopathogenic fungi showed pathogenicity against insect pests and culture filtrate of the fungi also shown to antimicrobial activity. In the future, we can use the entomopathogenic fungi and its secondary metabolites to control both insect pest control and plant pathogenic fungi simultaneously.

Biocontrol potential of an isolate of Helicosporium spp. against Rhizoctonia solani, Fusarium oxysporium and Phytophthora drechsleri was evaluated in vitro and in vivo. A selected biocontrol agent designated as Helicosporium 0635BP strongly inhibited growth and lysed mycelium of Rhizoctonia solani and Fusarium oxysporium on PDA. Autoclaved culture filtrate of the agent also completely inhibited growth of the turfgrass large patch pathogen, R. solani AG2-2 IV at the concentration of 50 ml $L^{-1}$. The pathogen was killed when dipped under the 20% filtrate for four hours or 50% for one hr. In a field trial, plots applied with the crude or times diluted culture filtrate showed 100% control efficacy of the turfgrass large patch as a chemical applied for a comparison. Results indicated that Helicosporium 0635BP is a promising biocontrol agent on control of the turfgrass large patch disease and its culture filtrate contained unknown heat suitable antifungal substance (s). Further studies on mass production, purification and identification of the unknown compound (s) are in progress for practical use.
Ozone is a gaseous molecule able to kill microorganisms, such as yeast, fungi, bacteria, and protozoa. However, ozone gas is unstable and cannot be used easily. In order to utilize ozone properly and efficiently, plant oil can be employed. Ozone reacts with C-C double bonds of fatty acids, converting to ozonized oil. In this reaction, ozonide is produced within fatty acids and the resulting ozonized oil has various biological functions. In this study, we showed that ozonized oil has antimicrobial activity against fungi and bacteria. To test the antimicrobial activity of ozonized oil, we produced ozonized olive oil. Ozonized olive oil was applied to Staphylococcus aureus, methicillin-resistant Staphylococcus aureus (MRSA), Staphylococcus epidermidis, Pseudomonas aeruginosa, Escherichia coli, and Candida albicans. Antimicrobial activity was assayed using the disk diffusion method following the National Committee for Clinical Laboratory Standards. Minimal inhibitory concentrations (MIC) were 0.25 mg for S. aureus, 0.5 mg for S. epidermidis, 3.0 mg for P. aeruginosa, and 1.0 mg for E. coli. Gram positive bacteria were more susceptible than Gram negative bacteria. We compared growth inhibition zones against S. aureus and MRSA, showing that the ozonized olive oil was more effective on MRSA than S. aureus. Furthermore, the ozonized olive oil killed C. albicans within an hour. These data suggested that ozonized olive oil could be an alternative drug for MRSA infection and could be utilized as a potent antimicrobial and antifungal substance.
To produce bioactivity-strengthen medicinal herbs, the 36 medicinal herbs which have antioxidation or blood circulation activity, were solid fermented using Phellinus baumii mycelium. Most of medicinal herbs, except Chrysanthemum indicum (flower), Zizyphus jujuba Miller (fructus), Aconitum koreanum R. Raymond (root), Magnolia denu-data (flower), and Polygonatum sibiricum Redt (root bark), showed good fermentation at $25^{\circ}C$ for 20 days under 90% of relative humidity. The poor fermentations of the herbs could be explained by lack of nutrient, structural rigidity, and the content of antifungal substance. After fermentation, the average water content of herbs were increased to $67.21{\pm}11.43%$ from $30.84{\pm}15.67%$, but the average pH and average methanol extraction ratio were slightly decreased to $11.16{\pm}7.06%$ and $4.83{\pm}0.73$ from $13.91{\pm}12.22%$ and $5.06{\pm}0.87$, respectively. The analysis of thrombin inhibition and DPPH scavenging activity of the methanol extracts of herbs showed that thrombin inhibition activities of the fermented Drynaria fortunei Kunze, Melia azedarach var. japonica, Prunus persica and Orostachys japonicus, and DPPH scavenging activities of the fermented Polygala tenuifolia, Scrophularia buergeriana, Angelica dahurica, Drynariafortunei Kunze, Cyperus rotundus, and Boschniakia rossica were increased as compared with those activities of non-fermented its cognate herbs. Our results suggest that the production of bioactivity-strengthen medicinal herbs is possible by solid fermentation of Phellinus baumii mycelium, as fermented Drynaria fortunei Kunze showed increased antioxidant and thrombin inhibitory activities than those of non-fermented herbs.
In order to investigate histopathologically the nature of varietal resistance and infection process of the rice bast fungus, Pyricularia oryzae. this experiment was undertaken by using the resistant cultivar Tongil and susceptible cultivars Norin No.6 and Jinheung in 1973 to 1974. 1) It was found that appressorium of the fungus forms not only at 4 hours after incubation under the favorable conditions but also peak of appressorial formation is at 48 hours treatment. Physical stimulus known to be a definite factor for appressorial formation. The optimum temperature range for appressorial infection was at $24^{\circ}C$ to $28^{\circ}C$, and pH was between 4.8 to 8.0 with 6.8 as the optimum. 2) Although percent of appressorial formation on the leaves of resistant Tongil and susceptiible Norin No.6 were only slightly different, there was a remarkable difference between resistant and susceptible cultivars with regard to percent of hyphal infection and index for hyphal extension. Index of hyphal extension was 1.6-2.7 in Tonyil. while in susceptible cultivar was 3.4-6.6. The rate of discoloration of infected cells, a indication of hypersensitivity, was greater in the resistant than in susceptible cultvar. 3) Therefore, it could be concluded that resistance of Tongil cultivar may be attributable to a higher degree of mechanical barriers as well as a higher level of antifungal substance accumulation.
