In order to evaluate the antimicrobial effect of 2, 3-disubstitued-1, 4-naphtoquinone derivatives we newly synthesized several 2-bromo-3-(substituted)-1, 4 naphthoquninones. Amination reaction of 2, 3-dihalo-1, 4 naphthoquinones with aryl and aliphatic amines in ethanol gave 2-halo-3-(N-alkyl or N-aryl)1, 4-naphtoquinone derivatives (1a, b-10a, b) i 60% 90%) yield. These derivatives subjected to antibacterial and antifungal activities. in vitro, against Bacilllus subtilis ATCC 6633 Candida albicans 10231 and local, Psudomonas aeruginosa NCTC10490, Staphylococcus aureus ATCC 6538p. Escherichia coli NIHJ Aspergillus niger KCTC 1231, Tricophyton mentagrophytes KCTC 6085. Among these derivatives 1b, 6b and 7a showed the potent antibacterial activities 1b, 8b and 9b have derivatives, 1b, 6b and 7a showed the potent antibacterial activities. 1b, 8b and 9b have the antifungal activities. 1b is most effective in preventing the growth of Bacillus subtilis and Psudomonas aeruginosa. Candida albicans. Aspergillus niger. Tricophyton mentagrophytes. The several of these compounds demonstrated a broad spectrum of activities in vitro.

Penicillium digitatum and P. italicum are the two important postharvest pathogens in citrus, causing about 90% of the total loss of citrus fruit during storage and transportation. Natural fungicides such as essential oils have been widely used instead of chemical fungicides for preventing and controlling postharvest diseases. In this research, p-anisaldehyde exhibited a strong inhibitory effect on P. digitatum and P. italicum, with the minimum inhibitory concentration and minimum fungicidal concentration values of both being 2.00 μl/ml. Additionally, p-anisaldehyde visibly inhibited both the green mold and blue mold development of citrus fruits inoculated with P. digitatum and P. italicum. The mycelia morphologies of these pathogens were greatly altered, and the membrane permeability and cell wall integrity of mycelia were severely disrupted under p-anisaldehyde treatment. These results suggest that the antifungal activity of p-anisaldehyde against P. digitatum and P. italicum can be attributed to the disruption of the cell wall integrity.
6-[(N-3,4-Dibromophenyl)amino]-7-chloro-5,8-quinolinedione(FCK13) was tested for antifungal activities. The MIC values were determined by the two-fold dilution method. The therapeutic potential of RCK13 had been assessed in comparison with ketoconazole and fluconazole against systemic infections with candida albicans in normal mice. RCK13 had ED50,0.80$\pm$0.21 mg/kg but ketoconazole had ED50, 8.00$\pm$0.73 mg/kg respectively. And administered RCK13 at the ED50 for 14 days improved survival rates as well as ketoconazole. Acute oral toxicity studies of RCK13 were carried out in ICR mice of both sexes. These acute oral toxicities of RCK13 were low and LD50 values were over 2,850 mg/kg in ICR mice. The genotoxicities of RCK13 had been evaluated. RCK13 was negative in Ames test with Salmonella typhimurium and chromosomal aberration test in CHL cells. The clastogenicity was tested on the RCK13 with in vivo mouse micronucleus assay. RCK13 did not show any clastogenic effect in mouse peripheral blood and was negative in mouse micronucleus assay. These results indicate that RCK13 has no genotoxic potential under these experimental conditions.
The antifungal activity of super reductive water (SRW) against plant pathogenic fungi was examined to extend its application to integrated pest management (IPM) for plant diseases. Diluted solutions ($\times$1/10, $\times$1/25, and $\times$1/50) of SRW inhibited fungal growth of kiwifruit soft rot pathogen, Diaporthe actinidiae, in a concentration dependent manner, When kiwifruits were inoculated on wounds with mycelium blocks, stock and diluted solutions successfully inhibited the disease development. In addition to the high pH of the SRW, fungistatic activity was also considered as the cause of the antifungal effect against the pathogen. Whereas conidial germination of Magnaporthe grisea was not affected by the diluted SRW solutions, appressorium formation was significantly inhibited in a concentration dependent manner, With little harmfulness to human health and environment SRW could be used to control plant pathogenic fungi, particularly appressorium-forming fungal pathogens.
Environment-friendly, commercially-available agricultural products were investigated for antimicrobial activity against Sclerotinia sclerotiorum, as a pathogen of sclerotium disease. Then ${\beta}$-thujaplicin from Chamaecyparis obtuse was investigated for antifungal activity against six kinds of pathogenic fungi. It showed a statistically significant (p <0.001) growth inhibition effect on Sclerotinia sclerotiorum as a pathogen of sclerotium disease, Rhizoctonia solani AG-4 as a pathogen of damping off, Phytophthora capsici as a pathogen of phytophthora blight, and Colletotrichum coccodes as a pathogen of anthracnose at a concentration of 50 ppm and on Stemphylium solani as a pathogen of spotting disease and Alternaria alternata as a pathogen of black mold at a concentration of 100 ppm. In conclusion, these results indicate that it may be possible to develop environment-friendly agricultural products using ${\beta}$-thujaplicin compounds.
The 4',7-dimethoxyisoflavone was isolated from the leaves of Albizzia lebbeck for the first time. This flavonoid showed antifungal activity against some plant pathogenic fungi tested in vitro, e.g., Alternaria melongenae, A. brassicicola, A. brassicae, Curvularia maculans, C. pallescens, C. lunata, Curvularia species, Colletotrichum species, Helminthosporium penniseti and H. speciferum. The sensitivity of different fungi to this chemical varied considerably. A. brassicae was most sensitive as complete inhibition of germination was observed in all the concentrations(100 to 1000 ppm) of the chemical. Similar effect on H. speciferum and Curvularia species was also recorded at 500 ppm, whereas H. penniseti did not germinate at 250 ppm. A. melongenae and A. brassicicola also did not germinate at 1000 ppm while 750 ppm was inhibitory to C. lunata and C. maculans. Germination in almost all fungi was significantly inhibited at each concentration in comparison to control except Curvularia sp. and H. speciferum. Use of 4',7-dimethoxyisoflavone to control some plant diseases under field conditions has been suggested.

Honey bees are important pollinators in agriculture, but are threatened by the pathogen Ascosphaera apis, which causes chalkbrood. Despite attempts to control this fungus using synthetic fungicides, none of them have been proven to be completely effective. Among 640 natural compounds that we tested, 4-hydroxyderricin (MIC=3.125, 6.25 mg/L after 24 h and 48 h growth, respectively) exhibited the strongest anti- Ascosphaera apis activity, followed by acetylshikonin (MIC=12.5 mg/L for 24 h and 48 h growth). 4-Hydroxyderricin showed selective growth inhibition of Ascosphaera apis and Rhizopus oryzae among tested fungus strains. Treatment 4-hydroxyderricin with miconazole revealed a synergistic effect (FICI=0.65±0.13 at 48 h incubation). These findings suggest that 4-hydroxyderricin, which has antifungal activity against Ascosphaera apis but few other fungal species, can effectively control infectious fungal diseases. Combined treatment of bees with 4-hydroxyderricin and miconazole could reduce cytotoxicity and improve the cost effectiveness of treatment.
Soil is the major source of plant-associated microbes. Several fungal and bacterial species live within plant tissues. Actinomycetes are well known for producing a variety of antibiotics, and they contribute to improving plant health. In our previous report, Streptomyces globisporus SP6C4 colonized plant tissues and was able to move to other tissues from the initially colonized ones. This strain has excellent antifungal and antibacterial activities and provides a suppressive effect upon various plant diseases. Here, we report the genome-wide analysis of antibiotic producing genes in S. globisporus SP6C4. A total of 15 secondary metabolite biosynthetic gene clusters were predicted using antiSMASH. We used the CRISPR/Cas9 mutagenesis system, and each biosynthetic gene was predicted via protein basic local alignment search tool (BLAST) and rapid annotation using subsystems technology (RAST) server. Three gene clusters were shown to exhibit antifungal or antibacterial activity, viz. cluster 16 (lasso peptide), cluster 17 (thiopeptide-lantipeptide), and cluster 20 (lantipeptide). The results of the current study showed that SP6C4 has a variety of antimicrobial activities, and this strain is beneficial in agriculture.
Furaneol, a key aroma compound found in strawberry, pineapple, and processed foodstuffs, has been known to possess various biological activities on animal models. In this study, the antimicrobial effects of furaneol against human pathogenic microorganisms were investigated. The results indicated that furaneol displayed a broad spectrum of antimicrobial activities against Gram-positive and Gram-negative bacteria and fungi without hemolytic activity on human erythrocyte cells. To confirm the antifungal activity of furaneol, we examined the accumulation of intracellular trehalose as a stress response marker on toxic agents and its effect on dimorphic transition of Candida albicans. The results demonstrated that furaneol induced significant accumulation of intracellular trehalose and exerted its antifungal effect by disrupting serum-induced mycelial forms. These results suggest that furaneol could be a therapeutic agent having a broad spectrum of antimicrobial activity on human pathogenic microorganisms.
In order to investigate structure-antibiotic activity relationships of brevinin-1 and thanatin containing Rana box composed of basic loop formed by disulfide bridge in their arboxy terminus, thanatin, brevinin 1 and their analogues (T-B1, T-B2 and B-T) in which their Rana box sequence exchanged was designed and synthesized by the solid phase method using Fmoc-chemistry. The basic sequence of Rana box of thanatin had more significant effect on both antibacterial and antifungal activity than that of brevinin 1. The tail sequence (QRM) of thanatin was found to be important in its antibacterial and antifungal activity. Rana box sequence of brevinin-1 did not have a significant effect on its antitumor and phospholipid vesicle-aggregating activities. Brevinin-1 showed stronger $\alpha$-helical structure in the membrane-mimicking environment such as SDS micelle than thanatin. A remarkable increase in a-helicity of bervinin-1 plays more important role in antibiotic activity than that of thanatin. Furthermore, antibacterial activity of thanatin against E. coli resulted from the disruptive effect against the outer cell membrane of E. coli.
