Recent Studies on the Chemical Constituents of Korean Ginseng (Panax ginseng C. A. Meyer) (고려인삼의 화학성분에 관한 고찰)

  • Panax ginseng C.A. Meyer(Araliaceae) has been traditionally used as an expensive and precious medicine in oriental countries for more than 5, 000 years. Ginseng saponin isolated from the root of Panax ginseng have been regarded as the main effective components responsible for the pharmacological and biological activities. Such as antiaging effects. antidiabetic effects anticancer effects. Protection against physical and chemical stress. Analgesic and antipyretic effects. Effects on the central nervous system, tranquilizing action and others. Thirty kinds of ginsenosides have been so far isolated from ginseng saponin and their chemical structures have been elucidated since 1960's. Among which protopanaxadiol type is 19 kinds. protopanaxatriol type. 10 kinds and oleanane type, one. Since ginsenosides are generally labile under acidic conditions ordinary acid hydrolysis is always accompanied by many side reactions, such as epimerization. hydroxylation and cyclization of side chain of the sapogenins Especially. it is well known that C-20 glycosyl linkage of ginsenoside was hydrolysed on heating with acetic acid to give an equilibrated mixture of 20(S) and 20(R) epimers. And also, the chemical transformations of the secondary metabolites have appeared during the steaming process to prepare red ginseng. Indicating demalonylation of malonyl ginsenosides, elimination of glycosyl residue at C-20 and isomerization of hydroxyl configuration at C-20. But these studies have not provided a comprehensive picture in explaning how these ginsenosides showed val'iotas pharmacological activities of ginseng. Though some of them have been involved in the mechanism of pharmacological actions. Recently, non-saponin components have received a great deal of attention for their antioxidant, anticancer antidiabetic, immunomodulating. anticomplementary activities and so on. To meet the demand for such wide applications, studies on the non-saponin components play an important role in providing a good evidence of pharmacological and biol ogical activities. Among the non-saponin constituents of Korean ginseng, polyacetylenes, phenols. Sesquiterpenes, alkaloids. polysaccharides oligosaccharides, oligopeptides and aminoglycosides together with ginsenosides of terrestrial part are mainly described.

Optimun Conditions for Production of Mycelia and Extracellular Polysaccharide from Inonotus obliquus and Their Immunomodulating Acitivities (차가버섯의 균사체 및 세포외다당체의 생산조건과 면역활성)

  • Optium conditions for the production of mycelia and extracellular polysaccharide (EXPS) from submerged mycelial culture of Inonotus obliquus and their immunomodulating activities were investigated. The optmium production of mycelia and EXPS from I. obliquus was observed in mushroom complete medium (MCM). The optimum pH, temperature, and agitation speed for the production of mycelia and EXPS were 5.5, $25^{\circ}C$, and 150 rpm, respectively. The culture period for maximum production of mycelia (10.89 g/l) and EXPS (1.25 g/l) in shake flask cultivation was 11 days. The anticomplementary activity of intracellular polysaccharide (INPS) and EXPS form I. obliquus increased in a dose-dependent manner. Lysosomal enzyme activity of EXPS and INPS increased by 2.0- and 2.2-fold at $100{\mu}g/ml$ concentration, respectively, compared to the control group.

Biological Activities of Substance Extracted from the Fruit Body of Formitopsis rosea (잔나비버섯 자실체로부터 분리한 물질의 특성 및 생리활성)

  • This study was conducted to investigate the characteristics and biological activities of the substance extracted from the fruit body of Formitopsis rosea. The substance was extracted by hot water and then it was separated high and low molecular weight fraction by ethanol precipitation, The high molecular weight fraction was found to be a proteoglycan composed of glucose, mannose, galactose, xylose, fructose, ribose and amino acids. The amino acids of proteoglycan were mainly threonine, isoleucine, glycine, aspartic acid. Anticomplementary activity of the high molecular weight fraction was higher than that of the low molecular weight fraction. And also, the high molecular weight fraction enhanced phagocytic activity and nitric oxide production of macrophage. In enzymatic lipid peroxidation reaction with $ADP-FeCl_3-NADPH$, the lipid peroxidation was inhibited 82.4% and 86.6% by high and low molecular weight fractions, in nonenzymatic lipid peroxidation reaction, it was 83.0% and 84.6%, respectively.

Micro-screening Method for the Anticomplement Substances from Natural Resources (천연유래의 항보체 활성물질 선발을 위한 미량탐색법)

  • To screen inhibitors on complement system from natural resources, micro-screening method was established by using hemolytic complement assay. Complement fixation reaction was carried out in the microplate system. For standard hemolysis (50% hemolysis) of the classical pathway (CP), hemolysin and complement serum were diluted to $1/75{\sim}1/100\;and\;1/80{\sim}1/120$, respectively, when sheep erythrocytes were $5.0{\times}10^8\;cells/ml$. In case of the alternative pathway (AP), complement serum was diluted to 1/5 and EGTA and $Mg^{2+}$ were added 4 mM, $4{\sim}8\;mM$, respectively, when rabbit erythrocytes were $4.0{\times}10^8\;cells/ml$. Dimethyl sulfoxide was used for the assay of non-aquous soluble compounds or extracts and its final concentration was not more than 1%. Three phenylpropanoids showed anticomplementary activities in proportion to the concentration for both pathways and rosmarinic acid exihibited the highest inhibitory activities: $5.4{\pm}3.6%(0.063\;mM){\sim}95.8{\pm}0.2%(0.5\;mM)\;and\;35.1{\pm}0.9%(0.063\;mM){\sim}95.6{\pm}1.1%(1\;mM)$ on the CP and the AP, respectively.

Studies on the Rapid Plasma Reagin(RPR) Card Test for the Diagnosis of Syphilis (매독진단(梅毒診斷)을 위(爲)한 신속(迅速)한 혈장항체검사법(血漿抗體檢査法)에 관(關)한 연구(硏究))

  • For the effective control of Syphilis, many investigators have developed a more rapid, simple and economical screening serological test which is adequately sensitive and specific. To fulfill the requirements of a more rapid serologic test for syphlis, a substitute for the conventional serum specimen was needed since considerable time and labor are involved in the processing of blood to serum. Burdon suggested the use of plasma in the serologic tests for syphilis as a substitute for serum. He noticed that plasma was more sensitive than serum in the Kline and Kahn tests, and attributed this to the presence of more antibody-like substance, "reagin" in plasma than in serum. However, to make plasma sufficiently sensitive, it was necessary to inactivate plasma by heating at a temperature of $56^{\circ}C$ for about 30 minutes. Heating of plasma resulted in the precipitation of fibrinogen which made centrifugation necessary to obtain dear plasma. Since the chief disadvantage to the use of unheated plasma(or serum) was a reduction in sensitivity of results-which probably was due to a labile factor such as complement-Portnoy et al began to consider rapid chemical methods of inactivation of plasma(or serum). They experienced that choline chloirde was shown to be anticomplementary which suggested its use as an inactivating agent for unheated plasma(or serum). In 1959 Portnoy et al reported the Rapid Plasma Reagin(RPR) Test for syphilis which is a more rapid, economical and simple. But still this test has many disadvantages as a rapid performing, field and office procedure, because it requires the usual laboratory equipments such as centrifuge, rotating machine, microscope etc. To substitute these disadvantages of the RPR test, in 1962, Portnoy et al developed the Rapid Plasma Reagin(RPR) card test for syphilis, which has the following advantages: a) Simplicity and rapidity of performance, b) Requires no laboratory equipments, c) Stable antigen suspension, d) Adequate sensitivity and specificity. This RPR card test can be used as a rapidly performing and screening test in field investigation, outpatient clinics, small laboratories and hospitals doing limited syphilis serology, and predonor in blood bank. Private clinic which has limited laboratory equipment and technic for syphilis serology can also use this RPR card test as a tool in the rapid diagnosis of syphilis. It was thought that this RPR card test is a useful tool in Korea for private physician and mass survey for syphilis diagnosis. But Portnoy patented the reagents needed for the performing the RPR card test. Therefore authors developed newly the reagents and according to Portnoy's method evaluated the newly developed. RPR card test compared with the VDRL, Kolmer CF, and RPCF tests. The RPR card and VDRL tests were performed plasma and serum from the total 1,132 cases. Among these 1,131 cases, 521 were syphilis suspected laboratory specimens, and 611 were syphilis unsuspected healthy young men. After screening with these two tests, the RPR card and VDRL tests, reactive specimens to the above one or both tests were retested by the Kolmer CF and RPCF tests.

Biological Activities of Polysaccharide Extracted from the Fruit Body and Cultured Mycelia of Phellinus linteus IY001 (목질진흙버섯 자실체와 배양 균사체 유래 ${\beta}-Glucan$성 다당류의 생리활성)

  • This study was conducted to investigate the biological activities of polysaccharide extracted from the fruit body and cultured mycelia of Phellinus linteus IY001. All fractions were extracted by hot water, in the next, Fr. I, Fr. II, Fr. III and Fr. IV were polysaccharide obtained by ethanol precipitation or ultrafiltration. The highest antitumor activity against sarcoma 180 in ICR mice was observed in Fr. III and Fr. IV at the level 85%, but the antitumor activity had no connection with their anticomplementary activity in vitro, it might probably be due to extraction of hot water. All fractions promoted the production of nitric oxide and $TNF-{\alpha}$ in macrophage, addition of Fr. I and Fr. II resulted in production of nitric oxide$3(5.9{\sim}37.6\;{\mu}M)$ and of the $TNF-{\alpha}$ production($8,696.2{\sim}9,420pg/ml)$. All fractions inhibited the lipid peroxidation induced by $AsA/Fe^{2+}$, $ADP/NADPH/Fe^{3+}\;and\;CCl_4/NADPH$ in rat liver microsomes, and Fr. III showed the electron donating ability stronger than tocopherol in assay system using DPPH. From these results, it is suggested that all fractions contain immunoregulatory components which may protect cellular materials from the oxidative damages by their radical scavenging activities.

Characterization of Immuno-stimulating Polysaccharides Isolated from Korean Persimmon Vinegar (감식초에서 분리한 면역활성다당의 특성)

  • In this study, polysaccharides were isolated from Korean persimmon vinegar to characterize the polysaccharides existing as soluble forms within traditional Korean fermented beverages, and their immuno-stimulating activities were examined. Three successive chromatographies were used to purify the main polysaccharide in the persimmon vinegar, PV-1b-I, to homogeneity from the crude polysaccharide (PV-0). The molecular mass of PV-1b-I was estimated as 110 kDa and it contained significant proportions of mannose (46.8%), galactose (28.5%) and arabinose (19.1%). PV-1b-I strongly reacted with ${\beta}$-glucosyl Yariv reagent, suggesting the presence of an arabino-3,6-galactan moiety. PV-1b-I also induced high levels of macrophage activation and mitogenicity on murine splenocytes in vitro. The intravenous administration of PV-1b-I significantly augmented NK cytotoxicity against YAC-1 tumor cells. PV-1b-I also showed potent anticomplementary activity in a dose-dependent manner. Finally, C3 activation products were identified by crossed immunoelectrophoresis using anti-human C3 and the anti-complementary activity of PV-1b-I under $Ca^{2+}$-free conditions, suggesting that this PV-1b-I causes complementary activations via both alternative and classical pathways. From these results, one can conclude that Korean persimmon vinegar contains select polysaccharides in addition to healthy components, and these polysaccharides appear to provide immuno-stimulating activities beneficial to human health.

Biological Activities and Bioactive Compounds in the Extract of Acer tegmentosum Maxim. Stem (산겨릅나무 줄기추출물의 생리활성 및 유효성분 분리)

  • Acer tegmentosum (Acereaceae) has been used a source of traditional medicines for the treatment of hepatic disorders in Korea. This research was conducted to determine biofunctional activities of A. tegmentosum stem extract and to identify its bioactive components. Methanolic extract from A. tegmentosum stem was partitioned by using organic solvents, including n-hexane, ethyl acetate, n-butanol, and water. Two compounds were isolated by using an ODS column chromatography from ethyl acetate soluble fraction shown to the strongest antioxidant activity ($RC_{50}=3.15\;{\mu}g/m{\ell}$) among the fractions. The isolated compounds were analyzed by $^1H$ and $^{13}C$ NMR, IR, UV/VIS, MS spectrum data and identified as catechin, ${\rho}-Hydroxyphenethyl$ alcohol $1-O-{\beta}-_D-(6'-O-galloyl)-glucopyranoside$. The compounds have shown strong antioxidant activity, with similar activity to BHA ($RC_{50}=2\;{\mu}g/m{\ell}$). Especially, ${\rho}-Hydroxyphenethyl$ alcohol 1-O-{\beta}-_D-(6'-O-galloyl)-glucopyranoside$ was shown strong anti-lipid peroxidative activity. However, the compounds were not shown antimicrobial activities. In antimicrobial activity assays, ethyl acetate soluble fraction was effective to bacterial inhibition, such as Escherichia coli and Klebsiella pneumonia, with minimum inhibitory concentrations in $125\;{\mu}g/m{\ell}$. Otherwise, antifungal activity against Candida albicans was shown in n-hexane soluble fraction exhibiting $63\;{\mu}g/m{\ell}$ of minimum inhibitory concentration. In anticomplementary activity assays, water soluble fraction was the most effective exhibiting 24% inhibitory activity.

Immunomodulatory Activities by Difference in Molecular Size of the Proteoglycan Extracted from Ganoderma lucidum IY009 (Ganoderma lucium IY009 유래 단백다당류의 분자량 차이에 따른 면역증강활성)

  • This study was conducted to investigate the immunomodulatory activities of proteoglycan extracted from cultured mycelia of Ganoderma lucidum IY009. The proteoglycan contained two polymer peaks, one was the higher MW peak of 2,000 kD and the other was low peaks of 12kD. To understand the part of strong pharmaceutical activity between two peak, the proteoglycan was separated by ultrafiltration and column chromatography and then examined the various pharmaceutical effects. High molecular weight fraction possesing high content of ${\beta}-linked$ glucan was exhibited high antitumor activity, against sarcoma 180 bearing ICR mouse. And also, anticomplementary activity was highly observed in high molecule fraction than low it fraction. When the raw 264.7 and murine peritoneal macrophage treated with low fraction, high fraction and other stimuli. The activities inducing tumor necrosis factor of the high factions were $2.2{\sim}2.5$ times stronger than that of low fraction.
