Technique of Drug Resistance Test of Clinical Value for the Selection of Antibacterial Agents (항균성제제(抗菌性製劑) 선택(選擇)에 있어 내성검사요령(耐性檢査要領)의 임상적응용(臨床的應用))

  • The authors carried out bovine mastitis test by California Mastitis test (CMT) for the milk from dairy cows in the suburbs of Daejeon. Among the CMT positive milk, Staphylococcus aureus were isolated and, for the proper selection of the commercial antibacterial agents, the strains were tested for the resistant test to the agents. The results obtained were summarized as follows. 1. The resistant test appeared graduate resistance is in such cases continuously repeat with the therapeutic antibacterial agents. 2. The result of resistant test appeared the weakly resistance, untill 4 times subculture in the Rifamastene, Neo-cillin, and 6 times in the Abimasten-100, Neo-masti, Special formular and mastalone. 3. In Tardomycel-L there was rare resistant reactions, untill 6 times subculture. 4. The author recommend that select the antibacterial agents by resistant test and use the selective agents in turn.

Experiments on Avoiding and Deodorizing and Antibacterial of Haemaphysalis longicornis (Ixodoidea: Ixodidae) of Complex Extracts Including Water-Soluble Phytoncide (수용성 피톤치드(water-soluble phytoncide)를 포함한 복합추출물의 작은소피참진드기(Haemaphysalis longicorins) 기피와 탈취 및 항균 실험)

  • The number of households with pets has increased in recent years. Consequently, interest in mites and their odors has increased. In this study, we investigated whether pyrethrum extract and water-soluble phytoncides are effective tick repellents, deodorizing agents, and antibacterial agents. In the tick-climbing test, the pyrethrum extract (23.33 points) and water-soluble phytoncide (22.00 points) showed a high repelling effect. To confirm the tick avoidance effect of the composite extract, a tick climbing test was conducted with a pest control extract, a composite extract (referred to as a sample) containing a water-soluble phytoncide, and a third-party product (control). The sample showed a high avoidance effect (13.00 points and Control 26.67 points. An indoor repellent efficacy test for ticks confirmed that the sample showed an excellent tick-repellent effect (88%) compared to the control (12%). As a result of a deodorization test, ammonia was shown to have a deodorizing effect of 98.3% and trimethylamine of 99.5%, whereas, in the antibacterial test, the E. coli, Staphylococcus aureus, and pneumonia groups showed an antibacterial effect of 99.9%. Combining the results of this study, it is judged that the complex extract including the above material has high utility value as a material that is effective in mite repellents, deodorization, and antibacterial activity.

The Dyeability and Antibacterial Activity of Fabrics Dyed with Chrysanthemum indicum L. Extract (국화 추출액을 이용한 염색직물의 염색성 및 항균성)

  • This study shows the dyeability and antibacterial activity about cotton and silk fabrics dyed with Chrysanthem indicum L. In the case of dyeability, in the first, K/S value was shown comparatively high numerical value. The value of all test sample was over 20. In the next, in the case of colorfastness, the light colorfastness showed the lowest grade, the other colorfastness was relatively good. However there are some differences according to fabrics and mordants. In the case of antibacterial activity to Klebsielia pneumoniae, the cotton fabrics had the most antibacterial effect at the dyed fabrics that had been mordanted $CuSO_4$.$5H_2O$, and the silk fabrics had the most antibacterial effect at the dyed fabrics that had been mordanted $CuSO_4$.$5H_2O$.$Al_2$ and $(SO_4)_3$.

  • Antibiotics polymer prepared by chemical bonding and simple blending of antibacterial into polymers have attracted much interest because of their long-lasting and antibacterial activity. Antibiotics polymer can significantly reduce losses associated with dissolution, photolytic decomposition and volatillization. Further more, increased efficiency safety and selectivity are additional benefits which may be realized. In this study, Antibiotics polymer was synthesized by chemical reaction of polyacrylic acid with sulfamethazine by N,N'-dicyclohexylcarbodiimide(DCC) method. Antibacterial susceptibility was determined against Streptococcus pyrogenes[gram(+)] and Esherichia coli.[gram(-)] using a standardized disc test. As a result, the synthetic antibiotics polymer exhibited the broad susceptibilty against Streptococcus pyrogenes and Esherichia coli. Especially, the antibiotic effect of antibacterial polymer against Gram negative(Esherichia coli) was much stronger than that against Gram positive(Streptococcus pyrogenes).

Antibacterial Activity of Ciprofloxacin-incorporated Central Venous Catheters and its Mechanism Against Planktonic Bacterial Cells

  • Bloodstream infections (BSI) are caused by planktonic microorganisms, sometimes leading to serious infections such as bacteremia and sepsis. BSI occurs more frequently to the patients wearing the central venous catheter (CVC). The ciprofloxacin-incorporated CVC (CFX-CVC) has been reported previously to possess antimicrobial activity. In this study, the antibacterial activity of CFX-CVC and its mechanism against planktonic BSI cells were explored by using the shake flask test and by examining the release rate of 260 nm-absorbing substances from the bacterial cells indicative of the membrane damage of the bacterial cells. CFX-CVC reduced more than 99.9% of the viable planktonic BSI cells demonstrating its potent antibacterial activity. It provoked bacteriolysis causing leakage of a large amount of 260 nm-absorbing materials from the planktonic bacterial cells like S. aureus and E. coli. These results provide evidence that the antibacterial activity of CFX-CVC came from the inhibition of the stability of the planktonic bacterial cells.

  • The water extracts of 83 oriental herbal medicines (Hanbang) which have been clinically used to treat bacterial infections in Korea were screened for in vitro antibacterial activity by the paper disc assay method. Two Gram positive bacteria, Staphylococcus aureus SG 511, Bacillus subtilis ATCC 6633, and two Gram negative bacteria, Escherichia coli 055, Pseudomonas aeruginosa 9027 were used as test organisms. Among the extracts tested, MeOH extract of Baenongtang showed remarkably potent antibacterial activity. Activity-guided chromatographic fractionations of the $CH_2Cl_2$ extract of Baenongtang afforded seven antibacterial compounds.

Natural Dyeing Properties and Antibacterial Activity of Nylon Fabric Dyed with Cochineal (코치닐에 의한 나일론직물의 천연 염색성과 항균성)

  • The purpose of this study was to investigate the dyeing-property and antibacterial activity on nylon fabric dyed with cochineal at variable dyeing conditions. Al, Cr, Fe, Cu and Sn were used as mordants and adsorption was compared with different mordanting methods. The maximum UV-visible absorption band of cochineal extract was 495 nm. The dyeability on nylon fabric was good because of having a amine group. The optimum dyeing conditions of nylon fabrics are dyeing concentration 1.5%(o.w.s), dyeing temperature $60^{\circ}C$, pH 3 and dyeing time 30 minutes. The pre-mordanting method is preferred for Al and Cr, and the post-mordanting one is preferred for Cu, Sn and Fe to achieve better dyeing. The optimum mordanting conditions of wool fabrics dyed with cochineal are mordanting concentration of 0.5%(o.w.s), mordanting temperature $60^{\circ}C$, and dyeing time 30 minutes. Nylon fabrics dyed with cochineal show a little antibacterial activity, but it was increased by Sn mordanting. MIC test results in antibacterial activities revealed that the antibacterial activity of Cu was the highest among mordants, but Sn mordant was the most effective in antibacterial activities after mordanting treatment of nylon fabric. The fastness properties of dyed nylon fabric showd a little worse or a similar level and there was no significant difference between a mordanted and non-mordanted fabric.

Synergistic Antibacterial Activity of Ecklonia cava Extract against Anti-biotic Resistant Enterococcus faecalis (항생제 내성 Enterococcus faecalis에 대한 감태(Ecklonia cava) 추출물의 항균 시너지 효과)

  • With continuing demand for the development of new, effective and safe therapies, an investigation was carried out to test the efficacy of an antibacterial agent derived from marine edible seaweed. The methanolic extract of Ecklonia cava from marine edible seaweed evinced potential antibacterial activity against Enterococcus faecalis. Among five solvent-soluble fractions of E. cava methanolic extract, the ethyl acetate soluble extract (EtOAc) exhibited the strongest antibacterial activity, with a MIC value of $128{\mu}g/mL $ against E. faecalis strains. Furthermore, a synergistic antibacterial effect between an antibiotic and the EtOAc fraction was assessed using fractional inhibitory concentration (FIC) indices. A combination of ciprofloxacin and the EtOAc fraction resulted in a ${\sum}FIC_{min}$ range of 0.188 and ${\sum}FIC_{max}$ of 0.508 to 563, suggesting that the ciprofloxacin-EtOAc fraction of E. cava combination resulted in an antibacterial synergy effect against E. faecalis.

  • Fluoroquinolone (FQ) antibiotics are an important class of synthetic antibacterial agents. These are the most extensively used drugs for treating bacterial infections in the field of both human and veterinary medicine. Herein, the antibacterial and pharmacological properties of four fluoroquinolones: lomefloxacin, norfloxacin, ciprofloxacin, and ofloxacin have been studied. The objective of this study was to analyze the antibacterial characteristics of the different fluoroquinolones. Also, the pharmacological properties of the compounds including the Lipinski rule of five, absorption, distribution, metabolism, and excretion, LD50, drug likeliness, and toxicity were evaluated. We found that among all four FQ molecules, ofloxacin showed the highest antibacterial activity through in silico assays with a strong interaction (-38.52 kJ/mol) with the antibacterial target protein (topoisomerase-II DNA gyrase enzyme). The pharmacological and pharmacokinetic analysis also showed that the compounds ciprofloxacin, ofloxacin, lomefloxacin and norfloxacin have good pharmacological properties. Notably, ofloxacin was found to possess an IGC50 (concentration needed to inhibit 50% growth) value of $0.286{\mu}g/L$ against the Tetrahymena pyriformis protozoa. It also tested negative for the Ames toxicity test, showing its non-carcinogenic character.

Sythesis of O-(p-Aminosalicylyl)-inositols and Their Antibacterial Test (O-(p-Aminosalicylyl)-inositol류의 합성과 항균시험)

  • Four stereoisomers of inositol p-aminosalicylate, that is hexaris-O-(p-aminosalicylyl)-myo-inositol, hexaris-O-(p-aminosalicylyl)-muco-inositol were synthesized by p-aminosalicylyl chloride with myo-, epi-, scyllo-, and muco-inositol respectively. Their antibacterial activity was tested against human type tubercle bacilli $H_{37}$/Rv, in contrast to 1.25.gamma./ml of p-aminosalicylic acid used as control. Hexaris-O-(p-aminoslicylyl)-muco-inositol showed the strongest antibacterial action at 0.625.gamma./ml, the other compounds more or less active than p-aminosalicylic acid.
