To detect and quantitation residual antibiotics and antibacterial agents in meats, we performed a biological assay employing the three microorganisms Bacillus subtilis ATCC 6633, Micrococcus luteus ATCC 9341, and Bacillus cereus var. mycoides ATCC 11778 for the screening purpose and developed a Gas Chromatography-mass Spectrometry(GC/MS) analysis for the confirmation and quantiation. In the biological assay (paper disk method), three test solution are used depending on the character of the residual antibiotics and antibacterial agents, follow by a simple clean up procedure which includes homogenization with Mcilvaine buffer, defatting with includes homogenization with Mcilvaine buffer, defatting with hexane, extraction with chloroform, clean-up by Sep-Pak $C_{18}$ and Bakerbond SPE carboxylic acid column. The chloroform layer is used for the analysis of sulfa agents. macrolides antibiotics and antibacterial agents, Adsorbed materials in the Sep-Pak $C_{18}$ were also employed for th analysis of penicillins and tetracyclines. Effluents from the Sep-Pak $C_{18}$ were cleaned-up one more by Bakerbond 10 SPE COOH column and employed for the analysis of aminoglycosides. In the instrumental analysis by using the GC/MSD, residual antibiotics and antibacterial agent were quantitated by selected ion monitoring (SIM) mode after derivatization. A simultaneous analysis of six residual antibiotic and antibacterial agent such as oxytetracycline, penicillin, ampicillin, choliraphenicol and thiamphenicol was developed with simple cleanup procedures revealing good recovery and reproducibility. Also, simultaneous detection of macrolides antibiotics such as erythromycin, spiramycin, and oleandomycin was developed after acid hydrolysis due to their large molecular structures. Because of the high reproducibility and selectivity of these two methods, it is very desirable that the combination of the two methods be used in the bioassay for the screening of residual antibiotics and antibacterial agent and that GC/MSD analysis be used for the confirmation and quantitation.
Purpose : In recent years, essential oils have been produced using natural extracts for various uses. Their functionality is currently being tested not only for cosmetics and perfumes but also for other categories of products. Therefore, this study verified their antibacterial effects on S. mutans, P. gingivalis, and L. gasseri which are the representative strains that cause oral diseases. Methods : Eighteen types of natural essential oils were made at a concentration of 50 % (v/v) using Tween 20, and their antibacterial effects were verified by applying S. mutans, P. gingivalis, and L. gasseri. The antibacterial effects were measured with the disc diffusion method. All the experiments were repeated three times, and the mean value of three measurement values for each variable was used for data analysis. A one-way analysis of variance was conducted using these mean values. Results : Of the eighteen types of essential oils tested, sixteen types showed antibacterial effects on S. mutans, and sixteen and fifteen types exhibited antibacterial effects on P. gingivalis and L. gasseri respectively. The types of essential oils with high-level antibacterial activities were geranium, may chang, and bergamot for S. mutans, lemongrass, bergamot, and eucalyptus for P. gingivalis, and lemongrass, machan, and geranium for L. gasseri in order of antibacterial effect. This result was statistically significant (p<.001). In addition, in the case of mandarin oil, it was found that there was no antibacterial activity in all three strains. Conclusion : This study proved the antibacterial activities of essential oils, which are used for various purposes in daily life, against dental caries and periodontal diseases. The study results will likely be applied to different prevention programs for oral health and broadly used to develop products such as oral care items and dentifrices.
The cDNA clone encoding the antibacterial peptide (SL-1) was isolated from the fat body of the common cutworm, Spodoptera litura, immunized with E. coli K12. The primary structure analysis revealed that its deduced amino acid sequence showed the characteristics of the cecropin family antibacterial peptides and that the amino acid residues highly conserved in the antibacterial peptides from moths and flies were also conserved, implying that SL-1 was a cecropin-like, and especially cecropin B-like, peptide. The predicted secondary structure of the mature SL-1 consists of three domains: (i) an amphiphilic ${\alpha}$-helical domain (Ile-4 to Gly-18); (ii) the hinge region (Gly-23 and Pro-24); and (iii) a hydrophobic domain (Ala-25 to IIe-38).
Water-soluble chitosan and water-insoluble chitosan with molecular weight of 2,000,000, 500,000, 80,000, and 40,000 with more than 90%of degree of deacetylation were produced to test antibacterial activity of chitosan against a pathogenic bacteria, Methicillin Resistant Staphylococcus aureus(MRSA). the AATCC Test Method 100and Modified AATCC Test Method 100 were used to evaluate the antibacterial activity of chitosan. Antibacterial activity of chitosan/acetic acid solution was the same when they were tested by two different methods, but those of polyester fabrics treated with chitosan/acetic acid solution were different in different antibacterial test. So several problems were found in the experimental methods. The AATCC Test Method 100 seems that excessive nutrition exists in inoculum solution by quantitative analysis on the basis the result of antibacterial activity on chitosan/acetic acid solution and amount of chitosan attached to the surface of treated fabrics.
Objectives: The purpose of this study was to evaluate the physical properties and antibacterial activity of denture base resin with added silver sulfadiazine. Methods: Specimens were made from self-curing denture base resin and silver sulfadiazine as an inorganic antibacterial agent. For physical evaluation of the specimens, surface roughness, surface hardness, and contact angle were measured. Bacterial growth was assessed by optical densityat 600 nm (OD600) and colony forming units (CFU) measurements to confirm antibacterial activity. Results: There was no significant difference in surface roughness, surface hardness, and contact angle in the experimental group containing silver sulfadiazine compared to the control group. In contrast, the experimental group showed a significant decrease in antibacterial activity compared to the control group in terms of OD value. Analysis of CFU confirmed a significant decrease in colonies in the experimental group compared to the control group. Conclusions: Denture base resin containing silver sulfadiazine, an inorganic antibacterial agent, exhibited enhanced antibacterial activity without physical changes. In conclusion, the use of denture base resin containing inorganic antibacterial agents may be expected in the future.
Essential oils extracted from Chamaecyparis obtusa were applied to make antibacterial corrugating liner. The quantitative analysis of the essential oils was carried out by GC-MS, which showed that the monoterpenes as a basic component of the essential oils extracted from Chamaecyparis obtusa in the domestic mountain were greater in amount than those in China. The antibacterial activity was then examined with Tyromyces palustris and gram-negative/positive bacteria. The papers treated with the essential oils desplayed great resistance against T. Palustris and gram bacteria but the best results were obtained with the handsheet formed by softwood bleached kraft pulp rather than the liner by OCC. The low antibacterial activity of the liner was considered to be due to starch components which could be readily attacked by the fungi.

This study was performed to find antibacterial substances contained in Scutellariae Radix (SR) using a systems pharmacological analysis method and to establish an effective strategy for the prevention and treatment of infectious diseases. Analysis of the main active ingredients of SR was performed using Traditional Chinese Medicine Systems Pharmacology (TCMSP) Database and Analysis Platform. 36 active compounds were screened by the parameter values of Drug-Likeness (DL), Oral Bioavailability (OB), and Caco-2 permeability (Caco-2), which were based on the drug absorption, distribution, metabolism, and excretion indicators. The UniProt database was used to obtain information on 159 genes associated with active compounds. The main active compounds with antibacterial effects were wogonin, β-sitosterol, baicalein, acacetin and oroxylin-A. Target proteins associated with the antibacterial action were chemokine ligand 2, interleukin-6, tumor necrosis factor, caspase-8,9 and mitogen-activated protein kinase 14. In the future, systems pharmacological analysis of traditional medicine will be able to make it easy to find the important mechanism of action of active substances present in natural medicines and to optimize the efficacy of medicinal effects for combinations of major ingredients to help treat certain diseases.
This research aimed to investigate the antibacterial activity of Lycoris radiata. The methanol extract and solvent fractions from L. radiata exhibited antibacterial activities against Escherichia coli, Staphylococcus aureus, and Helicobactor pylori. Open-column chromatography was used to isolate phytochemical constituents from L. radiata; spectroscopic analysis elucidated their structures as ${\beta}$-sitosterol (1), daucosterol (2), O-methyllycorenine (3), lycorenine (4), lycoricidinol (5), lycorine (6), and lycoricidine (7). Further testing of compounds 1 - 7 revealed antibacterial effects against E. coli, S. aureus, and H. pylori, which suggested the potential of these substances as antibacterial agents. We determined that compounds 1 and 2, isolated from the n-hexane fraction, were more effective against S. aureus and H. pylori. Compound 4, isolated from the methylene chloride fraction, exhibited noticeable antibacterial effects against E. coli. This study is the first report on the antibacterial activities of phytochemical constituents from L. radiata against E. coli, S. aureus, and H. pylori.
Fluoroquinolone (FQ) antibiotics are an important class of synthetic antibacterial agents. These are the most extensively used drugs for treating bacterial infections in the field of both human and veterinary medicine. Herein, the antibacterial and pharmacological properties of four fluoroquinolones: lomefloxacin, norfloxacin, ciprofloxacin, and ofloxacin have been studied. The objective of this study was to analyze the antibacterial characteristics of the different fluoroquinolones. Also, the pharmacological properties of the compounds including the Lipinski rule of five, absorption, distribution, metabolism, and excretion, LD50, drug likeliness, and toxicity were evaluated. We found that among all four FQ molecules, ofloxacin showed the highest antibacterial activity through in silico assays with a strong interaction (-38.52 kJ/mol) with the antibacterial target protein (topoisomerase-II DNA gyrase enzyme). The pharmacological and pharmacokinetic analysis also showed that the compounds ciprofloxacin, ofloxacin, lomefloxacin and norfloxacin have good pharmacological properties. Notably, ofloxacin was found to possess an IGC50 (concentration needed to inhibit 50% growth) value of $0.286{\mu}g/L$ against the Tetrahymena pyriformis protozoa. It also tested negative for the Ames toxicity test, showing its non-carcinogenic character.
