본 연구는 가시오가피 추출물의 UVB 조사에 따른 광노화에서의 피부 주름 형성 억제에 미치는 영향을 검토하기 위하여 진행하였다. 그 결과 가시오가피 추출물은 항산화 활성 및 콜라게네이즈 효소 억제에서 ascorbic acid보다 우수한 효능을 가지며, CCD-986SK 사람섬유아세포에서 콜라겐 생성을 유의적으로 증가시키는 것으로 나타났다. 또한 가시오가피 추출물의 실제적인 주름 형성에 미치는 영향을 알아보기 위하여 UVB가 조사된 무모 생쥐에서 경구투여를 실시한 결과, 피부조직에서 주름의 형성 및 자외선에 의한 콜라겐 조직의 파괴 반응을 억제하고 콜라겐 생성을 촉진시켜 자외선에 의한 피부주름을 예방하거나 완화할 수 있는 효과가 있음을 증명하였다.
The effects that ultraviolet rays elicit on collagen synthesis and degradation are the most common causes of wrinkle formation and photo-aging in skin. The objectives of this study were to evaluate the effects of Angelica acutiloba root ethanol extract (AAEE) to promote collagen synthesis and inhibit collagen degradation in human dermal fibroblasts. By examining total polyphenol and flavonoid contents, electron donating ability, radical scavenging activity, and superoxide dismutase-like activity, we found that AAEE exhibited fairly good antioxidant activity. Treatment with AAEE significantly increased type I procollagen production by cultured fibroblasts, as well as reduced ultraviolet-induced matrix metalloproteinase-1 (MMP-1) expression and MMP-2 activity in a dose-dependent manner (p < 0.05). In addition, AAEE significantly increased TIMP-1 mRNA expression (p < 0.05), although without an associated dose-dependent increase in TIMP-1 protein expression. In summary, we suggest that AAEE may be a potentially effective agent for the prevention or alleviation of skin-wrinkle formation induced by ultraviolet rays.
Background: In this study, the radix of Astragalus membranaceus Bunge extract fermented by Saccharomyces cerevisiae, Weissella cibaria, and Pediococcus pentosaceus to increase the levels of isoflavonoid aglycone contents. Methods and Results: In order to change the in isoflavonoids, we fermented the radix of A. membranaceus extracts with microorganisms that have β-glucosidase activity. Besed on the β-glucosidase activity, we selected three strains, Weissella cibaria, Pediococcus pentosaceus, and Saccharomyces cerevisiae. HPLC analysis revealed that the levels of isoflavonoid aglycones were increased in all fermentation cases, and the extracts fermented by S. cerevisiae showed the highest levels of isoflavonoid aglycones. We evaluated the antioxidant activity, anti-wrinkle effects and whitening effects of the S. cerevisiae-fermented extracts using the DPPH assay, tyrosinase inhibition activity assay, and collagenase inhibition activity assay. We confirmed higher activity in S. cerevisiae-fermented extracts than in control, with the half maximal inhibitory concentration (IC50) value of 565.1 ± 59.1 ㎍/㎖ in DPPH radical scavenging activity, tyrosinase inhibition rate of 78.4 ± 0.9%, and collagenase inhibition rate of 83.8 ± 1.1%. Conclusions: We selected three stains of microorganisms showing high β-glucosidase activity, W. cibaria, P. pentosaceus and S. cerevisiae. Isoflavonoid glycones in the radix of A. membranaceus were converted to isoflavonoid aglycones by fermentation. In addition, the fermented radix of A. membranaceus exhibited antioxidant activity, anti-wrinkle effect, whitening effect and radical scavenging activity.
This study is peformed to investigate the effect of water extract from Vaccinium uliginosum L., on melanin production in B 16 melanoma cells, procollagen production and matrix metalloproteinase-1(MMP-1) inhibition in human fibroblast cells. One hundred grams of the Vaccinium uliginosum L. was extracted with 2000 mL of water($90^{\circ}C$, 16h, 2times). The water extracts were lyophilized and stored at $4^{\circ}C$ until used. Dry weight yields of extracts of Vaccinium uliginosum L. were $3\%$(w/w). Extracts from Vaccinium uliginosum L. showed scavenger activities on DPPH radical, superoxide anion radical, hydroxyl radical, hydrogen peroxide and singlet oxygen radical. And these substances inhibited release of cyiokines from human keratinocyte after UV B exposure. Therefore we confirmed that extracts from Vaccinium uliginosum L. had antioxidative effect. These substances inhibited purified tyrosinase activity and melanogenesis in B 16 melanoma cells treated/untreated IL-$1{\alpha}$. Moreover this extract stimulated procollagen production and inhibited MMP-1 production in human fibroblast cells treated/untreated IL-$1{\beta}$. Therefore we confirmed that extracts from Vaccinium uliginosum L. had whitening effect. And these substances decreased degree of wrinkle in hairless mouse skin that induced by UV B irradiation. Therefore we confirmed that extracts from Vaccinium uliginosum L. had anti-wrinkle effect. From the above results, it is possible that Vaccinium uliginosum L. may be developed to be an anti-melanogenesis agent and anti-wrinkle agent.
To develop a new anti-wrinkle agent from medicinal plants, this study investigated the optimal conditions for extracting elastase inhibitor from Rheum undulatum L. Maximal extraction occurred by using 70% methanol at $50^{\circ}C$ for 24 hr; the elastase inhibitory activity was 60.4%($IC_{50}:6.7{\times}10^3{\mu}g/m{\ell}$). Systematic solvent extraction, thin layer chromatography, silica gel column chromatography, Sephadex LH-20 column chromatography, and reverse-phase high performance liquid chromatography were used in the partial purification of the elastase inhibitor. The compound was soluble in dimethylsulfoxide, methanol, and ethanol, and had maximum absorption spectra at 231.5 nm and 275.5 nm.

This study focused on the anti-oxidative and collagenase- and elastase inhibition effects of low molecular weight peptides (LMP) from commercial Jeju horse leg bone hydrolysates (JHLB) on pancreatin, via enzymatic hydrolysis. Cell viability of dermal fibroblasts exposed to UVB radiation upon treatment with LMP from JHLB was evaluated. Determination of the antioxidant activity of various concentrations of LMP from JHLB were carried out by assessing 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2-azino-bis-3-ethybenzothiazoline-6-sulphonic acid (ABTS) radical scavenging activity, ferric reducing antioxidant power (FRAP), and oxygen radical absorbance capacity (ORAC). The DPPH radical scavenging activity of LMP from JHLB (20 mg/mL) was 92.21% and ABTS radical scavenging activity (15 mg/mL) was 99.50%. FRAP activity (30 mg/mL) was $364.72{\mu}M/TE$ and ORAC activity (1 mg/mL) was $101.85{\mu}M/TE$. The anti-wrinkle potential was assessed by evaluating the elastase- and collagenase inhibition potential of these LMP. We found that 200 mg/mL of LMP from JHLB inhibited elastase activity by 41.32%, and 100 mg/mL of LMP from JHLB inhibited collagenase activity by 91.32%. The cell viability of untreated HS68 human dermal fibroblasts was 45% when exposed to a UVB radiation dose of $100mJ/cm^2$. After 24 h of incubation with $500{\mu}g/mL$ LMP from JHLB, the cell viability increased to 60%. These results indicate that LMP from JHLB has potential utility as an anti-oxidant and anti-wrinkle agent in the food and cosmetic industry. Additional in vivo tests should be carried out to further characterize these potential benefits.
This study was to investigate anti-skin wrinkle effect of Agastache rugosa Kentz extracts by extraction processes. In the comparison of the effect of the solvent extraction, the extracts by 70% ethanol (EE) showed better biological activities that those by hot water. Therefore, further fermented Agastache rugosa was applied to 70% ethanol extraction process (FEE). FEE showed higher DPPH scavenging activity of 62.98% than EE's 62.71% at $1.0mg/m{\ell}$, but there was no significant. Elastase inhibition was measured 23.0% from FEE at $1.0mg/m{\ell}$. Cytotoxicity showed the highest 16.26% from FEE, this value is safe in the cell experiment. Collagen production showed $113.1ng/m{\ell}$ from FEE, on the other hand EE was measured $77.4ng/m{\ell}$ in adding $1.0mg/m{\ell}$. MMP-1 production was observed $1398pg/m{\ell}$ from FEE and EE was measured $1632pg/m{\ell}$. These results were found the highest antioxidant and anti-wrinkle effect. As a result, it was also confirmed that anti-skin wrinkle activities of the Agastache rugosa Kentz extract was correlated with anti-oxidant activities.

The Rosa multiflora, a well-known plant belonging to Rosacea, is widely used in orthodox medicine in worldwide. However, its biological activity as a functional ingredient for cosmetic products have not yet been studied. Accordingly, an investigation of the above mentioned atrributes was performed on a 50% ethanol extract of Rosa multiflora. The antioxidant activities were determined by DPPH. Additionally, the contents of total phenols and flavonoids were analyzed. Also, the phenolic compounds were detected using HPLC. The melanogenesis regulatory effect was evaluated using melanin content and cellular tyrosinase activity in B16F10 melanoma cells. The elastase inhibitory activity assay was performed for anti-wrinkle effect. The antimicrobial activity was assessed using the disc diffusion assay. The DPPH radical scavenging ability, denoted by the $SC_{50}$ value was found to be $123.1{\mu}g/ml$, whereas that of positive control (ascorbic acid) was $27.5{\mu}g/mL$. The content of total polyphenol and flavonoid content were 202 mg/g and 86.77 mg/g, respectively. In addition, astragalin and gallic acid were identified in the extract. Also, the ethanol extract significantly inhibited ${\alpha}$-MSH-induced melanogenesis in B16F10 cells. For anti-wrinkle effect, elastase inhibition activity of the ethanol extract was 53.2% at a concentration of $100{\mu}g/ml$. The antimicrobial activity of the extract against S. aureus and E. coli was observed to be 0.5 - 5%, and no significant activity was noted against C. albicans. Therefore, the ethanol extract of Rosa multiflora can be used effectively for development of functional cosmetic materials.
도화를 기능성 화장품 소재로 활용하기 위하여 미백 및 주름개선 효과를 검증한 후 화장품 소재로서의 가능성을 검증하였다. 미백효과를 확인하기 위하여 tyrosinase 저해활성을 측정한 결과 도화 열수 및 에탄올 추출물 1,000ppm에서 각각 54.0, 58.3%의 저해활성을 나타내었으며, 멜라노마 세포에서의 tyrosinase 저해활성을 측정한 결과 도화 에탄올 추출물 100ppm에서 40.0%의 저해활성을 나타내였다. 또한 멜라노마 세포에서의 멜라닌 생합성 저해율을 확인한 결과 도화 에탄올 추출물 100ppm에서 56.5%의 효과를 나타내어 도화 에탄올 추출물의 미백효과가 가장 우수하였다. 주름개선 효과 측정으로 elastase 저해활성을 측정한 결과 도화 아세톤 추출물 1,000ppm에서 57.0%의 저해활성을 나타내었으며, collagenase 저해활성을 측정한 결과 도화 아세톤 1,000ppm에서 48.0%의 저해활성을 나타내었다. 또한 섬유아세포를 이용한 collagen 생합성량을 측정한 결과 도화 아세톤 추출물 100ppm에서 41.0%의 효과를 나타내어 도화 아세톤 추출물의 주름개선 효과가 가장 우수하였다. 이상의 결과로 미루어 보아 도화 추출물의 미백 및 주름개선 효과가 있음을 확인할 수 있었고, 기능성 화장품 소재로서의 가능성을 확인할 수 있었다.
부평초 추출물 및 부평초 분획물의 기능성 화장품 소재로서의 효능을 측정하기 위하여 항산화, 항주름 및 미백 활성을 평가하였다. DPPH와 ABTS+ 라디칼 소거 활성은 부평초 70% 에탄올추출물의 에틸아세테이트 분획물(SE-E) 100 ㎍/mL 농도에서 각각 44.2와 74.3%로 나타났다. 항주름 활성을 측정하기 위하여 세포내에서의 procollagen 생합성 활성과 MMP-1 저해 활성을 측정하였다. 그 결과 부평초 70% 에탄올추출물(SE)은 25 ㎍/mL 농도에서 48.5%, 같은 농도에서 SE-E는 가장 높은 57.8%의 생합성 활성을 나타내었다. SE와 SE-E의 MMP-1 저해활성은 25 ㎍/mL 농도에서 각각 13.4와 28.5%로 나타났다. 마지막으로 미백 효과를 검증하기 위해 melanin 생합성 저해 활성과 세포내 tyrosinase 저해 활성을 측정한 결과, 25 ㎍/mL 농도에서 SE는 각각 9.6과 13.8%의 저해활성을 나타내었으며 SE-E는 각각 15.4와 22.0%의 값을 나타내었다. 본 연구의 결과로부터 SE와 SE-E의 높은 항주름 및 미백 활성이 확인되어 우수한 기능성 소재로 개발 가능성이 높아 졌으며 추가적인 항균, 항염, 항암 등의 연구가 진행된다면 제약, 화장품, 기능성 식품 등의 분야에 효과적인 소재로 활용될 수 있을 것으로 사료된다.
