Anti-thrombotic Effects of Analogs of Protocatechuic Acid and Gallic Acid (Protocatechuic acid 및 Gallic acid 유도체들의 항 혈전작용)

  • Platelet anti-aggregating activities were tested with analogs of protocatechuic acid and gallic acid. Six of them which showed comparable inhibitory effects with aspirin against collagen induced platelet aggregation were selected and their anti-thrombotic effects were evaluated in the mouse thrombosis model and compared with those of aspirin and paeonol. At the dose of 50 mg/kg, p.o., ethyl gailate(13) treated group showed higher % of recovery within 6 min of thrombotic challenge and lower mortality within 5 min than aspirin treated group.

  • The number of geriatric patients seeking dental service is ever-rising because of increased life expectancy, also with problem of increased chronic medical conditions. One of them are patients on anti-thrombotic medication. Bleeding complication after minor oral surgery by anti-thrombotic agents is of concerns to dentists on dental management of these patients. Risk and benefit of the anti-thrombotic agents must be weighed before initiating dental procedures, which should be established as a treatment guideline. Purpose of the paper is to optimize the management of the dental patients on antithrombotic medication via standardization of treatment protocol of such a patient.

  • In the course of continous work for the discovery of anti-thrombotic constituents from tubers of Gastrodia elata, citric acid 1,5-dimethyl ester was isolated from ethylacetate soluble fraction prepared from the methanol extract. The structure of the compound was determined by the spectroscopic data. The anti-thrombotic effect of this compound was observed with prolonging the bleeding time in thrombin-induced thrombosis model of mice.

  • Both ethanol and water extracts of unossified velvet antler were found to exhibit a significant immuno-stimulating activity as measured by carbon clearance test in mice, a remarkable anti-fatigue effect in weight-loaded forced swimming performance in mice, a significant anti-stress effect on immobilization in rats. The antler extracts also showed a weak but significant anti-thrombotic activity. These findings are indicative of adaptogenic properties of antlers and their normalizing effects during stressful condition.

  • Objectives: Arachidonic acid is control the thromboxane A2 (TXA2) and prostacycline (PGI2) synthesis, TXA2 increase lead to thrombus produced by induces platelet aggregation and vasoconstriction. Angelicae gigantis radix (RAR) is mainly used blood deficiency and stagnation. In previous studies, RAR has been reported that a vasodilating and blood clotting delay effects. In this study, investigate that anti-oxidant and anti-thrombotic effects of RAR by heat-process.Methods: The heated angelicae gigantis radix sample were made by 140, 180, and 220 ℃ and 4, 6, 9 and 12 min using water or 30% ethanol. The anti-oxidant effects were measured by total polyphenol, total flavonoid, DPPH and ABTS radical scavening activation. Anti-thrombotic effect conducted in samples that are determined to be effective through the anti-oxidant experiment such as angelicae gigantis radix roasted 180℃, and 220℃ and angelicae gigantis radix roasted with 30% ethanol 180℃, and 220℃.Results: Anti-oxidant parameters were efficacious in high temperature roasted AR. Also AR and EAR increased a inhibitory activity of FXa compared with RAR. The blood coagulation time of administration groups were significantly increased compare with control group. The TXB2 was significantly decreased in AR and EAR.Conclusions : We confirmed that whether AR and EAR administration has anti-oxidant and anti-thrombotic effect or not. As the results, AR and EAR were improved anti-oxidant effects and blood biochemistry compare with control group. This study provides scientific evidence that AR and EAR are have an anti-oxidant effect and anti-thrombotic effect, it expected that there is no difference between the two.

Contrasting Correlation in the Inhibition Response of ADP-induced Platelet Aggregation and the Anti-coagulant Activities of Algal fucoidans Derived from Eisenia bicyclis and Undaria pinnatifida sporophylls (Mekabu)

  • Sulfated fucans are known to have both anti-thrombotic and anti-coagulant activities. In this study, the variation in platelet aggregation and anti-coagulant activities was investigated in vitro with regard to administered dose, molecular weight distribution, sulfate content, and sugar composition in two algal fucoidans from Eisenia bicyclis and Undaria pinnatifida sporophylls (Mekabu). The anti-coagulant activity largely correlated with sulfate content and with molecular weight distribution in a dose-dependent manner. However, both fucoidans demonstrated inhibitory responses to ADP-induced platelet aggregation in dose- and structure-dependent manners that contrasted with the anti-coagulant activity. Neither molecular weight distribution nor sulfate content greatly affected platelet-aggregation inhibition (PA-inhibition) by the fucoidan fractions, whereas anti-coagulant activity was sensitive to these structural factors. Interestingly, an E. bicyclis fucoidan fraction exhibited almost complete PA-inhibition at a treatment dose of 500 mg/mL while retaining weak anti-coagulant activity. In conclusion, these observations suggest that fucoidan may be a useful anti-thrombotic or anti-platelet agent in various arterial thrombotic disorders, including post-vascular intervention with controlled bleeding complications, due to its anti-coagulant modulating activity.

Anti-platelet and Anti-thrombotic Effects of Gastrodia elata (천마(Gastrodia elata Blume)의 항혈소판, 항혈전활성)

  • MeOH extract of Gastrodia elata was fractionated to five solvent fractions, hexane fr (fr I), 90% MeOH fr (fr II), EtOAc fr (fr III), BuOH fr (fr IV) and $H_2O$ fr (fr V). Among the five fractions, fr II, III and IV showed platelet anti-aggregating effects against ADP or collagen induced rat platelet aggregation in vitro. Fr II , III and IV were also tested in vivo, in the mouse and rat models of thrombosis. Oral administration of fr II, III or IV enhanced the recovery from the thrombotic shock in the mouse model of thrombosis to 32-40% from 17% of recovery with the control group of mice. Treatment with fr II, III or IV also attenuated the sudden reduction in the blood platelet count following intravenous collagen injection to rat. The above results were indicative of the presence of anti-platelet and anti-thrombotic components in this plant.

Comparative Study of Extracts from Rhubarb on Anti-Thrombotic and Anti-Platelet Activity (4종 대황(大黃)류 추출물의 항혈전 효능 비교)

  • Objective : The purpose of this study is to compare anti-thrombotic activities of 4 Rhubarb species. Methods : Rhubarb has been used as medical and edible resources worldwide for the therapy of cancer, constipation and inflammation, etc. To compare the anti-thrombotic activities among the species within rhubarb, we investigated and compared the inhibitory properties of water extracts from R. palmatum(RPE), R. officinale(ROE), R. nobile(RNE), and R. franzenbachii (RFE) on the fibrinolysis, blood coagulation and platelet aggregation. APTT and PT, blood coagulation time, were measured by apparatus of auto blood coagulation analysis that called ACL-7000. The measurement of fibrinolysis was measured and compares with the width of clear zone from melted fibrin plate. The platelet aggregation was measured and compared with inhibition rate of aggregation. Results : We found out the best species from rhubarb showing the significant fibrinolytic, anti-coagulant and anti-platelet aggregation activity. Our results show that Rheum nobile (RNE) has the most effective anti-thrombotic activity among 4 tested rhubarb. Conclusions : This study, therefore, RNE will be able to be used the development of drug which is thrombosis treatment.

Esters of Substituted Benzoic Acids as Anti-thrombotic Agents

  • Aliphatic esters of protocatechuic acid (PA, 1), vanillic acid (VA, 9) and gallic acid (GA, 18) were prepared and their anti-thrombotic effects were evaluated in the mouse model of thrombosis. The aliphatic groups included methyl, ethyl, n-propyl, i-propyl, n-butyl, i-butyl, n-amyl and cyclohexyl. n-Amyl ester of PA (7), i-propyl and cyclohexyl esters of VA (13 and 17 respectively) and ethyl ester of GA (20) treatment significantly lowered the death rate and increased the recovery from paralysis due to the thrombotic challenge. From the limited analogs available, it was tentatively concluded that the structural conformation, where carboxy oxygen (=O or -O) of the carboxyl group (COOH) at $C_1$ and the oxygen function at $C_3(either\; OH\; or\; OCH_3)$ are closely situated, is favorable for the esters of PA, VA and GA to be more antithrombotic.
