Evaluation of Anti-Oxidant from Natural Products (천연물로부터 유래한 천연 항산화제 규명)

  • In this study, we analyzed 80%MeOH extract of fruits of sorbaria sorbifolia var. stellipila MAX. to measure the total antioxidant capacity by oxygen radical absorbance capacity (ORAC) assay, individual flavonoid content by high-performance liquid chromatography (HPLC). n-Hexane ($1.02{\pm}0.036$), $CH_2Cl_2$ ($0.95{\pm}0.025$), EtOAc ($1.94{\pm}0.065$), n-BuOH ($1.98{\pm}0.054$), D.W. ($1.2{\pm}0.032$) fractions were examined antioxidative activity by ORAC assay. It was revealed that EtOAc($1.94{\pm}0.065$), n-BuOH($1.98{\pm}0.054$) fractions had significant antioxidative activity. The isolation and separation were facilitated using open column chromatography, while separation, purification and identification were accomplished by using high-performance liquid chromatography (HPLC) and nuclear magnetic resonance (NMR).

Design and Manufacturing of an Ultrasonic Waveguide for Nano-surface Treatment (나노표면개질 용 초음파 진동자 설계 및 제작)

  • In this article, a 20 kHz ultrasonic waveguide for nano-surface treatment was designed and manufactured. When designing the system, finite element analysis with ANSYS software was performed to find optimal dimensions of the waveguide, which can raise energy efficiency. Consequently an anti-resonance frequency of an Al waveguide with a piezoelectric actuator was 20 kHz, which predicted the experimentally obtained value of 18 kHz well. For the assessment of the performance, Steel Use Stainless (SUS) 304 and chromium molybdenum steel (SCM) 435 specimens were tested. Cross-sectional microscopies of SUS304 were taken and they showed that the treated thickness was $30{\mu}m$. Additionally, hardness tests of SCM435 were done and the hardness before the process was 14.0 Rockwell Hardness-C scale (HRC) and after the process was 20.5 HRC, respectively, which means 46% increase. Considering these results, the developed ultrasonic system is thought to be effective in the nano-surface treatment process.

Doxorubicin Binds to Un-phosphorylated Form of hNopp140 and Reduces Protein Kinase CK2-Dependent Phosphorylation of hNopp140

  • Human nucleolar phosphoprotein p140 (hNopp140) is a nucleolar phosphoprotein that can bind to doxorubicin, an anti-cancer agent. We have examined the interaction between hNopp140 and doxorubicin as well as the folding property of hNopp140. Also, the effects of ATP and phosphorylation on the affinity of hNopp140 to doxorubicin are investigated by affinity dependent co-precipitation and surface plasmon resonance methods. Doxorubicin preferentially binds to un-phosphorylated form of hNopp140 with a $K_D$ value of $3.3\;{\times}\;10^{-7}$ M. Furthermore, doxorubicin reduces the protein kinase CK2-dependent phosphorylation of hNopp140, indicating that doxorubicin may perturb the cellular function of hNopp140 by reducing the protein kinase CK2-dependent phosphorylation of hNopp140. Low contents of the secondary structures of hNopp140 and the fast rate of proteolysis imply that hNopp140 has a high percentage of flexible regions or extended loop structures.

A Study on Vowel Formant Variation by Vocal Tract Modification (성도 변형에 따른 모음 포먼트의 변화 고찰)

  • Vowels are classified by vocal tract shapes. These shapes form constriction points along the tract, which have an influence on such vocal tract resonance as $F_l,\;F_2,\;F_3$, and so on. This study reviews the perturbation theory of the tract and determines the corresponding formant frequencies from modified vocal tracts using vocal tract area function. Then, formant variation is observed from the theory. Finally, each set of $F_l,\;F_2,\;and\;F_3$ frequency is input to a speech synthesis software to make a vowel sound. Auditory impression of each sound without any modification of its vocal tract shape is almost the same as the corresponding phonetic symbol. Formant frequencies of $F_l,\;F_2,\;F_3$ vary according to the perturbation theory. Generally, constriction along the node causes formant values to decrease while constriction along the anti-node cause it to increase. Vocal tracts modified by more than $3\;cm^2$ change vowel qualities of /a/ and /i/ into those of f /v/ and /$\varepsilon$/, respectively. This study will be helpful in simulating sounds from modified vocal tracts before any operation. Further studies are desirable to compare vocal tract shapes of various languages and their sounds together.

  • Alzheimer's disease (AD) is a progressive neurodegenerative disorder. Neuropathological hallmarks of AD are amyloid plaques, dystrophic neurite, and alteration of subcellular organelles. However, the morpho-functional study of this degenerative process and ultimate neuronal death remains poorly elucidated. In this study, immunohistochemical and ultrastructural analyses were performed to clarify the abnormal morphological alterations caused by the progression of AD in APP/PSEN1 transgenic mice, express human amyloid precursor protein, as a model for AD. In transgenic AD mice brain, the accumulation of Amyloid ${\beta}$ plaques and well-developed dystrophic neurites containing anti-LC3 antibody-positive autophagosomes were detected in the hippocampus and cortex regions. We also found severe disruption of mitochondrial cristae using high-voltage electron microscopy and three-dimensional electron tomography (3D tomography). These results provide morpho-functional evidence on the alteration of subcellular organelles in AD and may help in the investigation of the pathogenesis of AD.

Antimicrobial Activity of Methyl Gallate isolated from the Leaves of Glochidion superbum Against Hospital Isolates of Methicillin Resistant Staphylococcus aureus

  • An antimicrobial compound has been isolated from the leaves of Glochidion superbum. The compound was determined as methyl 3, 4, 5-trihydroxybenzoate (methyl gallate), based on ultraviolet (UV), infrared (IR), nuclear magnetic resonance (NMR) and mass spectroscopy (MS) analysis. The isolated compound exhibited potent antimicrobial activity against three clinical isolates of methicillin resistant Staphylococcus aureus (MRSA) by qualitative agar disc diffusion method and quantitative broth dilution method. Agar disc diffusion was done in a dose-dependent manner for each bacterial isolate at disc potencies of 25, 50, 100, and $150{\mu}g/disc$. The zones of inhibition were on average equal to 12.27, 14.20, 15.43, and 24.17 mm respectively. The inhibition zones were compared with that of vancomycin disc at $30{\mu}g$ as a reference standard. The MIC and MBC values were $50{\mu}g/ml$ and $100{\mu}g/ml$ respectively. The results of anti MRSA activity were analyzed using one-way ANOVA with Turkey's HSD and Duncan test. In conclusion, methyl gallate which was isolated from G. superbum showed the inhibition activity against methicillin resistant S. aureus.

Control Effects of an Antibiotic Produced by Streptomyces sp. B25 on Tobacco Mosaic Virus and Determination of Its Molecular Structure

  • The culture filtrate of Streptomyces sp. B25, which was identified in this experiment, was tested for the control of tobacco mosaic virus (TMV) with the susceptible tobacco cultivar, NC 82, under the field conditions following the preliminary examination of its characters for TMV control. Control efficacy of the culture filtrate against TMV infection continued over 50% up to 6 days after treatment, and its systemic effect was about 30% of the direct effect. In field conditions control efficacy of the culture filtrate against TMV infection was 95.3 % at 2 weeks after TMV inoculation, and decreased to 58.3 % at 3 weeks after inoculation. Five fold-dilution of the culture filtrate showed about half of the control efficacy by the stock culture filtrate. Analysis of the antibiotic material responsible for the inhibition of TMV infection through nuclear magnetic resonance experiments revealed that the antibiotic is antimycin $A_1$, which is firstly reported as an anti-phytoviral antibiotic in this experiment.

High-Throughput Active Compound Discovery using Correlations between Activity and Mass Profiles

  • The active components in a plant extract can be represented as mass profiles. We introduce here a new, multi-compound discovery method known as Scaling of Correlations between Activity and Mass Profiles (SCAMP). In this method, a correlation coefficient is used to quantify similarities between the extract activity and mass profiles. The method was evaluated by first measuring the anti-oxidation activity of eleven fractions of an Astragali Radix extract using DPPH assays. Next, 15 T Fouriertransform ion cyclotron resonance (FT-ICR) MS was employed to generate mass profiles of the eleven fractions. A comparison of correlation coefficients indicated two compounds at m/z 285.076 and 286.076 that were strong antioxidants. Principal component analyses of these profiles yielded the same result. FT-ICR MS, which offers a mass resolving power of 500,000, was used to discern isotopic fine structures and indicated that the molecular formula corresponding to the peak at m/z 285.076 was $C_{16}H_{13}O_5$. SCAMP in combination with high-resolution MS can be applied to any type of mixture to study pharmacological activity and is a powerful tool for active compound discovery in plant extract studies.

Design of Z-directional gradient coil to improve gradient linearity for the nuclear magnetic resonace imaging(NMRI) (경사자장의 선형성 향상을 위한 핵자기공명 영상용 Z-방향 경사자기장 코일 설계)

  • Gradient coil offers the spatial informations of sample or patient in Nuclear Magnetic Resonance Imaging(NMRI) and its gradient field linearity over the field of view(FOV) has many influence on the MR imaging. Accurate and good quality MR imaging can be acquired by the high gradient field linearity over the FOV. So it is an important part to design of gradient coil with good linearity in the wide imaging range. Usually, Z-directional gradient field is generated by using the Helmholtz type coil which is consisted of one-pair loop with anti-current path. It gets less about 40% linearity of the diameter spherical volume(DSV). In this study, we calculated optimized geometrical parameters of two-pair loop system to cancel odd terms up to $B_7$ included effectively. we also analyzed and compared the gradient field distribution and linearity of the common Helmholtz coil with them of the two-pair loop system.

  • A linear ultrasonic motor was designed by a combination of the longitudinal and bending mode. Linear ultrasonic motors are based on an elliptical motion on the surface of elastic body, such as bar or plates. The corresponding eigen-mode of one resonance frequency can be excited twice at the same time with a phase shift of 90 degrees in space and time. That is excite symmetric and anti-symmetric modes. Then it determines the thrust and speed of the motor. Linear ultrasonic motors are investigated experimentally in according to be fabricated a general classification to motor structure and material characteristic. There was the first to simulate as use of finite element analysis ANSYS 9.0. The AL-T2W8-ARM14-LEG18-ANGLE80 motor has a maxim efficiency 17 [%] under the speed 0.14 [m/s], thrust 345 [gf] and preload 280 [gf], operating frequency is 57.6 [kHz].
