팔물탕의 항산화 효과와 자외선으로 유도된 각질형성세포 손상에 대한 보호효과 (Antioxidant and Protective Effects of Palmul-tang on Ultraviolet B (UVB)-induced Damage in Human Keratinocytes)

  • Objective : In this paper, we investigated the anti-oxidative capacities and protective effects of water extract of palmul-tang (PMT) against Ultraviolet B(UVB)-induced oxidative damage in human keratinocytes(HaCaT). Method : To evaluate the anti-oxidative activities of PMT, we measured scavenging activities on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation and reducing power of PMT. To give an oxidative stress to HaCaT cells, UVB was irradiated with $40mJ/cm^2$ to HaCaT cells. To detect the protective effects of PMT against UVB, we measured cell viability, apoptotic bodies and reactive oxygen species(ROS) generation. Results : PMT showed the anti-oxidative activities by scavenging DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation. Also PMT showed high reducing values. The UVB-induced oxidative conditions led to the cell apoptosis. However, treatment with PMT reduced oxidative stress conditions, including inhibition of cell apoptosis and expression of ROS. Conclusion : PMT had anti-oxidative activities and exhibited protective effects against UVB on HaCaT cells. PMT would be useful for the development of cosmetics treating UVB-induced skin aging.

  • In this paper, we studied the anti-oxidative capacities and protective effects of water extract of Gagam-Danguieumja(GDE) against Ultraviolet B(UVB)-induced oxidative damage in human keratinocytes(HaCaT). To evaluate the anti-oxidative activities of GDE, we measured scavenging activities on DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation and reducing power of GDE. To detect the protective effects of GDE against UVB, we irradiated with 40 mJ/㎠`s UVB to HaCaT cells then we measured reactive oxygen species(ROS) generation, apoptotic bodies and cell viability using DCFH-DA assay, Hoechst 33342 staining and MTT assay. GDE showed the anti-oxidative activities by scavenging DPPH radical, hydroxyl radical, hydrogen peroxide, superoxide anion, lipid peroxidation. Also GDE showed high reducing values. GDE reduced oxidative stress conditions by inhibition of ROS expression. Also the cell apoptosis by UVB-induced oxidative conditions was decreased by GDE treatment. These results could suggest that GDE had anti-oxidative activities and exhibited protective effects against UVB on HaCaT cells. GDE would be useful for the development of cosmetics treating UVB-induced skin aging.

Antioxidant and Hepatoprotective Effects of Tomato Extracts

  • The objective of present study was to investigate the anti oxidative and hepatoprotective effects of tomato extracts. Total antioxidant capacity and total antioxidant response were 5.5 and $19.8{\mu}g$ Trolox equivalent per mg of tomato extract, respectively. DPPH radical scavenging activity of tomato extracts ($10mg\;ml^{-1}$) was 70% as compared to 100% by pyrogallol solution as a reference. The effect of the tomato extracts on lipid peroxidation was examined using rat liver mitochondria induced by iron/ascorbate. Tomato extracts at the concentration of $0.5mg\;ml^{-1}$ significantly decreased TBARS concentration. Tomato extracts prevented lipid peroxidation in a dose-dependent manner. The effect of the tomato extracts on reactive oxygen species (ROS) generation was examined using cell-free system induced by $H_2O_2/FeSO_4$. Addition of $1mg\;ml^{-1}$ of tomato extracts significantly reduced dichlorofluorescein (DCF) fluorescence. Tomato extracts caused concentration-dependent attenuation of the increase in DCF fluorescence, indicating that tomato extracts significantly prevented ROS generation in vitro. The effect of tomato extracts on cell viability and proliferation was examined using hepatocyte culture. Primary cultures of rat hepatocytes were incubated with 1mM tert-butyl hydroperoxide (t-BHP) for 90 min in the presence or absence of tomato extracts. MTT values by addition of tomato extracts at the concentration of 2, 10, and $20mg\;ml^{-1}$ in the presence of t-BHP were 13, 33 and 48%, respectively, compared to 100% as control. Tomato extracts increased cell viability in a dose-dependent manner. These results demonstrate that tomato extracts suppressed lipid peroxidation and t-BHP-induced hepatotoxicity and scavenged ROS generation. Thus antioxidant and hepatoprotective effects of tomato extracts seem to be due to, at least in part, the prevention from free radicals-induced oxidation, followed by inhibition of lipid peroxidation.

Rhamnazin inhibits LPS-induced inflammation and ROS/RNS in raw macrophages

  • Purpose: The aim of this work was to investigate the beneficial effects of rhamnazin against inflammation, reactive oxygen species (ROS)/reactive nitrogen species (RNS), and anti-oxidative activity in murine macrophage RAW264.7 cells. Methods: To examine the beneficial properties of rhamnazin on inflammation, ROS/ RNS, and anti-oxidative activity in the murine macrophage RAW264.7 cell model, several key markers, including COX and 5-LO activities, $NO^{\cdot}$, $ONOO^-$, total reactive species formation, lipid peroxidation, $^{\cdot}O_2$ levels, and catalase activity were estimated. Results: Results show that rhamnazin was protective against LPS-induced cytotoxicity in macrophage cells. The underlying action of rhamnazin might be through modulation of ROS/RNS and anti-oxidative activity through regulation of total reactive species production, lipid peroxidation, catalase activity, and $^{\cdot}O_2$, $NO^{\cdot}$, and $ONOO^{\cdot}$ levels. In addition, rhamnazin down-regulated the activities of pro-inflammatory COX and 5-LO. Conclusion: The plausible action by which rhamnazin renders its protective effects in macrophage cells is likely due to its capability to regulate LPS-induced inflammation, ROS/ RNS, and anti-oxidative activity.

Effect of Structured Lipids Containing CLA on Hepatic Antioxidant Enzyme Activity in Rats Fed a Normal Diet

  • Conjugated linoleic acid (CLA) has been shown to have a range of biological activities, including anti-carcinogenic, anti-atherosclerotic, anti-adipogenic and anti-diabetogenic effects. Recent reports also showed that CLA has free radical scavenging capacity, which may have health benefits for human beings. The current study was performed to investigate the effect of structured lipid (SL)-containing CLA on plasma lipids and hepatic antioxidant enzyme activity. Sprague-Dawley mts were fed 5% and 10% SL-containing normal diet for 6 wks and these groups were compared to rats fed 5% and 10% corn oil. In plasma lipids, total-cholesterol was not affected by fat source or dietary fat level while triglyceride level decreased significantly in groups fed 10% fat diet compared to the other groups. Plasma thiobarbituric acid reactive substances (TBARS) level decreased significantly in the S5 and S10 groups compared to the C5 and C10 groups, although hepatic TBARS level was not altered by fat source. On the other hand, in terms of hepatic antioxidant enzyme activity, superoxide dismutase activity increased in the S10 group, whereas catalase activity decreased in the S10 group. Glutathione peroxidase activity decreased significantly in the SL groups compared to the C5 group. Glutathione reductase activity increased and glucose-6-phosphate dehydrogenase activity decreased in the C10 group compared to the C5 and C5 groups. In conclusion, the free radical scavenging activity of CLA seemed to suppress oxidative stress, which reduced lipid peroxidation resulting in lower hepatic antioxidant enzyme activity.

  • In this study, the anti-lipidperoxidative effects of G009, a polysaccharide extracted from Ganoderma lucidum IY009, was determined in ascorbic acid-$Fe^{2+}$-adenosine 5-diphosphate-intoxicated rat. In a model of ascorbic acid-Fe$^{2+}$-adenosine 5-diphosphate-induced hepatotoxicity in rat, G009 exhibited anti-lipidperoxidative effect in rat liver homogenate, and that malondialdehyde values of the liver homogenate inhibited from 48.1% to 74.8% in comparison to controls (p<0.05). The malondialdehyde formation in serum inhibited 66.5% at 100 mg/kg of G009. Also, serum levels of glutamic oxaloacetic transaminase and glutamic pyruvic transaminase in peroxidizer-induced rats treated with G009 was decreased compared with control. Especially, the formation of lipid peroxides in serum was related to glutamic pyruvic transaminase levels. These results suggest that G009 has a protective effect on ascorbic acid-$Fe^{2+}$-adenosine 5-diphosphate-induced hepatic injury through an inhibition of lipid peroxidation in liver.r.

가시오갈피 추출물의 항산화효과 (Antioxidant Effects of the Extracts of Acanthopanax senticosus)

  • Antioxidant properties of the extracts of Acanthopanax senticosus were investigated. The dried roots, stems or leaves were extracted with hot water or ethanol each. The ethanol extracts exhibited higher potency than aqueous extracts in scavenging free radicals and in inhibiting microsomal lipid peroxidation: the aqueous extracts of stems showed higher anti-oxidant effects than the root extracts. Copper-mediated LDL oxidation was also protected by the ethanol exlracts: antioxidant effects of the extracts tested were stronger than ascorbic acid, but not butylated hydroxytoluene. The activity of angiotensin converting enzyme was effectively suppressed by the aqueous extracts of the stems. However, in vivo antioxidant properties of the ethanol extracts of the stems did not seem to be significant, judged from the lipid peroxide values of serum and liver in normal mice. Thus, the ethanol extracts of the stems were shown to be more potent for protecting biological systems against various oxidant stresses in vitro, but not in vivo.

Structure-Activity Relationships of Gagaminine and Its Derivatives on the Inhibition of Hepatic Aldehyde Oxidase Activity and Lipid Peroxidation

  • In order to determine the structure-activity relationships for antioxidative effects of gagaminine, a steroidal alkaloid isolated from the roots of Cynanchum wilfordi (Asclepiadaceae), two derivatives identified as sarcostin and penupogenin were prepared from gagaminine by hydrolysis and reduction. These compounds were evaluated for the inhibitory effects on the aidehyde oxidase activity and on lipid perbxidation in vitro. Furthermore, their effects were compared with those of gagaminine and the related compounds, cinnamic acid and nicotinic acid. The results of this study prove that the cinnamoyl group in the structure of gagaminine is critical in inhibition of the aldehyde oxidase activity while the nicotinoyl group may be necessary for anti-lipid peroxidation of the compound.

  • Cyperus rotundus L. (Cyperaceae) has been used as an analgesic and antiiflammatory agent and in the treatment of menstrual disorder in folk remedies. Cyperi Rhizomata, processed and unprocessed, were extracted with methanol and fractionated with petroleum ether, chloroform. Butanol, water. The effect of unprocessed Cyperi Rhizoma and processed Cyperi Rhizoma on $CCl_4$ induced lipid peroxidation and hepatotoxity have been tested in rats. BuOH. Water fractions of unprocessed Cyperi Rhizome enhanced the inhibition of antilipid peroxidative effects in liver lipid. In chemical parameters obtained from serum analysis. Butanol fraction of unprocessed Cyperi Rhizoma showed significant decrease in hepatotoxicity. In result, unprocessed Cyperi rhizoma has significant antilipid peroxidative effect and hepatoprotective activity.

  • Some pharmacological activities of acanthoic acid, isolated from Acanthopanax koreanum root was investigated in animals. It is revealed that the compound had analgesic and anti-inflammatory activities without actions on central nervous system and showed inhibition of lipid peroxidation. The anti-inflammatory activity might be related to inhibition of prostaglandin E$_2$ synthesis in exudates of inflammation.
