뽕잎발효차 제조에 따른 in vivo 상에서의 S-180 항암 및 항알레르기 효과 (Anti-allergy Activity and in vivo for S-180 Solid Anti-cancer Effects in Manufacturing Fermented Mulberry Leaf Tea)

  • 본 연구는 식용식물로의 가치뿐만 아니라 약용으로 가치가 인정되는 뽕잎에 미생물 생균제를 이용한 뽕잎발효차 개발을 통하여 기능성 소재 개발 및 새로운 제품 개발의 방안을 제시하고자 뽕잎차 및 뽕잎발효차 추출물의 S-180 고형암, 항알레르기 활성을 비교분석 하였다. in vivo 상에서 S-180 고형암 억제 효과는 뽕잎차에서 16.67%, 뽕잎발효차에서 17.78%로 나타났다. 뽕잎차 및 뽕잎발효차 추출물의 항알레르기 효과를 검증한 결과 두 군 모두 에탄올 추출물보다 열수 추출물이 히스타민 분비 억제에 더 효과적이었고, HMC-1의 염증성 cytokine을 측정한 결과 추출물 군에서는 에탄올 추출물이 열수 추출물보다 염증성 cytokine의 억제율이 더 높았고 뽕잎발효차군이 뽕잎차군에 비해 더 효과적이었다. 뽕잎발효차 추출물의 기능성이 더 우수한 것은 발효를 통해 생리활성 물질이 생성된 것이 원인으로 사료되며 이 점에 있어 구체적인 연구가 필요하다고 판단된다.

Amygdalin Regulates Apoptosis and Adhesion in Hs578T Triple-Negative Breast Cancer Cells

  • Amygdalin, D-mandelonitrile-${\beta}$-D-glucoside-6-${\beta}$-glucoside, belongs to aromatic cyanogenic glycoside group derived from rosaceous plant seed. Mounting evidence has supported the anti-cancer effects of amygdalin. However, whether amygdalin indeed acts as an anti-tumor agent against breast cancer cells is not clear. The present study aimed to investigate the effect of amygdalin on the proliferation of human breast cancer cells. Here, we show that amygdalin exerted cytotoxic activities on estrogen receptors (ER)-positive MCF7 cells, and MDA-MB-231 and Hs578T triple-negative breast cancer (TNBC) cells. Amygdalin induced apoptosis of Hs578T TNBC cells. Amygdalin downregulated B-cell lymphoma 2 (Bcl-2), upregulated Bcl-2-associated X protein (Bax), activated of caspase-3 and cleaved poly ADP-ribose polymerase (PARP). Amygdalin activated a pro-apoptotic signaling molecule p38 mitogen-activated protein kinases (p38 MAPK) in Hs578T cells. Treatment of amygdalin significantly inhibited the adhesion of Hs578T cells, in which integrin ${\alpha}5$ may be involved. Taken together, this study demonstrates that amygdalin induces apoptosis and inhibits adhesion of breast cancer cells. The results suggest a potential application of amygdalin as a chemopreventive agent to prevent or alleviate progression of breast cancer, especially TNBC.

Effect of Apoptosis Induction of Ailanthus altissima on Human Lung Carcinoma Cells

  • We investigated the inhibitory effects of solvent extracts from Ailanthus altissima in A549 human lung cancer cell. A. altissima has been recognized as a traditional healthy food due to its various biological activities against hypertension, strokes, fever, pain, neuralgia, inflammation, and cancer effects. Recently, it has been reported that the extracts of various wild vegetables show strong anti-cancer properties by induction of apoptosis. However, the mechanisms of their cytotoxicity in human lung cancer cells have been poorly understood. The present study was investigated the effects of solvent extracts from A. altissima on cell growth and apoptosis on A549 human lung cancer cells. A treatment of A. altissima inhibited the growth of A549 cells in a dose-dependent manner by inducing apoptosis. Especially, the chloroform fraction showed the highest anti-cancer effect among five kinds of fractions. And also, induction of apoptosis by chloroform fraction were associated with down-regulation of Bcl-2, and up-regulation of pro-apoptotic Bax expression. From these results, A. altissima may have a therapeutic potential in human lung cancer cells and as a functional food.

Role of Pentacyclic Triterpenoids in Chemoprevention and Anticancer Treatment: An Overview on Targets and Underling Mechanisms

  • The incidences of cancer are continuously increasing worldwide, affecting life of millions of people. Several factors associated with the internal and external environment are responsible for this deadly disease. The key internal determinants like abnormal hormonal regulation, genetic mutations and external determinants such as lifestyle and occupational factors enhances onset of cancer. From the ancient time, plants were remained as the most trusted source of medicine for the treatment of diverse disease conditions. Extensive studies have been performed for the discovery of effective anticancer agent from the plant and still it is going on. Pentacyclic triterpenoids are biologically active phytochemicals having a different range of activities such as anti-inflammatory, hepatoprotective, anti-hypertensive, antiulcerogenic and anti-tumor. These compounds generally contain ursane, oleanane, lupane and friedelane as a chief skeleton of pentacyclic triterpenoids which are generally present in higher plants. Isoprene unit, phytochemical, with good antitumor/anticancer activity is required for the biosynthesis of pentacyclic triterpenoids. Mechanisms such as cytotoxicity, DNA polymerase inhibition, regulation of apoptosis, change in signal transductions, interfere with angiogenesis and dedifferentiation, antiproliferative activity and metastasis inhibition are might be responsible for their anticancer effect. Present review spotlights diverse targets, mechanisms and pathways of pentacyclic triterpenoids responsible for anticancer effect.

Hexane and Chloroform Fractions of Laetiporus sulphrueus var. miniatus Inhibit Thrombin-treated Matrix Metalloproteinase-2/9 Expression in Human Oral Squamous Carcinoma YD-10B Cells

  • Laetiporus sulphrueus var. miniatus is widely distributed worldwide, and has commonly been used as a medicinal mushroom. In the present study, we investigated the effects of water extract and solvent fractions from the Laetiporus miniatus as possible antioxidant, anti-thrombin and anti-invasive agents against phorbol 12-myristate 13-acetate (PMA)- or thrombin-induced matrix metalloproteinase-2 (MMP-2) and MMP-9 activities. Samples were fractionated into n-hexane, $CHCl_3$, ethyl acetate, n-butanol, and water fractions, and individually analysed. The water fraction had the highest extraction yield at 34.90% (w/w), while the n-butanol fraction demonstrated the highest anti-oxidative activity at 81.44%. In the thrombin inhibitory activity test, the water fraction exhibited the highest activity at 94.64%. Even at the concentration of $40{\mu}g/mL$, evaluation of anti-proliferating activity in YD-10B cells did not reveal any cytotoxic effects. Although MMP-9 expression in YD-10B cells increased after the addition of PMA and thrombin, MMP-2 did not. Additionally, MMP-2/-9 levels in PMA-treated YD-10B cells (i.e., both mRNA expression and protein activation) were highly inhibited in the hexane and chloroform fractions. Compared with MMP-2 levels, MMP-9 mRNA expression and proteolytic activity were inhibited to a greater extent by the hexane and chloroform fractions in thrombin-treated YD-10B cells. Taken together, these results support that thrombin induces tumor invasion through MMP-2/9 and suggest that the L. miniatus may act as an effective functional food, conferring anti-oxidative, anti-thrombotic and anti-cancer activities.

유근피 아세톤 추출물의 항산화 및 암세포 증식억제 활성 (Anti-oxidative and anti-proliferative activities of acetone extract of the cortex of Ulmus pumila L.)

  • 유근피 아세톤 추출물의 항산화 및 암세포 증식억제 활성을 확인하였다. 유근피 아세톤 추출물의 농도에 비례하여 유리라디칼 소거활성($EC_{50}=36.7{\mu}g/mL$)과 환원력($EC_{50}=53.2{\mu}g/mL$)이 증가하였다. 유근피 아세톤 추출물은 정상 세포인 인체 배아 폐상피세포(L132)에는 독성이 낮으면서 농도에 비례하여 인체 폐암세포(A549, $GI_{50}=74.3{\mu}g/mL$)와 대장암세포(SNU-C4, $GI_{50}=92.8{\mu}g/mL$)의 증식을 효과적으로 억제하였다.

Effects of Acanthopanax sessiliflorus on Immune Cells such as Thymocytes, Splenocytes and Macrophages in Mice

  • Objective : Immune potentiation including activation of T cells, B cells, macrophages, and dendritic cells is known to play a key role in prevention and treatment of patients with cancer. In this study, we investigated the effects of Acanthopanax sessiliflorus (AR) on the immune system, especially on thymocytes, splenocytes, and macrophages. Methods : We investigated the effects of AR on proliferation of splenocytes in normal mice, and the effects on proliferation of splenocytes and thymocytes in tumor-bearing mice. In addition, the effect of AR on NO production using macrophages was investigated. Results : Treatment with AR accelerated proliferation of splenocytes in vitro. AR also accelerated thymocyte proliferation, but did not affect splenocytes proliferation in normal mice. In contrast, AR accelerated proliferation of splenocytes and thymocytes significantly in tumor bearing mice. In addition, NO production level from macrophages was elevated by treatment with AR. Conclusion : These results demonstrate that AR has anti-cancer activities and related mechanisms are involved in immune potentiation such as acceleration of immune cell proliferation and elevation of NO production level in macrophages. In addition, we also demonstrate the possibilities of AR as complementary and alternative medicine to standard anti-cancer drugs.

가미보중익기탕이 항암활성 및 항전이효과에 관한 연구 (Study on Antitumor Activity of Kamibojungikgi-tang)

  • To explore the possible cancer agent from oriental prescriptions, we have examined its antitumor and anti metastatic activities of Kamibojungikgi-tang(KBIT). KBIT extracts exhibited cytotoxicity against P388, A549 and B16-F10 cell lines in a dose-dependent manner and showed antiadhesive effect of A549 cell to complex extracellular matrix at 1 ㎎/㎖ in vitro. In DNA topoisomerase I assay, KBIT extracts showed strong inhibitoty effect in a dose-dependent manner. In pulmonary colonization assay with B16BL6, a number of colonies in the lungs were decreased effectively in KBIT treated group as compared with control group. Moreover, in CAM assay, KBIT extracts significantly inhibited angiogenesis at 15㎍/egg as compared with control. The T/C% was 141% in KBIT treated group in S-180 bearing ICR mice. From the above results it was concluded that KBIT had antitumor and anti metastatic activities. So it is expected to be clinically helpful on the prevention and treatment of cancer, although it is still necessary to study its mechanism on molecular biology and immunology.

황기(黃芪)추출물의 항산화 효능과 에멀션 점탄성도에 미치는 영향에 관한 연구 (Study on Effects of Anti-oxidant and Viscoelastic on Emulsion by the Extract of Astragalus membranaceus)

  • Objectives : The Astragalus membranaceus has been used to diuretic, tonic, anti-viral, anti-oxidant activities in oriental herb medicine, and in each experiment, proved in this effects. Root of A. membranaceus includes according to flavonoid, saponin, polysaccharide, thus it has been studied for anti-oxidant, anti-inflammatory, anti-cancer activities. However, previous researches have been performed with being limited mainly to food industry. The present study was conducted anti-oxidative activity of A. membranaceus extracts. In addition, The objective of the present study was to determine the possibility for the application of emulsions in the development of A. membranaceus for cosmetics. Methods : In this study, aimed SOD like activity of A. membranaceus extract, effects of A. membranaceus extract on viscoelastic properties of emulsion was measured using rheometer. Results : In the results, 1, 3-butylene glycol extract of A. membranaceus efficiently reduce the active oxygen, SOD like activity of more than 40% decrement depend on concentration. And A. membranaceus extract increase the modulus of elasticity of emulsion, therefore A. membranaceus extract have not influence on stability loss. As such, the A. membranaceus extracts will remain stable when applied to emulsions or skin products. Conclusions : In consequence of this study, A. membranaceus extract can be effectively used in cosmetic emulsions when the relation between natural product extracts and formulation of cosmetics is elucidated. It is highly recommended that rheology be applied to determine the optimal extract concentration for cosmetic formulations.

Identification of Anti-Cancer Targets of Eco-Friendly Waste Punica granatum Peel by Dual Reverse Virtual Screening and Binding Analysis

  • Background: Punica granatum (family: Lythraceae) is mainly found in Iran, which is considered to be its primary centre of origin. Studies on pomegranate peel have revealed antioxidant, anti-inflammatory, anti-angiogenesis activities, with prevention of premature aging and reducing inflammation. In addition to this it is also useful in treating various diseases like diabetes, maintaining blood pressure and treatment of neoplasms such as prostate and breast cancer. Objectives: In this study we identified anti-cancer targets of active compounds like corilagin (tannins), quercetin (flavonoids) and pseudopelletierine (alkaloids) present in pomegranate peel by employing dual reverse screening and binding analysis. Materials and Methods: The potent targets of the pomegranate peel were annotated by the PharmMapper and ReverseScreen 3D, then compared with targets identified from different Bioassay databases (NPACT and HIT's). Docking was then further employed using AutoDock pyrx and validated through discovery studio for studying molecular interactions. Results: A number of potent anti-cancerous targets were attained from the PharmMapper server according to their fit score and from ReverseScreen 3D server according to decreasing 3D scores. Conclusion: The identified targets now need to be further validated through in vitro and in vivo studies.
