반복모형을 이용한 한우 초음파 측정형질의 유전모수추정 (Repeated Records Animal Model to Estimate Genetic Parameters of Ultrasound Measurement Traits in Hanwoo Cows)

  • 본 연구는 한우 암소 초음파 측정자료 특징을 알아보고, 측정형질에 대한 유전모수를 추정하여 육질 또는 육량 개량을 위한 기초자료를 제공하기 위해 수행되었으며, 2001년부터 2009년까지 한국종축개량협회에 의해 측정된 한우 암소 36,893두를 이용하였고, 그 중 반복기록이 있는 개체는 7,913두였다. 유전모수 추정을 위하여 반복개체모형을 이용하였으며, 유전모수 추정에는 REMLF90 (Miztal, 2001)을 이용하였다. 유전모수 추정모형의 설정을 위하여 흉위, 영양도 및 초음파 측정치에 대한 출생년도, 출생계절, 측정년도, 측정계절, 측정지역, 측정연령 등의 환경효과를 추정하였다. 반복개체모형으로 추정된 배최장근단면적, 등지방두께 및 근내지방도에 대한 유전력이 각각 0.31, 0.38, 0.27로 나타났고, 다형질개체모형으로 추정한 유전력은 각각 0.02, 0.09, 0.07로 낮게 추정되었다. 반복개체모형을 이용한 반복력은 배최장근단면적, 등지방두께, 근내지방도가 각각 0.46, 0.57, 0.39로 나타났다. 분석모형 간의 추정치의 차이를 비교 할 때 반복모형에서의 유전력과 반복력이 높게 추정되었다. 따라서 반복형질 값을 가진 형질들의 측정치를 표준화하여 한우암소개량을 위한 기초자료로 활용된다면 유전능력평가와 개량사업 비용절감에 도움이 될 것으로 기대된다.

Relationship of Slaughter Age and Carcass Traits of Hanwoo Cattle

  • Two experiments were carried out to investigate the relationship of slaughter age and carcass traits of Hanwoo cattle. A total number of 11,284 heads of cattle (4,624 bulls, 6,120 steers, and 540 females) were used in the experiment (Exp.) 1. In the Exp. 2, 3,887 heads of steers slaughtered at age of 24-36 mon were used. After 24 h post-slaughter chilling, the carcasses were weighed and evaluated by an official grader of carcass traits according to the Korean carcass grading standard. Backfat thickness was the main variable in yield index and grade whereas marbling score was the main variable in carcass quality grade. Meat color was an important factor affecting carcass quality grade of bulls; fat color was an important factor affecting those of steers and females; and maturity was an important factor affecting that of females. After 24-mon-old, extension of slaughter age did not automatically increase the quantity and quality of meat. However, 29 mon of age can be regarded as the optimum time to slaughter steers to attain the highest take home money for farmers.

  • c-MYC (v-myelocytomatosis viral oncogene homologue) is a transcription factor that plays important role in many biological process including cell growth and differentiation, such as myogenesis and adipogenesis. In this study, we aimed to detect MYC gene polymorphisms, their genotype frequencies and to determine associations between these polymorphisms and meat quality traits in Berkshire pigs. We identified a single nucleotide polymorphism (SNP) in intron 2 of MYC gene by Sanger sequencing, i.e., g.3350G>C (rs321898326), that is only found in Berkshire pigs, but not in other breeds including Duroc, Landrace, and Yorkshire pigs that were used in this study. Genotypes of total 378 Berkshire pigs (138 sows and 240 boars) were determined using Hha I restriction enzyme digestion after polymerase chain reaction. Observed allele frequencies of GG, GC, and CC genotypes were 0.399, 0.508, and 0.093 respectively. Statistical analysis indicated that the g.3350G>C polymorphism was significantly associated with $pH_{45min}$ and cooking loss (p<0.05), suggesting that g.3350G>C SNP can be used for pre-selection of $pH_{45min}$ and cooking loss traits in Berkshire pigs.

  • The aim of the study was to investigate pig reference families, generated from Korean native pigs (KNP) that were crossed with Yorkshire (YS) breeds, which were used to evaluate genetic markers to select breeding animals with superior pork quality. A set of five candidate genes (PRKAG3, MC4R, CAST, ESR, and PRLR ) was analyzed for association with pork quality traits. PRKAG3 (I199V) SNP genotypes were significantly associated with muscle moisture, protein, and fat contents. The MC4R D298N polymorphism was significantly associated with meat tenderness and color traits. The CAST polymorphism was significantly associated with muscle moisture and crude protein traits. These three genes have been associated with pork quality traits in other pig populations, and some of our results are consistent with earlier studies. In addition, two reproductive candidate genes (ESR and PRLR ) did not have significant associations. These results suggest that further study is warranted to investigate and develop more DNA markers associated with pork quality in our KNP-crossed pig families.

Identification of long non-coding RNA-mRNA interactions and genome-wide lncRNA annotation in animal transcriptome profiling

  • Protein-translated mRNA analysis has been extensively used to determine the function of various traits in animals. The non-coding RNA (ncRNA), which was known to be non-functional because it was not encoded as a protein, was re-examined as it was studied to actually function. One of the ncRNAs, long non-coding RNA (lncRNA), is known to have a function of regulating mRNA expression, and its importance is emerging. Therefore, lncRNAs are currently being used to understand the traits of various animals as well as human diseases. However, studies on lncRNA annotation and its functions are still lacking in most animals except humans and mice. lncRNAs have unique characteristics of lncRNAs and interact with mRNA through various mechanisms. In order to make lncRNA annotations in animals in the future, it is essential to understand the characteristics of lncRNAs and the mechanisms by which lncRNAs function. In addition, this will allow lncRNAs to be used for a wider variety of traits in a wider range of animals, and it is expected that integrated analysis using other biological information will be possible.

Association between PCR-RFLP Polymorphisms of Five Gene Loci and Milk Traits in Chinese Holstein

  • The objective of this study was to assess the association of polymorphisms in ${\kappa}$-cn, ${\beta}$-lg, ${\beta}$-lg 5′ flanking region, CSN1S2, and IGFBP-3 genes with milk production traits and mastitis-related traits in Chinese Holstein. Traits analyzed were 305 day standard milk yield, protein percentage, fat percentage, the ratio of fat percentage and protein percentage, pre-somatic cell count, somatic cell count, and somatic cell score, respectively. CSN1S2 locus was uninformative because only one genotype BB was found in Chinese Holstein. Allele frequencies of A and B in IGFBP-3 gene were 0.5738 and 0.4262 in Chinese Holstein population, which was different from reported Qinchuan cattle population. The genotypes of animals at IGFBP-3 locus significantly affected 305 day standard milk yield, protein percentage, and somatic cell score. The ${\beta}$-lg genotypes had a significant effect on protein percentage and the ratio of fat percentage and protein percentage. Polymorphism in ${\beta}$-lg 5′ flanking region was associated with 305 day standard milk yield, protein percentage, fat percentage, pre-somatic cell count, and somatic cell count. No significant associations of the polymorphism in ${\kappa}$-cn gene were observed for any trait.

Association of Candidate Genes with Production Traits in Korean Dairy Proven and Young Bulls

  • This study was performed to offer effective basic data for selection and improvement of Korean dairy cattle through identifying distributional properties among candidate genes (bovine butyrophilin, signal transducers and activators of transcription 5a, and prolactin hormone). In this study, polymorphisms of candidate genes were identified and the relationships between loci and production traits of each gene were analyzed using frozen semen of Holstein bulls (19 proven and 77 candidates). In butyrophilin (BTN) locus, polymorphisms information contents (PIC) value of BTN2 (0.372) was higher than those of others (BTN1; 0.155, BTN3; 0.254, BTN4; 0.169). As a result of analysis of genotyping STAT5a, using single stranded conformational polymorphism (SSCP) method and microsatellite locus, PIC values were 0.189 and 0.457, respectively. And PIC value of prolactin hormone gene was 0.176. In the relationships between genotypes and production traits, BTN3 was associated with 305-day production traits (p<0.05). PTAs for B allele were such as 110.43, 88.28 and 75.25 in BTN1, 3, 4 and these values were higher than those of A allele, but in the case of BTN2, A allele with 154.19 was higher than that of B allele. The results obtained from using candidate genes may be used as an useful index for the genetic improvement of dairy cattle population in Korea, and further studies are needed.

Polymorphism of Insulin-like Growth Factor-I Gene in 13 Pig Breeds and its Relationship with Pig Growth and Carcass Traits

  • The polymorphism of insulin-like growth factor-I (IGF-I) in 13 pig breeds (total n=559) was detected by PCR-Hha I- RFLP, and allele A (151 bp and 28 bp) or allele B (116 bp, 35 bp and 28 bp) were observed. In these pig breeds, it was found that European pig breeds carried high frequencies of allele B, while Chinese native pig breeds carried high frequencies of allele A. Meanwhile the role of porcine IGF-I was investigated in 117 Nanchang White pigs and 360 Large Yorkshire pigs. Eight traits about growth and carcass were recorded for analyzing the associations between IGF-I gene polymorphism and performance quantitative traits. In the Nanchang White pigs, those with AA genotype generally had higher birth weight than those with AB genotype (p<0.05), but all these genotypes had no significant effect on the other traits which had been analyzed. In Large Yorkshire pigs, those with BB genotype had higher 2 months and 6 months body weight than those with AA genotype (p<0.05), and had a thicker hind-back-fat thickness and mid-back-thickness than those with AB and BB genotypes (p<0.05). And those with BB genotype were the thinnest in Large Yorkshire. Furthermore, pigs with AA genotype had a lower lean percentage than those with AB and BB genotypes (p<0.01), and the lean percentage of those with BB genotype was the highest. Based on these results, it is possible to make the IGF-I gene locus into the application of marker-assisted selection programmes.

Effects of Dietary Supplemental $Megazone^{(R)}$ on Growth Performance, Nutrients Digestibility, Blood Characteristics, Meat Quality and Carcass Traits in Weaning-to-Finishing Pigs

  • This study was conducted to investigate the effects of $Megazone^{(R)}$ (a mineral mix) supplementation on growth performance, nutrients digestibility, blood characteristics, meat quality and carcass traits in weaning-to-finishing pigs. A total of 48 crossbred $(Landrace{\times}Yorkshire{\times}Duroc)$ pigs with initial body weight (BW) of $4.46{\pm}0.18kg$ were used in a 21 wks trial. Pigs were blocked by weight and allotted to two dietary treatments in a completly randomized design. There were 6 pens per treatment. Dietary treatments included: 1) Control (CON: basal diet) and 2) MT (basal diet+0.8% $Megazone^{(R)}$). Through the entire experimental period, there were no effects of dietary $Megazone^{(R)}$ supplementation on growth performance nutrients digestibility, blood characteristics and meat quality traits (p>0.05). Market weight and backfat thickness also had no differences between the two treatments (p>0.05). However, carcass weight and carcass ratio in MT treatment were improved significantly compared with CON treatment (p<0.05). In conclusion, supplmentation of $Megazone^{(R)}$ can increase carcass weight and carcass ratio in weaning-to-finishing pigs, however, it has no effects on growth performance, nutrients digestibility, blood characteristics and meat quality traits.

Genome-wide Association Study for Warner-Bratzler Shear Force and Sensory Traits in Hanwoo (Korean Cattle)

  • Significant SNPs associated with Warner-Bratzler (WB) shear force and sensory traits were confirmed for Hanwoo beef (Korean cattle). A Bonferroni-corrected genome-wide significant association (p< $1.3{\times}10^{-6}$) was detected with only one single nucleotide polymorphism (SNP) on chromosome 5 for WB shear force. A slightly higher number of SNPs was significantly (p<0.001) associated with WB shear force than with other sensory traits. Further, 50, 25, 29, and 34 SNPs were significantly associated with WB shear force, tenderness, juiciness, and flavor likeness, respectively. The SNPs between p = 0.001 and p = 0.0001 thresholds explained 3% to 9% of the phenotypic variance, while the most significant SNPs accounted for 7% to 12% of the phenotypic variance. In conclusion, because WB shear force and sensory evaluation were moderately affected by a few loci and minimally affected by other loci, further studies are required by using a large sample size and high marker density.
