Effects on Chemical Compositions and Digestibilities of the Bulking Agents as a Moisture Control and fermentation Methods of food Waste (음식찌꺼기의 발효사료화시 수분조절제와 발효방법이 화학적 조성분 및 소화율에 미치는 영향)

  • Studies were conducted to know effects of the bulking agents (saw dusts, mushroom waste, wheat bran coconut meal, rice hulls) adding o moisture control, fermentation methods (aerobic and anaerobic) and periods (1 to 20 days) of food waste fermentation for animal feeds on chemical compositions and in vitro DDM (digestibility of dry matter). Experiment designs were focussed basically to obtain extension service data. The NDF (neutral detergent fiber) composition in the oak and pine saw dust were 93.5% and 95.4% (DM basis) in respectively. Thus, the fermented food waste feeds using saw dust (50%) increased NDF(12%), and decreased in vitro DDM(48%) compared to those of raw materials before aerobic fermentation. The oak saw dust showed higher DDM compared to pine. Mushroom wastes which is a residues of mushroom culture mixed originally willow saw dust (80%) and wheat bran (20%) showed quite higher feed value compared to both saw dusts. It was found that an in vitro DDM and NDF composition in fermented feeds appeared highly dependent or the NDF composition in bulking agents. With an increase wheat bran ratio substitute mushroom waste showed linearly decreased NDF, and increased in vitro DDM in the fermented food waste feeds. The fermented feeds added bottling agents composed higher NDF resulted in higher NDF and lower in vitro DDM with prolonged fermentation time. The feeds from anaerobic fermentation appeared lower NDF and higher in vitro DDM compared to those of aerobic fermentation.

Effect on Instant Buckwheat Noodle on Digestibility and Lipids Profiles of Liver and Serum in Rats (인스탄트 메밀국소가 백서의 소화흡수율, 간장 및 혈청지질 농도에 미치는 영향)

  • To evaluate a possible effects of instant buckwheat noodle on hepatic and serum lipids profiles, apparent digestibilities and growth rate, male young rats were fed diets containing sucrose as a control group, instant 100% wheat flour noodle, or instant wheat noodle fortified with either 10% or 30% buckwheat flour as experimental groups for 4 weeks. Weight gain and feed intake in the two buckwheat noodle fed groups were similar to those of the wheat flour fed groups. In addition, no significant differences were found in the apparent digestibilities among the groups experimented. There were no differences in the hepatic and serum cholesterol levels due to the diets. However, Buckwheat noodle group showed a trend to decrease the liver and serum concentration of triacylglycerol, compared to the sucrose and 100% wheat flour groups. The addition of buckwheat flour to wheat flour up to 30% seemed to be effective in lowering the hepatic triacylglycerol level. After 10hr fasting, the serum glucose level of the buckwheat noodle group was similar to the levels of the other groups. The presented results indicate that instant noodle containing buckwheat at the level of 30% has a mild hypotriglyceridemic effects, and no adverse effects on the digestion of macronutrients in rats.

Changes in Rural Health and Environmental Factors (2001-2010) A Comparison between Agricultural and Nonagricultural Groups (농촌 환경ㆍ건강 지표의 변화 추이(2001~2010년) -농촌 거주 농업종사자와 비농업종사자의 비교-)

  • To verify welfare needs, rural health and environmental factors were compared between agricultural and nonagricultural groups over the past 10 years. Based on a survey of rural life by the Rural Development Administration, whose sample included agricultural (72.8%) and nonagricultural (27.2%) groups, all factors were analyzed using SAS ( 9.3). For rural home heating, the use of oil boilers (77.2%-78.0%) decreased (53.0%-53.7%) over the same period, whereas that of electric boilers increased (21.9% and 13.5% for agricultural and nonagricultural groups, respectively). The joint water- supply increased, and the use of flush toilets (52.4% to 84.5% in the agricultural group and 64.6% to 81.4% in the nonagricultural group) and hot-water bathing was higher in the agricultural group (79% to 92.6%) than in the nonagricultural group (72.2% to 87.6%) at the p<0.001 level. Incineration accounted for the largest portion of household waste until 2006, when it was replaced by other treatment methods such as the pay-per-treatment option (44.4% and 68.3% in the agricultural and nonagricultural groups, respectively) (p<0.001). Garbage disposal in 2001 was mainly burial or animal feed, but separate collection increased in 2010 (42.2% and 64.3% in the agricultural and nonagricultural groups, respectively) (p<0.001). The self-perception of health was lower in the agricultural group than in the nonagricultural group (2.96 to 2.74 on a five-point scale in the agricultural group and 3.07 to 2.98 in the nonagricultural group). Drinking decreased less in the agricultural group (criteria: one point once a week,; 1.48 to 1.20) than in the nonagricultural group (1.13 to 0.80) at p<0.001, and a similar pattern was found for smoking (p<0.001). Health screening decreased in the last 10 years (scoring based one point/1-2 times per year,: 0.94 to 0.64 in the agricultural group and 1.08 to 0.69 in the nonagricultural group;p<0.01). These results indicate that various health and environmental factors were less favorable to farmers except for bathing in comparison to nonagricultural living in the same area. This suggests that people in agriculture are less likely to live and work in favorable environments than those outside the sector and thus indicates a need for more attention to the national health welfare system for farmers for systematic realization.

Production and Characterization of Ethanol- and Protease-Tolerant and Xylooligosaccharides-Producing Endoxylanase from Humicola sp. Ly01

  • This paper reports the production and characterization of crude xylanase from the newly isolated Humicola sp. Ly01. The highest (41.8 U/ml) production of the crude xylanase was obtained under the optimized conditions (w/v): 0.5% wheat bran, 0.2% $KH_2PO_4$, and 0.5% peptone; initial pH 7.0; incubation time 72 h; $30^{\circ}C$; and 150 rpm. A considerable amount of the crude xylanase was induced using hulless barley bran or soybean meal as the carbon source, but a small amount of the enzyme was produced when supplementary urea was used as the nitrogen source to wheat bran. The crude xylanase showed apparent optimal cellulase-free xylanase activity at $60^{\circ}C$ and pH 6.0, more than 71.8% of the maximum xylanase activity in 3.0-30.0% (v/v) ethanol and more than 82.3% of the initial xylanase activity after incubation in 3.0-30.0% (v/v) ethanol at $30^{\circ}C$ for 2 h. The crude xylanase was moderately resistant to both acid and neutral protease digestion, and released 7.9 and 10.9 ${\mu}mol/ml$ reducing sugar from xylan in the simulated gastric and intestinal fluids, respectively. The xylooligosaccharides were the main products of the hydrolysis of xylan by the crude xylanase. These properties suggested the potential of the crude enzyme for being applied in the animal feed industry, xylooligosaccharides production, and high-alcohol conditions such as ethanol production and brewing.

Anti-Obesity Activity, Anti-Cancer Activity and Single Oral Dose Toxicity of Inonotus xeranticus Extracts (기와층버섯 추출물의 항비만활성, 항암활성 및 단회경구독성시험)

  • In this study, we investigated the in vitro anti-obesity, anti-cancer activity and single oral dose toxicity of Inonotus xeranticus extracted by methanol (INXM) or ethyl acetate (INXE). In order to investigate anti-obestity effect of Inonotus xeranticus extracts, the 3T3-L1 cells were treated with these extracts at various concentrations(1, 10, 100 and $300{\mu}g/ml$). It was observed that 3T3-L1 cells treated with $100{\mu}g/ml$ of Inonotus obliquus ethyl acetate extract (INOE), INXM and INXE, in the absence of differentiation cocktail (0.5mM isobutylmethylxanthine (IBMX) $1{\mu}M$ dexamethasone, $1{\mu}M$ insulin), differentiated at a rate of 78.5, 80.9, and 76.4% respectively. Differentiation rates of 86.6% and 83.4% were observed in 3T3-L1 cells which were treated with differentiation cocktail at $100{\mu}g/ml$ of INXM and INXE, respectively. The anti-cancer effect of Inonotus xeranticus extracts was investigated using a method of sulforhodamine B in sarcoma 180 cell line. The cells were treated with these extracts (1, 10, 100 and $300{\mu}g/ml$) for 48 hours. The growth of cells which were treated with $300{\mu}g/ml$ of INXM was inhibited by 80.1%. The growth of sarcoma 180 cells which were treated with 100 and $300{\mu}g/ml$ of INXE was inhibited by 74.7% and 64.5%, respectively. In single oral dose toxicity study, no differences were observed between control and treated groups in clinical signs, body weight gains, and feed and water consumptions. The results indicated that Inonotus xeranticus extracts did not show any toxic effects at 2,000mg/kg in mice, and the $LD_{50}$ of these extracts was found to be higher than 2,000 mg/kg in this experiment. From the above results, Inonotus xeranticus methanol and ethyl acetate extracts might have useful clinical applications in the management of cancer and obesity and may also be useful as a medicinal food.

Feed Value and Yield of Whole Crop Barley Varieties Depend on Organic Content (유기물함량이 총체보리 품종의 수량과 사료가치에 미치는 영향)

  • This experiment was carried out to evaluate the effects of organic content and to select lodging resistance for six varieties in paddy field of Chungnam province from 2006 to 2007. In this experiment, plant height was increased at the high fertile field and dry matter rate (%) per fresh matter was decreased at the high fertile soil condition but dry matter rate (%) of spike was increased. The dry matter (DM) yield showed higher in Sunwoo barley (13,520kg/ha) and Youngyang barley (12,780kg/ha) at the high fertile field. Fresh yield was also high in Wooho barely (34,720kg/ha) and Youngyang barely (33,670kg/ha) at the high fertile field but yield in high and normal fertile soil condition was not difference. Total digestible nutrients (TDN) content of Youngyang barley and Yuyeon barley was high as 69.5%, 70.3%, respectively, at the high fertile soil condition. According to this result Soman barley and Youngyang barley might be recommendable for whole crop barely in lower organic content condition of paddy rice field and Wooho barley and Soman barley with the lodging resistance and high yield is suitable for paddy field with high organic content.

Effect of Different Parts and Growing Stages of Miscanthus sacchariflorus as a non-Food Resource that does not Contribute towards Climate Change on Metabolic Availability in Ruminants (반추가축전용 기후변화대응 비식량자원 거대억새의 생육부위 및 시기에 따른 체내 이용가치 비교 연구)

  • Miscanthus sacchariflorus var. No. 1 has been newly developed in Korea. This study was conducted to assess the feed value of M. sacchariflorus var. No. 1 at different growth and harvesting time. Total 3 different miscanthus - 1y4m (first shoot and harvested at 4 month), 2y4m (second shoot and harvested at 4 month) and 2y8m (second shoot and harvested at 8 month). Two experiments were carried out, In vitro rumen simulated fermentation and In situ dry matter digestibility (DMD). Ruminal pH at in vitro fermentation were higher in M. sacchariflorus var. No. 1 treatments compared to the rice straw (RS). In volatile fatty acid production, 1y4m resulted in higher acetate production than the other M. sacchariflorus var. No. 1 at higher maturity stages. Significant differences among treatments were observed in propionate and total volatile fatty acid (VFA) productions at 9, 24 and 48 h of incubation times. Higher ammonia nitrogen productions were found as increased maturity of M. sacchariflorus var. No. 1. At In situ experiment, high DMD was detected in the order of RS (60.51%) > 1y4m (57.65%) > 2y4m (57.63%) > 2y8m (46.28%). The results from this study indicate that young and early harvested M. sacchariflorus var. No. 1 are able to improve its nutrient values in the ruminant animal.

Anthocyanin Stability and Silage Fermentation Quality of Colored Barley

  • This study was conducted to observe the fermentative quality and anthocyanin content in whole crop colored barley silage during storage periods and anthocyanin stability in in vitro ruminal fluid. Silages of colored barley cultivar "Boanchalbori" and normal barley cultivar "Yuyeonbori" were stored during 0, 2, 4, 6, and 12 months. The in vitro ruminal fluid was fermented for 0, 6, 12, 24, and 48 hrs. For the feed value, crude protein of colored barley silage was slightly increased in the silage compared to that of normal barley silage, and being increased up to 2 months after ensiling and thereafter maintained at the similar level. Neutral detergent fiber (NDF) and acid detergent fiber (ADF) contents of both the barley significantly increased by prolonged storage of 2 months (p<0.05), but they were maintained at the constant level after 2 months of storing silage. Whereas TDN (total digestible nutrients) contents of them were decreased by the prolonged storage of 2 months (p<0.05), then maintained at the constant levels. The fermentative quality and pH values in both the barley silages were slightly decreased during the storage time. Lactic acid and acetic acid contents were increased during prolonged storage period, but not significantly different among treatments. Butyric acid was not detected. In the colored barley silage, pH value showed slightly lower compared to that of the normal barley silage but not significant, and lactic acid content was significantly higher than the normal barley silage (p<0.05). The total anthocyanin content in the whole crop colored barley silage decreased to 42% after 2 months of ensilage, however maintained at the constant level until 12 months of ensilage. In the case of anthocyanin stability on in vitro ruminal fluid digestion, the pH value of the ruminal fluid was slightly lower at 6, 12, 24, 48h incubation time and the content of anthocyanin was at similar levels. These results indicated that the colored barley showed higher fermentation quality, and total anthocyanin content was maintained stable at 42% level of the first value in storing silage. As the anthocyanin had higher stability in the ruminal fluid, the colored barley has a potential as functional feeds for Ruminants.

Patterns of Waterbirds Abundance and Habitat Use in Rice Fields (논습지에 도래하는 수조류의 서식지 이용과 개체군 특성)

  • BACKGROUND: As natural wetlands are decreased by increment of human demand, the importance of rice fields as an alternative habitat for waterbirds is well documented. However, the relationship between waterbirds use and management practice of rice fields has not been fully understood. The present study attempted to understand the changes in temporal abundance of waterbirds and their preference for habitat types in rice fields all year round. METHODS AND RESULTS: Waterbirds census were conducted in rice fields around Asan bay in Korea during April 2009-March 2010 and April 2011-March 2012. In the bird counts, the locations of the observed birds on a 1/2,500 map were recorded along with the local habitat type (paddy, ditch, levee, road). Thirty five species of waterbirds recorded in the rice fields during the survey period and three major groups (shorebirds, herons, and waterfowls) were characterized according to season and micro-habitat use. Shorebirds visited a flooded paddy for feeding during their spring migration season (April-May), and herons used the rice field as feeding sites during their breeding periods (April-October). Most waterfowls were observed in a dry paddy to feed a fallen rice seed and stubs during the winter season (September-March). Waterbird groups selectively used micro-habitats in rice field. Shorebirds and waterfowls mainly preferred at rice paddies, while herons were attracted to most habitat types. CONCLUSION(S): Rice fields supported various waterbirds all year round and waterbird communities using the rice fields were dramatically changed according to seasonal change of rice field condition.

Improvement of Antibacterial Activities of Bacteriocidal Yeasts Using the GPD Promoter (GPD 프로모터를 이용한 항균활성 효모의 활성증진)

  • We have previously reported recombinant productions of bacteriocins using yeast expression plasmid pAUR123, which contains the alcohol dehydrogenase (ADH) promoter, in Saccharomyces cerevisiae cells and their antibacterial activities. In order to improve the antibacterial activities of bacteriocidal yeast cells, a strong glyceraldehyde phosphate dehydrogenase (GPD) promoter gene of S. cerevisiae was amplified and inserted upstream into bacteriocin genes such as the OR-7, Subpeptin JM4-A or JM4-B gene in the corresponding recombinant yeast plasmid. Yeast cells transformed by the recombinant plasmid containing the GPD promoter represented higher antibacterial activities against both Gram positive B. subtilis and Gram negative E. coli cells compared to those transformed by the corresponding recombinant plasmid containing the ADH promoter. Thus, yeast cells harboring the recombinant plasmid containing the GPD promoter constructed in this study could be applied in the food preservative or animal feed industries.
