The Effects of Serum Lipids and Blood Glucose on Retort Samgyetang Intake in Rats (흰쥐에 레토르트 삼계탕의 투여가 혈청지질 및 혈당에 미치는 영향)

  • This study presents the effects of serum lipids and blood glucose on retort Samgyetang intake in rats. For this purpose, we measured the ingredients and contents of Samgyetang, and produced a diet for experimental rats. The animal experiment was done in three different diet groups: control, Samgyetang(I) and Samgyetang(II). The results were as follows; 1) There was no sign of difference between the three groups in the body weight gain and food intake due to the effect of dietary component. 2) The density of serum triglyceride in the Samgyetang(II) group was decreased, but there was no sign of difference between the control group and Samgyetang(II). 3) There were no statistically significant differences of serum cholesterol and phospholipid among my of the diet groups. 4) The free fatty acid density and blood glucose level in the Samgyetang(I) and (II) groups were higher than in the control group. In the author's opinion, it is necessary to reexamine them for a long period through the animal experiment.

  • Holstein cows being raised in the Kangweon district were examined on the values of blood chemistry in infertile cows. The results obtained were as follows; 1. Total serum protein was 7.52$\pm$0.45g/100ml 2. Serum albumin was 3.06$\pm$0.28g/100ml 3. In values of serum globulin, $\alpha$1-globulin, $\alpha$2- globulin, $\beta$-globulin and ${\gamma}$-globulin were 0.79$\pm$0.14, 0.60$\pm$0.10, 0.86$\pm$0.13 and 2.21$\pm$0.35ng/100ml, respectively. 4. A/G ration was 0.69$\pm$0.09 5. Total cholesterol in serum was 119.73$\pm$34.52mg/100ml 6. Blood glucose was 55.24$\pm$6.46mg/100ml 7. Serum calcium was 9.27$\pm$0.87mg/100ml 8. Serum inorganic phosphorus was 5.74$\pm$0.86mg/100ml 9. Serum glutamic oxalacetic transaminase was 47.90$\pm$11.02I$\mu$/l 10. Serum glutamic pyruvic transaminase was 14.20$\pm$3.02I$\mu$/l

Comparisons of Growth, Mammary Development, Serum Proteins and Serum Concentrations of Estradiol, Growth Hormone and Prolactin between Korean Native and Holstein Heifers (한우와 홀스타인 암송아지의 성장, 유선발달, 혈청 단백질과 혈청 중 Growth Hormone, Prolactin 및 Estradiol의 농도 비교)

  • Growth, mammary development, serum concentrations of growth hormone, prolactin and estradiol which affect the growth and mammary development, were investigated in Korean Native heifers and Holstein heifers from 5 months old to 15 months old. From 5 months old to 15 months old, body weights in Korean Native heifers averaged 37%, lower than those in Holstein heifers of the same age (P<.01). Teat volumes in Korean Native heifers are average 67% smaller than those in Holstein heifers. Analysis of serum protein by secondary electrophoresis showed differences in protein pattern between the breeds and between the individuals. From 5 months old to 15 months old, serum concentration of growth hormone in Korean Native heifers was averge 47% lower than that in Holstein (P<.01), yet prolactin concentrations in serum were about the same between the two breeds. However serum concentration of estradiol in Korean Native heifers was average 20% higher than that in Holstein of same age (P<.01). This result showed evidently that the decrease of the growth hormone concentration in serum, while growing up, causes to retard the growth and mammary development in Korean Native heifers compared to Holstein heifers.

  • This study was undertaken to investigate the effects of ITS and EGF on embryonic development of in vitro matured and fertilized bovine oocytes in culture medium su, pp.ementing with or without calf serum. When fertilized oocytes were cultured in TCM-199 containing 0, 0.1, 0.5 and 1.0\ulcorner/ml ITS with 5% calf serum, the rates of development to blastocyst stage and the cell number of blastocysts were not significantly different among all treatments. And also monolayer of cumulus/granulosa cells prepared in containing calf serum and ITS were no beneficial effects of embryonic development. On the other hand, when EGF was su, pp.imented to TCM-199 containing calf serum or calf serum free, embryonic development rates(24.0 2.8% to 29.2 1.7% or 8% to 9%) and cell number of blastocysts(p<0.05) were significantly increased compared with EGF-free(22.1 2.1 or 1.0%, p<0.05). But when fertilized oocytes were cultured with cumulus/granurosa cells in TCM-199 containing EGF and calf serum, the rate of embryos development to the blastocyst stage and cell number of blastocysts were not significantly different compared with EGF-free and any concentrations. These results showed that ITS and EGF was not improved the development of bovine embryo in vitro matured and in vitro fertilized with calf serum and/or monolayer of cumulus/granulosa cells.

Effect of Suckling Systems on Serum Oxytocin and Cortisol Concentrations and Behavior to a Novel Object in Beef Calves

  • We investigated differences between effects of natural- and bucket-suckling methods on basal serum oxytocin (OT) and cortisol concentrations, and the effect of OT concentration on affiliative and investigative behavior of calves to a novel object. Ten Japanese Black calves, balanced with birth order, were allocated evenly to natural-suckling (NS) and bucket suckling (BS) groups. Blood samples were collected at the ages of 1 and 2 months (1 week after weaning) calves, and serum OT and cortisol concentrations were measured using enzyme-linked immunosorbent assay and enzymeimmunoassay tests, respectively. Each calf at the age of 2 months (2 weeks after weaning) was released into an open-field with a calf decoy, and its investigative and affiliative behaviors were recorded for 20 minutes. In 1-month-old calves, the basal serum OT concentration ($25.5{\pm}4.9$ [mean${\pm}$standard deviation, pg/mL]) of NS was significantly higher than that of BS ($16.9{\pm}6.7$) (p<0.05), whereas the basal cortisol concentration ($5.8{\pm}2.5$ [mean${\pm}$standard deviation, ng/mL]) of NS was significantly lower than that in BS ($10.0{\pm}2.8$) (p<0.05). Additionally, a negative correlation was noted between serum OT and cortisol concentrations in 1-month-old calves (p = 0.06). Further, the higher serum OT concentration the calves had at 1 month old, the more investigative the calves were at 2 months old but not affiliative in the open-field with a calf decoy. Thus, we concluded that the natural suckling method from a dam elevates the basal serum OT concentration in calves, and high serum OT concentrations induce investigative behavior and attenuate cortisol concentrations.

Effects of Dietary L-carnitine Supplementation on Growth Performance, Organ Weight, Biochemical Parameters and Ascites Susceptibility in Broilers Reared Under Low-temperature Environment

  • The objective of this study was to investigate the effects of L-carnitine on growth performance, organ weight, biochemical parameters of blood, heart and liver, and ascites susceptibility of broilers at different ages reared under a low-temperature environment. A total of 420 1-d-old male Ross 308 broilers were randomly assigned to two dietary treatments with fifteen replicates of fourteen broilers each. Treatment diets consisted of L-carnitine supplementation at levels of 0 and 100 mg/kg. At 11-d of age, low temperature stress was used to increase ascites susceptibility. Blood, heart and liver samples were collected at different ages for analysis of boichemical parameters. The results showed that, there was no significant difference in growth performance with L-carnitine supplementation, but the mortality due to ascites was significantly decreased. Dietary L-carnitine supplementation significantly reduced heart index (HI) and ascites heart index (AHI) on d 21, lung index (LUI) on d 35 and liver index (LI) on d 42. The broilers fed diets containing L-carnitine had significantly lower red blood cell counts (RBC), hemoglobin (HGB) concentration and hematocrit (HCT) on d 42. Dietary L-carnitine supplementation significantly reduced malondialdehyde (MDA) content of heart tissue on d 21 and 35, and significantly increased total superoxide dismutase (T-SOD) and Glutathione peroxidase (GSH-Px) activity of the heart on d 21 and 42. L-carnitine supplementation significantly reduced serum triglyceride (TG) content on d 28 and 35 and serum glucose (GLU) on d 35 and 42, and significantly increased serum total protein (TP) and globulin (GLO) content on d 42. L-carnitine supplementation significantly enhanced liver succinodehydrogenase (SDH), malic dehydrogenase (MDH) and $Na^+$-$K^+$-ATPase activity on d 28, and tended to reduce the lactic acid (LD) level of liver on d 35 (p = 0.06). L-carnitine supplementation significantly reduced serum uric acid (UA) content on d 28, 35 and 42. Based on the current results, it can be concluded that dietary L-carnitine supplementation reduced organ index, red blood cell counts and hematocrit, enhanced antioxidative capacity of the heart, enhanced liver enzymes activity involved in tricarboxylic acid cycle, and reduced serum glucose and triglyceride. Therefore, it is suggested that L-carnitine can potentially reduce susceptibility and mortality due to ascites.

NMR-based metabolomic profiling of the liver, serum, and urine of piglets treated with deoxynivalenol

  • Deoxynivalenol (DON), a Fusarium mycotoxin, causes health hazards for both humans and livestock. Therefore, the aim of this study was to investigate the metabolic profiles of the liver, serum, and urine of piglets fed DON using proton nuclear magnetic resonance ($^1H-NMR$) spectroscopy. The $^1H-NMR$ spectra of the liver, serum, and urine samples of the piglets provided with feed containing 8 mg DON/kg for 4 weeks were aligned and identified using the icoshift algorithm of MATLAB $R^2013b$. The data were analyzed by multivariate analysis and by MetaboAnalyst 4.0. The DON-treated groups exhibited discriminating metabolites in the three different sample types. Metabolic profiling by $^1H-NMR$ spectroscopy revealed potential metabolites including lactate, glucose, taurine, alanine, glycine, glutamate, creatine, and glutamine upon mycotoxin exposure (variable importance in the projection, VIP > 1). Forty-six metabolites selected from the principal component analysis (PCA) helped to predict sixty-five pathways in the DON-treated piglets using metabolite sets containing at least two compounds. The DON treatment catalyzed the citrate synthase reactions which led to an increase in the acetate and a decrease in the glucose concentrations. Therefore, our findings suggest that glyceraldehyde-3-phosphate dehydrogenase, citrate synthase, ATP synthase, and pyruvate carboxylase should be considered important in piglets fed DON contaminated feed. Metabolomics analysis could be a powerful method for the discovery of novel indicators underlying mycotoxin treatments.

Effects of Short-term Acute Heat Stress on Physiological Responses and Heat Shock Proteins of Hanwoo Steer (Korean Cattle)

  • This study was performed to evaluate the effect of heat stress on the status of physiological responses, blood parameter, serum T3 and cortisol, and heat shock proteins (HSP 27, 70, and 90) of Hanwoo cattle. Six Hanwoo steers (242.8 ± 7.2 kg of BW) were housed in the climate-controlled respiration chambers. The experiment consisted of 7 days (control; 0 day) at thermoneutral (air temperature (Ta) of 15℃ and relative humidity (RH) of 60%; temperature-humidity index (THI) = 64), and by 3 and 6 days (treatment groups) at heat stress (Ta of 35℃ and RH of 60%; THI = 87). Body temperature of each parts (frank, rump, perineum and foot) and rectal temperature elevated in heat stress groups (3 days and 6 days) than the control group (0 day). Respiration rates increased in 3 days and 6 days (88.5 ± 0.96 bpm and 86.3 ± 0.63 bpm, respectively) from 0 days (39.5 ± 0.65 bpm). Feed intake significantly decreased in heat stress groups (3 days and 6 days, 3.7 ± 0.14 kg and 4.0 ± 0.15 kg, respectively) than the control group (0 day, 5.0 ± 0.00 kg). In addition, final BW significantly decreased in heat stress groups (3 days and 6 days, 211.8 ± 4.75 kg and 215.5 ± 3.50 kg, respectively) than the control group (0 day, 240.0 ± 25.00 kg). However, heat stress has no significant effect on blood parameter, serum T3 and cortisol. Nevertheless, heat stress increased HSPs mRNA expression in liver tissue, and serum concentration of HSPs. Despite Hanwoo cattle may have high adaptive ability to heat stress, our results suggested that heat stress directly effect on body temperature and respiration rate as well as serum and tissue HSPs. Therefore, we are recommended that HSPs could be the most appropriate indicators of Hanwoo cattle response to heat stress.

Study of the Application of Fructooligosaccharides in Piglets

  • In this study, 90 crossbred weaned pigs(Duroc${\times}$Landrace${\times}$Large White)weighing - 7.86${\pm}$0.06 kg each were randomly allotted to one of three dietary treatments. Control pigs were a fed corn-soybean meal diet with no additives. The two treatment groups were fed the basal diet supplemented either with 75 mg/kg Aureomycin or 0.4% fructooligosaccharides (FOS) in order to study the effects on performance, serological indices, and enteric morphology in addition to examining the content of volatile fatty acids in intestinal digesta. The results indicate that the diets containing FOS and antibiotics had a significant effect on feed conversion ratios (FCR) and diarrhea incidence, as well as increasing the concentrations of isobutyric and butyric acid and total VFAs in the caecum, and acetic acid, isovaleric acid, and total VFAs in feces. Supplementation with FOS also resulted in significantly longer mucosal villi height and a higher percentage of goblet cells compared with the control. No difference was found in crypt depth among the three treatments. While serum glucose levels were significantly higher following FOS supplement, differences in serum total protein, albumin, globulin, and urea nitrogen levels were not significant.
