Effect of Intra-Uterine Infusion of Lipopolysaccharides on Reproductive Performance in Dairy Cows with Retained Placenta (후산정체 젖소에 있어서 Lipopolysaccharide의 처리가 번식 성적에 미치는 영향)

  • This study was carried out to investigate the effect intra-uterine infusion of lipopolysaccharides(LPS) on the reproductive performance of dairy cows with retained placenta, The LPS isolated from Bacteroids helcogenes and Fusobacterium varium was injected at the rate of 100 ${\mu}g$ with 30 ml of phospahte buffer saline in each cow(n=33) at day 20 postpartum. Twelve cows with retained placenta were not heated with LPS and served as a control. Conception rate after first service was 0% and 33,3% for control and LPS groups, respectively. Service period was $149.6{\pm}34.3\;and\;53.0{\pm}12.5$ days for control and LPS groups, respectively(p<0.05). Services per conception was $3.6{\pm}0.8\;and\;2.1{\pm}0.3$ for control and LPS groups, respectively(p<0.05). In conclusion, intra-uterine infusion of LPS in cows with retained placenta can reduce the service period and services per conception. Reduced service period can improve the dairy farm profitability by decreasing cost un feeding and management of cows.

  • Despite the extensive study of protein effect to man and domestic animals, protein effect of sunflower seed oil cake to the growth and value of erythrocyte in rats is still remained to be investigated. This study is to investigate the effect of protein of sunflower seed oil cake to the rat growth and value of erythrocyte as well as the liver weight, heart, kidneys and spleen of rats. Sixty to seventy-days-old, male Sprague-Dawley rats weighing 50-60g, were used in this experiment and 36 rats allotted to 4 groups: standard, 30% cake, 20% cake and low protein. The standard group was fed for 7 weeks on the ration supplemented with 17% casein, 30% and 20% cake groups with sunflower seed oil cake by 30% and 20%, respectively, and low protein group without any supplementation. To observe value of erythrocyte, blood was taken from heart. Value of erythrocyte, hemoglobin and hematocrit value were determined by usual method and oxygen consumption by the method of Watts and Gourley. The result of this experiment suggests that it is recommendable to supplement deficient amino acids in animal.

Comparison of Methane Production of Holstein Steers Fed Forage and Concentrates Separately or As a TMR (TMR과 조사료 농후사료 분리급여 방식의 홀스타인 거세우 메탄배출량 비교)

  • The objective of this study was to determine how feeding forage and concentrate separately (SF) or as a total mixed ration (TMR) affects enteric methane production of cattle. Six Holstein steers ($203{\pm}22.5kg$) were used in a $2{\times}3$ changeover design experiment. Experimental diets (TMR and SF) consisted of compound feed, timothy hay and soybean curd residue in a ratio of 40:48:12, respectively, and diets were fed at 10% of metabolic body weight, on an as-fed basis. There were no differences in dry matter intake and enteric methane production (g/d) between SF and TMR but the methane conversion rate (methane energy/GE intake) of TMR was significantly higher (p=0.05) than that of SF. The mean methane emission factor (kg/head/year) and conversion rate of the two treatments were 21.4 and 0.05, respectively. There was a strong relationship between metabolic body weight and enteric methane production (p<0.001). At the present time, further studies may be necessary in order to establish the effects of TMR and SF on enteric methane production.

Fatty Acid Biosynthesis of Chicken Fed Various Long Chain Fatty Acids (닭에서의 지방산(脂肪酸) 생합성(生合成)에 미치는 급여지방산(給與脂肪酸)의 영향(影響))

  • In order to investigate the effect of dietary long chain fatty acids on fatty acid biosynthesis of liver in birds, single comb White Leghron male chicks were fed a fat-free diet an diets containing margaric, stearic and linoleic acids and liver lipid components and liver and plasma fatty acid distributions were compared. Total lipids of tissues were extracted with a chloroform-methanol mixture. The lipid components were determined by thin layer chromatography and fatty acid distribution of lipid fractions were determined by gas liquid chromatography. Fatty acid feeding did not affect liver lipid components. When margaric acid(17 : 0), was fed, 17:0 and heptadecenoic acid(17:1) appeared in every lipid fractions of liver and plasma, and distribution values of these acids were not significantly different between the lipid fractions of liver. In blood plasma of the 17 : 0 fed chicks, however, significantly higher distribution values of 17 : 0 and 17.1 were observed in the triglyceride fraction and in the cholesterol ester fraction, respectively. Dietary stearic acid (18 : 0) did not show any effect on the distribution of 18 : 0 in every lipid fractions of liver but showed a significantly higher distribution value of 18 : 0 in the free fatty acid fraction of plasma. When linoleic acid (18 : 2) was fed, every lipid fractions of liver and plasma contained 18 : 2, especially a significantly higher distribution value was observed in the phospholipid fraction of liver. Dietary margaric and linoleic acids tended to decrease the distribution value of endogenously synthesized palmitoleic (16 : 1) and oleic (18 : 1) acids in liver.

Beneficial Effects of Dietary Antiacarcinogenic Conjugated Linoleic Acid(CLA) on the Performances of Laying Hens and Broilers

  • Effects of conjugated linoleic acid(CLA), known as an effective anticarcinogen in several aminal models, on the egg production and egg weight of laying hens, and the weight gains of broilers were investigated. CLA was synthesized from corn oil by the alkaline isomerization method and purified by the low-temperature precipitation method. Diets for laying hens and for broilers were synthesized to meet the specification of their NRC standard rationals. Two separated experiments(Experiment I and II) were conducted for laying hens. in experiment I, 45 hens(300 days of age) were divided into 15 hens per treatment group; each hen was housed in wired cage located in a temperature and humidity-controlled house and adopted to the control diet. One week later, each group was subjected to one the four treatment groups for 5 weeks : control, 1.0% CLA, 2.5% CLA and 5.0% CLA diets. Diet and water were ad libitum. The condition of experiment II was the same as that of experiment I except for the addition of 5% corn oil diet and the extension of feeding period to 7 weeks. Egg production, egg weight and feed intake were recorded every week. Forty-five broilers(10day of age) were adopted to the control diet for a week and then switched to the treatment diets for 5 weeks : control, 1.0% CLA, 2.5% CLA, 5% CLA and 5% corn oil, Body weight and feed intake of broilers were measured every week. Diets supplemented with various amounts of CLA enhanced the egg production and increased the egg weight regardless laying hen's age(150 days or 300 days) as compared to control diet. The most effective diet for the egg production and egg weight of young hens(150 days of age) was found to be 1.0% CLA diet, but relatively higher CLA diet(2.5% CLA) was required for old hens (300 day of age) to obtain similar results as seen in younger hens. All hens treated with CLA ate greater amount of feed than control hens. Broilers treated with various amount of CLA ate less feed as compared to control ones, but the body weight gain was greater than the control broilers. These results indicate that CLA enhanced the egg production and agg weight of laying hens, and increased the body weight gain of broilers with less diet consumption.

  • Type 2 diabetes mellitus (T2DM) is characterized by insulin resistance (IR). T2DM is correlated with obesity and most T2DM medications have been developed for enhancing insulin sensitivity. Silk protein fibroin (SPF) from spiders has been suggested as an attractive biomaterial for medical purposes. We generated transgenic rice (TR) expressing SPF and fed it to diabetic $BKS.Cg-m+/+Lepr^{db}$ mice to monitor the changes in blood glucose levels and adipose tissue proteins associated with energy metabolism and insulin signaling. In the present study, the adipocyte size in abdominal fat in TR-SPF-fed mice was remarkably smaller than that of the control. Whereas the adenosine monophosphate-activated protein kinase (AMPK)-activated protein kinase and insulin receptor substrate 1 (IRS1) protein levels were increased in abdominal adipose tissues after TR-SPF feeding, levels of six-transmembrane protein of prostate 2 (STAMP2) proteins decreased. Phosphorylation of AMPK at threonine 172 and IRS1 at serine 307 and tyrosine 632 were both increased in adipose tissues from TR-SPF-fed mice. Increased expression and phosphorylation of IRS1 at both serine 307 and tyrosine 632 in adipose tissues indicated that adipocytes obtained from abdominal fat in TR-SPF-fed mice were more susceptible to insulin signaling than that of the control. STAMP2 protein levels decreased in adipose tissues from TR-SPF-fed mice, indicating that STAMP2 proteins were reducing adipocytes that were undergoing lipolysis. Taken together, this study showed that TR-SPF was effective in reducing blood glucose levels in diabetic mice and that concurrent lipolysis in abdominal adipocytes was associated with alterations of AMPK, IRS1, and STAMP2. Increased IRS1 expression and its phosphorylation by TR-SFP were considered to be particularly important in the induction of lipolysis in adipocytes, as well as in reducing blood glucose levels in this animal model.

Effects of Food Quality and Temperature on life History Traits of Moina macrocopa Reared in Laboratory

  • Life table experiments were performed in order to examine the effects of food quality on Moina macrocopa fed with four kinds of algal foods, Botryococcus sp., Scenedesmus subspicatus, Selenastrum capricornutum, and Chlorella sp., at $20^{\circ}C$ . The temperature effects on M. macrocopa were also assessed, feeding Chlorella at $17^{\circ}C$,$20^{\circ}C$,$25^{\circ}C$, and a combination of $28^{\circ}C$ (light) and $25^{\circ}C$ (dark). The cartilaginous Botryococcs cells were Inappropriate food for Moina. Among the foods tested, Chlorella was the food of the best quality in all accounts of life history traits. Moina grown on Chlorella showed higher net reproductive rate ($R_0$), longer mean and maximum longevities, earlier mean age at maturity, longer mean carapace length at maturity, larger mean clutch size, and shorter mean time interval between clutch productions than those grown on Selenastrum and Scenedesmus. An optimal temperature for Moina was $20^{\circ}C$ . When Moina were grown on Chlorella at $20^{\circ}C$, they showed the highest r, the highest $R_0$, the shortest T, the longest mean longevity, the earliest mean age at maturity, the longest mean carapace length at maturity, and the largest mean clutch size. The results of life table experiments showed that the individual and population growth patterns were much more affected by low temperature $17^{\circ}C$, than by high temperature ($\geq 20^{\circ}C$). In the optimal condition, the r value was very high, 5.1 in $d^{-1}$. In conclusion, the food quality and the temperature are the most important factors to govern the size and continuity of Moina population, by which the individual growth rates and reproductivity of members in the population can be controlled to survive in their environment such as small and temporary water bodies in nature.

Protective Effects of Extracts of Protaetia brevitarsis on Carbon tetrachloride-induced Hepatotoxicity in the Mice (사염화탄소에 의해 유발된 흰쥐의 간독성에 미치는 흰점박이꽃무지 추출물의 간보호 효과)

  • To investigate the effects of Protaetia. brevitarsis extracts on the protection against liver damage by carbon tetrachloride($CCl_4$) in rat, two kinds of experiment were performed, firstly by the primary hepatocyte culture and secondly by the animal feeding. The primary hepatocyte culture with the extracts of P.brevitarsis showed significantly low activities of GPT, bile acid, and bilirubin, indicating an excellent protective effect against liver damage by $CCl_4$. Especially, below molecular weight 1,000 blew the water to have 32.1% recovery degree. In the seconde experiment, serum GPT activity was significantly decreased in water fraction of P. brevitarsis compared to $CCl_4$ treatment by 98.2%. Serum concentration of bile acid and bilirubin were tended to increased by $CCl_4$ treatment, but water fraction of P. brevitarsis and silymarin recovered the level. These consistent results in vitro and in vivo suggest that the extracts of P. brevitarsis may have strong protective effects against liver damage induced by the potential toxicants such as $CCl_4$.

Effect of Ensilage of Rye Treated with Formic Acid and Lactic Acid Bacteria Inoculant on Ruminal Fermentation Characteristics (개미산과 유산균제 첨가 베일 사일리지의 발효 차이가 반추위 발효 특성에 미치는 영향)

  • This study investigated the effects of silage additives on rumen fermentation characteristics of rye silage. Rye was harvested at ripening stage and treated with different additives in quadruplicate following: without additive (control), with either lactic acid bacteria inoculant (LAB), formic acid (FA), or Ca-formate (Ca-FA). Overall, ensiling characteristics of FA and Ca-FA silages contained 4-fold more (P<0.05) butyrate and 2-fold more (P<0.05) NH3-N concentration (% total nitrogen) than those of control and LAB silages. Cows fed LAB silage showed a diurnal trend with the highest values of propionate concentration compared to the control at 1, 2 and 3 hr after feeding. In contrast, FA and Ca-FA silages increased the proportion of butyrate significantly (P<0.05) at all sampling times compared to control and LAB silage. In conclusion, Forage rye treated with FA or Ca-FA showed different fermentation characteristics during ensilage and in the rumen compared to LAB silage. Further studies are needed to evaluate whether different fermentation characteristics in the rumen between LAB and FA silages had effect on partitioning of nutrients between milk production and body tissue.

Research on the Bacillus-fermented Enzamin Administration on Human NK Cell Activity and Bone Density of Hamster: A Pilot Study (Bacillus 발효액 Enzamin의 투여가 인체의 NK세포 활성도와 Hamster의 골밀도에 미치는 탐색연구)

  • Purpose: The purpose of this study was to investigate the effects of Bacillus fermentation broth (ENM) on the human NK cell activity, and bone matrix density, matrix content and area of hamsters fed ENM. Methods: NK cell activity was tested, and bone mineral density were measured by x-ray. Results: NK cell activity was significantly higher in the control group (644.71 pg/ml) and in the test group (1796.37 pg/ml) (p<.004). Test groups were significantly increased by 1110.37 pg/ml in the pre-test and 1796.37 pg/ml in the post-test(p<.001). Behavioral observations after feeding ENM to hamsters showed normal behavior with no difference between control and test groups. The hamster body weight of the control was 106 g at the initial and final 27 days. In the test group, the initial weight was 96.6 g and the final 27 days was 114 g. No morphological changes were observed in the X-ray photographs of the hamster hind legs. The bone matrix density was 0.059 g/cm3 in control and 0.062 g/cm3 in the test, which was increased by 0.003 g/cm3. The bone matrix content was 0.175 g in the control and 0.196 g in the test. The bone area was 2.95 cm2 in the control and 3.19 cm2 in the test, which was increased by 0.19 cm2. In the autopsy, neither the control nor the test group showed any remarkable abnormality, and each organ was normal. Conclusions: It is thought that ingestion of ENM is useful for immunity enhancement.
