  • The relationship of Oct-4 to pluripotent cells is suggested by its tightly restricted expression pattern during embryonic development. Just prior to implantation it is limited to pluripotent cells of the inner cell mass (ICM) that will form the embryo proper but is not expressed in the trophectoderm, the structure that will form the extraembryonic tissues. • To elucidate overall changes in DNA methylation that occurs by inappropriate epigenetic control during ageing, we compared fetal bovine fibroblasts and their aged neomycin-resistant versions using bisulfite-PCR technology. Reduction in DNA methylation was observed in euchromatic repeats (18S-rRNA/art2) and promoter regions of sing1e-copy genes (the cytokeratin/-lactoglobulin/interleukin-13 genes). Contrastingly, a stable maintenance of DNA methylation was revealed in various heterochromatic sequences (satellite I/IIalphoid and Bov-B). The differential inheritance modes of DNA methylation was confirmed through the analysis of individual neomycin-resistant clones. These global, multi-loci analyses provide evidence on the tendency of differential epigenetic modification between genomic DNA regions during ageing.

  • We recently demonstrated that satellite regions exhibit an aberrant DNA methylation in cloned bovine embryos. Here, we examined, using bisulfite -sequencing technology, whether the inefficient demethylation of cloned donor genomes could be rescued by the presence of oocytic nuclei. Both AciI digestion and sequencing analyses showed that satellite sequence was demethylated more efficiently in cloned tetraploid blastocysts than in diploid clones. When methyl -CpG density (the number of methyl-CpG sites per string) was scored, a significant decrease was observed In tetraploids (P<0.001). These results suggest that unknown mechanisms provided by oocytic nuclei could assist the demethylation of satellite sequences in tetraploid clones.

  • This study was conducted to investigate the effect of dietary domestic Skullcap (Scutellaria baicalensis) extracts (SCE) on laying performance and egg quality in laying hens. There were no significant differences in feed intake, egg production, or relative liver and spleen weights. The egg weights in groups fed diets containing SCE were significantly increased as compared with the control. The number of cecal total microbes in the group fed a diet containing 0.5% SCE was significantly reduced as compared with other groups. The malondialdehyde contents in stored eggs were significantly lowered by feeding SCE. The Haugh unit in the groups fed diets containing SCE tended to be increased after 2 weeks storage, but not significantly. This result indicated that dietary domestic SCE may delay lipid oxidation in eggs when added to laying hen diets.

  • The Asian-Australasian Association of Animal Production Societies (AAAP), the only international scientific organization in animal science representing Asia and Pacific region, showed a remarkable development since its inception in 1980. The number of member countries increased from 8 to 19 while 19 congresses were held in 11 different member countries. The AAAP also helped creating the official journal, Asian-Australasian Journal of Animal Sciences in 1988 with the title being changed to Animal Bioscience in 2021. It is an open access journal indexed by most global databases and has become one of the most respected global journals in animal science. Through scientific meetings and journal publication, the AAAP has made a tremendous contribution to the advancement of animal science and industry throughout its member countries over the last 40 years. This paper summarizes the advances in animal science in the Asia-Pacific region, together with the roles of the AAAP scientists and journals in developing animal science.

  • Sphingosine-1-phosphate(S1P) is one of the sphingolipid metabolites which affect a variety of cellular processes including the proliferation, differentiation, growth, survival, migration and gene expression. The present study was undertaken to investigate the effect of SIP on nuclear maturation of porcine oocytes. In vitro maturation frequency of porcine oocytes were compared in three different media; group Ⅰ: NCSU23+0.1% PVA, group Ⅱ: NCSU23+10% PFF(porcine follicular fluid), and group Ⅲ: NCSU23+10% PFF+10 ng/㎖ EGF+2.5 mM β-mercaptoethanol. Each group containing 0.1 ㎎/㎖ cysteine was divided into 4 sub-groups of SIP concentration(0, 50, 500 and 5000nM). Porcine oocytes were incubated in each maturation medium supplemented with hormones(10 IU/㎖ PMSG and 10 IU/㎖ hCG) for 22h and then further cultured in the same medium without the hormones for 22h. After completion of in vitro maturation, the oocytes were fixed and stained to examine nuclear maturation by using a rapid stain method. In the group Ⅰ, the proportions of metaphase Ⅱ stage among oocytes cultured in 0nM(control), 50 nM, 500nM and 5000nM S1P were 45.5%, 66.7%, 56.6% and 48.7%, respectively. (omitted)

  • This study was conducted to determine the effects of dietary level of whole flaxseed (WFS; 0, 10 and 15%) on performance, carcass characteristics and fatty acid composition of serum and subcutaneous, perirenal, and intramuscular adipose tissues of Korean Hanwoo cattle. The daily gains were not different among treatments. Dietary inclusion of WFS decreased (p<0.05) feed intake but improved (p<0.05) feed conversion ratio (feed/gain). Backfat thickness and marbling score were increased (p<0.05) by dietary WFS. Carcass weight, dressing percentage, loin-eye area, and carcass yield and quality were not different among treatments. The proportion of C18:3 in serum and, to a lesser extent, in adipose tissues were increased (p<0.01) by dietary WFS, indicating that lipids from WFS escaped ruminal biohydrogenation. Animals fed WFS had lower proportions of saturated fatty acid (SFA; C14:0 and 16:0) and higher proportions of polyunsaturated fatty acids (PUFA; C18:2. 18:3, 20:2, 20:4, 20:5 and 22:6) in perirenal and intramuscular fat than animals fed diets without WFS, resulting in an increased PUFA/SFA ratio. Furthermore, feeding WFS increased (p<0.01) proportions of $\omega$-3 and $\omega$-6 fatty acids in intramuscular fat but decreased (p<0.05) the $\omega$-6/$\omega$-3 ratio. Relative treatment effects were similar between 10 and 15% WFS. Feeding WFS can effectively alter composition of adipose tissues with enhanced feed conversion ratio.

  • Sperm capacitation refers to polymerization of filamentous (F)-actin from globular (G)-actin. While the role of actin-related protein 2/3 (Arp2/3) complex in actin polymerization is well appreciated, the underlying mechanism(s) and its relationship with capacitation are poorly understood. Therefore, to evaluate the potential role of Arp2/3 complex on capacitation, bovine spermatozoa were incubated with multiple doses (1, 10 and $100{\mu}M$) of CK-636, an inhibitor of Arp2/3 complex with heparin. The cellular localization of the Arp2/3 complex in spermatozoa was identified by immunohistochemistry, whereas western blot was also applied to detect the protein tyrosine phosphorylation of sperm proteins. Additionally, sperm motility and kinematic parameters were evaluated using a computer-assisted sperm analysis system. CK-636 resulted in significant changes in the ratio of Arp2/3 complex localization between acrosome and equatorial region of the spermatozoa. Short-term exposure of spermatozoa to $100{\mu}M$ of CK-636 significantly decreased sperm motility, however a non-detectable effect on protein tyrosine phosphorylation was observed during capacitation. On the basis of these results, we propose that Arp2/3 complex is associated with morphological changes during capacitation and compromised sperm motility.
