Effects of Samul-tang and Constituent Herbs on a Contracted Artery of Rabbit (사물탕(四物湯) 및 구성(構成) 약물(藥物)이 가토(家兎)의 수축혈관(收縮血管)에 미치는 영향(影響))

  • Objectives : To analyze the effects of Samul-Tang and combinations of constituent herbs on arterial contraction. Method : In order to investigate the effects, Angelicae gigantis Radix, Cnidii Rhizoma, Rehmanniae Radix, Paeoniae Radix, in which one of them, three of them, and all of them, were used to exam. Results : Samul-Tang significantly inhibited the contraction of artery induced by PE(phenylephrine), accordingly as the concentration of Samul-Tang increased and inhibited in both with intact and removed endothelium. Among the constituent herbs of Samul-Tang, Paeoniae Radix and Paeoniae Radix-Cnidii Rhizoma combination inhibited the PE-induced contraction of artery the most. Conclusions : Samul-Tang and constituent herbs of Samul-Tang inhibit the contraction of artery.

Effects of Angelica Gigantis Radix Extracts on the Collagenase Activity and Procollagen Synthesis in HS68 Human Fibroblasts and Tyrosinase Activity (당귀(當歸)의 사람 피부아세포의 콜라게나제 활성과 프로콜라겐합성과 티로시나제 활성에 미치는 영향)

  • Objectives : This study was designed to investigate the collagen metabolism and tyrosinase activity of Angelicae Gigantis Radix extracts (AGR). Methods : The effect of AGR on type I procollagen production and collagenase activity in human normal fibroblasts HS68 after UVB (312 nm) irradiation was measured by ELISA method. The tyrosinase activity after treatment of AGR was measured. Results : Type I procollagen production was recovered by AGR in UVB damaged HS68 cells. The increased collagenase activity after UVB damage was significantly recovered by AGR and the tyrosinase activity was significantly reduced. However, the L-DOPA oxidation was not changed. Conclusion : AGR showed the anti-wrinkle effects and whitening effects in vitro. These results suggest that AGR may have potential as an anti-aging ingredient in cosmetic herbs.

  • Twenty-four Oriental herbal medicines including Platycodi Radix and Scutellariae Radix, etc., which have been used for the cure of hyperlipidemia, coronary heart, disease were evaluated for the 3-hydroxy-3-methylglutaryl coenzyme A(HMG-CoA) reductase inhibition and antioxidant effect on a free radical, 1,1-diphenyl-2-picrylhydrazyl(DPPH) in vitro, and antihyperlipidemic effect on hyperlipidemic rats induced by Triton WR-1339 in vivo. 80% MeOH extract of eight herbs including Scutellariae Radix, Coptidis Rhizoma, Paeoniae Radix Rubra, Moutan Cortex Radicis, Angelicae Gigantis Radix, Bambusae Caulis in Taeniam, Cinnamomi Ramulus and Crataegi Fructus inhibited the HMG-CoA reductase activities and exhibited a radical-trapping action on a stable free radical, DPPH. On Triton WR-1339-induced hyperlipidemic rats, four herbs including Scutellariae Radix, Angelicae Gigantis Radix, Bambusae Caulis in Taeniam and Cinnamomi Ramulus showed respectively the significant suppression of serum total cholesterol, triglyceride, phospholipid and LDL-cholesterol levels and serum transaminase(ALT and AST) activities. From these results, it is suggested that each 80% MeOH extract of Scutellariae Radix, Angelicae Gigantis Radix, Bambusae Caulis in Taeniam and Cinnamomi Ramulus have effective antihyperlipidemic action against hyperlipidemia.

Comparative Studies on Concentration of Decursinol in Plasma after Oral Administration of Angelicae Gigantis Radix Extract and Combined Use of Decursin and Cnidii Rhizoma Extract or Bupleuri Radix Extract in Rats (당귀와 천궁 및 시호의 병용 경구 투여에서 흰쥐 혈중 Decursinol 농도의 비교 검토)

  • This study was performed to elucidate the influence on the plasma decursinol concentration after administration of combined decursin and Cnidii Rhizoma or decurisin and Bupleuri Radix extracts in comparison with single decursin administration. The identification of decursinol isolated from Angelica gigas was carried out by GC/MS and the concentration of decursinol in the plasma after oral administration was determined by HPLC. The value of area under the plasma concentration (AUC) and the maximal concentration (Cmax) of decursinol after administration of methanol and ether Angelicae Gigantis Radix extract was higher than those of single decursin-treated group. It was also found that the AUC and the Cmax of decursinol after combined administration with decursin and Cnidii Rhizoma were higher than those of decursin administration, whereas those of combined with decursin and Bupleuri Radix was lower. Moreover, the studies were performed with decursinol only or with combined decursinol and Cnidii Rhizoma/Bupleuri Radix, both of combined treatment increased the plasma decursinol concentrations compared with decursinol-treated group in rats. On the basis of these results, it is concluded that co-existing substances or co-administration with other drugs may influence the plasma levels of decurisinol after oral administration.

Effects of Angelicae Gigantis and Cynanchum wilfordii Hemsley Extract on the Changes of Cerebral Flow (당귀와 백수오 추출물이 뇌혈류 변화에 미치는 영향)

  • This study was designed to investigate the effects of Angelicae Gigantis and Cynanchum wilfordii Hemsley extract (AAC) on the changes of regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP) in normal rats, and in the rats with cerebral ischemia induced by middle cerebral artery occlusion, and further to determine the mechanisms. The results were as follows. AAC significantly increased rCBF but significantly decreased MABP in a dose-dependent manner in normal rats. The increase of AAC-induced rCBF was significantly inhibited by pretreatment with methylene blue (0.01 mg/kg, i.p.), an inhibitor of guanylate cyclase, and AAC-induced MABP was decreased by pretreatment with methylene blue. In cerebral ischemics, rCBF was stably improved by AAC (10 mg/kg, i.p.) during the period of cerebral reperfusion, which was contrasted with the findings of rapid and marked increase in the control group. These results suggest that AAC can increase rCBF in the normal state, as well as improve the stability of rCBF in cerebral ischemic state.

Analysis of Prescriptions from Somunsunmyungronbang and Nansilbijang (소문선명논방(素問宣明論方)과 난실비장(蘭室秘藏)의 본초구성 비교 분석)

  • Objectives : To quantitatively analyze the academic characteristics of Yuhagan and Idongwon and compare it to existing qualitative analysis. Methods : The prescriptions listed in Somunsunmyungronbang and Nansilbijang were entered into a database. Commonly used combination of medical herbs were researched throughout the literature. Results : In Nansilbijang, among the combinations of 8 medical herbs, Glycyrrhizae Radix, Cimicifugae Rhizoma, Bupleuri Radix, Ginseng Radix, Citri Pericarpium, Atractyodis Rhizoma, Astragali Radix, Phellodendri Cortex combination was used 11 times out of the 277 prescriptions. In Somunsunmyungronbang, combinations of 7 medical herbs: (Glycyrrhizae Radix, Platycodi Radix, Rhei Radix et Rhizoma, Menthae Herba, Atractylodis Rhizoma Alba, Gypsum Fibrosum, Forsythiae Fructus), (Glycyrrhizae Radix, Angelicae gigantis Radix, Paeniae Radix Alba, Atractylodis Rhizoma Alba, Gypsum Fibrosum, Cnidii Rhizoma, Scutellariae Radix), (Glycyrrhizae Radix, Platycodi Radix, Angelicae gigantis Radix, Rhei Radix et Rhizoma, Atractylodis Rhizoma Alba, Gypsum Fibrosum, Schizonepete Herba) were each used 7 times out of the 350 prescriptions. Conclusions : Nansilbijang shows more prescriptions applying Cimicifugae Rhizoma, Bupleuri Radix, Ginseng Radix, Citri Pericarpium, Atractyodis Rhizoma, Astragali Radix, Phellodendri Cortex. Prescriptions aimed to tonify ql with Ginseng Radix and Astragali Radix, to upraise the middle qi with Bupleuri Radix and Cimicifugae Rhizoma, and to regulate qi with Citri Pericarpium and Atractyodis Rhizoma were common. Somunsunmyungronbang preferred the use of Angelicae gigantis Radix, Platycodi Radix, Rhel Radix et Rhizoma, Gypsum Fibrosum, Schizonepete Herba, Forsythiae Fructus and Menthae Herba so its focus was on the aggressive management of febrile diseases.

Effects of Radix Angelicae Gigantis and Resina Ferulae on the Relaxation of Smooth Muscle and Expression of iNOS (당귀 및 아위가 평활근 이완과 iNOS 발현에 미치는 영향)

  • Objectives : Radix Angelicae Gigantis(RAG) and Resina Ferulae(RF) have been used in oriental medicine or folk medicine to increase stamina. The aim of this study was the characterization of the mechanism of action of RAG and RF on smooth muscle and macrophages in rats to find new substances for the treatment of erectile dysfunction, cardiovascular diseases and immune dysfunction. Methods : We investigated the effects of the water extracts of RAG and RF on phenylephrine or KCl-contracted rat endothelium-denuded aorta, the production of NO in vascular smooth muscle cell (VSMC) and the production of NO and induction of iNOS in the $IFN-{\gamma}-primed$ RAW 264.7 cells. Results : The water extracts of the RAG and RF showed significant concentration-dependent relaxation effects on phenylephrine or KCl-contracted rat endothelium-denuded aorta. It also reduced the tension of the rat endothelium denuded aorta which was contracted in $Ca^{2+}-free$ media. On the other hand, it increased production of NO in VSMC which was stimulated with $IL-{\beta}$ or $IL-{\beta}$ plus $IFN-{\gamma}$. The water extracts of RAG and RF increased production of NO and induction of iNOS in the $IFN-{\gamma}-primed$ RAW 264.7 cells. Conclusions : According to the above results, the water extracts of RAG and RF relaxed the smooth muscle effectively and increased the production of NO in VSMC and macrophages. So, these herbs can be applied to erectile dysfunction, hypertension, angina pectoris, artherosclerosis and a defense defect for virus or microbe.

Blockade of vascular angiogenesis by Aspergillus usamii var. shirousamii-transformed Angelicae Gigantis Radix and Zizyphus jujuba

  • The matrix metalloproteinases (MMP) play an important role in tumor invasion, angiogenesis and inflammatory tissue destruction. Increased expression of MMP was observed in benign tissue hyperplasia and in atherosclerotic lesions. Invasive cancer cells utilize MMP to degrade the extracellular matrix and vascular basement membrane during metastasis, where MMP-2 has been implicated in the development and dissemination of malignancies. The present study attempted to examine the antiangiogenic activity of the medicinal herbs of Aspergillus usamii var. shirousamii-transformed Angelicae Gigantis Radix and Zizyphus jujube (tAgR and tZj) with respect to MMP-2 production and endothelial motility in phorbol 12-myristate 13-acetate (PMA)- or VEGF-exposed human umbilical vein endothelial cells (HUVEC). Nontoxic tAgR and tZj substantially suppressed PMA-induced MMP-2 secretion. In addition, $25{\mu}g/mL$ tAgR and tZj prevented vascular endothelial growth factor-stimulated endothelial cell transmigration and tube formation. The results reveal that tAgR and tZj dampened endothelial MMP-2 production leading to endothelial transmigration and tube formation. tAgR and tZj-mediated inhibition of endothelial MMP may boost a therapeutic efficacy during vascular angiogenesis.

Development of Ultrafine Angelica Powder-Added Syrup (초미세 당귀분말 첨가 시럽의 제조 기술 개발)

  • Angelicae gigantis Radix (dried root of Angelica gigas) including major bioactives such as decursin and decursinol angelate provides rich flavors and several healthy benefits. Recent studies have shown that ultrafine powders of herbal medicines provide better physical properties and biological activities. Thus, ultrafine Angelica powder was added into the oligosaccharide syrup to provide flavors and healthy benefits in this study. Angelicae gigantis Radix was pulverized into d(0.1) = 3.220, d(0.5) = 7.822, and d(0.9) = 7.817 $\mu$m respectively using an air-flow mill. The ultrafine Angelica powder was added into the oligosaccharide syrup process with different ratios of water to oligosaccharide syrup at 1:5, 1:8, 1:11, and 1:14. The physicochemical properties such as viscosity and bulk density were measured. The Stokes' law was applied to predict the sedimentation velocity of the added Angelica powder in the syrup. The Angelica syrup prepared in this experiment showed good stability since the Angelica particles precipitated down slowly. The ratio of water to oligosaccharide syrup at 1:11 showed the optimal preparation in terms of the stability and the viscosity. The ultrafine-sized herbal powders such as Angelicae gigantis Radix have potentials for various food and pharmaceutical applications.
