This study was performed to investigate the characteristics of nutrient removal of municipal wastewater in the membrane bioreactor system with the different types of membrane. Membrane bioreactor consists of three reactors such as two intermittent anaerobic and the submerged membrane aerobic reactor with flat sheet and hollow fiber membrane, respectively. The removal efficiencies of $COD_{cr}$, BOD, SS, TN and TP on the flat sheet membrane bioreactor were 94.3%, 99.0%, 99.9%, 70.3% and 63.1%, respectively. In addition, The removal efficiencies of $COD_{cr}$, BOD, SS, TN and TP on the hollow fiber membrane bioreactor were 94.0%, 99.3%, 99.9%, 69.9% and 66.9%, respectively. The estimated true biomass yield, specific denitrification rate (SDNR), specific nitrification rate (SNR) and phosphorus removal content on the flat sheet membrane bioreactor were $0.33kgVSS/kgBOD{\cdot}d$, $0.043mgNO_3-N/mgVSS{\cdot}d$, $0.031mgNH_4-N/mgVSS{\cdot}d$, and 0.144 kgP/d, respectively. In addition, the estimated true biomass yield, specific denitrification rate (SDNR), specific nitrification rate (SNR) and phosphorus removal content on the hollow fiber membrane bioreactor were $0.30kgVSS/kgBOD{\cdot}d$, $0.067mgNO_3-N/mgVSS{\cdot}d$, $0.028mgNH_4-N/mgVSS{\cdot}d$, and 0.121 kgP/d, respectively. There was little difference between the flat sheet and hollow fiber on the nutrient removal efficiencies except SNR and SDNR. These differences between them were caused by the air demand to prevent the membrane fouling. The flux and oxygen demand for air scouring were $19.0L/m^2/hr$ and $2.28m^3/min$ for the flat sheet membrane, and $20.7L/m^2/hr$ and $1.77m^3/min$ for the hollow fiber membrane on an average.
Anaerobic ammonium oxidation (AMX) is a cost-efficient biological nitrogen removal process. The coexistence of ammonia-oxidizing bacteria (AOB) in an AMX reactor is an interesting research topic as a nitrogen-related bacterial consortium. In this study, a sequencing batch reactor for AMX (AMX-SBR) was operated with a conventional activated sludge. The AOB in an AMX bioreactor were identified and quantified using terminal restriction fragment length polymorphism (T-RFLP) and real-time qPCR. A T-RFLP assay based on the ammonia monooxygenase subunit A (amoA) gene sequences showed the presence of Nitrosomonas europaea-like AOB in the AMX-SBR. A phylogenetic tree based on the sequenced amoA gene showed that AOB were affiliated with the Nitrosomonas europaea/mobilis cluster. Throughout the enrichment period, the AOB population was stable with predominant Nitrosomonas europaea-like AOB. Two OTUs of amoA_SBR_JJY_20 (FJ577843) and amoA_SBR_JJY_9 (FJ577849) are similar to the clones from AMX-related environments. Real-time qPCR was used to quantify AOB populations over time. Interestingly, the exponential growth of AOB populations was observed during the substrate inhibition of the AMX bacteria. The specific growth rate of AOB under anaerobic conditions was only 0.111 d-1. The growth property of Nitrosomonas europaea-like AOB may provide fundamental information about the metabolic relationship between the AMX bacteria and AOB.
Purple non-sulfur bacteria, Rhodobacter sphaeroides KD131 grew to reach the maximum cell concentration in 45 hrs of incubation in the synthetic media containing (NH4)2SO4, L-aspartic acid and succinic acid as the carbon and nitrogen sources, respectively, at 30oC under 8 klux irradiance using halogen lamp. The strain produced hydrogen from the middle of the logarithmic growth phase and continued until the cell growth leveled out. The strain grew and produced hydrogen under the irradiance of 3-30 klux, but cell growth was inhibited over 100 klux. In addition, anaerobic/light culture condition was better than the aerobic/dark on the hydrogen production. Among various photo-bioreactors examined, the flat-vertical reactor manufactured using clear acrylic plastic material showed the best hydrogen production rate at the given culture condition.
Two mesophilic trickling bed bioreactors filled with two different types of media, hydrophilic- and hydrophobic-cubes, were designed and conducted for hydrogen production under the anaerobic fermentation of sucrose. Each bioreactor consisted of the column packed with polymeric cubes and inoculated with heat-treated sludge obtained from anaerobic digestion tank. A defined medium containing sucrose was fed by the different hydraulic retention time(HRT), and recycle rate. Hydrogen concentrations in gas-phase were constant, averaging 40% of biogas throughout the operation. Hydrogen production rate was increased till $10.5\;L{\cdot}h^{-1}{\cdot}L^{-1}$ of bioreactor when influent sucrose concentrations and recycle rates were varied. At the same time, the hydrogen production rate with hydrophobic media application was higher than its hydrophilic media application. No methane was detected when the reactor was under a normal operation. The major fermentation by-products in the liquid effluent of the both trickling biofilters were acetate, butyrate and lactate. In order to run in the long term operation of both reactor filled with hydrophilic and hydrophobic media, biofilm accumulation on hydrophilic media and biogas produced should be controlled through some process such as periodical backwashing or gas-purging. Four sample were collected from each reactor on the opposite hydrogen production rate, and their bacterial communities were compared by terminal restriction fragment length polymorphism (T-RFLP) analysis of PCR products generated using bacterial 16s rRNA gene primers (8f and 926r). It was expressed a marked difference in bacterial communities of both reactors. The trickling bed bioreactor with hydrophobic media demonstrates the feasibility of the process to produce hydrogen gas. A likely application of this reactor technology can be hydrogen gas recovery from pre-treatment of high carbohydrate-containing wastewaters.
수소생산 유효세균을 유지하기 위해 열처리한 혐기소화슬러지를 접종원으로 하여 PVA 담체를 활용한 고정화와 비고정화 방법에 의한 수소생산능 및 세균 군집을 비교분석 하였다. 고정화에 의한 수소생산성은 비고정화 방법과 비교하여 수소생산성에 영향을 미치지 않았다. 그러나 고정화 방법은 유기물 분해결과 발생하는 유기산 축적 및 pH 강하 등의 환경변화에 있어 세균의 성장과 활성에 안정적 환경을 제공할 수 있는 가능성을 나타내었다. DGGE 분석에 의한 세균군집분석에서는 비고정화와 고정화 반응조간에 형성된 세균군집이 차이가 있는 것으로 나타났으며, 상대적으로 고정화 반응조내의 유효 수소생산 세균 군집이 좀 더 안정적으로 유지되는 것으로 확인되었다. Droplet digital PCR에 의한 6종의 유효한 수소생산 세균 절대정량 분석결과, encapsulation에 관계없이 두 반응기에서의 우점세균수는 유사하였다. Firmicutes, Clostridium, Enterobacter, Ruminococcus 및 Escherichia가 $1{\times}10^5-1{\times}10^6$ copy number of ml-sample 수준에서 존재하였다.
An advanced anaerobic expanded granular sludge bed (AnaEG) and an internal circulation (IC) reactor, which were adopted to treat starch processing wastewater (SPW) and ethanol processing wastewater (EPW), were comprehensively analyzed to determine the key factors that affected the granules and microbial communities in the bioreactors. The granule size of $900{\mu}m$ in the AnaEG reactor was smaller than that in the IC reactor, and the internal and external morphological structures of the granular sludge were also significantly different between the two types of reactors. The biodiversity, which was higher in the AnaEG reactor, was mainly affected by reactor type. However, the specific microbial community structure was determined by the type of wastewater. Furthermore, the dominant methanogens of EPW were mainly Methanosaeta and Methanobacterium, but only Methanosaeta was a major constituent in SPW. Compared with the IC reactor, characteristics common to the AnaEG reactor were smaller granules, higher biodiversity and larger proportion of unknown species. The comparison of characteristics between these two reactors not only aids in understanding the novel AnaEG reactor type, but also elucidates the effects of reactor type and wastewater type on the microbial community and sludge structure. This information would be helpful in the application of the novel AnaEG reactor.
The objective of this study is to estimate the effect of the bed expansion and the characteristics of attached biomass in the start-up in the anaerobic fluidized bed reactor(AFBR). The fluidized bed reactor was operated with bacteria supported on the bed of granular activated carbon(GAC). The reactor was operated at 35$\circ$C, 5 kg $COD/m^3\cdot day$ at bed expansion varying from 0 to 100% with soluble glucose wastewater(5,000 mg/l). When the effluent reached a steady state at 100% of bed expansion, maximum COD removal efficiency of 87.3% and 0.031 $m^3CH_4/kg COD_{removed}$ were obtained. At higher bed expansion, COD removal efficiency, methane production rate and biogas production rate increased. Especially, at 50% of bed expansion, the efficiency of the treatment increasedg rapidly in the AFBR. The biomass colonized in the pits and crevices of the GAC particle and no complete biofilm was established in the bioreactor during the experiment.
A membrane bioreactor by sequentially alternating the inflow and by applying a two-stage coagulation control based on pre-coagulation was evaluated in terms of phosphorus removal efficiency and cost-savings. The MBR consisted of two identical alternative reaction tanks, followed by aerobic, anoxic and membrane tanks, where the wastewater and the internal return sludge alternatively flowed into each alternative reaction tank at every 2 hours. In the batch-operated alternative reaction tank, the initial concentration of nitrate rapidly decreased from 2.3 to 0.4 mg/L for only 20 minutes after stopping the inflow, followed by substantial release of phosphorus up to 4 mg/L under anaerobic condition. Jar test showed that the minimum alum doses to reduce the initial $PO_4$-P below 0.2 mg/L were 2 and 9 mol-Al/mol-P in the wastewater and the activated sludge from the membrane tank, respectively. It implies that a pre-coagulation in influent is more cost-efficient for phosphorus removal than the coagulation in the bioreactor. On the result of NUR test, there were little difference in terms of denitrification rate and contents of readily biodegradable COD between raw wastewater and pre-coagulated wastewater. When adding alum into the aerobic tank, alum doses above 26 mg/L as $Al_2O_3$ caused inhibitory effects on ammonia oxidation. Using the two-stage coagulation control based on pre-coagulation, the P concentration in the MBR effluent was kept below 0.2 mg/L with the alum of 2.7 mg/L as $Al_2O_3$, which was much lower than 5.1~7.4 mg/L as $Al_2O_3$ required for typical wastewater treatment plants. During the long-term operation of MBR, there was no change of the TMP increase rate before and after alum addition.
본 연구는 혐기조, 안정화조, 무산소조, 막분리호기조로 구성된 membrane bioreactor의 운영을 통하여 도시 하수의 영양염류 제거 특성을 규명하였다. 수리학적 체류시간(HRT)과 슬러지 체류시간(SRT) 및 운영 여과 플럭스의 평균값은 각각 6.2시간, 34.1일과 19.6 L/$m^2$/hr (LMH)이었으며, 공정을 운전한 결과, $COD_{Cr}$, SS, TN 및 TP의 평균 제거율은 각각 94.3%, 99.9%, 69.4% 및 74.6%이었다. 슬러지 생산계수, SDNR, SNR, SPPR 및 SPUR은 각각 0.653 kgVSS/kgBOD/d, 0.044 $mgNO_3$-N/mgVSS/d, 0.035 $mgNH_4$-N/mgVSS/d, 51.0 mgP/gVSS/d 및 5.4 mgP/gVSS/d였다. 또한, 생산된 슬러지의 평균 질소 및 인 함량은 각각 8.86%와 3.5%였다.
혐기성 암모늄 산화공정을 안정화시키기 전에 많은 양의 식종 미생물 투여가 필요하므로 혐기성 암모늄 산화균의 농후배양은 실규모의 혐기성 암모늄 산화 반응기를 운영할 때 필수적인 과정이다. 본 연구에서는 활성슬러지 미생물을 식종한 연속 회분식 반응기를 이용하여 혐기성 암모늄 산화균을 농후배양하고, 미생물 군집구조의 변화를 관찰하여 농후배양 결과를 검증하였다. 혐기성 암모늄 산화균의 농후배양은 70일간 시행되었고, 농후배양 후 활성시험에서 $NH_4\;^+$와 $NO_2\;^-$의 기질제거효율이 각각 98.5%와 90.7%로 관찰되어 혐기성 암모늄 산화균의 배양이 성공적으로 수행된 것으로 판단되었다. 계통분류학적 분지도 작성 결과, 다양하였던 Planctomycetes 문(phylum)의 미생물 군집구조가 농후배양 이후에 현저하게 단순해졌다는 것이 밝혀졌다. 농후배양 이후 발견된 36개의 clone들 모두가 혐기성 암모늄 산화균이었으며, Candidatus Brocadia (36%) 와 Candidatus Anammoxoglobus (64%) 속(genus)에 속하였다. RTQ-PCR (real-time quantitative PCR)을 통해 혐기성 암모늄 산화균을 정량한 결과, 혐기성 암모늄 산화 상향류식 연속 배양기에서 1년 이상 선택 배양된 붉은색 혐기성암모늄 산화 입상 슬러지에 비해 혐기성 암모늄 산화균의 16S rDNA 농도가 74.8%인 것으로 나타났다. 상기의 분자생물학적 분석을 통해 70일간 농후배양된 활성슬러지가 혐기성 암모늄 산화 실용화 공정의 접종 미생물로 활용 가능할 것으로 판단되었다.
