The nitrogen assimilation rate of nitrogenous nutrients by reservior phytoplankton was masured in the in situ condition in the euphotic layer of Lakes Soyang, Chuncheon and Uiam located on the upper reaches of the North Han River System in August, 1983, Korea. The assimilation rate of ammonia, nitrate and urea nitrogen in surface water was 13, 2 and $13{\mu}g$ at. $N{\cdot}m^{-3}{\cdot}(12:10~18:15)^{-1}$ in Lake Soyang, 325, 27 and $59{\mu}g$ at. $N{\cdot}m^{-3}{\cdot}(12:30~18:30)^{-1}$ in Lake Chuncheon, and 174, 12 and $45{\mu}g$ at. $N{\cdot}m^{-3}{\cdot}(12:30~19:30)^{-1}$ in Lake Uiam. Ammonia and urea were perferntially utilized by reservoir phytoplankton. The dark/light ratios of nitrate assimilation were much lower than those of ammonia and urea assimilation of nitrate showed little contribution. The primary productuin was estimated as 59mg $C{\cdot}m^{-2}{\cdot}day^{-1}$ and 6.9mg $N{\cdot}m^{-2}{\cdot}day^{-1}$ in Lake Spyang, 217mg C{\cdot}m^{-2}{\cdot}day^{-1}$ and 26mg N{\cdot}m^{-2}{\cdot}day^{-1}$ in Lake Chuncheon, and 110mg C{\cdot}m^{-2}{\cdot}day^{-1}$ and 13mg N{\cdot}m^{-2}{\cdot}day^{-1}$ in Lake Uiam, with production ratios of 8.6, 8.4 and 8,4, respectively. The turnover time o ammonia and urea in the upper euphotic layer was 2 to 47 days and 4 to 38 days, respectively. Nitrate required much longer periods. In the euphotic layer of reservoirs, ammonia and urea played signigicant roles in the biogeoKDICical nitrogen metabolism.
In order to detect the way of absorption and metaboism of the urea it is sprayed on the surface of the leaves of sunflower. The sunflowers used in this study are grown in different conditions such that the one in nittogen aboundant and the other in nitrogen deficient soil, respectively. The urea-N, ammonia-N, amide-N, and 80% alcohol soluble-N in the leaves were quantitatively determined. All of the nitrogenous components measured are generally tended to increased with rising the concentration of urea except only amide-N at 24 hours after sprayed, and these were highly significances. It seemed that hydrolizing of urea into ammonia and carbon dixide and the assimilation of ammonia into other organic nitrogenous constituents were rapid in the young leaves than in the mature. It is interest that the amide content, in the young leaves and nitrogen defieient one were enhanced with the increasing concentration of urea, although in the mature leaves it did not show any change in the urea treatment. It is presumed that the assimilation rate of ammonia and the urease activity were lower in the matture leaves than in the young and nitrogen deficient leaves. No significance at 5% level showed all of the nitrogenous components except total nitrogen between nitrogen abundant and deficent leaves. Urea content was a high peak at first 12 hours, ammonia at 48 hours, and amide and alcohol soluble nitrogen at 96 hours, whence decrease4d the content of these constituents gradually. The total nitrogen content is not incrased obviously by only one time of urea spray in this study. When the concentration of urea was relatively high there appeared the wilting spots on t도 edge of leaves. As a whole, it seemed that sprayed urea was rapidly absorbed and taken part in nitrogen metabolism within relatively short period.
Optimum temperature and pH of glutamine synthetase activity (E.C. 3.6.1.2.) of R. sphaeroides D-230 was $35^{\circ}C$ and 6.8, respectively. The adenylated state of GS in R. sphaeroides D-230 was stabilized by addition of 0.2mg/ml of cethyltrimethylammoniumbromide. Valine, histidine, proline, isoleucine, and lysine were good nitrogen source for the growth of R. sphaeroides D-230. The growth of R. sphaeroides D-230 in $N_2,\;NaNO_3\;or\;NH_4Cl$ as sole nitrogen source was lower than in any otherculture conditions. GS activity was inhibited, more or less, by various amino acid. THe relative inhibition rate of the enzyme by added 7mM arginine, $NH_4Cl,\;N_2,\;and\;NaNO_3$ was 63.8%, 26.79%, 6.24%, and 10.64%, drespectively. THe hydrogen evolution of R. sphaeroides D-230 grown in N-limited media was inhibited by 0.1mM MSX, irreversible GS inhibitor. GS activity was completely inhibited by 1.0mM MSX but ammonia released maximally at the same concentration of MSX. Ammonia release by added MSX was increased up to 1.0mM MS, but decreased above 1.0mM MSX. It is probably due to inhibition of nitrogenase actixity by MSX. Nitrogenase activity was not inhibited at low concentration of MSX. These results suggests that the inhibition of nitrogenase activity by ammonia is mediated by products of ammonia assimilation rather than by ammonia itself.
The effect of nitrate, nitrite and ammonia as inorganic nitrogen sources on the modulation of nitrogen metabolism of Botryococcus braunii UTEX.-572 has been studied under aeration. The primary process in the regulation of nitrogen metabolism by this alga has the nitrate uptake system. This uptake of nitrate operation was immediately inhibited by the presence of 0.5 mM of ammonium and reversed by 0.2∼0.3 mM ammonium. When cell were exposed to 5 mM of ammonium for 24 hours the activity of nitrate reductase became inactive.

The bacteria responsible for the reduction of ammonia concentration in a food-wastewater treatment facility were isolated and their characteristics were analyzed. The isolated bacteria were closely related to the bacteria belonging to genus Citrobacter, Enterobacter, Buttiauxella, Shigella, and Aeromonas, which were found in gut of animals, indicating the isolated bacteria may come from the butchery-byproduct of pigs which is the main component of wastewater. When we monitored the concentration of nitrite and nitrate in the process, it was relatively constant, indicating the isolated bacteria reduce ammonia concentration through ammonia assimilation. Based on the removal efficiency of ammonia by the isolated bacteria, we concluded that they play a role in the reduction of odorous compounds.
In order to understand the regulation of glutamate dehydrogenase(GDH) synthesis in Brevibacterium flavum, we have isolated a mutant lacking NADP-linked GDH activity by ethlmethane sulfonate treatment. The $gdh^-$ mutant was grown on the minimal plate with 1mM ammonium chloride and not that with 300mM ammonium chloride. The cell-free extracts from $gdh^-$ mutant and prototroph were also examined with glutamine synthetase(GS) and glutamate synthase (GOGAT) production by niteogen sources. The growth of $gdh^-$ mutant in presence of 20mM ammonium chloride means that GOGAT synthesis is sufficient to allow growth in this condition. GS production of $gdh^-$ mutant as well as parental strain was induced by 1mM urea and ammonium tartrate, but it was repressed by higher concentration of ammonia, and also induced by 20mM to 50mM glutamate as a substrate. It was special attention that GOGAT synthesis from $gdh^-$ strain was more repressed by higher concentration of ammonia than prototroph as described in E. coli system.
The effect of ammonia and glutamine on nitrogenase activity of Rhodopseudomonas sphaeroides was examined. The nitrogenase activity of this strain was inhibited by ammonia and glutamine. When ammonia and glutamine were exhausted, nitrogenase activity promptly resumed at its original rate. Methionine sulfoximine (MSX), irreversible glutamine synthetase (GS) inhibitor, is a structural analogue of glutamate. MSX was used in order to know whether the nitrogenase activity was inhibited by ammonia and glutamine directly or not. The ability of MSX to prevent nitrogenase switch-off by ammonia was found to be dependent upon the phase of culture. When the cells were sampled after 12 hour culture, $500{\mu}M$ MSX would not prevent the nitrogenase switch-off by ammonia. Twenty one percents of GS actibity was inhibited by $500{\mu}M$ of MSX and concentration of released ammonia decreased. But nitrogenase activiy was still inhibited by ammonia. However, nitrogenase switch-off after 20 hours would be prevented by $100{\mu}M$ of MSX. On the other hand, GS activity was ingibited completely by $100{\mu}M$ MSX and concentration of released ammonia somewhat increased. But nitrogenase activity was not inhibited. The data indicated that the inhibition of in vivo nitrogenase actibity of Rp. sphaeroides by ammonia seemed to be mediated by products of ammonia assimilation rather than by ammonia itself.
Rumen bacterial amino acids in sheep on urea diet were monitored to assess a possible change in amino acid synthesis as a long term response to high rumen ammonia environment. A sheep was fed a semipurified diet with soybean meal, followed by a diet with urea as a main nitrogen source. Mixed rumen bacteria were harvested from ruminal fluid taken 3 h after feeding (twice in soybean meal feeding and 6 times in urea feeding) and fractionated as cell wall, proteins and protein-free cell supernatant of monitor amino acids in each fraction. Ruminal ammonia concentration at the sampling ranged from 5.7 to 39.5 mgN/dl. Cell wall and protein fractions of mixed rumen bacteria were stable in their amino acid composition regardless of nitrogen sources of diet and the feeding duration. However, protein-free cell supernatant fraction showed a higher alanine proportion with urea feeding (18.6 and 28.2 molar % of alanine for samples from sheep fed soybean meal and urea, respectively) and its duration (20.6 and 32.9 molar % for samples from sheep on urea diet for 1 and 65 days, respectively). Total free amino acid level of bacteria was depressed in the initial period of urea feeding but restored on 65th day of the feeding. These results suggest that an alanine synthesizing system may develop in rumen bacteria as urea feeding becomes longer.
This study presents physiological rates of oxygen consumption, ammonia excretion, feeding rates, O/N ratio and assimilation efficiency of the blood cockle, Scapharca subcrenata, determined from specimens collected in Yeoja bay on the south coast of Korea. Physiological parameters were measured monthly under static, laboratory controlled conditions with ambient conditions, and measurements were performed seasonally in order to estimate scope for growth and its probable sources of variation. Oxygen consumption and ammonia excretion rates have been increased as temperature increased with the highest value of August, 2008. Feeding rate was the highest during April whereas the lowest was during August which is a period of gametogenesis with minimum biomass of phytoplankton around sampling area. Assimilation efficiency was not significantly different seasonally and O/N ratio decreased during July to August. The scope for growth was negative during high temperature months(July to August), reflecting the high temperature and low feeding rate, and had its highest positive values during spring and autumn. Data on the physiological parameters and scope for growth of Scapharca subcrenata obtained in this study will be used to assess the carrying capacity for blood cockle cultivation.
To investigate the effects of the dissolved oxygen concentration (DO) on Scope for growth (SFG) of the manila clam, Ruditapes philippinarum, we measured $LC_{50}$, filtration, respiration, ammonia excretion, and assimilation rates under $23\pm0.5^{\circ}C$ as a function of DO. The $LC_{50}$ of DO for R. philippinarum, was 2.4 mgDO $L^{-1}$. With decreasing DO, filtration and respiration rates of R. philippinarum decreased, while ammonia excretion rate increased, The assimilation rate was $68.2\%$ at 6.5 mgDO $L^{-1}$, decreased to $29.8\~39.3\%$ at 3.5 mgDO $L^{-1}$. R. philippinarum had positive SFG's at the $DO{\geq}2.5mgDO\;L^{-1}$.
