Ultraviolet Spectrophotometric Analysis of 2-Aminonaphtalene Sulfonic Acids (2 - 아미노나프탈렌술폰산류의 자외선분광 광도법에 의한 분석)

  • The sulfuric acid sulfonation mixtures of 2-aminonaphthalene, contained 2-aminonaphthalene, Dahl's acid, Broenner's acid, amino-F-acid and Baden acid, can be determined quantitatively by multicomponent spectrophotometric analysis. The analysis was performed in diluted sodium hydroxide aqueous solution and based on the ultraviolet absorption of the sodium salts of each isomers. The determination of quantity of each isomers was performed by subjection the absorbances of the unknown mixture and of its constituents, gathered at a large number of wavelengths, to a least square treatment by an electronic personer computer. This method provided a rapid analysis of such complex mixture, and the standard deviation was ${\pm}1.65$ mole %.

Enhanced Release of Cholecystokinin by Dietary Components in Chicks (사료성분에 의한 닭의 혈중 Cholecystokinin 농도의 상승)

  • The effect of dietary components on cholecystokinin (CCK) release into plasma was investigated in chicks by feeding a meal through a stomach tube, followed by the CCK determination with specific CCK-8 antibody. In experimental 1, the results showed that both isolated soya protein and an amino acid mixture simulating the amino acid composition of the soya protein increased the release of CCK, though to a lesser extent with a delayed response in the former, when added to a protein-free diet. Among amino acids added singly to the protein-free diet, phenylalanine was more efficient than arginine and valine, exerting a response almost identical to the complete amino acid mixture. In experimental 2 and 3, by feeding the protein diets supplemented SBTI, piasma CCK level was promptly increased and this response was in a dose dependent fashion during the measurement time, being higher at 1000 than at 100 mg/kg diet. Since the SBTI supplementation did not affect crop emptying rates significantly, it was concluded that SBTI by itself enhanced CCK release into circulation.

  • Wistar Strain male rats were divided into four different diet groups and the control group was fed on seven percent casein diet (C), the second with a mixture of 80% of rice and 20% of buckwheat (RW), the third with rice only (R), and the fourth with a mixture of rice and buckwheat of equal nitrogen level (RWS). Each diet group consisted of six rats was fed for three weeks by the adlibitum feeding method. The results of the experiment, i, e, the growth gain and change in the level of branch-amino acids in the plasma determined by the micro-bioassay method were as follows. 1. The group C showed the largest growth gain and the rest did in the order of RW group, R group, and RWS group. 2. It was shown that rats fed on diet of higher protein score tend to have higher level of free branch-amino acids in the plasma. 3. Thus, the protein score may be estimated based on the level of the free branch-amino acids in the plasma.

Amino Acids Supplemented with Culture Medium Stimulated On Development of Porcine Embryos

  • This study was carried out that to investigate the effects of amino acids supplemented with culture medium on development of porcine embryos cultured in vitro. Cumulus oocyte complexes (COCs) were cultured in the maturation medium containing hormones (0.5$\mu\textrm{g}$/$m\ell$ LH, 0.5$\mu\textrm{g}$/$m\ell$ FSH and 1$\mu\textrm{g}$/$m\ell$ estradiol-17${\beta}$) for 20-22 h at 39$^{\circ}C$ in an atmosphere of 5% CO$_2$in air. Subsequently, COCs were cultured in hormone-free maturation medium for 20-22 h. After maturation for 40-44h, oocytes were removed cumulus cells by pipetting and cultured with epididymal sperm for 5 h in the mTBM. Embryos obtained were divided in 4 groups (1) cultured in NCSU 23 containing 0.4% BSA to blastocyst stage(Control), (2) essential amino acids (EA), (3) non-essential amino acids (NA), (4) mixture of essential and non essential amino acid (EA+NA). All treated groups(2-4) were used a glucose free NCSU 23 medium supplemented with pyruvate (0.33 mM), lactate (4.5 mM) to morula stage. From morula to blastocyst stage embryos of all treated groups were cultured in NCSU 23 containing 0.4% BSA. The rates of cleaved oocytes at 48 h after IVF were from 82% to 88% in the groups of control, EA, NA and EA+NA, respectively. The in vitro developmental rates into blastocysts in the groups of EA and EA+NA were significantly (P<0.05) higher than those of group of control (35.1, 35.4 vs. 19.4%, respectively), however, no significant (P<0.05) between control and NA. In conclusion, supplemented with essential amino acid or mixture of essential and non essential amino acid in the culture medium at morula stage increased the rate of development to blastocyst on in vitro produced porcine embryos.

  • Silkworm and silkworm-chrysalis as an edible insect have been investigated. Through this investigation, the following results are obtained : 1) Silkworm and silkworm-chrysalis as a good source of protein-rich compound was examined, and they contained 54.9% to 56% protein as a crude protein. 2) Except tryptophan, almost all the essential amino acids contained. The amount of the amino acids was on the increased as much as the silkworm growth. 3) The following amino acids was not detected on the silkworm fibron analysis, aspartic acid, glutamic acid, cystine and methionine. But the following amino acid were relatively high amount : glycine, alanine, and tyrosine. 4) Sucrose, fructose, and glucose as a carbohydrates was investigated, the results are from 0.312% to 0.313% as an invert sugar; 0.289% to 0.305% as a glucose; 0.333% to 0.412% as a fructose. 5) The acidity and pH of the milk was examined according to the silkworm-chrysalis powder added. added material was not effected as far as the milk was change. Therefore, the milk and the silkworm-chrysalis mixture are being a good drinkable things.

Quality Characteristics of the Kochujang Prepared with Mixture of Meju and Koji during Fermentation (메주와 고오지를 혼용하여 담금한 고추장 숙성중의 품질특성)

  • Quality characteristics of kochujang prepared with meju, koji and mixture of the two(meju+koji) were investigated during fermentation to improve quality of kochujang. During fermentation of kochujang, moisture content was in the range of $53.4{\sim}66.5%$, salt was $8.3{\sim}10.1%$, crude protein was $8.3{\sim}19.3%$ and pH of kochujang was $4.6{\sim}5.4$. Amino-nitrogen content increased during fermentation and the levels were $230{\sim}270\;mg%$ after 150 days of fermentation. The highest amino-nitrogen content was found in mixed kochujang at the beginning stage of fermentation, but in koji kochujang after 30 days of fermentation. Maximum reducing sugar content was $15.0{\sim}19.5%$ at 60th day of fermentation. The highest amino acid content of kochujang protein was found in meju kochujang followed by koji kochujang at the first stage of fermentation. The content of glutamic acid, a major amino acid was $1.38{\sim}3.66%$ of total amino acid content. High levels of aspartic acid, leucine, arginine, alanine and phenylalanine were found in the kochujangs. Mixed kochujang showed the highest L value among the samples until 30 days of fermentation. After that, the highest L value was found in meju kochujang. The highest degree of redness was observed in meju kochujang until 60 days of fermentation and in koji kochujang during $90{\sim}120$ days of fermentation.

  • The purpose of this study was to determine protein amino acid contents of some Korean foods by gas-liquid chromatography, and to evaluate this technique as a procedure for the quantitative determination of amino acids in foods. The crude protein content of foods was also estimated from the nitrogen content. 1. Nitrogen content of each food sample was determined previously to adjust the amount of sample for GLC analysis 2. In the analysis of 17 known amino acids, a linear relationship was found between the weight of 13 amino acids of 17 amino acids, the internal standard as well as the injection volume of a mixture and the detector responses for the derivatives of the amino acids. No response for arginine, cystein, histidine, and tyrosine was observed. 3. The relative molar response (RMR) values for the 13 amino acids of standard solution relative to glutamic acid as '1.00' were obtained under normal operating conditions with a hydrogen flame ionization detector. 4. The recovery of amino acids from their mixtures with natural food materials was carried out. The recoveries were essentially quantitative except threonine and serine. An overall mean recovery of 11 amino acids was $101.4{\pm}8.4$ per cent before hydrolysis and $98.1{\pm}8.7$ per cent after hydrolysis of samples. 5. The comparative analysis of the acid hydrolysates of two food samples by gas-liquid and ion-exchange chromatographic analysis were carried out. In white-bait pemmican, only threonine and asparagine amounts by GLC analysis had similar values to those obtained by ion-exchange chromatography. The other seven amino acids gave higher values as measured by GLC than by ion-exchange. With the food sample, soybean, alanine, valine, asparagine, and glutamic acid were in good agreement in two analysis, while leucine, proline, threonine, phenylalanine, and lysine were found in slightly higher concentrations in the GLC analysis. 6. Grant variations of amino acid content were found among samples analyzed. The amino acid contents of each sample were compared with the values found in the literature.

Separation of Two Amino Acids by Microemulsion Bulk Liquid Membrane

  • In this research work the potentialities of microemulsion bulk liquid membrane for the selective transport of L-tryptophan (L-Trp) and L-tyrosine (L-Tyr) are investigated at 298.15 K. Reversed micelle formed by sodium bis(2-ethylhexyl)sulfosuccinate (AOT) in dichloroethane, was used as mobile carrier to transport amino acids between a source and a receiving aqueous phase. The effects of pH, surfactant concentration and initial amino acid concentration on the extraction efficiency and transfer rate of the amino acids were studied. It is verified that for a mixture of two amino acids, L-Trp can be extracted selectively by using this type of the bulk liquid membrane with optimized condition.

Recommendations for the Selective Labeling of [$^{15}N$]-Labeled Amino Acids without Using Auxotrophic Strains

  • The strategy to incorporate [$^{15}$ N]-labeled amino acids were discussed. Instead of using specific auxotrophic strains for selective labeling, the prototrophic strain, BL2l(DE3), was used with a plasmid, pLysS, and found to be very effective for several amino acids including alanine, lysine, leucine, and threonine. Isoleucine, valine, glutamine, and tyrosine were also found to be effective despite some diffusion into other amino acids. Interesting result was obtained when [$^{15}$ N]-labeled glycine was tried: only glycines were labeled when amino acid mixture was added in the growth medium, and serines were co-labeled when amino acids were omitted. These results can be used as a guideline when selective labeling strategy is considered, and when the resulting data are interpreted.

Development and Applications of a Chemical Method for Sequential Analysis of Reducing Oligosaccharides

  • A new method based on the chemical reaction has been devised for the sequential analysis of reducing oligosaccharides using 8-amino-2-naphthalenesulfonic acid (ANS), a fluorescent precolumn derivatization reagent for reducing saccharides. The procedure established includes 1) the derivatization of a reducing oligosaccharide to produce a Schiff base, 2) the reduction of the base with sodium cyanoborohydride $(NaBH_3/CN), 3)$ the methoxycarbonylation of the resultant secondary amino group, 4) the cleavage of the glycoside bond next to the reducing end, based on the intramolecular acid hydrolysis by the action of a sulfonic acid group of the ANS derivative, 5) the identification of the liberated reducing end by high-performance liquid chromatography (HPLC), and finally 6) the recovery of the resultant oligosaccharide fragment from the cleavage reaction mixture. The extensive examination of the conditions for the sequential analysis of reducing oligosaccharides resulted in the procedure of simplicity , high selectivity and high recovery. This procedure was found to be useful for the sequential analysis of di-, tri- and tetrasaccharides.
