A Facile Synthetic Method of 2-Oxaxolidinones and 1,3-Oxazine-2-ones, Essential Moieties of New Antiulcer Agent

  • 2-Oxazolidinones and 1,3-oxazine-2-ones, key moieties of new antiulcer agents, were prepared successfully by treating corresponding hydroxyamide with N-bromosuccinimide (NBS) and silveracetate in acetonitrile. From the fact that the methods for the preparation of hydoxy amides are versatile and such amides could be converted to the corresponding 2-oxazolid-iones and 1,3-oxazine-2-one under our reaction condition, we think that our method is very practical one for the preparation of such compounds. In addition, the above synthetic example affords a good evidence of the synthetic applicability of our improved Hofmann rearrangement.

A New Phenolic Amide from Lycium chinense Miller

  • A new phenolic amide, dihydro-N-caffeoyltyramine (1) was isolated from the root bark of Lycium chinense Miller, along with known compounds, trans-N-caffeoyltyramine (2), cis-N-caffeoyltyramine (3), and lyoniresinol $3{\alpha}-Ο-{\beta}-D-glucopyranoside$ (4). Their structures were determined by spectroscopic analysis. A NBT superoxide scavenging assay revealed that three phenolic amides showed potent antioxidative activity.

Efficacy of dental local anesthetics: A review

  • The objective of this review was to investigate the efficacy of dental local anesthetics, as it is well known among clinicians that local anesthesia may be challenging in some circumstances. Therefore, the focus of this review was on the efficacy of the products used in dental local anesthesia. In a Pubmed database literature search conducted, a total of 8646 articles were found to be related to dental local anesthetics. After having applied the inclusion criteria (human research, performed in the last 10 years, written in English language, and focus on dental local anesthetics) and having assessed the quality of the papers, 30 were deemed eligible for inclusion in this review. The conclusion of this review is that none of the dental local anesthetic amides provide 100% anesthesia. The problem appears to be more pronounced when mandibular teeth are attempted to be anaesthetized and especially if there is irreversible pulpitis involved. The authors conclude that this finding suggest exploration of more efficient techniques to administer dental local anesthesia, especially in the mandible, to establish a 100% efficacy, is needed.

  • An efficient process for the solution-phase synthesis of biaryl amides has been developed. Girard's reagent T. an inexpensive scanvenger. was found to be very efficient in trapping excess aromatic acid chlorides. resulting in water soluble by-products. which were easily removed from the products by liquid-liquid extraction. The ease of use. and the excellent purity of the amide libraries obtained are important features of this protocol. (omitted)

  • The reaction of the sodium salts of 4-methoxy and 4, 7-dimethoxy 6-hydroxy benzofuran-5-carboxylic acid with ethyl chloroformate yields the corresponding dicarbethoxy derivatives. The N-substituted amides were obtained by treating the latter compounds with amines. The corresponding hydrazides were synthesized by the reaction of hydrazine hydrate on the dicarbethoxy derivatives which spontaneously cyclized to 5-substituted-2, 3- dihydro-1, 3, 4, -oxadiazol-2-one. Also the reaction with phenyl hydrazine has been studied. The dicarbethoxy derivatives and N-substituted amides were tested against Gram positive and Gram negative bacteria in vitro. Most of the compounds posses moderate or slight activity against Gram positive bacteria.

키랄 보조제로서의 C2-대칭성 피롤리딘 아미드의 합성과 광학 순도 결정 (Synthesis and Determination of Optical Purity of C2-Symmetric Pyrrolidine Amides as Chiral Auxiliaries)

  • 쉽게 얻을 수 있는 1,2:5,6-di-O-isopropylidene-D-mannitol (1)로부터 $C_2$-대칭성 피롤리딘 아미드 (8)을 합성하였다. 벤질 아민과 dimesylated hexitol (4)를 고리화 반응 시키면 트랜스 형태의 $C_2$-대칭성 피롤리딘 아민(5)가 주생성물로 얻어지나, 이와 함께 이성질화가 일어나 시스 이성질체인 화합물(6)도 생성된다. 피롤리딘 아민 (5,6)을 탈보호된 아민 (7)을 거쳐 아미드 (8,9)로 변형시켜 이를 순수한 형태로 분리하고, 이 $C_2$-대칭성 피롤리딘 아미드(8)의 광학 순도를 측정하기 위해 Mosher 유도체(13,14)를 합성하였다.$^1H$$^{19}F$ NMR 실험으로 Mosher 유도체 (13,14)가 단일 물질임이 밝혀졌고, 이로서 피롤리딘 아미드 (8)이 광학적으로 순수한 화합물임을 확인하였다.

이부프로펜의 피페라진 유도체에 대한 약리활성연구 (Studies of Pharmacological Activity on the Piperazine Derivatives of Ibuprofen)

  • To enhance the activity of ibuprofen, amides of ibuprofen, 1-piperazinyl-2-(4-isobutylphenyl)propionamide(Ibu-P.A.) and 1-(4-methylpiperazinyl)-2-(4-isobutylphenyl)propionamide (Ibu-M.P.), were synthesized and the pharmaceutical properties and the pharmacological activities of the amides were studied. The lipid:water partition coefficients and pKa values were examined in vitro, and the antiinflammatory effect, analgesic effects, acute toxicity, and intestinal absorption were studied for the amides and compared with ibuprofen in vivo. The results are summarized as belows; 1) The lipid:water partition coefficients of Ibu-M.P. were higher than those of ibuprofen. 2) The calculated pKa values of ibuprofen and Ibu-M.P. were 5.49 and 8.66, respectively. 3) The antiinflammatory effects of ibuprofen, Ibu-P.A., and Ibu-M.P. were same intensity, but the duration of the effects of Ibu-P.A. and Ibu-M.P. were longer than that of ibuprofen. 4) The analgesic effect of Ibu-M.P. was more potent than those of ibuprofen and Ibu-P.A. in the acetic acid-induced writhing test. 5) The $LD_{50}$ was 495 mg/kg for ibuprofen, 187 mg/kg for Ibu-M.P., and over 1250 mg/kg for Ibu-P.A.. 6) The absorption rate constants(k) and half-life($t_{1/2}$) were 0.74($hr^{-1}$) and 0.94(hr) for ibuprofen, and 0.72 ($hr^{-1}$) and 0.96 (hr) respectively for Ibu-M.P..

D-Glucose-Glycine 계의 Maillard 반응생성물 및 그 생성기구 (Maillard Reaction Products Formed from D-Glucose-Glycine, System and Their Formation Mechanism)

  • D-glucose-glycine 계를 사용하여 Maillard 반응에 의하여 생성되는 저분자 휘발성성분을 비롯하여 amide 화합물의 생성 및 그 기구를 검토하였다. 그 결과, 동정된 저분자 휘발성성분 중에서, headspace gas 중의 휘발성성분은 furan, acetone, 2-methylfuran, 2,5-dimethylfuran 2-butanone 2,3-pentanedione, diacetyl 등이었다, 이 중에서 diacetyl의 생성량이 가장 많아, 전 peak 면적의 약 $70\%$를 차지하였다. 또한, 에테르 추출물중의 주요반응생성물은 초산, furfuryl alcohol 2,5-dimethylpyrrole 2-acetylpyrrole 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one 등이었고, ethyl acetate 로 추출한 산성획분중에는 N-acetyl-glycine와 N-methylacetamide 등 2종류의 amide 화합물의 생성이 밝혀졌다. 이들 amide 화합물적 생성기구를 밝히기 위하여, Mailiard 반응초기생성물인 diacetyl 및 glyoxal을 각각 butylamine과 반응시킨 결과, Schiff 염기의 산화적 분해로 N-butylacetamide 및 N-butylformamide의 생성이 인정되었다. 따라서 N-acetylglycine 및 N-methylacetamide는 glucosylamine의 2,3-enol 화 및 ${\beta}-elimination$에 의한 탈수의 진행으로 생성된 dicarbonyl 화합물이 glycine과 반응하여 Schiff 염기를 형성하고, 이 Schiff 염기가 산화적분해를 받아서 N-acetylglycine이 생성되고, N-methylacetamide는 N-acetylglycine의 탈탄산에 의해서 생성된다고 생각한다.
