Mutagenicity of Human Urine Excreted after Ingestion of Roast Beef (구운고기를 섭취한 후 배설된 사람 뇨의 돌연변이 유발성물질(Mutagenicity)에 관하여)

  • This study was undertaken to observe the mutagenic occurrence in urine excreted after the ingestion of roast beef. Two healthy nonsmoker persons of both sex were selected for this test, employing two strains(TA98, TA100) of Salmonella typhimurium according to Ames' method. The mutagenic activity began to appear in urine of both sex three hours after ingestion of 300g of roast beef, gradually increasing until 6 hours and declining thereafter.

  • Gamma irradiation at 20 kGy was applied to Kanjang (soy sauce), Doenjang (soybean paste), Kochujang (hot pepper pasts) and Chungkukjang for their possible genotoxicity. The genotoxicity of 20 kGy-irradiated samples was evaluated by Salmonella typhimurium reversion assay. The Salmonella tester strains included TA98, TA100, TA1535 and TA1537 in the absence and presence of an exogenous metabolizing system (59 mix). All samples were negative in the bacterial reversion assay with S. typhimurium TA98, TA100, TA1535 and TA1537. The results indicated that 20 kGy of gamma irradiation on water-soluble fraction of Kanjang, Doenjang, Kochujang and Chungkukjang were not shown mutagenicity.

  • The mutagenicities and antimutagenicities of butanol (PL I) and water (PL II) extracts from the filtrate of the cultured broth of Phellinus linteus were examined using the Ames/Salmonella test. No mutagenic activity of PL I and PL II was found in Salmonella typhimurium strains TA98 and TA100, either with or without S9 activation. In contrast, PL I and PL II showed inhibitory effects on the mutagenic activities induced by the directly-acting mutagens, 4-nitro-o-phenylenediamine (NPD) using the tester strain TA98 and sodium azide (NaN3) using the tester strain TA 100 in the absence of S9 mix. PLI and PL II also showed inhibitory effects on the mutagenicities of the indirectly-acting mutagens, 2-aminofluorene (2-AF) using the tester strain TA98 and benzo[a]pyrene (B[a]P) using the tester strain TA 100 in the presence of S9. These results suggest that P. linteus has an antimutagenic activity and may play a role in the prevention of cancer.

Modifying Action of Chitosan Oligosaccharide on 2-Amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx)-induced Mutagenesis

  • The mutagenic activity of chitosan oligosaccharide and its antimutagenic effect against 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline (MeIQx) were investigated using the Salmonella/Ames test. No mutagenic activity was found in the Salmonella typhimurium strains TA 98 and TA 100, either with or without S9 activation. In contrast, chitosan oligosaccharide showed an inhibitory effect on the mutagenic activity of the cooked food mutagen, MeIQx, in the presence of S9. The influence of chitosan oligosaccharide on the genotoxicity of MeIQx was examined using a host-mediated assay in mice. The oligosaccharide was administered for 14 consecutive days (intragastric application at doses of 0.1 or 0.5 g/kg body wt) to mice. S. typhimurium TA 98 was given intravenously before an oral dose of MeIQx (4.5 mg/kg body wt.). The number of $his^+$ revertants were determined from the Ever of mice. The intragastric application of oligosaccharide led to a 47% reduction in the number of mutants induced by MeIQx (p<0.05). These results suggested that chitosan oligosaccharide had antimutagenic properties against MeIQx in vitro and in vivo.

Antimutagenic Activity and Cytotoxicity of the Whole Plant of Rumex acetosa (수영의 항돌연변이 활성 및 세포독성)

  • The extract and fractions of Rumex acetosa (Polygonaceae), a Korean medicinal plant, were examined for their cytotoxicities against five cultured human tumor cell lines, i.e. A549 (non-small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCY15 (colon), using the SRB (sulforhodamine-B) method in vitro and antimutagenic activities by Ames test with Salmonella typhimurium TA98 and TA100 and SOS chromotest with E. coli PQ37. Among the tested samples, the methylene chloride fraction strongly inhibited the proliferation of each examined tumor cell line with $IC_{50}$ values ranged from 13.2 to $18.7\;{\mu}g/ml$ and showed potent antimutagenic activities with 96.1% and 96.7% at the concentration of 1 mg/plate against the mutagens, NPD and sodium azide, respectively. Its antigenotoxic activity was also very effective at the final concentration of $10\;{\mu}g/reaction$ tube against the mutagens, MNNG and NQO by SOS chromotest.

  • This study describes aerodynamic characteristics for the HAWT (Horizontal Axis Wind Turbine) rotor blade using general CFD(Computational Fluid Dynamics) code. The boundary conditions for analysis are validated with the experimental result by the NREL (National Renewable Energy Laboratory)/NASA Ames wind tunnel test for S809 airfoil. In the case of wind turbine rotor blade, complex phenomena are appeared such as flow separation and re-attachment. Those are handled by using a commercial flow analysis tool. The 2-equation k-$\omega$ SST turbulence model and transition model appear to be well suited for the prediction. The 3-dimensional phenomena in the HAWT rotor blade is simulated by a commercial 3-D aerodynamic analysis tool. Tip vortex geometry and Radial direction flows along the blade are checked by the analysis.

An Influence of Pretreatment Conditions on Mutagen Binding of Lactobacillus paracasei subsp. tolerans JG22 against MNNG and 2-NF

  • The objectives of this study were to investigate the effect of Lactobacillus paracasei subsp. tolerans JG22 isolated from pepper leaf jangajji on the mutagenic activity of N-methyl, N'-nitro, N-nitrosoguanidine (MNNG) and 2-nitrofluene (2-NF) and to evaluate the effect of physico-chemical pretreatment on the antimutagenic activity of the strain. The viable cells of JG22 strain displayed a significantly high (p <0.05) antimutagenic activity against both mutagens tested. The antimutagenic effect of JG22 strain seems to be positively correlated with the amounts of the cells in the incubation time. This strain produced the antimutagenic activity of the maximum levels after preincubation for 30 min. The binding of this strain against the mutagenic compounds might be mainly present in the cell wall fraction rather than the cytosol fraction. Pretreatment with proteolytic enzymes and simulated gastric and intestinal juices and at different pH values had no significant effect on two mutagens removal by the viable cells. However, the binding activity of the mutagen by the strain seems to be affected by heating, enzymes including $\alpha$-amylase and lysozyme, divalent ions, and sodium metaperiodate. Thus, carbohydrates consisting of the cell walls may be important elements responsible for the binding of MNNG and 2-NF by this strain. In conclusion, the binding of the mutagens to cells of JG 22 strain may play a vital role in suppressing the process of mutagenesis induced by mutagens.

Pharmacological Analysis of Vorinostat Analogues as Potential Anti-tumor Agents Targeting Human Histone Deacetylases: an Epigenetic Treatment Stratagem for Cancers

  • Alteration of the acetylation status of chromatin and other non-histone proteins by HDAC inhibitors has evolved as an excellent epigenetic strategy in treatment of cancers. The present study was sought to identify compounds with positive pharmacological profiles targeting HDAC1. Analogues of Vorinostat synthesized by Cai et al, 2015 formed the test compounds for the present pharmacological evaluation. Hydroxamte analogue 6H showed superior pharmacological profile in comparison to all the compounds in the analogue dataset owing to its better electrostatic interactions and hydrogen bonding patterns. In order to identify compounds with even better high affinity and pharmacological profile than 6H and Vorinostat, virtual screening was performed. A total of 83 compounds similar to Vorinostat and 154 compounds akin to analogue 6H were retrieved. SCHEMBL15675695 (PubCid: 15739209) and AKOS019005527 (PubCid: 80442147) similar to Vorinostat and 6H, were the best docked compounds among the virtually screened compounds. However, in spite of having good affinity, none of the virtually screened compounds had better affinity than that of 6H. In addition SCHEMBL15675695 was predicted to be a carcinogen while AKOS019005527 is Ames toxic. From, our extensive analysis involving binding affinity analysis, ADMET properties predictions and pharmacophoric mappings, we report Vorinostat hydroxamate analogue 6H to be a potential candidate for HDAC inhibition in treatment of cancers through an epigenetic strategy.

In Vitro Antimutagenic Effects of Alaternin and Isorubrofusarin Gentiobioside from roasted Cassia tora

  • The antimutagenic activity of a methanol extract of roasted Cassia tora seed against aflatoxin $B_1(AFB_1)$ was demonstrated with the Salmonella typhimurium assay. The numbers of revertants per plate decreased significantly when this extract was added to the assay system using Salmonella typhimurium TA100 and or TA98. The MeOH extract was then sequentially partitioned with $CH_2Cl_2$, EtOAc, n-BuOH, and $H_2O$. The $CH_2Cl_2$ and n-BuOH fractions possessed antimutagenic activity, but the EtOAc and $H_2O$ fractions were inactive. Both the MeOH extract and its fractions were capable of inhibiting the indirect-acting mutagen $AFB_1$, suggesting that these fractions may prevent the metabolic activation of $AFB_1$, or scavenge the electrophilic intermediate capable of inducing mutations. Column chromatography using silica gel yielded pure alaternin from the $CH_2Cl_2$ a fraction, and adenosine and isorubrofusarin gentiobioside from the n-BuOH fraction. Alaternin and isorubrofusarin gentiobioside demonstrated significant antimutagenic activities.

Functional Activities of Ethanol Extracts from Flammulina velutipes (팽이버섯 에탄올 추출물의 기능적 특성)

  • This study was conducted to evaluate the antioxidative effect and antimutagenic capacity of ethanol extracts of Flammulina velutipes by employing biological and biochemical assays. The $IC_{50}$ of MDA with BSA conjugation reaction, lipid peroxidation and scavenging effect on DPPH radical in ethanol extracts of Flammulina velutipes was found to be 28.39 mg/assay, 9.33 mg/assay and 144.61 mg/assay respectively. Therefore, the most effective antioxidative capacity of ethanol extracts of Flammulina velutipes was $Fe^+$-induced linoleate peroxidation, among the method used this study. The indirect and direct antimutagenic effects of the ethanol extracts of Flammulina velutipes were examined by the Ames test using Salmonella typimurium TA98 and TA100. The inhibition rates on indirect mutagenicity mediated by 2-anthramine and on direct mutagenicity mediated by sodium azide in Salmonella typimurium TA100 and mediated by 2-nitrofluorene in Salmonella typimurium TA98 were 0%, respectively. These findings indicate that ethanol extracts of Flammulina velutipes have no effects on indirect and direct mutagenicity. Based on these results, it believed that the ethanol extracts of Flammulina velutipes has antioxidative capacities, and is a the candidate for the prevention and dietetic treatment of chronic diseases and the development of antioxidative functional food.
