RAPD analysis was performed from four host-specific toxin (HST) producing Alternaria, i.e., A. kikuchiana, A. mali, a. longipes and A. Longipes and A. alternata f. sp. lycopersici, nonpathogenic A. alternata and A. brassicicola to assess their phylogenetic relationship. DNA polymorphism was detected among species (pathotypes) of HST producing Alternaria by PCR amplification and differentiation of the species was recognized by RAPD analysis. Primer OPA-02 was the most profitable among 7 notificated primers for differentiation of the HST producing Alternaria species. UPGMA analysis of the RAPD bands from alternaria spp. revealed that HST producing Alternaria and nonpathogenic a. alternata are closely related.
국내에서 분리한 20종의 Alternaria 277개 균주를 사용하여 Alternaria 진균소독인 altenuene(ALT), altertoxin-I(ATX-I) 및 tenuazonic acid(TA)의 in vitro에서의 생성능과 그 분포를 조사하였다. 각 사용균주를 200g의 살균된 쌀배지에 접종하여 $25^{\circ}C$에서 3주간 배양하였다. Alternaria 배양체들을 methanol로 추출하고 용매분획과 TLC 및 HPLC분석을 통하여 순화하였다. A. alternata와 그와 형태적으로 유사한 A. kikuchiana, A. longipes 및 A. mail는 비록 종간 및 종내의 균주간에 생성량의 차이는 매우 다양하였지만 TA를 비롯한 5종의 진균독소를 모두 생성하였다. A sesami와 A. sesamicola는 4종의 진균독소(AOH, AME, ALT, ATX-I)를 생성하였고 A. cucumerina, A. dauci, A. macrospora, A. porri, A. solani, A. tagetica와 A. zinniae와 같은 대형분생포자와 긴 beak를 형성하는 7종의 Alternaria균들은 AOH와 AME만을 생성하였다. A. brassicicola, A. helianthi, A. panax, A. radicina 및 A. raphani등 5종의 Alternaria는 5종의 진균 독소를 모두 생성하지 않았다.
Four series (S, M, R, and W) of Alternaria longipes isolates were obtained based on consecutive selection with Dimethachlon (Dim) and ultraviolet irradiation. These isolates were then characterized according to their tolerance to Dim, sensitivity to osmotic stress, and phenotypic properties. All the selected Dim-resistant isolates showed a higher osmosensitivity than the parental strains, and the last generation was more resistant than the first generation in the M, R, and W series. In addition, the changes in the Dim resistance and osmotic sensitivity were not found to be directly correlated, and no distinct morphologic characteristics were found among the resistant and sensitive isolates, with the exception of the resistant isolate K-11. Thus, to investigate the molecular basis of the fungicide resistance, a group III two-component histidine kinase (HK) gene, AlHK1, was cloned from nineteen A. longipes isolates. AlHK1p was found to be comprised of a six 92-amino-acid repeat domain (AARD), HK domain, and response regulator domain, similar to the Os-1p from Neurospora crassa. A comparison of the nucleotide sequences of the AlHK1 gene from the Dim-sensitive and -resistant isolates revealed that all the resistant isolates contained a single-point mutation in the AARD of AlHK1p, with the exception of isolate K-11, where the AlHK1p contained a deletion of 107 amino acids. Moreover, the AlHK1p mutations in the isolates of each respective series involved the same amino acid substitution at the same site, although the resistance levels differed significantly in each series. Therefore, these findings suggested that a mutation in the AARD of AlHK1p was not the sole factor responsible for A. longipes resistance to dicarboximide fungicides.
Twenty universal rice primers (URPs) were used to detect PCR polymorphisms in 25 isolates of six different Alternaria species producing host specific toxins (HST). Eight URPs could be used to reveal PCR polymorphisms of Alternaria isolates at the intra- and inter-species levels. Specific URP-PCR polymorphic bands that are different from those of the other Alternaria spp. were observed on A. gaisen and A. longipes isolates. Unweighted pair-group method with arithmetic mean (UPGMA) cluster analysis using 94 URP polymorphic bands revealed three clustered groups (A. gaisen group, A. mati complex group, and A. logipes group).
In the course of survey of endophytic fungi from Bangladesh pumpkin seeds in 2011~2012, two strains (CNU111042 and CNU111043) with similar colony characteristics were isolated and characterized by their morphology and by molecular phylogenetic analysis of the internal transcribed spacer, glyceraldehydes-3-phosphate dehydrogenase (gpd), and Alternaria allergen a1 (Alt a1) sequences. Phylogenetic analysis of all three sequences and their combined dataset revealed that the fungus formed a subclade within the A. alternata clade, matching A. burnsii and showing differences with its other closely related Alternaria species, such as A. longipes, A. tomato, and A. tomaticola. Long ellipsoid, obclavate or ovoid beakless conidia, shorter and thinner conidial size ($16{\sim}60[90]{\times}6.5{\sim}14[{\sim}16]{\mu}m$) distinguish this fungus from other related species. These isolates showed more transverse septation (2~11) and less longitudinal septation (0~3) than did other related species. Moreover, the isolate did not produce any diffusible pigment on media. Therefore, our results reveal that the newly recorded fungus from a new host, Cucurbita maxima, is Alternaria burnsii Uppal, Patel & Kamat.
Narceine methyl ester and narceine are potent alkaloids which were isolated from Corydalis longipes were found effective in vitro at very low concentration, i.e., $100{\sim}500\;ppm$ against spore germination of some test plant pathogenic fungi (Alternaria solani, A. tagetica, Cercospora abelmoschi, Curvularia maculans, Erysiphe cichoracearum, E. pisi, Fusarium udum, Helminthosporium oryzae, H. penniseti, Ustilago cynodontis). Among the test, phytopathogens the spores of F. udum, C. maculans and H. penniseti were highly sensitive at 200 ppm. However, spores of E. pisi, A. solani and A. tagetica were less sensitive at low concentration followed by other test fungi. Most of the fungi showed zero or nearly zero percent spore germination at 400 and 500 ppm.
Antifungal activity of ChiCW and synergistic interactions between ChiCW with fungicides were investigated. Conidial germinations of phytopathogenic fungi, Alternaria brassicicola, Botrytis elliptica, and Colletotrichum gloeosporioides, were inhibited by ChiCW but A. longipes was not. In addition, ChiCW showed synergistic effect with fungicides Switch (cyprodinil+fludioxonil) and tebuconazole to inhibit fungal conidial germinations. The level of synergism of ChiCW with tebuconazole was higher than that with Switch. The results indicate that ChiCW may exhibit a higher level of synergism with fungicides that have a primary effect upon membranes.
Chaerophylline and berberine hydroxide, both being alkaloids, isolated from Corydalis chaerophylla and C. longipes respectively were assayed against spore germination of some fungi, e.g. Alternaria solani, A. brassicola, A. brassicicola, Helminthosporium penniseti, Helminthosporium sp., Heterosporium sp., Curvularia penniseti, C. maculens and C. palliscens. While chaerophylline inhibited spore germination of most of the fungi at 1000 ppm, being also effective at 50, 100, 200 and 500 ppm, berberine hydroxide was significantly effective at much lower concentration, i.e., 400 ppm against several fungi. This compound was also effective against some fungi at 50, 100, 150, 200 ppm. There was 100% inhibition of spore germination in several fungi at highest concentration of both the compounds. Some of the fungi showed similar results even at lower concentrations.
A fungal strain, R9, was isolated from the South Atlantic sediment sample and identified as Aspergillus fumigatus. An antifungal protein, AfAFPR9, was purified from the culture supernatant of Aspergillus fumigatus R9. AfAFPR9 was identified to be restrictocin, which is a member of the ribosome-inactivating proteins (RIPs), by MALDI-TOF-TOF-MS. AfAFPR9 displayed antifungal activity against plant pathogenic Fusarium oxysporum, Alternaria longipes, Colletotrichum gloeosporioides, Paecilomyces variotii, and Trichoderma viride at minimum inhibitory concentrations of 0.6, 0.6, 1.2, 1.2, and 2.4 μg/disc, respectively. Moreover, AfAFPR9 exhibited a certain extent of thermostability, and metal ion and denaturant tolerance. The iodoacetamide assay showed that the disulfide bridge in AfAFPR9 was indispensable for its antifungal action. The cDNA encoding for AfAFPR9 was cloned from A. fumigatus R9 by RT-PCR and heterologously expressed in E. coli. The recombinant AfAFPR9 protein exhibited obvious antifungal activity against C. gloeosporioides, T. viride, and A. longipes. These results reveal the antifungal properties of a RIP member (AfAFPR9) from marine-derived Aspergillus fumigatus and indicated its potential application in controlling plant pathogenic fungi.
A novel antifungal protein produced by the fungal strain Penicillium citrinum W1, which was isolated from a Southwest Indian Ocean sediment sample, was purified and characterized. The culture supernatant of P. citrinum W1 inhibited the mycelial growth of some plant pathogenic fungi. After saturation of P. citrinum W1 culture supernatants with ammonium sulfate and ion-exchange chromatography, an antifungal protein (PcPAF) was purified. The N-terminal amino acid sequence analysis showed that PcPAF might be an unknown antifungal protein. PcPAF displayed antifungal activity against Trichoderma viride, Fusarium oxysporum, Paecilomyces variotii, and Alternaria longipes at minimum inhibitory concentrations of 1.52, 6.08, 3.04, and $6.08{\mu}g/disc$, respectively. PcPAF possessed high thermostability and had a certain extent of protease and metal ion resistance. The results suggested that PcPAF may represent a novel antifungal protein with potential application in controlling plant pathogenic fungal infection.
