Streptozotocin, Ploxamer 407, $CCl_4$ 및 D-Galactosamine으로 유도한 병태모델 흰쥐에 대한 산마늘 추출물의 약리효과 (Pharmacological Effects of the Allium victorialis var. platyphyllum Extracts on the Rats Induced by Streptozotocin, Poloxamer-407, $CCl_4$ and D-Galactosamine)

  • To develop Allium victorialis var. platyphyllum (Liliaceae) for an available functional food, pharmacological experiments on the extracts of this plant were undertaken in the rat treated with streptozotocin, poloxamer-407 and $CCl_4$. The two MeOH extracts were obtained from the leaves and the bulbs, respectively. The three agents, streptozotocin, poloxamer-407 and $CCl_4$, were treated to induce diabetes mellitus, hyperlipidemia and hepatic injury, respectively. Treatment with the leaf extract lowered blood glucose by 24.9% at 200 mg/kg (p.o.) in the STZ-treated rat and prohibited the increase of body weight, water consumption and food intake. This exσact also significantly decreased not only thε plasma cholesterol and triglyceride in the poloxamer-407-treated rat by 35.3% but also serum ALT by 49.0%. The potency was found by overall estimation on the experimental results as followings: 200 mg/kg leaf extract> 200 mg/kg bulb extract> 100 mg/kg leaf exσact> 100 mg/kg bulb extract. These results suggested that this plant might have the availability for a functional food. It was also suggested that the leaves are more effective than the bulbs for the functional vegetable, especially in hyperlipidemia and hepatic injury.

Determination of Flavonoids from Allium victorialis var. platyphyllum and Their Effect on Gap Junctional Intercellular Communication

  • This study was carried out to identify and quantify the flavonoids from 6 different plant parts of Allium victorialis var. platyphyllum (AVP), including the flower, leaf, root, stem, flower stalk, and flower seed, using liquid chromatography/ mass spectrometry. Two major flavonoids were structurally identified as quercetin (3,5,7,3'4,'-pentahydroxyflavone) and kaempferol (3,5,7,4'-tetrahydroxyflavone) at contents of 11.8-25.8 and $6.0-64.4\;{\mu}g/mL$, respectively. In particular, the flower and root plant parts contained the highest amounts of quercetin and kaempferol compared to the other parts. We also assessed the recovery effects of each plant-part extract of AVP on gap junctional intercellular communication (GJIC) in WB-F344 cells by the scrape-loading and dye transfer (SL/DT) method. According to the results, GJIC was reduced by approximately 70.2% ($62.3{\pm}12.5$ cells) compared to the control ($209{\pm}9.5$ cells, 100%) when 12-O-tetradecanoylphorbol-13-acetate (TPA) was treated alone in the WB-F344 rat liver epithelial cells. However, the stem extract (0.2 mg/mL) restored GJIC to basal levels (92%, $204{\pm}2.3$ cells, p<0.01) and the flower extract (0.2 mg/mL) stimulated GJIC to 82.5% ($172.6{\pm}8.3$ cells, p<0.05), when applied together with the TPA.

고지방식이 마우스에서 산마늘의 간과 신장 보호효과 (Hepatoprotective and Nephroprotective Effects of Allium victorialis Leaf Extracts on the High Fat Diet Supplied Mice)

  • 고지방식이로 유발된 간과 신장의 손상에서 산마늘 메탄올 추출물의 효과를 알아보고자 하였다. 12주 동안 산마늘 메탄올 추출물 62.5, 125, 250 mg/kg/day를 투여한 결과 고지방식이로 유발된 혈청중의 AST, ALT, BUN, creatinine의 증가 억제와 간의 조직병리학적 변화를 감소시켜 지방간염과 신장의 손상이 현저하게 감소하였다. 또한, 항산화 효소인 superoxide dismutase, glutathione, catalase의 증가와 malondialdehyde의 감소로 항산화방어체계(antioxidant defense system)를 강화시켰다. 산마늘 메탄올 추출물 125 mg/kg 투여군은 대조군인 silymarin 100 mg/kg 투여군과 유사한 효과를 나타내었다. 결론적으로 산마늘 메탄올 추출물은 고지방식이로 유발되는 간과 신장의 손상을 완화하는 유용한 작용을 하는 것으로 나타났다.

산마늘 기능성 활성물질 탐색 및 자원화 (Effect of Allium victorialis var.platyphyllum Leaves on Triton WR-1339-induced and Poloxamer-407-induced hyperlipidemic Rats and on Diet-Induced Obesity Rats)

  • The herb of Allium victorialis var. platyphyllum (Liliaceae) has been used as an edible wild herb and to treat heart failure and gastritis. We have already reported antihyperlipidemic anti-tumor effects of this plant. To enlarge the commercial availability of this food, it was investigated whether the extracts of A. victorialis var.platyphyllum reduce hyperlipidemia and obesity or not. The plants tested in this experiment were collected from two eco-types of IS. Ullung and Mt. Odae cultivated at Pyongchang. Extracts were prepared by extracting the fresh leaves and those dried at $36^{\circ}C$ and $90^{\circ}C$, respectively. Pretreatment with the ethanolic extracts for two weeks (p.o.) reduced serum triglyceride-, total cholesterol- and LDL-cholesterol contents in rats induced by Triton WR-1339, respectively. Furthermore, oral administration of the extracts also inhibited the hyperlipidemia induced with oral diet of 30% corn oil. In the other attempt to find to alleviate the obesity, the model rats with obesity were induced by the high fat-diet for six weeks. Post-treatment with the extracts for two weeks significantly reduced the hyperlipidemia. Retroperitoneal-, epidymal- and total abdominal fat pad weights were considerably reduced at 100 mg/kg oral administration of the extracts. Increased feces lipid contents were also found in the rat treated with the extracts. The extract of Mt. Odae eco-type showed more potent activity than that of Is. Ullung one. These results suggest that use of the fresh leaves may lead to the higher activity in treatment of hyperlipidemia and obesity than of the dried one.

Antihepatotoxic effect of Heat-treated Allium victorialis var. platyphyllum in $CCl_4-induced$ Rats and the Gas Chromatographic Analysis of Volatile Sulfur Substances

  • The ethanolic extracts of the leaves and bulbs of Allium victorialis var. platyphyllum (Liliaceae) collected from Daegwallyoung (D) and Ullung Island (U) in Korea were obtained using three different extracting methods. The first extracts, DL-1 DB-1, UL-1 and UB-1, were obtained from leaves (L) and bulbs (B) dried at $90^{\circ}C$, respectively, and the second extracts, DL-2, DB-2, UL-2 and UB-2, were obtained by extracting the leaves and bulbs of fresh plant parts. The third extracts DL-3, DB-3, UL-3 and UB-3 were obtained by incubating leaves and bulbs at $36^{\circ}C$. The six extracts obtained from A. victorialis var. platyphyllum at Daegwanllyoung (cultivated site) were orally administered to examine for a possible antihepatotoxic effect in $CCl_4-induced$ rats. DL-1 exhibited the most pronounced effect. The extracts inhibited serum ALT, AST, SDH, ${\gamma}-GT$, ALP and LDH activities elevated by $CCl_4$ injection and attenuated decreased glutathione S-transferase, glutatione reductase and ${\gamma}-glutamylcysteine$ synthetase activities and a decreased hepatic glutathione. However, the extracts obtained from Ullung Is. (native site) were less active than the extracts from Daegwallyoung, suggesting that A. victorialis var. platyphyllum from the cultivated site is more useful for functional food than of native site. These results also suggest that the antihepatotoxic effect is due to a higher content of hepatic glutathione. Gas chromatography of the twelve extracts showed significantly different sulfides, disulfides or trisulfides contents belonging to volatile sulfur substances (VSS). Nine components were identified on the basis of their mass spectra, namely, dimethyl disulfide, dimethyl trisulfide, diallyl disulfide, dipropyl disulfide, allyl methyl sulfide, allyl methyl trisulfide, 2-vinyl-4H-1,3-dithiin, 3,4-dihydro-3-vinyl-1,2-dithiin, and allithiamine. Extract DL-1 had the highest VSS content. Dried plant materials contained larger amounts of the VSSs than other extracts, and the leaves contained larger amount than the bulbs. These results suggest that heat treatment increases the antiheaptotoxic ability of A. victorialis var. platyphyllum by increasing the proportion of VSSs.

Constituents and the Antitumor Principle of Allium victoriallis var. platyphyllum

  • To search for cytotoxic components from Allium victoriallis , MTT assays on each extract and an isolated component, gitogenin 3-O-lycotetroside, were performed against cancer cell lines. Cytotoxicities of most extract were shown to be comparatively weak, though $IC_50$ values of $CHCl_3$fraction was found to be <31.3-368.4 $\mu\textrm{g}/ml$. From the incubated methanol extract at $36^{\circ}C, eleven kinds of organosulfuric flavours were predictable by CG-MS performance. The most abundant peak was revealed to be 2-vinyl-4H-1,3-dithiin(1) by its mass spectrum. Further, this extract showed significant cytotoxicities toward cancer cell lies. Silica gel column chromatography of the n-butanol fraction led to the isolation of gitogenin 3-O-lycotetroside (3) along with astragalin (4) and kaempferol 3, 4'-di-O-$\beta$-D-glycoside (5). This steroidal saponin exhibited significant cytotoxic activities ($IC_50$, 6.51-36.5 $\mu\textrm{g}/ml$) over several cancer cell lines. When compound 3 was incubated for 24 h with human intestinal bacteria, a major metabolite was produced and then isolated by silica gel column chromatography. By examining parent and prominent ion peak in FAB-MS spectrum of the metabolite, the structure was speculated not to be any of prosapogenins of 3, suggesting that spiroketal ring were labile to the bacterial reaction. These suggest that disulfides produced secondarily are the antitumor principles.

  • The effects of angiogenesis inhibitor from the extract libraries of Korean and Chinese medicinal plants were investigated using a protein microarray chip. Protein chip was constructed by immobilization of integrin ${\alpha}_5{\beta}_1$ on protein chip base plates and employed far screening active extracts that inhibit the integrin-fibronectin interaction from the extract libraries. The 100 extracts of medicinal plants were obtained from extract bank of National Institute of Crop Science, RDA. The 14 extracts among 100 extract libraries were shown efficient inhibition activity for the interaction between integrin-fibronectin. The medicinal plants of 14 extracts were Vitex negundo var. incisa (Lam.) C.B. Clarke, Epimedium koreanum Nakai, Cedrela sinensis A. Juss, Ipomea aquatica Forsk, Schisandra chinensis Baill, Pulsatilla koreana Nakai, Paeonia lactiflora Pall. var.hortensis Makino, Oenothera odorata, Allium chinense, Allium victorialis var. platyphyllum MAKINO, Polygonatum odoratum Druce var. pluriflorum Ohwi, Hosta lancifolia, Agrimonia pilosa L. var. japonica Nakai and Potentilla chinensis SER. The Paeonia lactiflora, Oenothera, and Agrimonia pilosa from these 14 extracts libraries were shown strong inhibition activity of integrin ${\alpha}_5{\beta}_1$.

산마늘 추출물의 항산화활성 (Antioxidative Activity of A. victorialis var. platyphyllum Extracts)

  • 울릉도 자연산 및 강릉산 재배 산마늘(Allium victorialis var. platyphyllum)의 물 추출물과 70% 에탄올 추출물의 항산화 활성을 조사하였다. 총 폴리페놀 함량은 강릉산보다 울릉도산에서, 인경보다 잎에서 높게 나타났으며, 강릉산 잎의 70% 에탄올 추출물에서 52.57 mg/g으로 가장 높게 나타났다. 플라보노이드 함량은 잎의 에탄올 추출물에서 유의성 있게 높게 나타났으며, 인경에서는 산지별, 추출용제별 차이가 없었다. 울릉도산 잎의 70% 에탄올 추출물에서 73.30 mg/g으로 가장 높게 나타났다. 전자공여능은 울릉도산은 70% 에탄올 추출물에서, 강릉산은 물 추출물에서 높았고, 인경보다 잎에서 높게 나타났다. 울릉도산 잎의 알코올 추출물에서 55.13%로 가장 높게 나타났다. SOD 유사활성은 강릉산보다 울릉도산에서 높게 나타났고, 울릉도산 잎의 에탄올 추출물이 92.00%로 가장 높게 나타났다. Hydroxy radical 소거활성은 잎의 에탄올 추출물에서, 인경보다는 잎에서, 울릉도산 보다는 강릉산 잎에서 높게 나타났다. 강릉산 잎의 70% 에탄올 추출물에서 58.85%로 가장 높았다. 지질과산화 억제활성은 울릉도산과 강릉산 모두 잎과 에탄올 추출물에서 높게 나타났고, 울릉도산 잎의 70% 에탄올 추출물에서 73.33%로 가장 높았다. 본 연구 결과로 산마늘 추출물은 항산화 관련 건강기능성 식품 등으로 널리 응용될 수 있을 것이다.
